BACKGROUND: Recurrent Pregnancy Loss (RPL) is a multifactorial reproductive disorder influenced by genetic, immunological, and metabolic disturbances. Altered trace element and heavy metal levels increasingly contribute to oxidative stress, inflammation, and impaired methylation, potentially affecting pregnancy outcomes.
METHODS: A case-control study was conducted including RPL patients (n = 48) and healthy controls (n = 48). Serum levels of Cu, Zn, Pb, Fe, Cr, Se, Cd, As, and Mg were assessed using Atomic Absorption Spectrophotometry. Inflammatory and metabolic biomarkers MMP-2, TNF-α, Vitam

BACKGROUND: One-carbon metabolism is implicated in psychotic disorders, yet most evidence relies on peripheral markers that are susceptible to confounding by antipsychotic exposure and illness chronicity. Direct cerebrospinal fluid (CSF)-based assessment at the onset of psychosis provides insight into central metabolic alterations largely independent of peripheral confounding; however, such evidence remains limited. We examined CSF markers of methylation capacity in patients with drug-naïve first-episode psychosis (FEP).
METHODS: In this cross-sectional comparative study, CSF concentrations o

Amino acid metabolism serves as a central hub linking retinal energy supply, neurotransmission, and cell signaling, which is critical for maintaining retinal structure and function. This review summarizes the molecular mechanisms by which abnormal amino acid metabolism contributes to retinal diseases. The major mechanisms include: 1) excitotoxicity caused by disruption of glutamate-glutamine cycle; 2) mitochondrial oxidative stress and epigenetic changes due to accumulation of branched-chain and sulfur-containing amino acids; 3) remodeling of immune microenvironment due to altered tryptophan a

This study introduces a rapid and environmentally sustainable High Performance Liquid Chromatography method to simultaneously quantify free thiols and monitor their disulfide exchange kinetics with cystine. This method is based on the derivatisation of thiols with 5,5’-dithiobis(2-nitrobenzoic acid) (DTNB, also known as Ellman’s reagent). The method utilises a mobile phase consisting of 0.1% v/v Trifluoroacetic acid in water and methanol, with Ultraviolet detection at 248 nm. The method exhibited high sensitivity (Limit of Quantification 0.78 µM; Limit of Detection 0.39 µM) and selectivity for

Peptide self-assembly is a promising strategy for developing bioactive nanomaterials with improved drug delivery and therapeutic performance. However, artificial design approaches may not fully incorporate natural paradigms and typically avoid cysteine-rich sequences due to their inherent instability. Challenging this convention, we established a natural medicine-inspired discovery pipeline to identify self-assembling thiol-rich peptides. From pangolin scales, we identified a novel natural peptide, Ac-KCSQLNVNCKG, containing two free cysteine thiols that spontaneously self-assembles into chira

While copper is an essential redox cofactor, excess copper, particularly Cu(I), is highly toxic and therefore requires precise control of metal speciation in the bacterial periplasm. CopI is a cupredoxin-like protein implicated in copper tolerance under microaerobic conditions and contains multiple histidine- and methionine-rich regions, yet its metal-binding properties remain poorly understood. Here, we deconstructRubrivivax gelatinosusCopI into its constituent metal-binding modules and provide the first quantitative, domain-resolved analysis of their Cu(II) coordination chemistry. The aim of the study was to explore the precise mechanisms involved in maternal mercury exposure induced neural tube defects (NTDs). We established a mouse model of mercury chloride (HgCl2)-induced embryo NTDs using ICR mice, and investigated the roles of amino acid transport and associated methionine metabolic disorders in the placenta and fetus. Oral administration of 0, 1.5, 3, and 4.5 mg/kg/d HgCl2 to pregnant ICR mice was performed between gestation days 5.5 and 9.5, with assessments conducted on gestation day 10.5. The incidence of NTDs was significantly elevated in the 3 mg/kg/d group w

BACKGROUND: Cancer, including head and neck squamous cell carcinoma (HNSCC), induces changes to metabolism that drive the disease. Regional metabolomics allows understanding of metabolic variation across the tumour, including in the tumour core, where hypoxia is likely more pronounced.
METHODS: Ultra-high performance liquid chromatography-mass spectrometry metabolomics was applied to regionally distinct patient tissue samples: tumour edge, tumour core and adjacent non-tumour. Statistical, correlation and pathway enrichment analyses were performed.
RESULTS: Markers of hypoxia or pseudohypoxia

Dampening neutrophil-driven inflammation in the airways remains a challenge in treating cystic fibrosis (CF) lung disease. Myeloperoxidase (MPO) is a neutrophilic enzyme that produces reactive oxygen species and is highly concentrated in CF airways. Greater MPO concentrations have previously been correlated with increased mucus plugging in bronchiectasis, suggesting that MPO could impair mucociliary transport. As such, we evaluated the impact of MPO treatment on barrier integrity, mucin production, mucus viscoelasticity and mucociliary transport in fully differentiated human airway epithelial

Betaine-homocysteine methyltransferase (BHMT) is an enzyme involved in one-carbon metabolism and plays a crucial role in maintaining liver health. In this study, we investigated the impact of liver-specific deletion of BHMT on liver dysfunction using a mouse model. We generated BHMT floxed mice and bred them with albumin Cre to generate liver-specific BHMT knockout (BHMT LKO) mice. Liver tissues harvested from six-month-old chow-fed BHMT floxed and LKO mice were characterized through histological, biochemical, and molecular analyses. BHMT LKO mice displayed a complete loss of hepatic expressio

Cysteine is a sulfur-containing amino acid that plays a central role in skin physiology through thiol-mediated redox regulation and glutathione (GSH) synthesis. It critically influences melanogenesis, collagen homeostasis, and wound healing. However, its clinical application is limited by poor stability and bioavailability. In this review, we provide a mechanistic and comparative analysis of cysteine and its derivatives, including N-acetylcysteine (NAC), cysteinamide (C-NH2), GSH, and related compounds. These derivatives regulate melanogenesis by modulating dopaquinone pathways and tyrosinase

Selenoproteins, a unique class of proteins critical for cellular antioxidant defense, are characterized by the incorporation of selenocysteine (Sec) in their active sites. Sec is co-translationally inserted into proteins via a specialized mechanism that reprograms the UGA codon to encode Sec, involving a specific RNA structure designated the Sec insertion sequence (SECIS) element and several essential enzymes. Although numerous selenoproteins have been identified in prokaryotes (primarily bacteria), the detection of selenoprotein genes in these organisms remains challenging, largely due to dif

D-Cysteine (D-Cys) is critically involved in regulating redox homeostasis within the brain, and its dysregulation has been closely related to the pathogenesis of various neurological disorders. However, the precise mapping of cerebral D-Cys levels in situ remains a formidable challenge due to the lack of specific detection tools. Herein, we report a new class of fluorinated firefly luciferase prosubstrates for sensitive and noninvasive detection of D-Cys in the brain via nitrile-aminothiol bioorthogonal reaction. The lipophilic 7'F-CBT exhibits enhanced blood-brain barrier (BBB) permeability a
