Circulating neuronal proteins are elevated in severe autism and associated with patient-specific neuronal dysfunction and behavioral deficits.

Autism spectrum disorder (ASD) is a complex neurodevelopmental condition with a multifactorial etiology involving both genetic and environmental factors. Although previous studies have identified altered levels of circulating proteins in individuals with ASD, the functional significance of these peripheral biomarkers remains largely unclear. In this study, we recruited a well-characterized cohort of young Arab children with ASD (n = 100) and matched controls (n = 60), aged 2-4 years, in Qatar. We used a multimodal approach, integrating data from proteomics, transcriptomics, induced pluripotent

Plasma proteomic signatures predict incident benign prostatic hyperplasia: a prospective cohort study of 20 996 men.

BACKGROUND: Benign prostatic hyperplasia (BPH) is a prevalent disease in elderly men. However, the plasma proteomic signatures for incident BPH are absent, hindering early prediction and risk stratification. We aimed to identify plasma proteins associated with incident BPH and the implicated biological pathways. METHODS: This prospective cohort was constructed based on the UK Biobank, enrolling 20 996 BPH-free males at baseline. We used the Olink Explore platform to determine the abundances of 2920 plasma proteins. We used Cox regression models to evaluate associations and the Extreme Gradien

A novel homozygous ABCB6 variant causing the Lan null phenotype and anti-Lan-mediated hemolytic disease of the fetus and newborn: A case report and a review of anti-Lan in pregnancy.

BACKGROUND: The Lan antigen is a high-prevalence red blood cell antigen encoded by the ATP binding cassette subfamily B member 6 (ABCB6) gene. Variants in this gene are responsible for Lan null and variant phenotypes. Individuals lacking the Lan antigen may develop anti-Lan antibodies following sensitizing events such as transfusion or pregnancy. Hemolytic disease of the fetus and newborn (HDFN) caused by anti-Lan antibodies is extremely rare. CASE PRESENTATION: We report a case of a Chinese pregnant woman carrying a novel homozygous variant in exon 17 of the ABCB6 gene (NM_005689.4:c.2312_23

Immunometabolism drives metabolic and infectious disease progression through bidirectional metabolic reprogramming and epigenetic remodeling.

Immunometabolism has become a central mechanism governing innate and adaptive immune responses. The field has moved from cataloguing metabolic shifts during immune activation to understanding how specific pathways control immune cell fate and function. This review brings together current knowledge on immunometabolic regulation in metabolic and infectious diseases, emphasizing bidirectional crosstalk between these domains. We address controversies over whether inflammation causes insulin resistance or follows from it, examine the distinct metabolic programs of different immune cell types, and e

Specific cleavage of 5' overhangs of non-target strand by Cas9 activated by target DNA binding.

The clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 system has become a powerful genome-editing tool that uses RNA-DNA pairing to cleave target DNA with protospacer adjacent motif (PAM) sequences. While its primary function is well-studied, secondary activities remain poorly understood, causing unintended off-target effects. This study reports for the first time that Cas9 specifically cleaves the 5' overhang of the non-target strand (NTS) in target double-stranded DNA. This specific cleavage requires an additional PAM element at the NTS 5' region, is mediated by Cas9's

The deubiquitinase CYLD inhibits thrombin-induced p38 MAPK-p65 NF-κB signaling and inflammatory cytokine production to suppress triple-negative breast cancer progression.

Triple-negative breast cancer (TNBC) is an aggressive subtype characterized by chronic inflammation with limited therapeutic options. Although thrombin activation of protease-activated receptor-1 (PAR1) promotes ubiquitin-dependent signaling and inflammatory responses in endothelial cells, its contribution to inflammatory effects in TNBC remain poorly understood. Here, we show that loss of the deubiquitinase cylindromatosis (CYLD) markedly increases basal phosphorylation of p38 mitogen-activated protein kinase and p65 nuclear factor kappa B subunit to levels comparable to thrombin stimulation,

Heme-mediated liver-heart axis promotes myocardial pyroptosis in db/db mice.

Diabetes, a systemic metabolic disorder, often involves multi-organ interactions in its complications. Although the pathogenesis of diabetic cardiomyopathy (DCM) is complex, the role of the liver in this process remains unclear. Using the db/db mouse model, this study demonstrated that metabolic dysfunction associated steatotic liver disease (MASLD) reduced hepatic synthesis of hemopexin (HPX) and impaired Kupffer cell function, leading to systemic heme accumulation. This circulating heme was taken up by cardiomyocytes via the membrane protein feline leukemia virus subgroup C receptor 2 (FLVCR

Astrocyte-Derived Exosomal miR-211-5p Alleviates Blood-Brain Barrier Injury in a Rat Model of Traumatic Brain Injury.

BACKGROUND: Traumatic brain injury (TBI) triggers a cascade of secondary damage, including neuroinflammation, astrocyte activation, and disruption of the blood-brain barrier (BBB), all of which contribute to long-term neurological deficits. Astrocyte-derived exosomes have emerged as a promising therapeutic avenue; however, the specific contributions of their molecular cargo remain poorly understood. This study explores whether astrocyte-derived exosomal delivery of microRNA-211-5p (miR-211-5p) can attenuate secondary injury and enhance functional recovery following TBI. METHODS: Primary astro

Modified Huanglian Wendan Decoction improves prediabetic renal injury by activating the AMPK/ULK1 pathway to restore autophagy and suppress inflammation.

BACKGROUND: Prediabetes represents an intermediate metabolic condition between normal blood glucose and diabetes, characterized by insulin resistance, metabolic dysfunction, and low-grade inflammation, collectively contributing to early renal injury. However, manifestations such as glomerular hyperfiltration, increased urinary protein excretion, and subclinical tubular injury are often overlooked during this stage. Therefore, elucidating the underlying molecular mechanisms and implementing early interventions may significantly attenuate subsequent renal function decline. In this context, altho

Healthy lifestyle modifies the association between per- and polyfluoroalkyl substances and hyperlipidemia: A propensity score-matched case-control study.

The impact of per- and polyfluoroalkyl substances (PFAS) on hyperlipidemia and the potential modifying role of overall lifestyle remain underexplored. We examined six serum PFAS and a composite lifestyle score among 192 propensity score-matched case-control pairs. Individual and joint associations were evaluated using conditional logistic regression, quantile g-computation, and Bayesian kernel machine regression. A composite healthy lifestyle score was constructed based on smoking, alcohol drinking, physical activity, body shape, and diet. Per interquartile range (IQR) increase in ln-transform

Mapping disease-linked AOPs to inform bioassay selection for chemical hazard assessment.

Modern societies synthesize, distribute, and utilize thousands of chemicals, but their contribution to the global burden of non-communicable diseases remains insufficiently characterized. Improved availability of robust hazard data would enable more systematic assessment, prioritization, and management of chemical-related health risks. New Approach Methodologies (NAM) are promising because they provide high-throughput, mechanistically informative, and animal-free testing that improves current regulatory toxicity testing. However, selecting biologically relevant in-vitro assays capable of predi

Investigating Novel Inflammatory Biomarkers and Pathways in Cardiac Cachexia.

BACKGROUND: Cardiac cachexia (CC) is an indicator of advanced heart failure (AHF), with significant unintentional weight loss (UWL) and poor prognosis. Its diagnostic criteria incorporate upregulation of established inflammatory cytokines. Identification of novel biomarkers, for example, via proteomic analyses, could unearth unstudied mechanisms behind CC formation with future applications in disease therapeutics. This study, therefore, aimed to explore the presence of contemporary, novel biomarkers, and mechanistic pathways in CC using targeted proteomics. METHODS: Patients with AHF or stabl

Oleanolic Acid Ameliorates Hepatic Lipid Metabolism and Autophagy in Type 2 Diabetic Mice via the STAT3 Signaling Pathway.

Type 2 diabetes mellitus (T2DM) poses a major global health challenge due to its considerable disability and mortality rates. The complex etiology of T2DM involves hepatic steatosis and autophagy dysfunction. Previous research indicates that oleanolic acid (OA) ameliorates T2DM, with signal transducer and activator of transcription 3 (STAT3) serving as a crucial regulator in numerous cellular processes. Therefore, this study aims to explore whether the therapeutic effects of OA are associated with STAT3 activation and its potential role in hepatic lipid metabolism and autophagy in T2DM. T2DM w

Anti-Aβ3-10 monoclonal antibody 7B8 improves cognitive function and protects the blood-brain barrier in APP/PS1 mice by regulating the HMGB-1/RAGE/NF-κB pathway.

BACKGROUND: Alzheimer's disease (AD), the most prevalent dementia, is primarily underpinned by the amyloid cascade hypothesis. Passive Aβ immunotherapy effectively reduces cerebral Aβ deposition but is limited by severe side effects, including cerebral amyloid angiopathy (CAA), microhemorrhage, and amyloid-related imaging abnormalities (ARIA). Here, we investigated the efficacy and safety of a novel anti-A3-10 monoclonal antibody (7B8) in APP/PS1 double-transgenic mice, with a focus on its impacts on amyloid clearance, neuroinflammation, and blood-brain barrier (BBB) integrity. METHODS: 7B8 w

ATR inhibition potentiates the antitumor efficacy of HER3-DXd in HER3-positive/HR-positive breast cancer by increasing DNA damage.

BACKGROUND: Endocrine resistance remains a major challenge in hormone receptor-positive (HR+) breast cancer (BC), where up to 70% of tumours overexpress HER3, a receptor associated with poor prognosis and therapeutic resistance. HER3-DXd (patritumab deruxtecan) is currently under clinical investigation for HER3-expressing metastatic BC. However, strategies to further enhance its efficacy, particularly in endocrine therapy-resistant settings, are urgently needed. We hypothesised that targeting ATR, a key regulator of DNA damage repair (DDR), potentiates HER3-DXd in HER3+/HR+ BC, including tamox
