Assessing causal relationships between iron level, disorders of iron metabolism, and hepatocellular carcinoma: a Mendelian randomisation study : The first author.

BACKGROUND: Emerging evidence indicates that elevated systemic iron levels and dysregulated iron metabolism are closely associated with the pathogenesis of hepatocellular carcinoma (HCC). Observational studies have linked iron overload (e.g., hereditary hemochromatosis) to an increased risk of HCC, potentially mediated through iron-induced oxidative stress, DNA damage, and chronic inflammatory responses. However, the causal relationship between specific iron-related factors-such as organ-specific iron accumulation, systemic iron metabolism disorders, or exogenous iron exposure (dietary/drug ir

Inherited microcytic anemias due to disorders of iron and heme metabolism: An updated clinical review.

Microcytic anemia is among the most common hematological abnormalities in clinical practice and is usually attributable to iron deficiency, thalassemia traits, or anemia of inflammation. A small but clinically important subset of patients, however, has inherited disorders of iron metabolism or heme synthesis presenting with persistent, unexplained, familial, or iron-refractory microcytosis. Over the past decade, advances in the understanding of the hepcidin-ferroportin axis, erythroid iron regulation, standardized hepcidin assays, and next-generation sequencing have substantially refined diagn

A review - Prescribing iron salts: Comparison of efficacy and tolerance of different iron complexes.

Iron is an essential element as it participates in a wide variety of metabolic processes, including oxygen transport, deoxyribonucleic acid synthesis and electron transport. However, as iron can form free radicals, its concentration in body tissues must be tightly regulated because, in excessive amounts, it can lead to tissue damage. Disorders of iron metabolism are among the most common diseases of humans with diverse clinical manifestations. Iron deficiency is the most common mineral deficiency, affecting approximately 20% of the population. Patients may present with an iron deficiency witho

A mitochondrion-targeted poly(N-isopropylacrylamide-coacrylic acid) nanohydrogel with a fluorescent bioprobe for ferrous ion imaging in vitro and in vivo.

An imbalance in iron homeostasis contributes to mitochondrial dysfunction, which is closely linked to the pathogenesis of various diseases. Herein, we developed a nanosensor for detecting mitochondrial ferrous ions in vitro and in vivo. A poly(N-isopropylacrylamine)-coacrylic acid nanohydrogel was synthesized, and ferrous ions were detected using the fluorescent probe FeRhonox-1 embedded within it. (3-Carboxypropyl)-triphenylphosphonium bromide was chemically conjugated to the hydrogel matrix to enable mitochondrial targeting. The developed nanosensor showed a narrow particle size distribution

Identifying haemochromatosis patients with C282Y homozygosity from inpatient electronic patient records in England using a novel algorithm: a retrospective observational study.

INTRODUCTION: Hereditary haemochromatosis (HH) is the most common genetic condition among populations of northern European ancestry, but it does not have a specific International Classification of Diseases 10th revision (ICD-10) diagnosis code. HH is commonly assigned the ICD-10 code E83.1 defined as 'disorders of iron metabolism'. However, the E83.1 diagnosis code is also applied to patients with transfusion-related iron overload and hyperferritinaemia from non-iron loading conditions. Venesection is the main treatment option for patients with HH and is assigned the Office of Population, Cens

Disorders of Iron Metabolism: A "Sharp Edge" of Deoxynivalenol-Induced Hepatotoxicity.

BACKGROUND/OBJECTIVES: Deoxynivalenol (DON), known as vomitoxin, is one of the most common mycotoxins produced by Fusarium graminearum, with high detection rates in feed worldwide. Ferroptosis is a novel mode of cell death characterized by lipid peroxidation and the accumulation of reactive oxygen species. Although it has been demonstrated that DON can induce ferroptosis in the liver, the specific mechanisms and pathways are still unknown. The aim of this experiment was to investigate that DON can induce iron metabolism disorders in the livers of mice, thereby triggering ferroptosis and causin

Advances in Ferroptosis in Chronic Inflammatory Diseases of the Upper and Lower Airways.

BACKGROUND: The pathogenesis of chronic inflammatory diseases in the upper and lower airways is common and mutually influential and should be managed systematically. Disorders of iron metabolism and redox homeostasis imbalance, which lead to the accumulation of reactive oxygen species (ROS) and lipid peroxidation, are considered key common characteristics linking ferroptosis to chronic inflammatory diseases of the upper and lower airways. Recognizing the mechanism of ferroptosis and how it contributes to chronic inflammatory diseases of the upper and lower airways enables a more comprehensive

Early pathological changes in the liver and kidney of non-obese diabetic (NOD) mice: involvement of iron accumulation and ferroptosis.

Disorders of iron metabolism and ferroptosis play an important role in the development of diabetes and related pathologies. The involvement of ferroptosis in type 1 diabetes has mainly been investigated in animal models with chemically induced diabetes. Our aim was to examine the involvement of iron homeostasis disturbances and ferroptotic events in liver and kidney damage in non-obese diabetic (NOD) mice in the early phase of spontaneous development of diabetes (15 days of stable hyperglycemia). We found an accumulation of iron and lipid peroxides in the proximal tubule epithelial cells (PTEC

Neurodegeneration With Brain Iron Accumulation and Ferroptosis Disorders in Children and Adults: An Imaging Review.

Neurodegeneration with brain iron accumulation (NBIA) refers to a group of rare genetic disorders characterized by abnormal iron deposition in the basal ganglia and brainstem due to impaired iron homeostasis. Disease severity and manifestations vary according to the underlying genetic mutation and age of presentation; however, most subtypes share progressive neurological features such as dystonia, Parkinsonism, spasticity, cognitive decline, and intellectual disability. In this review, we first outline the physiological role of iron in the central nervous system, emphasizing its importance for

Eryptosis in renal anemia: mechanisms, clinical implications, and therapeutic targeting.

Renal anemia is one of the most common complications of chronic kidney disease (CKD) and is associated with serious clinical consequences. Its prevalence increases significantly as renal function declines, affecting over 90% of dialysis patients. Traditionally, the pathophysiology of renal anemia has been centered on two primary mechanisms: absolute or relative deficiency of erythropoietin (EPO) and disorders of iron metabolism. However, even with treatment using erythropoiesis-stimulating agents (ESAs) and iron supplements, approximately half of the patients exhibit a hypo-responsive or subop

Characterization of ferroportin disease and SLC40A1-related hemochromatosis - Results from the EASL non-HFE registry.

BACKGROUND & AIMS: Pathogenic variants in the cellular iron exporter ferroportin (SLC40A1) cause hepatic and splenic iron overload. Low to normal transferrin saturation (TSAT) and iron accumulation in Kupffer cells with high splenic iron distinguish ferroportin disease (FD) from SLC40A1-related hemochromatosis (SLC40A1-HC), which are both caused by variants in SLC40A1. The aim of our study was to describe pathogenic mutations in SLC40A1, phenotypic variability in affected patients and compare outcomes with HFE-related hemochromatosis (HFE-HC). METHODS: The international EASL non-HFE hemochrom

Human umbilical cord mesenchymal stem cells protect against ferroptosis in acute liver failure through the IGF1-hepcidin-FPN1 axis and inhibiting iron loading.

Acute liver failure (ALF) is a significant global issue with elevated morbidity and mortality rates. There is an urgent and pressing need for secure and effective treatments. Ferroptosis, a novel iron-dependent regulation of cell death, plays a significant role in multiple pathological processes associated with liver diseases, including ALF. Several studies have demonstrated that mesenchymal stem cells (MSCs) have promising therapeutic potential in the treatment of ALF. This study aims to investigate the positive effects of MSCs against ferroptosis in an ALF model and explore the underlying mo

Magnetically attracting hydrogel reshapes iron metabolism for tissue repair.

Ferroptosis, caused by disorders of iron metabolism, plays a critical role in various diseases, making the regulation of iron metabolism essential for tissue repair. In our analysis of degenerated intervertebral disc tissue, we observe a positive correlation between the concentration of extracellular iron ions (ex-iron) and the severity of ferroptosis in intervertebral disc degeneration (IVDD). Hence, inspired by magnets attracting metals, we combine polyether F127 diacrylate (FDA) with tannin (TA) to construct a magnetically attracting hydrogel (FDA-TA). This hydrogel demonstrates the capabil
