Mucopolysaccharidoses (MPS) represent a group of rare inherited lysosomal storage disorders, of which neurosurgical issues arise during childhood and adolescence. The literature however remains fragmented in terms of these specific outcomes. Correspondingly, we sought to synthesize a primer for neurosurgeons in regard to MPS, summarizing the neurosurgical manifestations and management reported in the contemporary literature. A systematic interrogation of the literature was performed according to PRISMA guidelines. Electronic databases were searched from inception through June 2026. Studies rep

Mucopolysaccharidoses (MPS) are a group of inherited metabolic diseases associated with impaired lysosomal degradation of glycosaminoglycans. The aim of this manuscript is to report a case of a patient with mucopolysaccharidosis type IIIC (MPS IIIC) diagnosed at 12 years of age - to raise awareness among pediatricians, pediatric neurologists, and endocrinologists about the possible misdiagnosis. The patient was initially diagnosed with autism spectrum disorder and hyperactivity with normal motor development. During follow-up, psychomotor regression with moderate intellectual disability was obs

Mucopolysaccharidosis type Ⅰ was a rare inherited metabolic disorder caused by a deficiency of alpha-L-iduronidase, resulting in the incomplete degradation of glycosaminoglycans that accumulate in lysosomes, leading to tissue damage and organ dysfunction. The main treatment options for mucopolysaccharidosis type Ⅰ were hematopoietic stem cell transplantation and enzyme replacement therapy. Patients who did not receive early replacement therapy often struggle to survive into adulthood, and cases of enzyme replacement therapy initiated in adulthood were extremely rare. This article summarized an

Mucopolysaccharidoses (MPS) are lysosomal storage disorders caused by enzyme deficiencies that impair glycosaminoglycan degradation and cause multisystem disease. We generated two human induced pluripotent stem cell (iPSC) lines from the KOLF2.1J background using CRISPR/Cas9-mediated knock-in of pathogenic variants: TRNDi045-A-40 carries a homozygous SGSH c.734G > A (p.Arg245His) variant associated with severe Sanfilippo syndrome type A (MPS IIIA), and TRNDi045-A-41 harbors a homozygous IDUA c.1205-1206 GG > AA (p.Trp402Ter) variant causing Hurler syndrome (MPS I). These lines model MPS diseas

Mucopolysaccharidosis type IIIB (Sanfilippo syndrome type B) is a neurodegenerative lysosomal storage disorder characterized by cognitive decline, behavioral abnormalities, and neuroinflammation, for which no effective central nervous system (CNS)-targeted therapy currently exists. Increasing evidence indicates that inflammatory processes contribute substantially to disease progression. In this study, we evaluated the effects of resveratrol on behavioral outcomes and neuroinflammatory markers in an MPS IIIB mouse model, including both sexes. Behavioral analyses demonstrated anxiety-like behavi

Mucopolysaccharidosis Type I (MPS I) is a rare autosomal recessive lysosomal storage disorder caused by mutations in α-ʟ-iduronidase (IDUA) enzyme, a glycosylated hydrolase responsible for glycosaminoglycan (GAG) degradation. Pathogenic variants are associated with a broad clinical spectrum, from severe Hurler syndrome to attenuated Scheie phenotype. The missense mutations L490P and P533R cause attenuated-intermediate MPS I, retaining residual enzyme activity. To investigate the structural mechanisms underlying these phenotypes, we performed all-atom molecular dynamics simulations -18 trajecto

Mucopolysaccharidoses (MPS) are a group of pediatric disorders that are distinguished based on a lysosomal enzyme deficit leading to accumulation of glycosaminoglycans (GAGs). One of the GAGs is heparan sulfate (HS) which accumulates in cells inside the body, but the most devastating symptoms are caused by its accumulation in the central nervous system. Resveratrol, a natural compound, reduces the HS accumulation in both MPS I and MPS IIIB. In this study, we investigated the effects of resveratrol on HS metabolism in cellular models of MPS I and MPS IIIB, with particular focus on its role in G

Mucopolysaccharidosis type I (MPS-I) is a hereditary lysosomal storage disorder caused by deficiency in the lysosomal enzyme α-L-iduronidase (IDUA) leading to pathogenic over-accumulation of glycosaminoglycans (GAGs), major constituent of connective tissue and mediators of cell signaling. We tested the therapeutic efficacy of the autolysosomal activator GHF201 in the Idua -/- mouse model of MPS-I. We show that GHF201 reduced GAGs and increased cathepsin activity in Idua -/- mouse embryonic fibroblasts (MEFs) and brain cells. In vivo GHF201 improved aberrant locomotion and behavior, reduced int

Vacuolar protein sorting 16 (VPS16) functions in endolysosomal trafficking. Biallelic VPS16 variants cause a mucopolysaccharidosis-like syndrome reported in only four patients, with neurological involvement ranging from mild developmental delay to severe impairment with epilepsy and pyramidal signs. We report an additional patient biallelic for VPS16 NM_022575.4:c.2272-18 C > A, with profound developmental delay, early-onset epilepsy, and spasticity. Multisystem involvement included recurrent infections, neutropenia, serosal effusions, and tubular proteinuria. Brain MRI showed white-matter abn

Bi-allelic variants in IDUA classically cause mucopolysaccharidosis type I (MPS I), a multisystem lysosomal storage disorder. This study describes 14 individuals from 12 families presenting with retinitis pigmentosa, later attributed to bi-allelic IDUA genotypes. Notably, seven individuals, despite comprehensive systemic assessments, showed no features consistent with MPS I at last assessment (aged 46-73 years). Leukocyte α-L-iduronidase activity was reduced but not abolished across all individuals assessed (n = 9), supporting a shared mechanistic basis for the mild, retina-predominant, or exc

New, rapid and inexpensive syntheses of 4-methylumbelliferyl fluorogenic substrates for α-L-iduronidase and iduronate-2-sulfatase, have been developed from a common β-D-glucuronosyl fluoride intermediate. The syntheses feature an efficient purple light-promoted, CCl4-free, one-pot Ferrier photobromination/radical debromination sequence for the key C-5 epimerization. The substrates are useful chemical tools for studying mucopolysaccharidosis type I (MPS I) and type II (MPS II).

BACKGROUND: Mucopolysaccharidosis type II (MPS II) is caused by a deficiency of the lysosomal enzyme iduronate-2-sulfatase. This study aimed to evaluate audiological characteristics and longitudinal hearing changes in patients with MPS II.
METHODS: We retrospectively reviewed 17 patients with MPS II whose hearing thresholds could be examined using pure-tone audiometry (PTA) at our institution for at least two years. Hearing was evaluated using auditory brainstem responses (ABR) and PTA. Longitudinal changes in hearing were analyzed to assess hearing progression and explore treatment-related e

Accurate optical-property extraction in turbid media requires reliable sensing across both near- and far-field scattering regimes, where classical diffuse reflection spectroscopy (DRS) is not equally valid. We present an integrated DRS and iterative multi-plane optical properties extraction (IMOPE) framework for characterizing turbid media across scattering regimes. Seven-plane phase retrieval enabled IMOPE analysis, while camera-based DRS provided an objective method for phase-image segmentation. Experiments were performed using tissue-like phantoms with reduced scattering coefficients of 0.3

Hunter syndrome (mucopolysaccharidosis type II, MPS II) is an X-linked lysosomal storage disorder caused by deficiency of iduronate-2-sulfatase (IDS), leading to lysosomal accumulation of glycosaminoglycans (GAGs), dermatan sulfate and heparan sulfate. Although traditionally viewed as an inherited metabolic disease, increasing evidence indicates that the most severe clinical burden of MPS II arises from central nervous system (CNS) dysfunction driven by convergent neurobiological mechanisms relevant to neuropsychiatric and neurodegenerative disorders. GAGs accumulation disrupts lysosomal integ

UNLABELLED: Mucopolysaccharidosis Type II (MPS-II) is a rare X-linked lysosomal storage disorder, characterized by deficiency of the lysosomal hydrolase iduronate 2-sulphatase (IDS). Cardiac impairments contribute to mortality in patients. Enzyme replacement therapy (ERT) with Idursulfase is a promising treatment for the cardiac dysfunction. We compared the proteome changes following intravenous (IV) with and without adjunct nebulized ERT in IDS-KO mice. Male mice were assigned to 4 groups: 1) WT IV + nebulized saline (WT; n=8); 2) IDS-KO IV + nebulized saline (KO; n=5); 3) IDS-KO IV Idursulfa

Mucopolysaccharidosis type IVA (MPS IVA) is an autosomal recessive lysosomal storage disorder caused by deficient activity of N-acetylgalactosamine-6-sulfatase (GALNS). In this single-center study, we analyzed the clinical and genetic characteristics of 82 pediatric MPS IVA patients, including 68 severe, 12 intermediate, and 2 mild cases. The most common clinical manifestations were pectus carinatum (72%), short stature (56%), and genu valgum (45%). All patients showed markedly reduced GALNS activity, although no significant difference was observed across phenotypic subgroups. Genetic analysis

Mucopolysaccharidosis IVA, also known as Morquio syndrome, is a rare autosomal recessive lysosomal storage disorder caused by a deficiency of N-acetylgalactosamine-6-sulfate sulfatase. Spinal involvement, particularly at the craniocervical junction, is a major contributor to morbidity and mortality due to atlantoaxial instability and cervical spinal stenosis. Early diagnosis and timely surgical intervention are essential to prevent irreversible neurological deterioration. In this report, we present the case of a 10-year-old girl with progressive quadriparesis due to severe cervical myelopathy

Mucopolysaccharidoses (MPS) are lysosomal storage disorders characterized by the accumulation of glycosaminoglycans (GAGs), which can lead to cytoplasmic alterations in leukocytes. The objective of this study was to characterize leukocyte inclusions in patients with different types of MPS and assess their diagnostic relevance. A total of 69 Brazilian patients diagnosed with five MPS types were evaluated through cytological analysis of peripheral blood smears. Distinct leukocyte inclusion patterns were observed in four subtypes. In MPS Type II, small violet cytoplasmic inclusions were present i
