BACKGROUND: Leishmaniosis is a vector-borne disease caused by Leishmania parasites transmitted by infected phlebotomine sand flies. In the epidemiological study reported here, we investigated exposure to Leishmania infantum in sheep (Ovis aries) and goats (Capra hircus) in mainland Portugal.
METHODS: A cross-sectional serosurvey was conducted in three geographical regions of Portugal (Trás-os-Montes, Centre and South), with sampling in 19 municipalities and 87 localities.
RESULTS: A total of 2124 small ruminants were tested (1820 sheep and 304 goats). Overall seroprevalence was 21.3% (453/21

BACKGROUND: Toxoplasma gondii is an apicomplexan parasite which infects nearly all warm-blooded animals and humans, causing zoonotic toxoplasmosis. Pork infected with T. gondii is considered a significant source of human infection. Currently, no commercial vaccines are available for porcine toxoplasmosis globally, thus a safe and effective vaccine is urgently needed. This study evaluated the immuno-protective effects of two T. gondii gene knockout attenuated strains, RHΔtkl1 and PruΔpp2a-c, in pigs.
METHODS: Pigs immunized by intramuscular injection with 1 × 107 tachyzoites of attenuated RHΔt

Chagas disease (CD) remains a major public health challenge in Latin America, largely due to persistent underdiagnosis, particularly in endemic and resource-limited settings. Rapid diagnostic tests (RDTs) offer a pragmatic approach to expand serological screening; however, their performance varies across epidemiological contexts and parasite genetic backgrounds. The TR Chagas Bio-Manguinhos test, based on recombinant chimeric antigens, has shown high sensitivity in previous evaluations, but data from Southern Cone remain limited. We conducted a cross-sectional diagnostic accuracy study using 3

BACKGROUND: Toxoplasma gondii is a major congenital parasitic pathogen that can be transmitted vertically and is associated with a wide spectrum of adverse fetal and neonatal outcomes. In Iran, data on concurrent maternal-neonatal serological assessments remain limited, particularly in hospital-based settings. Neonatal serology is often interpreted without parallel maternal data or contextual information on awareness and risk factors. This hospital-based cross-sectional study in Yasuj, southwestern Iran, aimed to determine the seroprevalence of anti-Toxoplasma IgG and IgM among hospitalized in

BACKGROUND: Unlike mouse models of congenital toxoplasmosis, pregnant sheep models provide the opportunity to evaluate treatment strategies that more closely resemble clinical practice in pregnant women, including chemotherapeutic interventions initiated after specific IgM seroconversion. BKI-1748, which targets Toxoplasma gondii CDPK1 and MAPKL-1 protein kinases, has demonstrated an excellent safety profile and efficacy when administered repeatedly to pregnant sheep, starting at 2 and 7 days after challenge.
METHODS: In this study, treatment was initiated at day 14 post-infection (p.i.), fol

Fever is a hallmark of malaria. Several studies have linked febrile temperatures to reduced parasite viability, but also to increased cytoadhesion, a key driver of pathology. However, different mechanisms have been proposed to cause changes in cytoadhesion and parasite sensitivity to heat. Here, we demonstrate that exposure of Plasmodium falciparum-infected red blood cells (iRBCs) to physiologically relevant febrile heat stress (39 °C), derived from patient data, enhances cytoadhesion through increased trafficking of the major virulence factor PfEMP1 to the iRBC surface. This phenomenon is not

AIM: To determine the cost-benefit of a control programme that used serological testing of young heifer replacement calves to reduce Neospora caninum seroprevalence in two dairy herds.
METHODS: Data were collected between 2016 and 2023 from replacement heifer calf cohorts that were tested at a young age for N. caninum antibodies, on two adjacent Waikato dairy farms with endemic neosporosis. Calf serology (ELISA) results were interpreted using cut-off values adjusted to compensate for the presence of maternal antibodies, to classify calves as seronegative, inconclusive, or seropositive. Linear growth stunting increases a child's risk for mortality and lifelong morbidities. Understanding and preventing the causes of childhood stunting is a global health priority. The leading contributors to stunting are undernutrition and infection. Schistosomiasis, a neglected tropical disease, has long been associated with childhood stunting, but there are limited reviews summarizing the evidence. This narrative review examines cross-sectional associations between infection and linear growth, longitudinal impacts of schistosomiasis treatment on growth and the influence of schistosomiasis on

Malaria caused by Plasmodium falciparum remains a leading cause of childhood mortality in sub-Saharan Africa. The intraerythrocytic parasite can evade complement-mediated destruction through recruitment of human Factor H (FH). In vitro, Factor H-related protein 1 (FHR-1) competes with FH for binding sites, suggesting a potential role in modulating complement activation during infection. Because one-third of Africans carry a CFHR3/1 deletion, we examined the impact of FHR-1 deficiency on malaria severity. We analyzed plasma from 500 Ghanaian children presenting with different clinical manifesta

Malaria, caused by Plasmodium species, remains a major global health burden. The spleen is the central organ for clearance of infected red blood cells and regulation of immunity, yet paradoxically also serves as a site of parasite sequestration. Splenic fibroblasts may contribute to this process through adhesion mechanisms, but their role remains poorly defined. This study investigated P. yoelii tryptophan-rich antigen 7 (PyTRAg7), a member of the TRAg protein family, in splenic parasite-host interactions. TRAgs constitute a conserved protein family present in multiple Plasmodium species, incl

Core-xylosylated N-glycans represent a ubiquitous class of N-glycans widely distributed across nature. Recently, core-xylosylated N-glycans have attracted considerable attention in the medical field, as they could elicit strong immune responses. Notably, antibodies targeting core-xylosylated N-glycans could kill Schistosoma in vitro, and thus process potentials for protecting host from Schistosoma infections. However, the structural complexity has posed significant challenges to access structure-defined core-xylosylated N-glycans. Here, a powerful synthetic platform enabling the efficient prep

Visceral leishmaniasis is a disease of major public health importance in Brazil, where dogs act as the main reservoir in urban transmission cycles. The immunochromatographic rapid test (RT) is currently used as a screening tool for canine visceral leishmaniasis (CVL), with the enzyme-linked immunosorbent assay (ELISA) serving as a confirmatory test. Dogs with non-reactive RT results are classified as seronegative, whereas those with reactive RT and ELISA results are classified as seropositive. Dogs with discordant results are considered uninfected according to the current diagnostic protocol.

Canine leishmaniosis, caused by Leishmania infantum, is a vector-borne disease in which early treatment monitoring remains challenging. This prospective study evaluated short-term changes in serological, inflammatory, and serum protein electrophoretic parameters during first-line therapy with meglumine antimoniate plus allopurinol. Eighteen dogs with confirmed clinical disease were enrolled at diagnosis, and fifteen dogs completing all follow-up visits (Days 0, 7, 14, 21, and 29) were included. Infection was confirmed by compatible clinical and/or laboratory abnormalities, high anti-Leishmania

Schistosomiasis ranks as the second most significant parasitic illness in terms of socioeconomic impact, after malaria. The hepatitis C virus (HCV) is a primary cause of serious liver disease, such as chronic hepatitis, cirrhosis, and hepatocellular carcinoma. Schistosomiasis increases the probability of HCV co-infection, exacerbates liver damage, and reduces the efficacy of antiviral treatments. The study aimed to compare the prognostic significance of serologically identified versus biopsy-confirmed hepatic schistosomiasis (bilharzial hepatic granuloma) in chronic HCV patients treated with i

Merozoite surface protein 8 (MSP8) is a conserved blood-stage antigen implicated in erythrocyte invasion and vaccine-induced protection, but its role in coordinating host immunity during malaria remains unclear. Here, we combined MSP8 immunization, CRISPR/Cas9-mediated deletion of Plasmodium yoelii (P. yoelii) msp8, and perturbation of chemokine-receptor signaling to define how MSP8 shapes disease outcome in the lethal murine model of malaria. Mouse antisera against Plasmodium falciparum MSP8 partially inhibited merozoite invasion of human erythrocytes in vitro, and PyMSP8 immunization conferr

Toxoplasma gondii-the causative agent of toxoplasmosis-is a zoonotic pathogen of warm-blooded hosts. Infection causes mild-to-severe symptoms, including lethargy, fever, muscle pain, abortion, ocular disease, and encephalitis. Toxoplasma affects many vertebrate species, although felids are the only known definitive hosts. Seroprevalence in wildlife is often assessed using cross-sectional data, but few studies have tracked individual-level infections through time. We present a 4-yr dataset from white-tailed deer (Odocoileus virginianus) with repeated sampling of individuals that highlights chal

BACKGROUND: Studies have linked toxoplasmosis to neuropsychiatric disorders. However, no previous reports exist on parasite tissue cyst frequency in suicide or violent death autopsies, or its variation among brain behavior-associated regions (amygdala or hippocampus).
METHODS: Amygdala, hippocampus, prefrontal, and occipital areas from forensic brain tissues were examined using real-time PCR with T. gondii RE sequence and indirect immunofluorescence antibody test (IFAT) on brain tissue using anti-BAG1 monoclonal antibodies. CD45R0 was analyzed by immunohistochemistry. Serum postmortem samples

BACKGROUND: D-dimer has been evaluated to assess the severity of disease in sepsis and COVID-19 but its usefulness in the stratification of malaria severity is unknown. We aimed to assess the D-dimer level in imported malaria as a predicting marker of Plasmodium falciparum severe malaria.
METHODS: We performed a retrospective monocentric study including all adult patients with a diagnosis of P. falciparum malaria and with a D-dimer test performed within 24 h from diagnosis. Sensitivity, specificity and accuracy of D-dimer in the diagnosis of severe malaria were estimated. The association betw

The liver flukes Fasciola hepatica and Fasciola gigantica cause fasciolosis, a zoonotic disease of major health and livestock importance. To evade host immunity, these parasites secrete immunomodulatory molecules, including fatty acid-binding proteins (FABPs), which are essential for parasite survival by mediating fatty acid uptake and transport, and by exerting anti-inflammatory effects that promote immune evasion and chronic infection. This study investigates the roles of F. hepatica FABPs' in nutrient acquisition, immune modulation, and parasite survival, assessing their potential as vaccin
