Biofunctional characterization and preclinical evaluation of the hypolipidemic potential of putative probiotic Pediococcus pentosaceus GWS5 and Lactiplantibacillus plantarum KD11 isolated from ethnic Himalayan Gundruk and Kinema.

Hyperlipidemia remains a significant global health concern, necessitating novel therapeutic approaches. This study aimed to isolate Lactic acid bacteria (LAB) from Gundruk and Kinema, the traditional fermented foods of Darjeeling, Kalimpong and Sikkim and to screen them for their in vitro cholesterol lowering ability. Isolates demonstrating high cholesterol reduction were further characterized for probiotic attributes, molecularly identified and evaluated for their hypolipidemic efficacy in vivo. Pediococcus pentosaceus GWS5 and Lactiplantibacillus plantarum KD11 exhibited the greatest cholest

Genetic evidence for a potential role of cytoskeleton-associated protein 2 in lung squamous cell carcinoma: a bidirectional mendelian randomization study.

BACKGROUND: Cytoskeleton-associated protein 2 (CKAP2) plays a role in tumor proliferation, but its causal relationship with lung cancer remains unclear. This study employed a bidirectional two-sample Mendelian randomization (MR) analysis to investigate this association. METHODS: Using genome-wide association and FinnGen data, we assessed the potential causal relationship between genetically predicted plasma CKAP2 levels and lung cancer risk, including its subtypes. The inverse-variance weighted (IVW) method served as the primary analysis, supplemented by MR-Egger and weighted median. Sensitiv

The zebrafish model tackles anti-P2Y12 variability in humans: a translational approach.

Antiplatelet drugs are a pillar in the treatment strategy to prevent ischemic events in patients with cardiovascular disease. However, the action of existing therapies is nonuniform, and the causative mechanism for this variability remains poorly understood. The differences in the microRNA (miRNA) profiles among individuals have been suggested to impact platelet reactivity and treatment outcomes. In several previous clinical reports, miR-150 has been associated with platelet function variability and cardiovascular events. Therefore, we initiated our analysis by examining the mechanistic role o

The prognostic potential of an immunology score in patients treated with curative surgery for non-small cell lung cancer.

PURPOSE: Curative surgery is the best treatment for patients with early-stage non-small cell lung cancer (NSCLC), but around 40% experience recurrence impacting survival. Does the individual immunological status affect the prognosis? This study investigated four immunology markers in relation to recurrence and survival after curative surgery for NSCLC. METHODS: Natural Killer cell activity (NKA), C-reactive protein (CRP), Interleukin 6 (IL6) and Transforming growth factor beta (TGFβ) were analysed before surgery and at three months after surgery for NSCLC. RESULTS: Of the 103 enrolled patien

Astrocyte lipid metabolism: From physiological homeostasis to reactive phenotype plasticity in neurodegeneration.

Astrocytes are one of the most abundant types of glial cells in the central nervous system (CNS) and play pivotal roles in metabolic support, synaptic regulation, and neurotransmitter homeostasis. Astrocytic dysfunction is closely associated with the pathogenesis of neurodegenerative disorders such as Alzheimer's disease (AD), and lipid metabolism constitutes a central axis for maintaining astrocyte function and CNS homeostasis. Here, we provide a systematic review of recent advances in astrocyte lipid metabolism, summarizing the fundamental roles of astrocytes in the CNS and focusing on the b

Inulin- and Butyrate-Mediated HIF-1α Butyrylation Prevent Diabetic Ferroptosis and Cardiomyopathy by Maintaining Iron Homeostasis.

UNLABELLED: Abnormal iron metabolism and ferroptosis contribute to diabetic cardiomyopathy (DCM); however, the regulatory mechanisms remain unclear. Inulin improves systemic metabolism and cardiac function, but whether it alleviates myocardial ferroptosis and DCM is unknown. Using two distinct type 2 diabetes (T2D) mouse models-the high-fat diet combined with streptozotocin-induced T2D model and the db/db mouse model-in combination with an iron chelator, we found that imbalanced iron homeostasis and ferroptosis drive DCM. Inulin restored cardiac iron homeostasis, inhibited ferroptosis and DCM,

What Proportion of Antibody-Negative Type 1 Diabetes Cases Have a Nonautoimmune Etiology? A Genetics-Based Estimate.

UNLABELLED: Among patients with young-onset type 1 diabetes, 10-15% are autoantibody (aAb) negative. How many of these are autoimmune versus nonautoimmune is unknown. To estimate this proportion, we used the only high-confidence evidence of autoimmunity in these cases, the presence of an aAb(+) sibling. In the 2,500 affected sibling pairs of the Type 1 Diabetes Genetics Consortium cohort, we compared the Type 1 Diabetes Genetic Risk Score (GRS2) between 379 aAb(-) patients with an aAb(+) sibling (autoimmune cause) and 253 aAb(-) patients with no aAb(+) siblings (unknown cause), under the assum

A Blood-Derived Factor Rescues ALS: Platelet Factor 4 Activates OPTN-Dependent Autophagy to Clear SOD1 Aggregates Independently of PINK1.

Peripheral factors that systemically regulate amyotrophic lateral sclerosis (ALS) have remained elusive-until now. Here, by integrating population-scale epidemiology with mechanistic dissection, we identify platelet factor 4 (PF4) as the central driver of a circulating neuroprotective axis that restores proteostasis and rescues ALS. In a prospective cohort of >500 000 UK Biobank participants, platelet indices were strongly associated with ALS risk, and serum PF4 levels were significantly reduced in ALS patients. Systemic administration of recombinant PF4 in hSOD1G93A mice produced dramatic the

Pharmacokinetics and efficacy of tank-water-administered dabrafenib, a BRAF inhibitor, in a zebrafish model of melanoma.

Zebrafish models are widely used to study BRAF-mutant melanoma biology. However, long-term treatment of adult fish with small-molecule BRAF inhibitors remains challenging, and pharmacokinetic data to inform rational dosing strategies are largely lacking for most small molecules in zebrafish. Here, we assessed the pharmacokinetics, metabolism and efficacy of continuously tank-water-administered dabrafenib, a BRAF inhibitor, in adult zebrafish. Dabrafenib was rapidly absorbed from tank water, reaching efficacious plasma levels within 1 h, but also showed fast elimination kinetics with a half-lif

Pharmacist-Led Continuous Glucose Monitoring for Prediabetes: Protocol for a Prospective Cohort Study.

BACKGROUND: Prediabetes represents a critical intervention window to reduce progression to type 2 diabetes. The current standard of care for treatment of prediabetes is weight loss and lifestyle modification, which are both highly effective, although limited by real-world adherence. It is well recognized that continuous glucose monitoring (CGM) improves glycemic and behavioral outcomes in people with diabetes, although outcomes among people with prediabetes are not well described. OBJECTIVE: The aim of this study is to evaluate the impact of pharmacist-led CGM on glycemic control and health-r

TXNIP and NLRP3 inflammasome markers in major depressive disorder: Associations with remission status and verbal memory.

BACKGROUND: Oxidative stress plays a role in the pathophysiology of depression and cognitive dysfunction. Oxidative stress induced NOD-like receptor 3(NLRP3) inflammasome and Thioredoxin-interacting protein (TXNIP) activation may contribute to neuroinflammation in depression. As oxidative stress contributes to insulin resistance, it may represent a shared biological pathway linking depression and diabetes. This study compared NLRP3, TXNIP, and insulin resistance in remitted and non-remitted patients with major depressive disorder (MDD) and healthy controls, and their associations with cognitiv
