High Immunophenotyping of Human PBMCs Using Lyophilized Antibodies.

Immunophenotyping of human peripheral blood mononuclear cells (PBMCs) is a critical technique for understanding immune responses in both health and disease. Traditionally, this process relies on the use of liquid antibody cocktails, which are time-consuming to prepare, prone to laboratory error, and have limited shelf lives. Recent advancements in lyophilization of antibodies and other reagents have addressed these challenges by enhancing reagent stability, extending shelf life, and simplifying transport and storage. Dried antibody cocktails have been shown to perform comparably to liquid cock

Evaluation of itch intensity scales in atopic dermatitis: Differential measurement properties and associations with relevant cytokines.

Itch is the cardinal symptom contributing to patient burden in atopic dermatitis. Multiple validated itch scales are used in clinical trials, generating heterogeneous datasets. In addition, recent studies suggest an association between cytokine levels and disease severity in atopic dermatitis. This study aimed to compare the performance of different validated itch instruments and their relationship to blood cytokine profiles. Forty-nine adults with severe atopic dermatitis and severe itch were treated with 300 mg dupilumab for 16 weeks. At the initial assessment and after treatment, itch inten

Exploring the link between fatty acids, FADS2 expression, and ADHD using structural equation modeling.

Fatty acids have been linked to attention-deficit/hyperactivity disorder (ADHD), but the biological basis of this association remains unclear. This study examined dietary and plasma fatty acid profiles and fatty acid desaturase-2 (FADS2) in children with ADHD. This cross-sectional study included 85 children with ADHD and 85 controls aged 6-12 years. ADHD symptoms were assessed using the Conners' Parent Rating Scale-Revised: Short Form (CPRS-R: S). Dietary fatty acid intake was estimated from three non-consecutive 24-hour recalls. Plasma fatty acids were measured by GC-MS. Plasma FADS2 protein

The scientific rationale for long-acting biologics in dermatology.

The evolution of biologic therapies in dermatology increasingly emphasizes extending drug half-life to optimize pharmacokinetics and therapeutic durability. Mechanisms such as FcRn-mediated recycling, Fc engineering, albumin binding, PEGylation, and amino acid polymer technologies enhance systemic persistence and stabilize drug exposure. These advances enable sustained inhibition of key inflammatory pathways, including IL-23/T-helper 17 and IL-4/IL-13, reducing disease relapse and improving long-term outcomes. Extended half-life biologics also improve adherence, reduce immunogenicity, and less

Structural insights into TNF-α inhibition by bioactive compounds found in plants of North East India: in vitro validation and in silico investigations using QSAR, molecular docking, and dynamics simulations.

Medicinal plants from the Eastern Himalayas of Northeast India are integral to traditional therapies for inflammation. This study evaluated the anti-inflammatory potential of 23 reported phytochemicals isolated from this region by targeting TNF-α, a critical mediator of autoimmune disorders. TNF-α production was induced in human peripheral blood mononuclear cells using 100 ng/mL lipopolysaccharide for 24 h and quantified using ELISA. The IC50 was calculated by probit regression, with methotrexate as the reference standard (IC50 = 1.96 µM; 94% inhibition at 3.5 µM). Rutin emerged as the most po

The Initial Therapeutic Response in Patients with Unresectable Hepatocellular Carcinoma Treated with Atezolizumab plus Bevacizumab Contributes to the Change of Liver Function during Treatment: A Multicenter Analysis.

INTRODUCTION: Therapeutic response in patients with unresectable hepatocellular carcinoma (HCC) receiving multikinase inhibitors is associated with changes in liver function. However, limited data are available for patients treated with atezolizumab plus bevacizumab (ATZ/BEV). METHODS: We retrospectively enrolled 151 patients with unresectable HCC who received ATZ/BEV as first-line systemic therapy. Liver function was assessed using albumin-bilirubin (ALBI) score at baseline (pre-ALBI) and 6 weeks after the initiation of ATZ/BEV (6W-ALBI). We evaluated the dynamic relationship between initial

The insulin-like growth factor system and the progression of renal complications of type 2 diabetes.

AIMS: Chronic kidney disease is associated with dysregulation of the insulin-like growth factor (IGF) system. Alterations in IGF bioavailability, driven by IGF binding proteins (IGFBPs), may influence the decline in renal function. However, the longitudinal relationship between baseline IGF-I, IGF-II, IGFBPs and progression of kidney dysfunction remains unclear. We evaluated how IGF/IGFBP profile relates to longitudinal changes in urinary albumin/creatinine ratio (ACR) and serum creatinine level in people with type 2 diabetes (T2D). METHODS: Individuals' measurements were performed in 436 ind

SORBS2 regulates diastolic function through cytoskeletal networks and calcium handling.

Diastolic dysfunction, defined by impaired relaxation and increased ventricular stiffness, is central to heart failure with preserved ejection fraction, yet cardiomyocyte-intrinsic mechanisms remain incompletely understood. Here, we show that SORBS2 is a vital component of murine cardiomyocyte adhesion complexes. Its genetic ablation in mice causes progressive diastolic dysfunction with preserved systolic function, accompanied by atrial enlargement and reduced survival. Postnatal cardiomyocyte-directed re-expression of SORBS2 restores diastolic indices and significantly improves longevity. Mec

SPI1 promotes endothelial cell ferroptosis and intracranial aneurysm progression via EP300-mediated FOSL2 acetylation.

This study investigates the role of SPI1 in the pathogenesis of intracranial aneurysm (IA). Results confirmed that SPI1 was significantly upregulated in both an in vivo mouse IA model and an in vitro model of angiotensin II-stimulated human cerebral microvascular endothelial cells. Functional studies demonstrated that SPI1 overexpression promoted vascular endothelial cell ferroptosis, as evidenced by dysregulation of key markers (p53, GPX4, SLC7A11, ACSL4, TFRC), elevated lipid peroxidation, and mitochondrial damage, whereas SPI1 knockdown attenuated these effects. Mechanistically, SPI1 upregu

Germ layer-specific interaction of ion channels and transporters controls membrane potential and left-right patterning in Xenopus tropicalis.

Emerging evidence indicates that ion channels and transporters are critical for normal embryonic development. They are essential for establishing membrane potential (Vm), a property of all cells. However, it is uncertain if embryos have germ-layer specific regional differences in electrical behavior that play a role in defining local cell fates. In our study, by manipulating ion concentrations and applying a wide spectrum of inhibitors, we demonstrate that various ion channels and transporters differentially determine Vm in specific germ layers of the Xenopus tropicalis embryo. Altering the el

A Comprehensive Review of PCSK9 Inhibitor Therapy.

Proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitors are a key component of lipid-lowering therapies for atherosclerotic cardiovascular disease risk management in high-risk primary and secondary prevention populations. PCSK9 monoclonal antibodies such as evolocumab and alirocumab were the first agents approved and are the preferred agents, given the phase 3 trial data supporting improved cardiovascular outcomes. Subgroup analyses of these trials have highlighted high-risk populations that may especially benefit from PCSK9 inhibitor therapy, although demographic disparities persist

Anti-capsid nanobodies function as HIV-1 capsid inhibitors by intracellular capsid degradation.

Antiretroviral therapy (ART) strategies against HIV-1 have been successful in suppressing HIV-1 replication and preventing disease progression. However, drug-resistance development, side-effects and life-long adherence of current drugs have driven the development of novel inhibitors against HIV-1. Inhibition of capsid production by targeting the highly conserved capsid protein (CA) has shown to be promising in blocking viral replication. Here, we developed a capsid-targeting biological composed of a single-domain Llama VHH that binds CA, fused to human IgG1 Fc to activate TRIM21-mediated degra

Clinical evaluation of plasma D-dimer concentrations as a marker for thromboembolism in cattle with inflammation.

Plasma D-dimer concentrations were measured in 15 cases with thromboembolism and 19 cases without thromboembolism among bovine patients with inflammatory diseases, and the clinical usefulness of plasma D-dimer as a thromboembolic marker was evaluated. No significant difference in plasma D-dimer concentrations was observed between the two groups. A receiver operating characteristic curve was created, and with a plasma D-dimer cutoff value of 0.38 mg/L, the sensitivity and the specificity were 86.7% and 31.2%, respectively. Furthermore, there was a large variability in plasma D-dimer concentrati
