The Prothrombotic Phenotype of Thirdhand Electronic Cigarette Exposure is Sex Independent and Involves Systemic Mediated Effects on Platelet Function: Evidence from a Mouse Model.

Electronic cigarettes (e-cigs) are major contributors to inflammatory-mediated responses, which are implicated in a vast array of pathophysiological conditions, including cardiovascular disease (CVD). More recently e-cigs have been recognized as a source of thirdhand exposure (THEC); the process by which expelled toxins settle on materials (i.e., carpets, curtains, clothes etc.) and undergo chemical reactions, rendering them more harmful overtime. Herein, mice were exposed to THEC for four months, and platelet reactivity, systemic mediated effects on platelet function, and cytokine expression

Multimodal activity-affinity assay of ADAM-10 extracellular vesicles in untreated plasma reveals metastatic stage of colorectal cancer.

Metalloproteinases (MPs) such as a-disintegrin and metalloproteinase-10 (ADAM-10) are key drivers of extracellular matrix remodeling during tumor progression, yet MP-based liquid biopsy tests have not reached clinical utility. Here, we show that active ADAM-10 is selectively enriched on the surface of circulating extracellular vesicles (EVs) in the plasma of colorectal cancer patients. Our findings further suggest ADAM-10+ EVs are locally enriched in dense pre-metastatic tumor extracellular matrices and subsequently accumulate in blood post-metastasis. To capture these unique signatures of dis

Bacterial constipation: Mucin-degrading intestinal commensal bacteria cause constipation.

The contribution of gut microbes to constipation remains mechanistically underexplored, despite constipation being one of the most prevalent gastrointestinal disorders. Here, we identify cooperative induction of constipation by two mucin-degrading gut commensals: Akkermansia muciniphila and Bacteroides thetaiotaomicron. In constipated patients with Parkinson's disease (PD) and chronic idiopathic constipation (CIC), we observed that A. muciniphila and B. thetaiotaomicron were increased. Gnotobiotic mice colonized with either bacterium exhibited no constipation, whereas mice co-colonized with bo

Interactions between small leucine-rich proteoglycans and proteases: implications for tissue homeostasis and pathology.

Small leucine-rich proteoglycans (SLRPs) are essential regulators of extracellular matrix (ECM) organization and cell signaling. Beyond their structural roles, SLRPs modulate protease activity or serve as their substrates, thereby influencing key physiological processes such as collagen fibrillogenesis, wound healing, skeletal development, and immune surveillance. On the other hand, the dysregulation of SLRP-protease interactions, in pathologies such as fibrosis, chronic inflammation, and cancer, results in aberrant ECM remodeling, immune activation, and altered cell behavior. Here, we review

Independent validation of circulating microRNAs as biomarkers in a case-control study of adolescents with type 1 diabetes for more than 8 years.

MicroRNAs (miRNAs) are epigenetic regulators of gene activity. Analysis of circulating miRNAs enables minimal-invasive studies of disease mechanisms and identification of novel disease biomarkers. The aim of this case-control validation study was to investigate previously identified circulating miRNAs in Swedish adolescents with long-duration (8.0-16.5 years) type 1 diabetes (T1D), and healthy matched controls to confirm their utility as biomarker candidates to diagnose and monitor the progression of T1D. Quantitative PCR analysis of 23 previously reported miRNAs was performed in 24 T1D and 24

Metabolic Inflammation as a Common Thread in Cardio-Endocrine Diseases: Toward a Unified Therapeutic Framework.

Metabolic inflammation, or metaflammation, has emerged as a unifying mechanism linking cardiovascular and endocrine disorders. This narrative review aimed to synthesize mechanistic, clinical, and therapeutic evidence on how inflammation bridges these domains and to explore prospects for a unified therapeutic framework. We systematically searched PubMed, Scopus, and Web of Science for English-language articles published between 2010 and 2024, yielding 256 relevant articles, of which 115 were included after detailed screening. Studies addressing adipose tissue inflammation, gut microbiota dysbio

Integrative transcriptomic and machine learning analysis identifies CDH17 and HOXC13 as robust candidate prognostic biomarkers in uveal melanoma.

BACKGROUND: Uveal melanoma (UM), the most common form of ocular melanoma, represents poor prognosis, with approximately 50% of patients developing metastatic disease that significantly reduces survival rates. There is an crtitical need for accurate prognostic biomarkers to better stratify patients by risk and guide personalized treatment strategies. METHODS: We employed an integrative, multi-step strategy to analyze UVM transcriptomic data, combining Cox proportional hazards survival analysis, stage-dependent differential expression, and a multi-layer perceptron (MLP) model to identify candid

An Integrative Proteotranscriptomics Approach Reveals New ADAM9 Substrates and Downstream Pathways.

A Disintegrin And Metalloproteinase 9 (ADAM9) is a cell-surface protease that can shed the ectodomain of membrane protein substrates. Dysregulated ADAM9 activity has been implicated in several diseases, such as solid tumors, autoimmunity, inflammatory diseases, and coronavirus disease 2019. Despite its importance, the substrates and targets of ADAM9 in normal and pathological processes are poorly understood. Here, we developed an integrative proteotranscriptomics approach to systematically identify the transcriptional and post-transcriptional targets of ADAM9 in HCT116 cells, which have a stab

Length and density of α2-3 sialyllactose-containing chains on glycopolymers determine receptor binding of avian influenza viruses.

Influenza A viruses use host sialoglycans for attachment via hemagglutinin (HA) and hydrolyze them upon budding via neuraminidase (NA). The HAs of some human isolates prefer extended and branched glycans for binding; however, the preference of avian influenza viruses (AIVs) for these glycans is poorly understood. This study addressed the glycan-binding preferences of HA and AIV particles by using a series of sialoglycopolymers with various glycan representations and densities. Glycan extension was mimicked by linking the terminal sialyllactose with subsequent polyethylene glycol chains, and gl

Ligand-complex-based quantification of β2-integrin-mediated affinity and avidity of murine T cells.

BACKGROUND: Integrins facilitate binding to the extracellular matrix and other cells. Their subunit β2 is exclusively expressed by leukocytes, binds to the intercellular cell adhesion molecule 1 (ICAM-1), and is pivotal for their recruitment to sites of inflammation such as the atherosclerotic plaque. METHODS: To investigate β2-integrin-mediated adhesiveness, a well-established assay for human whole blood was adapted for the analysis of murine T cell subsets. Changes in avidity and affinity were assessed by incubation of murine complexes ICAM-1 in murine whole blood and consecutive stimulatio

TREM1-PET imaging maps whole-body innate immune responses in a mouse model of metastatic melanoma.

Poor treatment response in brain metastases is largely attributed to anti-tumor T-cell suppression through the modulation of tumor-associated myeloid cells (TAMCs), resulting in immune evasion. Triggering receptor expressed on myeloid cells-1 (TREM1) is a membrane receptor highly expressed on TAMCs that is associated with poor clinical outcomes and of interest as a potential imaging biomarker of myeloid cell function, prognosis, and treatment response. Here we evaluate TREM1-targeted positron emission tomography (PET) tracer, [64Cu]TREM1-mAb, for TAMC detection in a murine model of intracrania

Abietane-Type Diterpenoids from the Resin of Pinus yunnanensis and Their Potential Anti-Renal Fibrosis Activities.

Chronic kidney disease (CKD) has emerged as a pressing global public health concern, making the identification of renal fibrosis inhibitors a key research focus. In this study, seven undescribed abietane-type diterpenoids, pinusyunins A-G (1, 2, 4, and 7-10) and three known analogues (3, 5, and 6), were isolated from Pinus yunnanensis resin, which were identified by spectroscopic analyses and quantum computational chemistry methods. Biological evaluation showed that all the isolates exhibited inhibitory activity against the expression of collagen I, fibronectin, and α-SMA in transforming growt

Gut integrity, systemic inflammation and exacerbations of COPD: the role of I-FABP.

BACKGROUND: High plasma intestinal-fatty acid binding protein (I-FABP) levels reflect gut epithelial integrity loss. Its interplay with systemic inflammation remains poorly understood in COPD and exacerbations of this disease (ECOPD). Therefore, the aim of this study was to investigate the association between plasma I-FABP levels, inflammatory markers and severity of COPD (including severity and frequency of ECOPD), and to evaluate I-FABP as potential clinical marker. METHODS: Data from the 2-year observational ICE-AGE study were analysed post hoc. Plasma I-FABP levels, and inflammatory param

Islet-derived T cells from both mice and humans recognize conserved insulin A-chain peptides presented by HLA-C*03:04.

Type 1 diabetes (T1D) is an autoimmune disease in which T cells mediate the elimination of the insulin-producing beta cells in the pancreatic islets, resulting in the need for exogenous insulin. Studies of T1D in both patients and the nonobese diabetic (NOD) mouse model of the disease illustrate that beta cell-specific CD8+ T cells are central contributors to the beta cell destruction characterizing the disease. Compared to HLA-A and HLA-B, relatively little is known about the participation of HLA-C-restricted T cells in T1D. To tackle this question, we developed and characterized an NOD-based

Structure-activity relationships of steroid and sterol neuromodulators on inflammatory markers in a murine microglial cell line.

Neuroactive steroids are allosteric modulators of GABAA receptors and are implicated in the etiology and treatment of neuropsychiatric disorders. Existing treatments are helpful but have drawbacks. Neuroactive steroids recently gained attention as rapidly acting antidepressants in postpartum depression and other indications. Unlike other GABAA receptor modulators, neurosteroids may possess anti-inflammatory actions, potentially contributing to therapeutic benefit. Here we seek to understand neuroactive steroid structure-activity relationships relevant to these anti-inflammatory effects. We use

Sotagliflozin, suPAR, and Cardiovascular Outcomes in Patients With Diabetes and Heart Failure: A SOLOIST-WHF Ancillary Study.

OBJECTIVE: Sotagliflozin, a dual sodium-glucose cotransporter 1 and 2 (SGLT1/2) inhibitor, improves kidney and heart failure (HF) outcomes through incompletely understood mechanisms. Soluble urokinase plasminogen activator receptor (suPAR), an immune-derived glycoprotein, is implicated in kidney and cardiovascular disease pathogenesis. We investigated whether sotagliflozin reduces suPAR levels; whether baseline suPAR predicts cardiovascular outcomes; and whether baseline suPAR modifies sotagliflozin's treatment effect. RESEARCH DESIGN AND METHODS: We measured suPAR levels at baseline and at 1

Plasma growth differentiation factor-15 is associated with cardiovascular events in patients hospitalized for acute exacerbation of COPD.

Patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD) are at increased risk of major adverse cardiovascular events (MACE) and death. Growth differentiation factor-15 (GDF-15), a marker of cellular stress and inflammation, and Syndecan-1, a marker of endothelial dysfunction, have been suggested as prognostic biomarkers in plasma for MACE. We aimed to assess their association with a combined outcome of MACE or all-cause mortality over a 5-year period. This sub-study was embedded within the randomized controlled trial CORTICOsteroid reduction in COPD (CORTICO-COP), wh

Anaplasma phagocytophilum invasin AipB interacts with the β2 integrin β-subunit CD18 to orchestrate infection.

Anaplasma phagocytophilum is an obligate intracellular bacterium that invades neutrophils to cause the potentially severe infection, human granulocytic anaplasmosis. The A. phagocytophilum invasin-host cell receptor interactions that facilitate bacterial uptake and hence disease are incompletely defined. Here, we show that EPHNCH_1611, which we refer to as A. phagocytophilum invasion protein B (AipB), mediates bacterial entry but not adhesion. The AipB host cell-interacting domain was narrowed down to amino acids 109 to 129 and is conserved among Anaplasma and Ehrlichia species. AipB109-123 an
