Combining Genomics With Lipid and Inflammatory Biomarkers to Predict Coronary Artery Disease Risk: UK Biobank Study.

BACKGROUND: Coronary artery disease (CAD) polygenic risk score (PRS), low-density-lipoprotein cholesterol (LDL-C), lipoprotein(a) (Lp(a)), and high-sensitivity C-reactive protein (hsCRP) are biomarkers that predict CAD. It is unclear whether integrating genomics with lipid and inflammatory biomarkers could complement traditional risk scores in identifying people at risk of CAD. OBJECTIVES: This study assesses the predictive value of CAD PRS, LDL-C, Lp(a), and hsCRP for incident CAD across different age and sex groups. METHODS: Participants (n = 215,695) from the UK Biobank aged 40 to 69 year

Long-term prognostic value of apolipoprotein B levels in patients with very premature (≤40 years) acute coronary syndrome.

BACKGROUND: Apolipoprotein B (apoB) is a recognized risk factor for acute coronary syndrome (ACS); however, its prognostic value in secondary prevention and superiority over other lipid biomarkers, especially in younger populations, remains uncertain. OBJECTIVE: To investigate whether elevated baseline apoB predicts recurrent cardiovascular events in patients who experienced an ACS at ≤40 years of age and compare its incremental predictive value with that of other lipid biomarkers. METHODS: We recruited 405 consecutive patients who survived an ACS at ≤40 years of age. Clinical endpoints incl

A cohort study in Southern Xinjiang, China, 2018-2023 on the association of metabolic syndrome components and their interactions with cardiovascular disease risk.

The association between metabolic syndrome (MetS) and its components with cardiovascular disease (CVD) incidence represents a key global public health focus. However, given the unique dietary patterns of the population in southern Xinjiang, China, the differential effects of various Mets components on CVD onset and the interactions among these components remain poorly elucidated. This retrospective cohort study was conducted from 2018 to 2023, enrolling 144,966 participants from a county in southern Xinjiang with a median follow-up duration of 5.0 years. Cox proportional hazards regression and

Premature Coronary Artery Disease and Familial Dyslipidemia in Patients Presenting With Acute Coronary Syndrome: A Tertiary Cardiac Center Registry.

BACKGROUND: Premature coronary artery disease (CAD) in patients presenting with acute coronary syndrome (ACS) is an increasing clinical concern driven by genetic predisposition and lifestyle factors. This study compared clinical and laboratory characteristics of premature versus non-premature CAD and identified predictors of early-onset disease. METHODS: This was a cross-sectional observational study of 2,000 patients admitted with confirmed ACS. Premature CAD was defined as males <55 years and females <65 years. Clinical, laboratory, and cardiological variables were compared between groups.

Cascade screening for elevated Lp(a) in relatives of children who visited the pediatric lipid clinic: yield of daily clinical practice.

AIMS: Cascade screening can identify individuals with elevated lipoprotein(a) [Lp(a)], a causal risk factor for atherosclerotic cardiovascular disease (ASCVD). The aim of this study is to explore the effectiveness of cascade screening with asymptomatic children as index cases to identify family members with elevated Lp(a). METHODS: In this retrospective study we used our database consisting of all children referred for a tentative diagnosis of hereditary dyslipidemia to the Amsterdam University Medical Centers pediatric lipid clinic (1989-2023). Elevated Lp(a) was defined as ≥30 mg/dL or ≥75

Predictive value of high-density lipoprotein cholesterol and the cardio-ankle vascular index on cardiovascular outcomes in subjects with cardiovascular risks: the COUPLING Study.

The impacts of lipid abnormalities on the development of cardiovascular events (CVEs) in relation to the cardio-ankle vascular index (CAVI) have not been fully elucidated. We sought to determine whether the association between high-density lipoprotein cholesterol (HDL-C) levels and cardiovascular outcomes varies depending on the CAVI status in patients at risk for cardiovascular disease (CVD). Of 5109 patients enrolled in the multicenter, prospective COUPLING study (CardiOvascUlar Prognostic coupLING Study in Japan), the present analyses included 4,569 patients (51% male; mean age 68.5  ±  11.

The low-density lipoprotein receptor LDLR mediates cellular entry of nonenveloped hepatitis A virus.

Hepatitis A virus (HAV) is an unusual picornavirus with two types of extracellular virions: nonenveloped particles (nHAV) shed in feces and quasi-enveloped particles (eHAV) circulating in blood. Both enter cells by clathrin-dependent endocytic pathways merging in late endolysosomes with capsid binding to ganglioside receptors. Phosphatidylserine receptors facilitate eHAV endocytosis, but no protein receptor has been identified for nHAV. Here, we show low-density lipoprotein receptor (LDLR) is such a receptor. LDLR knockout did not alter viral attachment to cells, but restricted cellular uptake

Evinacumab with or without lipoprotein apheresis in homozygous familial hypercholesterolaemia.

AIMS: Homozygous familial hypercholesterolaemia (HoFH) is characterized by markedly elevated low-density lipoprotein cholesterol (LDL-C). Most individuals with HoFH do not reach LDL-C targets with standard lipid-lowering therapies (LLTs). However, low density lipoprotein receptor (LDLR)-independent LLTs have demonstrated effectiveness in these individuals. METHODS: This post hoc subanalysis examined concomitant use of two LDLR-independent treatments, lipoprotein apheresis (LA) and evinacumab (an angiopoietin-like 3 inhibitor), in participants with HoFH from three clinical studies of evinacuma

Proteomics and metabolomics biomarkers for predicting the onset and progression of diabetic complications: A systematic review and bioinformatics integration.

Diabetic complications are major drivers of morbidity and mortality, leading to progressive cardiovascular, renal, retinal, and neurological damage. Early intervention is hindered by the lack of reliable biomarkers for prediction and disease staging. Advances in proteomics and metabolomics provide powerful tools to elucidate molecular mechanisms and identify clinically actionable signatures. This systematic review included 67 cohort and 41 cross-sectional studies published between 2012 and August 2025, comprising 73,580 participants. Most studies involved type 2 diabetes (T2D) populations (n =

Evaluating the Whole Patient: Lessons from the Pre-CKM Era Toward Integrated Cardio-Kidney-Liver-Metabolic Care.

Before the American Heart Association introduced the cardiovascular-kidney-metabolic (CKM) syndrome concept in 2023, clinical care was largely organ-specific. This retrospective study analyzed diagnostic patterns and gaps in 406 patients with hypertension referred to and evaluated at the University Hospital Basel Hypertension Centre in 2017, 2019, or 2022 to identify blind spots in the assessment of cardio-kidney-liver-metabolic health. Electronic health records were used to assess CKM-relevant diagnostics, including lipid profiles, N-terminal pro-B-type natriuretic peptide (NT-proBNP), echoca

Lipoprotein(a) and acute coronary syndrome: Associations with sex, age and traditional cardiovascular risk factors.

INTRODUCTION AND OBJECTIVES: Lipoprotein(a) [Lp(a)] is recognized as an independent risk factor for coronary disease owing to its atherogenic, proinflammatory, and prothrombotic properties. Current guidelines recommend a single measurement in adults to refine cardiovascular (CV) risk assessment. We aimed to characterize Lp(a) levels in patients with acute coronary syndrome (ACS) and explore associations with sex, age, comorbidities, and traditional cardiovascular risk factors. METHODS: We conducted a cross-sectional study of patients with ACS admitted to our center between January 2022 and De

Tirzepatide mitigates atherosclerosis progression and modulates oxLDL-mediated proatherogenic effects in macrophages: evidence for M1/M2 homeostasis restoration.

Tirzepatide (TZP), a novel dual agonist of glucagon-like peptide (GLP)-1/glucose-dependent insulinotropic polypeptide (GIP) receptors (GLP-1R/GIPR), has been shown to reduce cardiovascular (CV) risk in patients with diabetes or obesity. This study investigated anti-atherosclerotic effects of TZP and the underlying mechanisms using apo E-/- mice and cultured macrophages. In the present study, apo E-/- mice were fed a high fat/high cholesterol (HF) diet with or without TZP treatment for 12 weeks. Atherosclerotic lesions, metabolic parameters, and M1/M2 macrophage homeostasis were assessed. In vi

Prognostic value of the atherogenic index of plasma in patients with multivessel coronary disease undergoing CABG: a retrospective cohort study.

BACKGROUND: The Atherogenic Index of Plasma (AIP), a composite lipid index based on triglycerides (TG) and HDL-C, has been linked to cardiovascular risk. However, the prognostic relevance of AIP after coronary artery bypass grafting (CABG) in patients with multivessel coronary disease (MVD) is not well established. Therefore, we examined whether AIP predicts long-term outcomes in our cohort. METHODS: Consecutive patients with MVD undergoing CABG (2015–2020) were included. The baseline and 1-year AIP were used to define tertiles. Major adverse cardiovascular and cerebrovascular events (MACCEs)

Astrocytic APOE3-Christchurch expression ameliorates brain amyloid-β pathology in 5xFAD mice.

The rare APOE3-Christchurch (APOE3Ch) variant is linked to resistance against PSEN1 p.E280A-driven autosomal dominant Alzheimer's disease (AD). Recent studies in AD mouse models have demonstrated an effect of APOE3Ch in reducing tau pathology and tau propagation, yet its effects on amyloid pathology and related toxicity are not fully understood. While prior studies have reported reduced amyloid pathology with APOE3Ch, we extended this knowledge by investigating how astrocyte-specific expression of APOE3Ch impacts amyloid pathology and related responses in 5xFAD mice, an amyloid mouse model. Association between metabolic signatures of predicted VAT mass and risk of MASLD and other chronic liver diseases.

BACKGROUND AND AIMS: Visceral adipose tissue (VAT) plays a key role in metabolic dysfunction, and it is increasingly recognised as a contributor to metabolic dysfunction-associated steatotic liver disease (MASLD) and other chronic liver conditions. However, the systemic metabolic pathways linking VAT to liver disease remain unclear. This study aimed to identify metabolic signatures associated with VAT and examine their potential role as mediators in the relationship between VAT accumulation and the risks of MASLD, cirrhosis and hepatoma. METHODS: This prospective study included 269,018 UK Bio

Lipoprotein(a) levels in Finnish adults: distribution and associations with other cardiovascular risk factors and their awareness and control.

BACKGROUND: Elevated lipoprotein(a) [Lp(a)] is an important genetic risk factor for cardiovascular diseases (CVDs). Because Lp(a)-lowering therapies are limited, prevention focuses on identifying individuals with elevated Lp(a) and optimizing other modifiable risk factors. We aimed to assess the distribution of Lp(a) levels in Finnish adults and examine its association with other CVD risk factors, as well as the awareness, treatment, and control of dyslipidemia. METHODS: Data were derived from the Healthy Finland health examination survey conducted in 2023, comprising a nationally representat

The functional landscape of coding variation in the familial hypercholesterolemia gene LDLR.

Variants in the familial hypercholesterolemia gene LDLR-the most important genetic driver of cardiovascular disease-can raise circulating low-density lipoprotein (LDL) cholesterol concentrations and increase the risk of premature atherosclerosis. Definitive classifications are lacking for nearly half of clinically encountered LDLR missense variants, limiting interventions that reduce disease burden. We tested the impact of ~17,000 (nearly all possible) LDLR coding variants on both LDLR cell-surface abundance and LDL uptake, yielding sequence-function maps that recapitulate known biochemistry,

Familial Hypercholesterolemia Screening in Childhood and Early Adulthood: A Cost-Effectiveness Study.

IMPORTANCE: Heterozygous familial hypercholesterolemia (FH), a genetic condition, results in lifelong increased low-density lipoprotein cholesterol (LDL-C) and increases lifetime cardiovascular disease (CVD) risk. Most individuals with FH remain undiagnosed, so early FH identification and treatment could lower CVD burden. OBJECTIVE: To evaluate the projected cost-effectiveness of population sequential FH screening (lipid testing followed by genetic testing after a high LDL-C measurement) at 10 or 18 years of age. Normalization strategies for protein quantification in plasma astrocyte-derived extracellular vesicles: a clinical applicability study.

BACKGROUND: Astrocyte-derived extracellular vesicles (ADEVs) have emerged as a novel research tool in the field of central nervous system disorders. However, significant differences in yield and purity exist among extracellular vesicles (EVs) isolated by different methods, leading to considerable heterogeneity in clinical study outcomes. Therefore, establishing appropriate normalization strategies to enhance comparability across results is a key prerequisite for their clinical translation. METHODS: This study included 15 patients with major depressive disorder (MDD) and 15 healthy controls (HC
