ODC1 Polyamine Metabolism Drives Prostate Cancer via AKT and Splicing.

Prostate cancer is an aggressive disease with limited quantifiable biomarkers. One gene of interest is ODC1, which encodes ornithine decarboxylase, the rate-limiting enzyme converting ornithine to putrescine in polyamine metabolism. Although ODC1 is known to be involved in prostate cancer development, exactly how it drives the disease mechanistically is not fully understood. To explore this, we created a prostate cancer cell model with reduced ODC1 expression and examined its effects on tumour behaviours. Knocking down ODC1 significantly slowed cell growth and movement while increasing cell de

Study on the Function and Mechanism of Neutrophil Extracellular Traps in Regulating Necroptosis Following Traumatic Brain Injury.

PURPOSE: Traumatic brain injury (TBI) remains a major global public health challenge with high morbidity and mortality, and secondary injury characterized by neuroinflammation, brain edema, and neuronal cell death is a critical determinant of patient prognosis. Neutrophil extracellular traps (NETs) and necroptosis are involved in TBI pathology, but their crosstalk remains unclear. Here, we used NETs inhibitors (Cl-amidine and DNase I) and the necroptosis inhibitor Necrostatin-1 (Nec-1) to investigate the roles of NETs and necroptosis in neuronal injury following TBI. METHOD: Male C57BL/6J mic

Integrated Genome-wide Association and Single-cell Transcriptomic Analysis Identifies OAT as Therapeutic Targets for Periodontitis.

Periodontitis is a chronic inflammatory disease affecting tooth-supporting tissues. Beyond microbial dysbiosis, host metabolic regulation plays a critical role. This study identified ornithine aminotransferase (OAT), a mitochondrial enzyme in amino acid metabolism, as associated with altered fibroblast phenotypes and metabolic profiles in periodontitis. Integrative genetic analysis showed a putative causal relationship between increased OAT expression and disease risk. Single-cell RNA-Seq revealed OAT enrichment in fibroblasts, especially in subsets with inflammatory and matrix-remodeling char

Impact of long-term nitrogen scavenger therapy on clinical outcome in individuals with urea cycle disorders.

Principles of long-term medical management in individuals with urea cycle disorders (UCDs) encompass (1) a low protein diet, (2) supplementation of arginine and/or citrulline along with essential amino acids, nutrients, vitamins and trace elements, and (3) use of nitrogen scavenging agents to reduce recurrent hyperammonemic events (HAEs). These principles aim at providing metabolic stability, elimimation of chronic complications, and achievement of normal development as well as growth. A retrospective comparative analysis was performed by studying 138 individuals with male ornithine transcarba

Current Insight into Human Ornithine Aminotransferase: A Review.

Human ornithine aminotransferase (hOAT) is a mitochondrial matrix pyridoxal-5'-phosphate enzyme (PLP) that catalyzes the reversible transfer of the δ-amino group of L-ornithine (L-Orn) to α-ketoglutarate (α-KG) yielding glutamate-5-semialdehyde (GSA) and glutamate. GSA is prone to cyclize to Δ1-pyrroline-5-carboxylate. Human OAT holds significant clinical and scientific interest because (i) its dysfunction causes gyrate atrophy (GA) of the choroid and retina, a rare autosomal recessive disease, and (ii) it is recognized as a potential target for chemotherapeutic drug development, being overexp

Role of Aspartate in Immune Response and Mortality in a Polymicrobial Sepsis Model: Insights from Metabolomics and Transcriptomics.

Sepsis is a life-threatening syndrome characterized by dysregulated host responses to infection. In addition to early hyperinflammation, many patients develop profound immune suppression, and multiple targeted immunotherapies have failed to improve outcomes, highlighting the need for actionable biomarkers and new therapeutic strategies. Here, we integrated metabolomic and transcriptomic profiling of peripheral blood mononuclear cells (PBMCs) and splenocytes in rat models of polymicrobial sepsis to identify metabolites associated with immune dysfunction. Candidate findings were validated using

Ubiquitously expressed prefoldin-like chaperone (UXT) regulates putrescine metabolism and promotes colorectal cancer progression.

Colorectal cancer (CRC) is a prevalent and increasingly common malignancy that poses significant threats to patient survival and quality of life. This study investigates the role of ubiquitously expressed prefoldin-like chaperone (UXT) in regulating polyamine metabolism, particularly putrescine, and its impact on CRC progression. Through comprehensive bioinformatics analysis, UXT was identified as a key factor positively correlated with putrescine abundance in CRC cell lines. Clinical samples confirmed upregulation of UXT and its positive correlation with putrescine levels. Functional assays r

The Multi-dimensional Mechanisms and Transformation Prospects of the Intratumoral Microbiota-Arginine Metabolism Axis in Tumor Progression and Immune Regulation.

The intratumoral microbiota, as an important component of the tumor microenvironment (TME), impact tumor progression by regulating the arginine-ornithine metabolic axis. It has become a new frontier in tumor research. Arginine is a crucial amino acid in TME, and its metabolites, ornithine and polyamines, directly promote tumor proliferation and induce immunosuppression. Intratumoral microbiota mainly exert their effects through two direct pathways: 1) arginine depletion, such as Streptococcus in gastric cancer. Specific intratumoral microbiota highly express arginine deiminase (ADI) or arginas

ARG1-polyamine axis: cell-type-specific functions in disease pathogenesis and therapeutic targeting.

ARG1 catalyzes the conversion of L-arginine to L-ornithine, urea, polyamines, and L-proline, thereby balancing nitrogen detoxification with tissue-specific roles in proliferation and immunity. This review delineates the context-dependent functions of ARG1 across diverse cell types-including tumor cells, immune cells, endothelial cells, keratinocytes, and stem cells. In tumors, ARG1 drives immunosuppression and metabolic reprogramming but can paradoxically suppress tumorigenesis. Immune modulation via ARG1-polyamine crosstalk regulates T cell differentiation, macrophage polarization, and microb

Serum metabolite patterns of adipose tissue distribution and body composition subphenotypes.

BACKGROUND: Obesity prevalence is increasing globally, accompanied by increases in obesity-related diseases. While obesity is usually defined by BMI, the development of obesity-related disease might be better characterized by specific adipose tissue (AT) distributions or body composition subphenotypes. Serum metabolite patterns reflecting AT distribution could provide insights into potential underlying pathophysiological pathways and the interplay between AT depots. We therefore aim to identify metabolite signatures associated with specific AT depots and body composition subphenotypes. METHOD

Novel paracellular marker based on ᴅ-dipeptide structure for highly sensitive quantification with UPLC-MS/MS for in vitro and in vivo blood-brain barrier permeability analysis.

BACKGROUND: The blood-brain barrier (BBB) paracellular permeability must be evaluated in various contexts in vitro and in vivo, including pharmacological evaluation of drug candidates, investigations of pathological changes in disease and model development. However, most available paracellular marker substances are either radioactive, lack sensitivity and thereby require large sample volumes, or they can influence the BBB themselves via osmotic pressure. Moreover, in drug permeability studies, an adequate paracellular marker should be detectable in the same sample as the compound being tested,

Energy-Metabolism-Enhancing Probiotics Enhance the Therapeutic Response to a Glucagon-like Peptide-1 Receptor Agonist.

Background/Objectives: Glucagon-like peptide-1 receptor agonists (GLP-1RAs) are effective treatments for obesity, but substantial weight regain is common after therapy is discontinued. This study investigated whether probiotic strains with anti-obesity effects could enhance GLP-1RA-induced weight loss and attenuate post-treatment weight rebound. Methods: Candidate lactic acid bacteria were screened for anti-obesity efficacy in a high-fat-diet (HFD)-induced obese mouse model, and the selected strain was further characterized using metabolomic profiling of culture supernatants. Luteolin ameliorates Escherichia coli-induced intestinal injury by modulating gut microbiota, metabolites and the TLR4/MyD88/NF-kB signaling pathway.

Luteolin, a naturally occurring flavonoid abundantly found in various fruits and vegetables, possesses anti-inflammatory and antioxidant properties. Its biological activities, including modulating immune responses and alleviating oxidative stress, make it a promising therapeutic candidate for inflammatory diseases. However, the precise role of this compound in mitigatingEscherichia coli induced (E. coli-induced) intestinal inflammation remains largely unexplored. More specifically, the mechanistic underpinnings by which it preserves intestinal mucosal barrier integrity, fine-tunes the activati

Arginase 2 regulates cholesterol biosynthesis in endothelial cells.

BACKGROUND: Arginase 2 (Arg 2) is a mitochondrial enzyme that hydrolyzes L-arginine to L-ornithine and urea, influencing endothelial nitric oxide (NO) bioavailability and vascular function. Although Arg 2 is implicated in endothelial dysfunction under hypercholesterolemic and oxidative stress conditions, its potential role in endothelial cholesterol metabolism remains unknown. METHODS: Arg 2 was deleted in immortalized human umbilical vein endothelial cells (HUVECs) using CRISPR/Cas9, followed by transcriptomic analyses. Differential gene expression was validated by quantitative RT-PCR and im

Maternal-prenatal gut microbiome-systemic metabolome perturbations and TH2-skewed immunity link to offspring gut microbiome disruption and atopic dermatitis susceptibility.

BACKGROUND: Emerging evidence suggests that maternal-prenatal gut microbiome disturbances shape offspring allergic outcomes through modulation of the in utero immune environment. Yet, no comprehensive clinical studies in human mother–offspring dyads have deconvoluted the maternal-prenatal gut microbiome and systemic immune-metabolome signatures underlying offspring allergic predisposition. METHODS: We performed a longitudinal nested case–control study involving 128 well-characterized mother–offspring dyads from defined cases (offspring with atopic dermatitis (AD); n = 64) and controls (offspri

Gut microbiota reshapes host energy metabolism to modulate depressive behaviors.

Disturbances in energy metabolism are a key pathophysiological feature of major depressive disorder (MDD). The gut microbiota, as a critical regulator of host metabolism, may influence systemic energy homeostasis and contribute to depression. To investigate this, we performed a multi-omics analysis integrating targeted metabolomics and shotgun metagenomics on samples from 100 MDD patients and 68 healthy controls. MDD patients exhibited significant disruptions in central energy pathways (glycolysis, TCA cycle, and ornithine cycle), which correlated with symptom severity and cognitive impairment

Serum Proteomic Analysis Using Gel-Based Liquid Chromatography Tandem Mass Spectrometry Reveals Differences Between Canine Oral Malignancies and Non-Malignant Conditions.

BACKGROUND: Canine oral cancers are difficult to manage due to complex biology and a lack of non-invasive biomarkers. Proteomic approaches, particularly gel-based liquid chromatography-tandem mass spectrometry (GeLC-MS/MS), have been used on tissue and saliva, but serum remains obscure despite its clinical accessibility and ability to reflect systemic disease. OBJECTIVES: This study evaluated GeLC-MS/MS for serum proteomic profiling in canine oral malignancies, compared to benign and healthy conditions. METHODS: We analysed 62 serum samples from dogs with oral melanoma (OM, n = 28), oral squ

Dietary advanced glycation end products and risk of cardiometabolic disorders in older adults: cross-sectional data from the 2007-2021 Korea National Health and Nutrition Examination Survey (KNHANES).

BACKGROUND: Advanced glycation end products (AGEs) have been identified as potential dietary risk factors for cardiometabolic disorders. Despite studies in Western populations, little is known about the impact of AGEs in elderly Asian with different dietary patterns and aging dynamics. This study aimed to examine the association between dietary intake of three representative AGEs—Nɛ-(carboxymethyl)lysine (CML), Nɛ-(1-carboxyethyl)lysine (CEL), and Nδ-(5-hydro-5-methyl-4-imidazolone-2-yl)-ornithine (MGH1)—and the risk of cardiometabolic disorders among older Korean adults. Pedigree and Functional Analysis of Two Cryptic OTC Variants Causing Ornithine Transcarbamylase Deficiency in Two Unrelated Chinese Male Patients.

BACKGROUND: Ornithine transcarbamylase deficiency (OTCD, MIM#311250) is a rare X-linked urea cycle disorder causing hyperammonemia. While around 600 pathogenic OTC variants have been reported, cryptic changes like synonymous or in-frame variants remain poorly characterized and are easily overlooked in routine screening. METHODS: We analyzed the clinical and genetic profiles of two unrelated Chinese male patients clinically diagnosed with OTCD. Whole-exome sequencing (WES) was performed to identify the genetic etiology. A minigene assay was then used to assess the splicing effect of the detect

Sensory nerve desensitisation exerts differential effects on the severity of acute pancreatitis in rodent models.

Acute pancreatitis (AP) is a common gastroenterological disorder characterized by severe abdominal pain and inflammation. Despite its high mortality rate, the pathomechanism of AP remains incompletely understood, although neurogenic inflammation - where nerve-derived mediators trigger or amplify inflammatory responses - also appears to contribute to its pathogenesis. The aim of this study was to further investigate the role of neurogenic inflammation in AP and the impact of sensory nerve desensitisation on disease severity. Sensory afferent neurons were ablated using resiniferatoxin (RTX) prio
