ODC1-Mediated Ornithine Metabolism Exacerbates Obesity by Disrupting AMPK/ACC Pathway.

Ornithine (OR) is a key intermediate metabolite; however, its molecular role in obesity remains unclear. This study aimed to investigate the effects of OR and its rate-limiting enzyme, ornithine decarboxylase 1 (ODC1), on lipid metabolism using high-fat diet (HFD)-induced obese C57BL/6 mice and C3H10T1/2 cell models. The results showed that OR supplementation and ODC1 overexpression exerted similar effects, including significantly aggravated HFD-induced obesity, elevated serum polyamine levels, impaired glucose tolerance, reduced oxygen consumption, and hepatic steatosis. Transcriptomic analys

Efficacy and safety of pegzilarginase in patients below 2 years of age with arginase 1 deficiency: a phase 3, open-label, multi-centre study.

BACKGROUND: Arginase 1 Deficiency (ARG1-D) is a rare metabolic disorder characterized by marked hyperargininaemia and progressive neurological impairment. Dietary protein restriction alone is often insufficient to normalize plasma arginine (pArg) or prevent disease progression. Pegzilarginase, a recombinant human ARG1 enzyme therapy, has demonstrated pArg normalization and improved clinical outcomes in patients ≥2 years. This study evaluated safety, pharmacokinetics (PK), and activity of pegzilarginase in patients <2 years. METHODS: In this phase 3, open-label, single-arm study (NCT06582524,

Disrupted erythrocyte S1P-eNOS axis promotes hypoxia, hypertension and fibrosis in obstructive sleep apnoea-hypopnoea syndrome.

BACKGROUND AND AIMS: Obstructive sleep apnoea-hypopnoea syndrome (OSAHS) has emerged as a global epidemic with profound cardiovascular and renal consequences, yet its early pathogenic mechanisms remain poorly understood. Whether red blood cells (RBCs) act as the primary hypoxia sensor that transduces intermittent apnoea into irreversible outcomes remains enigmatic. This study aims to define the pathogenic nature of RBCs during the progression of OSAHS with a goal of identifying early biomarkers and targeted treatments to prevent detrimental outcomes. METHODS: A large OSAHS cohort and matched

Deciphering underground decarboxylase activity towards Nε-modified lysine derivatives in enterobacteria.

Thermal food processing generates diverse compounds interacting with the gut microbiota. Despite their abundance, the microbial turnover of diet-borne Nε-modified lysine derivatives remains largely unexplored. We demonstrate that the enterobacterial ornithine decarboxylase SpeC degrades the prevalent advanced glycation end product Nε-carboxymethyllysine (CML) to carboxymethylcadaverine via an underground activity (∼4 molecules/enzyme/min). This promiscuity extends to additional Nε-modified lysine derivatives - namely formylated (FmL), monomethylated (MML) and dimethylated (DML) lysine - yieldi

Functional editing of the OTC locus by targeted integration with phenotype correction and restoration of endogenous expression patterns.

Here, we report highly efficient functional repair of the ornithine transcarbamylase (OTC) locus in mutant mouse and human hepatocytes in vivo using a dual adeno-associated virus system delivering CRISPR-Cas9 editing reagents and a promoterless donor for targeted integration. The approach was mutation agnostic and targeted intronic sequences to prevent inadvertent inactivation of hypomorphic alleles. Notably, in a murine model, we corrected the metabolic defect and simultaneously achieved liver-wide restoration of physiological metabolic zonation of Otc expression by capturing native cis-actin

Gut metaproteomics reveals activated arginine catabolism and impaired arginine biosynthesis in systemic lupus erythematosus.

UNLABELLED: The resilience of microbial metabolic functions during gut microbiome dysbiosis depends on functional redundancy across taxa. However, this ecological principle remains largely unexplored in human autoimmune diseases such as systemic lupus erythematosus (SLE). Here, we utilized quantitative metaproteomics to analyze fecal samples from 103 SLE patients and 62 healthy controls. Analysis of 30,124 protein groups revealed a protein-abundance-based shift in microbial arginine pathway capacity. Specifically, argininosuccinate synthase (ArgG), the committed enzyme for arginine biosynthesi

Serum metabolomics profiling in head and neck squamous cell carcinoma: a multicentre diagnostic study.

BACKGROUND: Head and neck squamous cell carcinoma (HNSCC) is frequently diagnosed at advanced stages, when outcomes are poor. Biomarkers for HNSCC detection remain needed. METHODS: We performed serum metabolomics profiling in 938 participants, including 457 patients with HNSCC, 451 controls, and 30 patients with oral potentially malignant disorders (OPMDs). Discovery and external validation cohorts were analysed using LC-MS/MS. Machine learning-based feature selection was used to develop an 8-metabolite diagnostic model. Disease specificity was assessed in an independent third-centre cohort.

The Role of Urea Cycle Functional Studies in Preclinical Research.

Inborn errors of metabolism affecting the urea cycle are rare severe conditions caused by impaired nitrogen detoxification, leading to hyperammonemia and neurological morbidity across a broad clinical spectrum. Current biochemical diagnostics largely rely on quantification of metabolites. While indispensable for diagnosis and acute management, these snapshot readouts are highly dependent on external factors and often fail to reflect urea cycle function in metabolically compensated states, limiting their usefulness for disease stratification, therapeutic monitoring, and preclinical evaluation o

C5a-C5aR1 axis activation drives neutrophil extracellular trap formation in SARS-CoV-2 N protein-mediated acute kidney injury.

The mechanism of COVID-19-associated acute kidney injury (AKI) remains incompletely understood. This study aimed to investigate the regulatory role of the C5a-C5aR1 axis in neutrophil extracellular trap (NET) formation in SARS-CoV-2 nucleocapsid (N) protein-induced AKI. An AKI mouse model with kidney-specific N protein expression was established using ultrasound-microbubble-mediated technology, and interventions with the C5aR1 inhibitor DF2593A or DNase I (Pulmozyme) were administered to detect renal neutrophil infiltration, NET deposition, C5a-C5aR1 expression, and renal function indices. Polyamine metabolic reprogramming in ependymal cells promotes endogenous repair after spinal cord injury.

The mechanisms underlying the activation of endogenous stem cells following spinal cord injury (SCI) remain a fundamental challenge in neural regeneration. Ependymal cells, which constitute a resident stem/progenitor population, possess latent regenerative potential, yet the metabolic cues governing their activation remain poorly understood. In this study, by integrating single-cell RNA sequencing with computational metabolic profiling, we systematically characterized the temporal transcriptional and metabolic reprogramming of injury-responsive ependymal cells after SCI. We found that SCI indu

Systematic verification of saliva-based biosignatures for oral cancer via targeted metabolomics.

Oral squamous cell carcinoma (OSCC) remains a major global health challenge, and early detection is essential for improving patient outcomes. To identify reliable salivary biomarkers, we performed a targeted metabolomics study using 13C2/12C2-dansylation, integrating candidate metabolites from published OSCC metabolomics datasets and systematically verifying them through LC-MRM-MS. A panel of 26 metabolites was quantified in saliva samples from 299 subjects, including healthy controls (n = 98), oral potentially malignant disorders (OPMD I, n = 50; OPMD II, n = 53), and OSCC patients (n = 98).

Rapid improvement of cystoid macular edema in a patient with gyrate atrophy.

Gyrate atrophy (GA) is a rare autosomal recessive disease caused by a defect in the enzyme ornithine aminotransferase (OAT). We report a case of a 4-year-old boy with GA of the choroid and retina. Following initiation of an arginine-restricted diet and targeted supplementation, there was rapid improvement in his cystoid macular edema (CME) and foveoschisis. We also report clinical and imaging findings from his 2-year-old sister, who carries the same genetic variant. This report highlights the potential for early metabolic intervention to produce rapid anatomical improvement in GA-associated CM

Low-Dose Ionizing Radiation and Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS): A Review of Recent Evidence and Future Research Directions Toward the Elucidation of a Metabolic, Immunologic, and Signaling Cascade.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is an idiopathic, multisystem disorder marked by debilitating fatigue, post-exertional malaise, cognitive dysfunction and neuroinflammation. Its etiology remains unclear, yet emerging evidence implicates a complex interplay between immune dysregulation, metabolic impairment, mitochondrial bioenergetics, and environmental stressors such as viral infection or low-dose ionizing radiation (LDIR). To develop treatments for ME/CFS, it is essential to identify suitable targets for therapy. In this narrative review, we discuss recent findings

Evidence of Hypoxia Signaling and Endothelial Activation in Migraine: Relationships Between HIF-1α, VEGF-A, and Arginine Metabolism.

Background/Objectives: Migraine is a common neurovascular disorder associated with substantial disability. Increasing evidence suggests that hypoxia-related signaling, endothelial dysfunction, and nitric oxide metabolism contribute to its pathophysiology. This study investigated the relationships between hypoxia-inducible factor-1 alpha (HIF-1α), vascular endothelial growth factor A (VEGF-A), and arginine pathway metabolites in chronic migraine. Methods: In this observational study, fasting ethylenediaminetetraacetic acid (EDTA) plasma samples were obtained from 28 patients with chronic migrai

Hypoxia-Associated Remodeling of the Arginine-Citrulline-Ornithine Axis in Parkinson's Disease and Restless Legs Syndrome: A Targeted LC-MS/MS and HIF-1α Profiling Study.

Background and Objectives: Hypoxia-inducible factor-1 alpha (HIF-1α) is a central regulator of cellular responses to hypoxia and has been implicated in the pathophysiology of several neurological disorders. Parkinson's disease (PD) and restless legs syndrome (RLS) have both been associated with alterations in oxygen sensing, mitochondrial dysfunction, and disturbances in amino acid metabolism; however, the relationship between HIF-1α and amino acid metabolic pathways in these disorders remains incompletely understood. The present study investigated circulating HIF-1α concentrations and amino a

L-citrulline treatment of nitric oxide deficiency in MELAS a phase I dose-finding and safety study.

There is evidence that nitric oxide deficiency occurs in mitochondrial encephalopathy, lactic acidosis, and stroke-like episodes (MELAS) and may result in impaired blood perfusion in small blood vessels, contributing to stroke-like episodes (SLEs). The primary aim of this phase one study was to estimate the maximum tolerated dose (MTD) of L-citrulline in adults with MELAS. The primary safety outcome was the occurrence of a dose limiting toxicity (DLT) in the first eight weeks after treatment initiation. Secondary outcomes included changes in cerebral blood flow (CBF), cerebrovascular reactivit

Carbonyl stress by glyceraldehyde promotes glycation of nuclear non-muscle myosins in DNA-protein crosslink assemblies.

DNA-protein crosslinks are bulky lesions that block DNA replication and transcription and are increasingly recognized in metabolic disorders characterized by carbonyl stress. Glyceraldehyde is a highly reactive triose that forms advanced glycation end-products, but its contribution to nuclear DNA-protein crosslink formation has not been defined. We examined glyceraldehyde-induced DNA-protein crosslinks in rat cardiomyoblast-derived H9c2 cells using an advanced recovery of potassium sodium dodecyl sulfate precipitates (ARK) assay combined with liquid chromatography-tandem mass spectrometry. Relationship between elevated arginine and pathophysiology of progressive disease in arginase 1 deficiency.

BACKGROUND: Arginase 1 deficiency (ARG1-D; OMIM #207800) is a rare urea cycle disorder caused by loss of arginase activity, preventing the hydrolysis of arginine to ornithine and urea. The resulting hyperargininaemia leads to accumulation of neurotoxic metabolites including guanidino compounds and nitric oxide in the central nervous system. Nervous system structures are affected, including the corticospinal tracts, cerebral and cerebellar white matter, basal ganglia, hippocampus, and corpus callosum. Clinically, patients present with developmental and neurological impairment, including growth

Clinical and genetic analysis of Liver-Predominant ornithine transcarbamylase deficiency caused by a novel de novo OTC sequence variant in a female child.

Ornithine transcarbamylase deficiency (OTCD) is the most common urea cycle disorder. Although the classical presentation is dominated by hyperammonemia-related neurological manifestations, some patients may present with atypical liver-predominant phenotypes, which can delay early diagnosis. Here, we identified a 3-year-6-month-old female patient who presented primarily with liver dysfunction, characterized by elevated transaminases, mild coagulation abnormalities, and increased hepatic parenchymal echogenicity, without overt jaundice or typical neurological symptoms. Metabolic mass spectrometr
