Immune checkpoint inhibitor-based chemotherapy for granulocyte-colony stimulating factor-producing advanced esophageal squamous cell carcinoma: a report of three cases.

INTRODUCTION: Granulocyte-colony stimulating factor (G-CSF)-producing esophageal squamous cell carcinoma (ESCC) is a rare and highly aggressive malignancy. Currently, no specific treatment approach has been established for G-CSF-producing tumors, including those in the lungs, esophagus, and other organs. Here, we present three cases of advanced G-CSF-producing ESCC treated with immune checkpoint inhibitor (ICI) combination chemotherapy. CASE PRESENTATION: Case 1: A 52-year-old man was diagnosed with G-CSF-producing ESCC with multiple lymph node and lung metastases. First-line chemotherapy wit

Maternal-fetal type I interferon signaling drives TREM2 dysregulation and synaptic dysfunction in neurodevelopmental disorders.

Maternal infections during gestation cause perturbed neuroimmune interactions and increased risk of neurodevelopmental disorders in the offspring. Using a maternal immune activation model with polyinosinic:polycytidylic acid (poly(I:C)), we investigated how maternal infections reshape the progeny hippocampal landscape. Poly(I:C) increased maternal type I interferon (IFN-I), disrupting neuronal transcriptional trajectories and excitatory synapse homeostasis in juvenile offspring. Poly(I:C) offspring microglia exhibited reduced expression of triggering receptor expressed on myeloid cells 2 (TREM

The NLRP3/IL-6/EGR2/CSF-1 Axis Regulates Macrophage Differentiation and Inflammatory Resolution in Infection-Associated Wound Healing : NLRP3 Axis Controls Macrophage Fate in Wound Infection.

Chronic Pseudomonas aeruginosa (P. aeruginosa) infection impairs wound healing, yet the underlying inflammatory mechanisms remain poorly understood. This study investigates how the NLRP3 inflammasome orchestrates macrophage differentiation and inflammatory resolution during infection-associated wound healing. A murine P. aeruginosa wound infection model was established. Macrophage polarization dynamics were assessed by flow cytometry (CD11b/F4/80/Arg1/iNOS/CD206/CD86) and histology. Wound healing outcomes were compared between wild-type and NLRP3-/- mice via closure kinetics, ECM analysis (Mas

Dentate gyrus immaturity and region-specific synaptic and autophagy alterations across hippocampal subfields in schizophrenia.

BACKGROUND: Hippocampal abnormalities are consistently observed in schizophrenia. We previously identified a histomorphological phenotype in the dentate gyrus (DG) suggestive of developmental arrest, alongside region-specific alterations in autophagy markers. However, how these features relate to synaptic organisation and neuronal maturity markers remains unclear. METHODS: Post-mortem hippocampal tissue from older individuals with chronic schizophrenia (n = 10) and matched controls (n = 12) was examined using immunohistochemistry. Expression of synaptophysin and CD56 was assessed across the D

Haematopoietic Cells Deficient for Calreticulin or Expressing Its Oncogenic Variants Maintain Functional Antigen Presentation.

Intact antigen presentation in cancer cells is a key prerequisite for endogenous anticancer T cell responses and a starting point for immunotherapy. Calreticulin was postulated as a chaperone and integral part of the peptide-loading complex of MHC-I. Oncogenic mutations of calreticulin allow surface expression in complex with the thrombopoietin receptor, leading to ligand-independent activation, cell proliferation, and myeloproliferative neoplasms. We hypothesized that mutated calreticulin could also corrupt the function of MHC-I, as previous reports revealed decreased surface expression, pept

SFTSV infection drives platelet-associated fibrinogen accumulation and platelet-mediated coagulopathy.

BACKGROUND: Severe fever with thrombocytopenia syndrome (SFTS) is an emerging viral hemorrhagic fever characterized by high mortality, thrombocytopenia, and coagulation abnormalities. Unlike other inflammatory conditions where plasma fibrinogen rises, patients with SFTS exhibit normal fibrinogen levels despite evidence of consumptive coagulopathy and hepatocyte injury. OBJECTIVES: The aim of this study was to investigate whether platelets contribute to fibrinogen dysregulation and coagulopathy in SFTS. METHODS: We enrolled 102 patients with SFTS stratified by clinical symptoms (mild vs sever

[IGFBP-4 improves synovitis by reducing M1 polarization of macrophages via the Wnt/β-catenin pathway].

Objective To investigate the mechanism by which insulin-like growth factor binding protein 4 (IGFBP-4) ameliorates synovial inflammation in osteoarthritis (OA) by inhibiting M1-type polarization of synovial macrophages via the Wnt/β-catenin signaling pathway. Methods The OA model was established in male C57BL/6 mice by intra-articular injection of sodium iodoacetate. Adenoviral vectors carrying IGFBP-4 overexpression or knockdown sequences were injected into the knee joint cavity for in vivo intervention. Micro-computed tomography (Micro-CT) was used to observe the subchondral bone changes in

An inducible co-stimulator ligand (ICOSL)-based peptide therapy for proteinuric kidney diseases targeting αvβ3 integrin.

Dysregulated αvβ3 integrin activation in podocytes contributes to proteinuric kidney disease by disrupting podocyte architecture and the glomerular filtration barrier. However, no treatments currently exist that therapeutically target this integrin. We previously identified inducible co-stimulator ligand (ICOSL) as an endogenous antagonist of αvβ3 integrin that protects kidney function through its arginine-glycine-aspartic acid (RGD) motif, independent of its canonical role in immunity. Leveraging the therapeutic advantages of small peptides and the renoprotective function of ICOSL, we used a

Long-term immunoprophylaxis by TLR9 ligand mediates macrophage-ILC cross-talk against respiratory viral infection.

Rapid development of host immunity is critical during the emergence of viral outbreaks when vaccines are not yet available. Here, we demonstrate that CpG oligodeoxynucleotide-schizophyllan complex (K3-SPG) induces antigen-independent, long-term protection against lethal respiratory virus infection. A single intranasal administration successfully enhanced host resistance to infection for as long as 100 days posttreatment. Mechanistically, we observed a lung-restricted biphasic innate activation of macrophages and innate lymphoid cells. Early protection was mediated by CD11b+CD11c+ macrophages (

Secondary pulmonary alveolar proteinosis mimicking progression of interstitial lung disease during treatment for adult-onset still's disease: An autopsy case report.

RATIONALE: Secondary pulmonary alveolar proteinosis (sPAP) is a rare disorder caused by impaired surfactant clearance due to alveolar macrophage dysfunction and is often associated with hematologic disorders or immunosuppressive therapy. In patients with connective tissue disease-associated interstitial lung disease (CTD-ILD), worsening respiratory findings are frequently interpreted as progression of the underlying ILD, which may delay recognition of sPAP, particularly when anti-granulocyte-macrophage colony-stimulating factor (GM-CSF) antibodies are negative. In the present case, preexisting

Psilocybin's effect on human brain synaptic plasticity.

Psychedelics such as psilocybin have been linked to enhanced neuroplasticity and symptom relief in affective disorders, but the neurobiological mechanisms and impact of environmental context remain unclear. Here, we tested whether a single dose of psilocybin (0.3 mg/kg) alters synaptic density in healthy individuals and whether setting-dependent subjective experience shapes this effect. Fifteen healthy participants had a psilocybin-induced psychedelic experience either inside an MRI scanner or in a therapeutic-like room. We assessed synaptic density changes by measuring the Synaptic Vesicle gl

Adeno-Associated Virus Vector-Mediated Expression of Neuroligin 2 in the Mouse Hippocampus Attenuates the Progression of Pentylenetetrazol-Induced Seizures.

Gene therapy can target specific brain regions and may be an effective therapeutic tool for drug-resistant epilepsy. Neuroligin 2 (NL2) is a postsynaptic cell-adhesion protein involved in the formation of inhibitory synapses. Since epileptic seizures are attributed to excessive brain neuronal activity, we hypothesized that NL2-mediated inhibitory signaling suppresses seizures. Male C57BL/6J mice were bilaterally injected into the hippocampus (HPC) with an adeno-associated virus (AAV) vector expressing NL2 under the control of synapsin I promoter (AAV-NL2). Mice treated with AAV-NL2 were intrap

Spatio-temporal expression of E-cadherin mediates trophoblast lineage transitions during human placentation.

Human placentation relies on the precise differentiation of embryo-derived trophoblasts into specialized lineages, a process strictly regulated by the spatio-temporal expression of key genes. Herein, we report that the expression of the transmembrane glycoprotein, known as E-cadherin, fluctuates dynamically across distinct trophoblast subpopulations rather than acting as a static structural tether. Along the villous pathway, E-cadherin expression initially increases to facilitate the aggregation of cytotrophoblast cells (CTs) and is subsequently downregulated via transcriptional repression, en

Immunohistochemistry for c-MET, HLA class II, and CD68 does not show prognostic value in early-stage classic Hodgkin lymphoma patients in the EORTC-GELA H9 intergroup randomised trial.

BACKGROUND: In classic Hodgkin lymphoma (cHL), identifying high-risk patients is crucial for tailored treatment. Clinical risk factors are useful, but there is room for improvement. Although many diagnostic tissue-based biomarkers have been described to correlate with prognosis, none have achieved the accuracy to be included into daily clinical practice. METHODS: The most promising tissue biomarkers, c-MET, HLA class II, and CD68, were tested in early-stage cHL for prognostic validation, individually and in combination with each other and established clinical risk factors. All biomarkers were

Immunoregulatory artificial lymph nodes restore antigen-specific tolerance in advanced multiple sclerosis.

Tolerogenic dendritic cell (tolDC) vaccines are a promising antigen-specific therapy for multiple sclerosis (MS), yet their efficacy is limited by poor homing to secondary lymphoid organs and rapid loss of the tolerogenic phenotype in native inflamed lymph nodes. Here, we developed a 3D-printed tolerogenic artificial lymph node (tol-aLN) that creates a protected immune niche for durable tolerance induction. tolDCs were loaded onto a porous gelatin methacrylate scaffold engineered for sustained C-C motif chemokine ligand 21 (CCL21) release, recruiting naïve CD4+ T cells and shielding tolDCs fro

Continuous Glucose Monitoring and Long-Term Glycemic and Kidney Outcomes.

IMPORTANCE: Real-time continuous glucose monitoring (rtCGM) improves short-term glycemic control in diabetes. However, scant evidence exists regarding rtCGM's longer-term benefits for glycemia or kidney health. OBJECTIVE: To evaluate whether rtCGM initiation and sustained use is associated with improved glycemic control and kidney markers in high-risk adults with type 2 diabetes (T2D). DESIGN, SETTING, AND PARTICIPANTS: This comparative effectiveness research study compared glycemic control and kidney markers among high-risk adults with T2D initiating and sustaining use of rtCGM (rtCGM arm)

Trans- and cis-interaction dynamics of cadherin complexes on membranes visualized by high-speed atomic force microscopy.

Classical cadherin-mediated cell-cell adhesion is important for maintaining the animal body and defects in adhesion cause severe diseases. The combination of trans- (between cells) and cis- (on the same membrane) interactions of the extracellular domains of cadherins is necessary for cadherin clustering in cell-cell junctions, which is essential for robust tissue formation. However, the solution structure and interaction dynamics of trans/cis-binding cadherin complexes have not been observed because of technical limitations; therefore, the cadherin clustering mechanism is not well understood.

Mesenchymal stem cell-derived extracellular vesicles modulate Th1- and STAT1-Associated inflammatory responses in peripheral blood mononuclear cells from patients with osteoarthritis.

BACKGROUND: Osteoarthritis (OA) is increasingly recognized as a chronic inflammatory disease, with dysregulated immune responses contributing to cartilage degeneration and disease progression. Immune dysregulation, including alterations in Th1-associated responses, may contribute to the chronic inflammatory environment observed in osteoarthritis. MSC-derived exosomes have been highlighted as potential cell-free therapeutic agents due to their potent immunomodulatory properties; however, their effects on Th1-associated immune responses in OA remain poorly understood. METHODS: Extracellular ves

Myasthenia gravis: from recognition of heterogeneity to a paradigm shift toward precision therapy.

Myasthenia gravis (MG) is a heterogeneous neuroimmune disorder in which pathogenic autoantibodies impair neuromuscular junction transmission and produce fluctuating skeletal muscle weakness. Although conventional therapy with acetylcholinesterase inhibitors, corticosteroids, and broad immunosuppressants remains effective for many patients, its limitations are increasingly evident, including delayed onset, cumulative toxicity, and inadequate control in refractory disease. These challenges have accelerated the transition from non-selective immunosuppression to biologic and other targeted therapi

Clusterin is enriched in plaque-associated astrocytes and localizes to synapses in Alzheimer's disease.

INTRODUCTION: The clusterin gene (CLU) is a top genome-wide association studies (GWAS) risk locus for Alzheimer's disease (AD) and elevated clusterin levels have been reported in AD biofluids and brain tissue. Despite growing interest in clusterin as a biomarker and therapeutic target, its cellular and subcellular localization in the human brain remains incompletely understood, in part due to a lack of well-characterized reagents. METHODS: We generated and validated monoclonal antibodies (mAbs) that distinguish secreted and intracellular clusterin, and applied these tools to human biofluids,
