Effects of biguanides on the intermediate metabolism of glucose in normal and portal-strictured rats.

Phenformin and metformin treatments may be complicated by lactic adidosis. This metabolic complication seems favoured by preexistent hepatic disease. We have therefore compared the metabolic effects of phenformin and metformin on non fasting normal and portal-strictured rats. The latter group is characterized by impaired hepatic passage of these drugs without hepatocellular lesions. Given orally to normal rats over 5 days, phenformin (20 mg/kg/24 h) and metformin (150 mg/kg/24 h) decreased blood glucose levels and increased blood urea and the substrates of gluconeogenesis (alanine, glutamine,

Gluconeogenesis from alanine in patients with progressive malignant disease.

We have studied by tracer technique the conversion of the carbon skeleton of alanine to glucose in patients with progressive malignant disease. These data have been compared to similar studies done in patients with chronic undernutrition from other causes. The results show increased conversion of alanine to glucose in the overnight fasting state as compared to the control group. Whereas the percentage increases are comparable to those found with pyruvate-glucose cycling in such subjects, the total amount of carbon conversion is considerably less (alanine carbon, 5.6 mmol/hr, versus pyruvate ca

Nutritional and metabolic aspects of fatty liver disease in poultry.

Summary There are two metabolic disorders of major commercial importance in poultry that involve the occurrence of fatty deposits in the liver. Fatty Liver and Kidney Syndrome (FLKS) affects young birds and the main manifestations, lipid infiltrations into liver and many other organs, are apparently secondary effects of the primary lesion that lies in carbohydrate metabolism. Although several nutritional and environmental factors influence FLKS, the main factor is the vitamin, biotin. [Activity of gluconeogenetic enzymes of rat kidney cortex during acute hypoxia].

The activities of gluconeogenic enzymes of the rat kidney cortex was studied after exposure to lowered atmospheric pressure (200 mm Hg) for 3 hours. The hypoxic stress was found to cause an increase in the activities of phosphoenolpyruvate carboxykinase and alanine aminotransferase, but failed to affect significantly the activities of fructose-1,6-diphosphatase, glucose-6-phosphatase, and aspartate aminotranspherase. The ratio of glucose-6-phosphatase/hexokinase activities was increased under these conditions.

Induction of rat kidney gluconeogenesis during acute liver intoxication by carbon tetrachloride.

1. Glucose production from L-lactate was completely inhibited 24h after carbon tetrachloride treatment in liver from 48h-starved rats. The activities of phosphoenolpyruvate carboxykinase, fructose diphosphatase and glucose 6-phosphatase were decreased by this treatment in fed and starved rats, whereas lactate dehydrogenase activity was only decreased in fed animals. 2. The production of glucose by renal cortical slices from fed rats previously treated with carbon tetrachloride was enhanced when L-lactate, pyruvate and glutamine but not fructose were used as glucose precursors. Renal phosphoeno

[Hepatic energy metabolism disorders in emotionally-painful stress and prevention of these disorders with sodium gamma-hydroxybutyrate].

Content and biosynthesis of glycogen in rat liver tissue as well as transition of 14C from main substrates of tricarboxylic acid cycle to CO2, examined in liver slices, were studied under conditions of stress caused by pain and emotion. The stress was accompanied by mobilization of the glycogen deposits; glycogen synthesis in liver tissue and oxidation of pyruvate, asparate succinate or alpha-ketoglutarate in tricarboxylic acid cycle were inhibited. At the same time, the stress was accompanied by ulcerous impairments of gastric mucosa. The phenomena observed appear to be due to increased activ

Lactic acidosis.

Lactic acid is generated as the end product of anaerobic metabolism of glucose and is disposed by gluconeogenesis or oxidation. Changes in the lactate pyruvate ratio are not necessarily indicative of tissue hypoxia. The plasma lactate concentration is the result of lactate production and lactate removal (hepatic and renal gluconeogenesis; oxidation by muscle, liver and kidney). Lactic acidosis is defined as a state of metabolic acidosis (arterial pH less than 7.3) due to an increase in the blood concentration of lactate (greater than 2 mEq/l). Lactic acidosis may occur with evidence of tissue

Possible sites of interaction of acute renal failure with amino acid utilization for gluconeogenesis in isolated perfused rat liver.

Experiments were performed to elucidate the mechanisms involved in the enhanced conversion of amino acids to glucose in acute uraemic rats. Increased gluconeogenesis from a mixture of serine, threonine, lysine, glutamate, ornithine and citrulline was confirmed using a non-recirculating perfusion system. Stimulation was concentration dependent, being most pronounced at physiological amino acid concentrations. Stimulation of glucose and urea formation could be mimicked by using serine alone whereas with lactate and pyruvate inhibition of gluconeogenesis was observed. Serine dehydratase activity

A biochemical explanation for the fatty liver and kidney syndrome of broilers: its alleviation by the short-term use of dietary fat.

1. Fatty livers and kidney syndrome (FLKS) was induced in young broiler chickens by giving them a diet composed principally of wheat and meat meal. 2. FLKS resulted in reduced growth and increased liver weight; fasting for 18 h increased mortality, liver lipid and the specific activity of hepatic ATP-citrate lyase compared with birds fed on a commercial diet. The specific activities of hepatic fructose-1,6-diphosphate-1-phosphohydrolase and pyruvate carboxylase were reduced in birds suffering from FLKS and fasted for 18 h. 3. Feeding of the FLKS-inducing diet supplemented with 150 g animal tal

Regulation of proline oxidase activity by lactate.

We found that proline oxidase, the first enzyme of the proline degradative pathway, is inhibited by lactate. The Km of the enzyme for proline increases with increasing concentrations of lactate. Since proline can be a source for gluconeogenesis, regulation of proline degradation by lactate may serve as a mechanism for allocation of metabolic fuel sources. The marked inhibition of proline oxidase at levels of lactate that commonly occur in both genetic and acquired lactic acidosis may cause the previously unexplained hyperprolinemia seen in these metabolic disorders.

Streptozotocin-induced diabetes in the Chinese hamster. Biochemical and endocrine disorders.

Streptozotocin treatment (125 mg/kg) in the Chinese hamster induced hyperglycaemia, hypoinsulinaemia, hyperglucagonaemia and changes in body, liver, pancreas, stomach, kidney and adipose tissue weights. The pancreatic reserves of insulin and glucagon in the diabetic animals were low, but stomach glucagon high. These animals showed high levels of phosphoenolpyruvate carboxykinase and low levels of glucokinase, hexokinase, isocitrate dehydrogenase and malic enzyme, but normal levels of pyruvate kinase in the liver. Increases in lactate dehydrogenase subunit B and isozymes 2, 3 and 4 were also ob

The influence of branched chain aminoacids and their ketoderivatives on renal gluconeogenesis.

Isolated kidney tubules served as a model to investigate the direct effect of branched chain aminoacids and their ketoderivatives on gluconeogenesis. The data presented in this paper demonstrate that the ketoderivatives rather than the branched chain aminoacids themselves inhibit renal glucosesynthesis from various precursors entering the glucogenic pathway at different levels. Though the point of the inhibitory attack of ketoacids could not be localized, an impairement of the kidney cortex to respond to metabolic acidosis with an increase of gluconeogenesis is evident from the present data. Hyperalaninemia hyperpyruvicemia and lactic acidosis due to pyruvate carboxylase deficiency of the liver; treatment with thiamine and lipoic acid.

A 16-month-old female infant with severe mental and motor retardation, clinically diagnosed as Leigh's encephalomyelopathy, forms the basis of this study. This infant was found to have lactic acidosis, low cerebrospinal fluid glucose, hyperalaninemia, and increased levels of urine lactate, pyruvate and alanine. These laboratory studies suggested an inborn error in gluconeogenesis. Further investigation revealed a low level of hepatic pyruvate carboxylase activity. The patient's elder sister who also had mental and motor deterioration was then also found to have an elevated blood lactate. These

The mechanism of hypoglycemia due to semistarvation in the rat.

A quantity of diet providing 50% of the energy and protein requirement was fed to 4 week old rats. After 3 weeks, the amount of food was reduced to 25% and resulted in hypoglycemia in most of the rats within 4 to 7 days. The rats were divided into three groups: 1) normal controls; 2) malnourished, exhibiting blood sugar levels more than 40 mg/100 ml; 3) malnourished with blood sugar concentration less than 40 mg/100ml. The plasma concentrations of energy providing substrates: free fatty acids, glycerol, pyruvic acid, lactic acid and alanine were significantly lower in the hypoglycemic group th

Effect of urine metabolites from healthy and uremic subjects on gluconeogenesis in slices of rat kidney cortex and liver.

A high molecular weight fraction was obtained by extended dialysis of urine of healthy and uremic subjects. After addition to the incubation medium, this fraction inhibited gluconeogenesis by rat kidney cortex slices. From the six subfractions extracted by gel chromatography (Sephadex G 100) fraction IV caused a decrease of glucose formation. The activity of PEP-carboxykinase but not of pyruvate carboxylase was reduced, indicating a decreased formation of phosphoenol pyruvate. The total high molecular weight fraction stimulated glucose release by liver slices from fed but not from starved rats
