Novel Roles for the Ectoenzyme CD38 in the Maintenance of Transcriptional and Metabolic Homeostasis in Astrocytes.

CD38 is an ectoenzyme that converts NAD+ to NAM to help maintain bioenergetic homeostasis. CD38 dysregulation and gene variation is reported in neurodegenerative conditions such as Parkinson's disease (PD) and Alzheimer's disease (AD), highlighting the need to better understand CD38 biology within the brain. Here, we demonstrate enrichment of Cd38 in midbrain astrocytes and describe how CD38 deficiency influences brain metabolism, astrocytic gene expression, and bioenergetics. We demonstrate increased NAD content, decreased NAM content, and increased NAD/NAM in the midbrain and striatum of CD3

ADAM10 and its role in Alzheimer disease.

INTRODUCTION: The amyloidogenic pathway of amyloid precursor protein (APP) processing is well known in the pathogenesis and therapeutics of Alzheimer disease (AD), whereas the non-amyloidogenic pathway has been less studied. ADAM10 is the main α-secretase responsible for this pathway in the human brain. CONTENT: ADAM10 belongs to a family of transmembrane proteins with catalytic activity. It acts as an α-secretase on APP and many other substrates, some of which are particularly relevant in the central nervous system. The ADAM10 gene has been identified in genome-wide association studies of pa

Clinical Significance of Mucin Signatures in Inflammatory Bowel Diseases: A Systematic Review of Their Expression Patterns, Polymorphisms, and Post-translational Modifications.

BACKGROUND: The intestinal mucosal barrier plays an important role in the pathophysiology of inflammatory bowel disease (IBD), with mucins being key components of this barrier. Classified as either transmembrane or secreted, these highly glycosylated proteins protect the mucosal barrier while also influencing barrier integrity. Given their indispensable role in maintaining the intestinal mucosal barrier, mucins are compelling candidates for the evaluation of barrier dysfunction. Numerous studies have investigated mucins in the context of IBD, but a clear consensus regarding their expression, p

Methods and Guidelines for Analyzing the Conformational Dynamics of Proteins in the Antigen Presentation Pathway.

Endoplasmic reticulum aminopeptidase 1 (ERAP1) digests antigenic precursors up to the optimum length for binding to the major histocompatibility complex (MHC) class I. This zinc-dependent aminopeptidase has been found in a catalytically active closed conformation and in open conformations that expose its substrate-binding cavity to the bulk solvent. Therefore, its activity has been linked to catalytic cycles between conformational states that accommodate substrate binding, peptide hydrolysis and product release. During the last decade, X-ray crystallography in conjunction with biophysical and

Feedback regulation between histone H3 lysine 18 lactylation and TROP2-mediated glycolysis drives metastatic progression of colorectal cancer.

BACKGROUND: TROP2, a critical cell surface oncogenic signal transducer, is increasingly linked to refractory metastatic colorectal cancer (CRC) and other solid tumours. Robust lactate accumulation within metastatic niches correlates with pathological metastatic progression. Anti-TROP2 antibody-drug conjugates (ADCs) are clinically available but show limited efficacy in advanced metastatic CRC. Elucidating how TROP2 signalling orchestrates molecular and cellular programs enabling CRC metastatic progression would help improve metastasis therapies. METHODS: Tissue microarray, immunohistochemistr

Stimulant use disorder indicative of increased serum soluble intercellular adhesion molecule-1 concentrations with altered brain reward and interoceptive processing.

Stimulant use disorders (STIM): (1) elicit compulsive behaviors that can lead to altered brain structure and function; and (2) trigger inflammatory responses by inducing the release of immune molecules, suggesting potential adverse effects. Our previous findings show that amphetamine use disorder is associated with altered neural processing during reward, interoception, and inhibitory control tasks. To extend these findings, we investigated links between neural processing and inflammation that may contribute to STIM. Participants from the first half of the Tulsa-1000 (T1000) study (n = 500) me

Imiquimod-induced colitis: A novel ROS/ERK-driven model of intestinal inflammation and barrier dysfunction.

Psoriasis and inflammatory bowel disease (IBD) are chronic immune-mediated disorders affecting the skin and gut, respectively, and share underlying mechanisms involving immune dysregulation and microbiota alterations. Imiquimod (IMQ), a compound commonly used to induce psoriasis-like skin inflammation in mice and exacerbate dextran sulfate sodium (DSS)-induced colitis. Accumulated research findings indicate that IMQ-induced reactive oxygen species (ROS) play a crucial role in biological functions and inflammation. In this study, we investigate the role of ROS in the pathogenesis of intestinal

Proteomic signatures in triple-negative breast cancer.

An aggressive and heterogeneous malignancy, referred to as triple-negative breast cancer, is characterised by the absence of estrogen receptor, progesterone receptor, and human epidermal growth factor receptor 2. Treatment options remain limited, relying primarily on chemotherapy due to the lack of well-defined therapeutic targets, which is linked with poor prognosis and high recurrence rates. Proteomics and other high-throughput technologies have significantly advanced TNBC research by enabling the identification of protein-based biomarkers with potential applications in diagnosis, prognosis,

Identification of CADM1 as an Immunotherapeutic Target and Evaluation of a Novel CADM1-Targeting Antibody-Drug Conjugate in Preclinical Osteosarcoma Models.

Due to the paucity of validated cell-surface osteosarcoma-specific targets, patients with this condition have long been excluded from the benefits of antibody-drug conjugate (ADC) therapy observed in patients with several solid and hematologic malignancies. Our comprehensive surfaceome profiling approach previously identified osteosarcoma-specific cell-surface antigens that are highly expressed in osteosarcomas but minimally expressed in normal tissues. As a result, one such antigen, CADM1, was selected for the generation of an ADC. We tested a CADM1-targeting ADC with a tesirine payload (SG32

Normalization strategies for protein quantification in plasma astrocyte-derived extracellular vesicles: a clinical applicability study.

BACKGROUND: Astrocyte-derived extracellular vesicles (ADEVs) have emerged as a novel research tool in the field of central nervous system disorders. However, significant differences in yield and purity exist among extracellular vesicles (EVs) isolated by different methods, leading to considerable heterogeneity in clinical study outcomes. Therefore, establishing appropriate normalization strategies to enhance comparability across results is a key prerequisite for their clinical translation. METHODS: This study included 15 patients with major depressive disorder (MDD) and 15 healthy controls (HC

Mechanistic diversity of Bacteroides fragilis toxins and neutralization with single domain antibody.

Enterotoxigenic Bacteroides fragilis (ETBF) promotes colonic inflammation by secreting metalloenzyme toxins (BFTs). Understanding BFT mechanisms and developing neutralization strategies is critical. Here, we have solved the structures of BFT-1 and BFT-2, revealing that residue 357 in the active site of the catalytic domain explains the diversity of function observed in BFT subtypes. We demonstrate that BFTs can directly cleave human epithelial-cadherin at extracellular domain 4, with BFT-2 possessing the highest activity. Using an alpaca antibody library, we identified a single-domain antibody

Immunopathogenic role of the HLA-B∗51:01-specific immunopeptidome in Behçet's disease.

OBJECTIVE: The immunopeptidome, the collection of peptides bound to human leukocyte antigens (HLAs), plays a key role in initiating immune responses. HLA-B∗51:01 is an allele associated with Behçet's disease (BD). However, the role of the immunopeptidome bound to HLA-B∗51:01 in the pathogenesis of BD remains unclear. METHODS: We profiled the HLA-bound immunopeptidome using plasma samples from HLA-B∗51:01-positive BD and healthy controls (HCs). HLA-class I molecules were immunoprecipitated, and liquid chromatography-tandem mass spectrometry was performed to compare HLA-bound peptides between H
