Hepatocellular carcinoma (HCC) is a major cause of cancer deaths with few treatment options. Cuproptosis, a copper-dependent cell death pathway, is a promising therapeutic target for HCC. In the present study, cuproptosis-related genes in HCC were identified from GeneCards, with dihydrolipoamide dehydrogenase (DLD) emerging as the top hub gene via STRING and Cytoscape analysis. DLD was markedly overexpressed in HCC tissues compared to normal liver tissues (P = 4.6 × 10⁻⁵) and was associated with advanced TNM stage, fibrosis, and reduced overall survival in both the TCGA (P = 0.039) and ICGC (P

The expression of glycine N-methyltransferase (GNMT), a critical regulator of S-adenosylmethionine (SAM) levels, is down-regulated in humans with metabolic dysfunction-associated steatotic liver disease (MASLD) and hepatocellular carcinoma (HCC). In low-fat-fed mice, GNMT knockout (KO) induces liver steatosis that progresses to HCC. This is accompanied by increased SAM and a shunting of tricarboxylic acid (TCA) cycle intermediates away from gluconeogenesis to other biosynthetic pathways that support lipid accretion and tumorigenesis. The objective of this study was to test whether this metabol

Gluconeogenesis, a dual-purpose pathway in type 2 diabetes mellitus (T2DM), not only synthesizes glucose but also clears metabolic waste via the Cori cycle (lactate recycling through LDH, PC, PEPCK) and Alanine cycle (nitrogen disposal via ALT, GDH, urea cycle), preventing acidosis, ROS accumulation, and ammonia toxicity. Metformin, the cornerstone T2DM therapy, inhibits gluconeogenesis by targeting mitochondrial complex I, elevating AMP/ATP ratios, and activating AMPK-PKCι/λ signaling to repress CREB-CRTC2-driven PEPCK/G6Pase expression, disrupting these cycles. This leads to lactate, pyruvat

BACKGROUND: Cannabidiol (CBD) has shown potential benefits in managing symptoms associated with autism spectrum disorder (ASD), although its underlying mechanisms of action remain unclear. This study investigated whether CBD improves ASD-like behavior in association with changes in lipid metabolic disturbances and apoptosis-related signaling in a prenatal valproic acid (VPA) rat model.
METHODS: Male offspring from VPA-exposed rats received CBD (10 mg/kg, intraperitoneal, twice daily) for 7 days starting on postnatal day 21. Behavioral testing evaluated hyperactivity, social interaction, and r

This study aims to investigate, from the perspective of pathologic-symptom toxicology, the effects of salt processing on Psoraleae Fructus(BGZ)-induced liver injury in a rat model of kidney-Yin deficiency(Yin) and to elucidate the underlying mechanisms through metabolomics. A Yin model was established in rats and treated with BGZ or salt-processed BGZ(YBGZ). Yin manifestations were evaluated based on body weight, anal temperature, and the serum cyclic adenosine monophosphate(cAMP)/cyclic guanosine monophosphate(cGMP) ratio. Liver injury was assessed based on serum alanine aminotransferase(ALT)

AIMS/HYPOTHESIS: Dysregulation of hepatic glucose production, primarily driven by aberrantly elevated gluconeogenesis, is a core pathological feature of type 2 diabetes. However, the molecular mechanisms governing this process remain incompletely understood. Protein disulfide isomerase A5 (PDIA5) is ubiquitously expressed, but its specific function in hepatic metabolism is unknown. We hypothesised that PDIA5 plays a critical role in regulating hepatic gluconeogenesis, and that its dysregulation contributes to hyperglycaemia.
METHODS: We assessed hepatic PDIA5 expression in liver biopsies from

The kidney is a high-energy-consuming organ, and glucose is one of its principal fuel sources. It has been documented that disturbed renal glucose metabolism occurs in acute kidney injury (AKI), but whether these disturbances are consistent in different types of AKI, and how they change during the transition from AKI to chronic kidney disease (CKD), remain to be addressed. In this study, we used AKI models induced by cisplatin (20 mg/kg, 48 h), lipopolysaccharide (10 mg/kg, 48 h), and ischemia-reperfusion (48 h) to mimic distinct etiologies, and the catalytic enzymes involved in glucose transp

Cardiac metabolism relies on glycogen conversion by glycolysis. Glycolysis intersects fatty acid oxidation and often directs a signal crosstalk between redox metabolites. Myocardium with ischemia/reperfusion significantly diverts from normal metabolism. Prospectively, peroxisome lies central to metabolism and redox changes, but mechanisms underlying in ischemia/reperfusion remain undefined. This work aims at investigating the potential effects and mechanisms of Salvianolic acid B (Sal B) in cardioprotection through metabolic remodeling. Following experiments, we found that Sal B is absorbed in

Buffaloes play a crucial role in Asian agriculture, enhancing food security and rural development. Their distinct metabolic needs drive tissue-specific mitochondrial adaptations, regulated by both mitochondrial and nuclear genomes. This study explores how nuclear-encoded mitochondrial genes involved in lipid and carbohydrate metabolism vary across tissues-an area with significant implications for buffalo health, productivity, and human health. We hypothesize that tissue-specific variations in metabolic pathways are reflected in the expression of nuclear-encoded mitochondrial genes, which are t

Gestational Diabetes Mellitus (GDM) is an emerging maternal health problem with increasing incidences. The lack of complete understanding of its pathophysiological mechanisms and novel regulatory biomarkers makes early diagnosis difficult. High-throughput RNA sequencing and computational bioinformatics analyses were conducted to identify novel hub genes, and their regulatory mechanisms were validated through qRT-PCR, western blot, and siRNA-mediated knockdown studies. Intermediate metabolites and circulatory levels of amino acids in the serum of GDM patients and healthy controls were measured.

Age-related hearing loss (ARHL) is a complex condition with genetic, aging, and environmental influences. Sirtuins, particularly SIRT1, are NAD-dependent protein deacetylases critical to aging and stress responses. SIRT1 is modulated by nicotinamide N-methyltransferase (NNMT) and its product, N1-methylnicotinamide (MNAM), which influence ARHL progression. While SIRT1 is protective under certain conditions, its overexpression may paradoxically exacerbate hearing loss. This study examines MNAM supplementation's impact on SIRT1 expression and ARHL in low-fat diet (LFD)-fed B6 and CBA mice. Mice w

Yak meat is in high demand due to its unique flavor. Thus this research utilized GC × GC-ToF-MS to discover important flavor compounds in yak meat raised during the cold season under different feeding systems: traditional grazing (TG), grazing-based supplementation (GS), and stall-feeding (SF). Meat quality results showed that SF significantly improved meat's lightness and tenderness (P < 0.05), as compared to TG. Intramuscular fat (2.7 g/100 g) was highest in the SF, followed by the GS (2.46 g/100 g) and the TG (1.57 g/100 g), whereas protein content was similar in the GS and TG, but again hi

The leaves of Annona muricata Linn. have long been utilized in traditional medicine for diabetes treatment, and there is no study that has employed a metabolomics approach to investigate the plant's effects in managing the disease. We aimed to explore the antidiabetic effects of the standardised A. muricata leaf extract on diabetes-induced rats by alloxan monohydrate (Ax) and nicotinamide (NA) using a proton nuclear magnetic resonance (¹H-NMR)-based metabolomics approach. Absolute quantification was performed on the leaf extract using ultra-high-performance liquid chromatography-tandem mass sp

Schistosomiasis is a parasitic disease that imposes a significant burden on society. The eggs are the primary pathogenic factor in schistosomiasis, and their accumulation in liver could lead to the formation of granulomas and liver fibrosis. However, the metabolic changes in liver resulting from schistosomiasis remain poorly understood. We established a mouse model of schistosomiasis japonica, where the eggs accumulate in the liver and form egg granulomas. We used mass spectrometry imaging to analyze the differences in metabolites among various liver regions, including the liver tissue from no

Hepatic deletion of methionine adenosyltransferase-1a (Mat1a) in mice reduces S-adenosylmethionine (SAMe), a key methyl donor essential for many biological processes, which promotes the development and progression of metabolic dysfunction-associated steatotic liver disease (MASLD). Hyperglycemia and reduced MAT1A expression, along with low SAMe levels, are common in MASLD patients. This study explores how Mat1a-knockout (KO) hepatocytes respond to prolonged high glucose conditions, focusing on glucose metabolism and lipid accumulation. Hepatocytes from methionine adenosyltransferase-1a-knockou

Excessive lipid accumulation can lead to obesity, metabolic-associated fatty liver disease, and type 2 diabetes. However, there are currently few drugs that could effectively and safely inhibit the accumulation of intracellular lipids. In this study, we observed that baicalin significantly altered cellular respiration by reducing mitochondrial oxygen consumption while enhancing glycolytic flux, accompanied by increased phosphorylation of AMPK and ACC, suggesting an adaptation to altered energy availability. Baicalin effectively reduced lipid droplet formation and intracellular triglyceride lev

Metabolic disorders and complications induced by high-fat diets (HFDs) are a hot research topic in aquatic animal nutrition and health, but the mechanism of gut microbes and their metabolites on muscle homeostasis is not yet clear. In this study, a 16-week HFD (Con, 6% fat and HFD, 12% fat) rearing experiment was conducted with a freshwater drum (20.88 ± 2.75 g, about 20,000 fish per pond) to investigate the underlying regulation of gut microbes on muscle nutrient and metabolism. Results revealed that HFD had no remarkable effect on proximate nutrients (moisture, ash, crude protein, and crude
