Cuproptosis and the bioinorganic basis of copper-induced cell death.

Copper is an essential yet potentially toxic biological trace element, whose functions in cells arise from its unique redox and coordination chemistry. The same chemical properties that make copper indispensable for a variety of fundamental biological processes, including respiration and antioxidant defense, also render it highly toxic when intracellular concentrations exceed homeostatic levels. Recently, a copper-dependent form of regulated cell death (RCD) termed cuproptosis has been identified, in which excess copper triggers cytotoxicity by directly binding the metabolic machinery of mitoc

Overaccumulation of miR-483-3p exerts acute toxic effects on ovarian granulosa cells by impairing cell proliferation, mitochondrial function, and METTL3-mediated m6A modification.

BACKGROUND: Diminished ovarian reserve (DOR) is a common cause of female infertility. Prior high-throughput sequencing showed specific follicular fluid exosomal miRNA profiles in DOR patients, implicating exosomal miRNAs in DOR pathogenesis. Using miRNA PCR arrays, we found miR-483-3p was significantly upregulated in DOR follicular fluid exosomes. Bioinformatics and experiments indicated METTL3 as a potential miR-483-3p target, suggesting excess miR-483-3p exerts cytotoxicity and suppresses METTL3-mediated N6-methyladenosine (m6A) modification. This study thus investigated miR-483-3p effects o

Real-world assessment of elevated serum alkaline phosphatase and hyperphosphatemia as predictors of mortality in non-dialysis chronic kidney disease patients: a hospital-based prospective cohort study.

BACKGROUND/OBJECTIVE: Disordered mineral metabolism is associated with adverse outcomes in dialysis populations, but its prognostic significance in non-dialysis CKD is less well defined. This study evaluated elevated serum alkaline phosphatase (ALP) and hyperphosphatemia as predictors of mortality and renal outcomes in non-dialysis CKD. METHODS: In this prospective cohort study, patients from a tertiary renal clinic were followed for >12 months. Elevated ALP was defined as >105 U/L in females or >130 U/L in males; hyperphosphatemia as phosphate >4.5 mg/dL. The primary outcome was mortality, a

Pseudomonas sp. UW4 ameliorates continuous cropping by optimizing soil nutrients and combating garlic oxidative damage.

Continuous cropping obstacles have emerged as a major global challenge to sustainable agricultural production, inducing severe soil degradation and ecosystem imbalance. While root-associated microbiota have demonstrated ecological potential in ameliorating continuous cropping disorders, the specific application mechanisms of Plant Growth-Promoting Bacteria (PGPB) in garlic (Allium sativum L.) under continuous cropping stress remain insufficiently explored. This comprehensive study systematically investigated PGPB-mediated improvements in garlic’s morpho-physiological traits, antioxidant defens

Metabolic Dysfunction Associated Steatotic Liver Disease and Bone Turnover Markers in Postmenopausal Women: A Cross-Sectional Study in Central Nepal.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease, a condition of fatty infiltration in the liver, is mostly observed in women. Women after menopause are more likely to develop insulin resistance, hyperlipidemia, and visceral fat storage, which are all recognized to be risk factors for MASLD. Chronic low-grade inflammation in hepatocytes, which is a hallmark of MASLD, may significantly affect bone metabolism. This study aimed to evaluate the relationship between MASLD and bone markers in postmenopausal women. METHODS: The study is a cross-sectional study, carried o

Reporting a Novel Disease Causing Variant in PGAP3 Associated With Hyperphosphatasia and Intellectual Disability: A Case Report and Comprehensive Literature Review.

BACKGROUND: A rare autosomal recessive disorder known as hyperphosphatasia with impaired intellectual development syndrome (HPMRS), also referred to as Mabry syndrome, is caused by a deficiency in glycosylphosphatidylinositol (GPI). Elevated blood alkaline phosphatase (ALP) levels, cognitive impairment, and epileptic seizures are among its key features. These pathways are involved in the synthesis of GPI and the transfer of GPI anchor to the proteins, fatty acid remodeling, and transport of GPI-anchored proteins (GPI-APs). METHODS: Using exome sequencing (ES), the cause of hyperphosphatasia a

A phase 2 trial of burosumab for treatment of fibroblast growth factor-23-mediated hypophosphatemia in children and adults with fibrous dysplasia.

Fibrous dysplasia (FD) is a rare disorder associated with fractures and deformities. FD lesions produce excess phosphaturic hormone fibroblast growth factor 23 (FGF23), leading to hyperphosphaturia in most patients, and hypophosphatemia in those with high FD burden. Skeletal complications are associated with both low-normophosphatemia and frank hypophosphatemia. Burosumab is approved for other forms of FGF23 excess, but there is little evidence to inform use in FD. A phase 2 study investigated the safety and efficacy of burosumab in patients with FD. The primary endpoint was the proportion of

Inositol Thiophosphates as Inhibitors of Mammalian, Plant, and Fungal Phytases.

Inositol phosphates constitute a conserved family of signaling molecules that regulate a wide range of cellular processes such as energy metabolism, calcium release, and signal transduction. In mammalian cells, the endoplasmic reticulum-localized enzyme, multiple inositol polyphosphate phosphatase 1 (MINPP1), is the sole phytase known to hydrolyze myo-inositol hexakisphosphate (IP6). MINPP1 is critical for maintaining intracellular inositol phosphate homeostasis, and its dysfunction is linked to severe neurodevelopmental disorders. Beyond mammals, phytases are widely distributed across bacteri

Tidal Rhythm Regulates the Adverse Outcome Pathways of Microplastics on Coastal Microbial Mat Multifunctionality.

Coastal microplastic pollution is seeing increased concerns, yet the multifunctional response of microbial mats at the sediment-seawater-atmosphere multiphase interface to various microplastics remains poorly understood, especially incorporating long-term tidal regimes. This study utilized a simulation platform coupling 180 tidal cycles to investigate differential impacts of nondegradable (polyethylene, PE) and degradable (polylactic acid, PLA) microplastics on coastal microbial mats. PE at 50 mg/kg impaired microbial mat species richness and evenness, significantly inhibiting nitrate reductas

Assessing causal relationships between iron level, disorders of iron metabolism, and hepatocellular carcinoma: a Mendelian randomisation study : The first author.

BACKGROUND: Emerging evidence indicates that elevated systemic iron levels and dysregulated iron metabolism are closely associated with the pathogenesis of hepatocellular carcinoma (HCC). Observational studies have linked iron overload (e.g., hereditary hemochromatosis) to an increased risk of HCC, potentially mediated through iron-induced oxidative stress, DNA damage, and chronic inflammatory responses. However, the causal relationship between specific iron-related factors-such as organ-specific iron accumulation, systemic iron metabolism disorders, or exogenous iron exposure (dietary/drug ir
