Infection with the protozoan parasite Trypanosoma cruzi can lead to Chagas disease, and diagnosis of infection mostly relies on serological testing, although no gold standard exists. The evaluation of treatment efficacy is also challenging as seronegativization takes decades to be detected. The Multi-Cruzi platform, which provides a serological profile against 15 parasite antigens, has been proposed as promising alternative for confirmatory diagnostic and monitoring of treatment response. To further evaluate this platform, we performed epitope mapping of the Multi-Cruzi antigens with peptide m

The spleen orchestrates anti-malarial immunity, with its macrophages phagocytizing Plasmodium-infected blood cells (iRBCs). While regulated cell death (RCD) has been extensively studied in various diseases, the relationship between regulatory cell death of splenic macrophages and the progression of malaria infection remains unexplored. In this study, we identify that Plasmodium infection triggers ferroptosis in splenic macrophages characterized by aberrant HMOX1 upregulation concomitant with HIF-1α downregulation. Mechanistically, the loss of HIF-1α signaling fails to restrain HMOX1 expression

The cattle tick Rhipicephalus microplus is an ectoparasite of major economic importance to livestock, particularly in tropical and subtropical regions. The lack of effective vaccines against tick infestation has hindered its control and the identification of new molecular targets for recombinant vaccine development arises as an alternative strategy. Tick saliva is known as a reservoir of potential vaccine targets, including several proteins involved in attachment, immune response modulation and feeding that targeting might affect tick infestations. Among these proteins, histamine binding prote

Helminth infections remain a major public health problem in the Lao People's Democratic Republic (Lao PDR), particularly in rural communities where foodborne trematodes and soil-transmitted helminths are co-endemic. Opisthorchis viverrini is a well-established risk factor for cholangiocarcinoma, while Strongyloides stercoralis can cause severe and potentially fatal disease. This study aimed to determine the prevalence and patterns of helminth infections in southern Lao PDR using combined parasitological and serological diagnostic approaches. A community-based cross-sectional study was conducte

BACKGROUND: Alveolar echinococcosis (AE), caused by Echinococcus multilocularis, is a lethal zoonotic disease. Population screening for early diagnosis is crucial for effective treatment, and novel diagnostic antigens are needed for diagnostic purposes.
METHODS: E. multilocularis serine protease inhibitor 1 (EmSPI-1) was cloned and expressed in Escherichia coli. Quantitative real-time PCR (qRT-PCR) and Western blotting were used to assess gene expression at different life-cycle stages of the tapeworm. The diagnostic potential of recombinant EmSPI-1 (rEmSPI-1) was evaluated by enzyme-linked im

OBJECTIVES: Sickle Cell Disease (SCD), characterized by persistently low hemoglobin (Hb) and hematocrit levels, is among the leading causes of under-five mortality in high prevalence countries. Sub-Saharan Africa bears the highest burden of both sickle cell disease (SCD) and malaria. Subclinical malaria (SCM), defined as parasitemia without overt clinical symptoms, may worsen anemia in SCD, but its clinical significance remains uncertain. We evaluated the prevalence, predictors, and clinical associations of SCM using a hierarchical diagnostic framework.
METHODS: We conducted a cross-sectional

Natural killer (NK) cells contribute to early immunity against Plasmodium falciparum by recognizing and eliminating infected red blood cells (iRBCs), a process mediated in part by the integrin LFA-1. However, the cognate parasite ligand for LFA-1 has remained unknown. Here, we identify glycophorin binding protein-130 (PfGBP-130) as a surface-expressed ligand on iRBCs that binds the I-domain of LFA-1 (LFA-1 αI). Using an LFA-1 αI-Fc fusion protein, we demonstrate stage-specific binding to iRBCs, and LC-MS/MS analysis of immunoprecipitates of αI-Fc bound to iRBC revealed PfGBP-130 as a high-conf

BACKGROUND: Visceral leishmaniasis (VL), caused by Leishmania infantum, is the most severe form of leishmaniasis, prevalent in many countries, but still with limitations in diagnosis for both human and canine hosts. Serological assays based on recombinant proteins are the most efficient diagnostic alternatives, with the rapid diagnostic test (RDT) being the most cost-effective. The previously described chimeric Q5 is a recombinant protein derived from three native L. infantum antigens, which is potentially useful for both human and canine VL diagnosis, through preliminary enzyme-linked immunos

Plasmodium vivax (Pv) forms non-replicating liver stages called hypnozoites, which activate after primary infection, and cause relapses of symptomatic blood-stage malaria. We hypothesize that hypnozoites must actively suppress schizogony to maintain a quiescent state. Differential transcriptome prospecting identifies two hypnozoite-expressed transcripts encoding putative RNA-binding proteins. We assess the functional role of the two encoded proteins in Plasmodium yoelii (Py), a rodent malaria parasite that naturally does not form hypnozoites. Strikingly, individual expression of each protein i

BACKGROUND: Canine leishmaniosis (CanL) is endemic in Italy and is an important veterinary and public health concern. Integrated preventive strategies combining vaccination and vector control are recommended in endemic areas, and field data supporting the safety and effectiveness of this concurrent use are limited.
METHODS: A longitudinal field study conducted on Favignana Island (Sicily, Italy) evaluated the concurrent use of a vaccine (LETIFEND®, Leti) and sandfly repellent collar (SCALIBOR®, MSD Animal Health). Client-owned dogs (154) were prescreened by clinical examination, serology, qua

Feline heartworm disease, caused by Dirofilaria immitis, is an emerging concern in endemic regions, although its epidemiology remains poorly defined due to diagnostic limitations. In Italy, available data on feline D. immitis infection are limited and largely based on serological investigations. The aim of the present study was to provide an epidemiological assessment of infection in cats across Sardinia, a region where canine dirofilariosis is endemic. A total of 141 cats were evaluated using a multimodal diagnostic approach including echocardiography, antigen detection, and the modified Knot

The zoonotic cestode Spirometra mansoni (Cestoda: Diphyllobothriidae), characterized by a complex life cycle involving plerocercoid larvae (spargana) in amphibians, reptiles, and mammals, as well as adult intestinal stages in carnivorous mammals, particularly felids and canids, presents significant risks to both veterinary and public health. To address diagnostic challenges in feline infections, potential serodiagnostic antigens were characterized using an integrated immunoproteomic and molecular approach. Four subcutaneous infections in nude mouse demonstrated plerocercoid larval migration fr

INTRODUCTION: Chagas disease, caused by the protozoan parasite Trypanosoma cruzi, remains a major public health problem, affecting approximately seven million people worldwide. Although historically restricted to endemic regions, migration has led to its global distribution, increasing the relevance of congenital transmission in non-endemic areas. If not timely diagnosed and treated, infection can progress to chronic Chagas cardiomyopathy in nearly 30 % of patients, generating substantial social and healthcare burdens. Diagnosis requires concordant results from two or three serological assays

BACKGROUND: As Southeast Asian countries advance toward malaria elimination, challenges such as residual transmission reservoirs and zoonotic spill-over require high-resolution micro-surveillance. While conventional diagnostics often miss hidden transmission foci in low transmission settings, serological assays provide an essential supplement by measuring both historical and recent exposure. This scoping review assesses the implementation of antibody surveillance across Southeast Asian countries and highlights its potential in the concerted effort towards regional malaria elimination.

BACKGROUND: Gastrointestinal (GI) parasitic infection remains one of the most prevalent and economically significant health challenges in equine populations. The infective stages of GI parasites can activate systemic inflammatory responses and disrupt lipid metabolism leading to exacerbation of parasitic infection. Moreover, age-related low-grade chronic inflammation may further contribute to the persistent infection which impacts the health status of individual horses.
AIMS/OBJECTIVE: This study examined serum levels of the inflammatory marker C-reactive protein (CRP) and lipid profile param

This study aimed to investigate the role of interleukin (IL)-25 in the pathogenesis of angiostrongylosis. We used ICR mice as non-permissive hosts, divided them into groups and experimentally infected them with infective larvae of the Angiostrongylus cantonensis. Two groups of infected mice were injected intraperitoneally with either mouse IL-25 or an anti-IL-25 monoclonal antibody (mAb) at 3 days post-infection (dpi), followed by booster injections at the same dose every 5 days. Serum samples and brain tissues were collected weekly from each group for immunological and pathological examinatio

Toxoplasmosis, a globally prevalent zoonosis caused by Toxoplasma gondii (T. gondii), poses major threats to both human and animal health, leading to reproductive losses in livestock and severe disease in immunocompromised individuals. Although enzyme-linked immunosorbent assay (ELISA) and PCR are widely used for diagnosis and surveillance, they may be limited by turnaround time, laboratory instrumentation, and, in the case of serological assays, the need for species-specific reagents. To enable rapid, equipment-minimal detection applicable to multiple host species, we developed a point-of-car

Marine seafood-derived anisakid nematodes (e.g., Anisakis spp. and Phocanema spp. [formerly Pseudoterranova spp.]) may cause gastrointestinal and allergic manifestations. Here, we report an unusual case of persistent, asymptomatic peripheral eosinophilia following ingestion of raw or lightly marinated seafood, raising the possibility of subclinical antigenic stimulation. The patient remained entirely asymptomatic with normal total IgE levels, and parasite serology (including Angiostrongylus cantonensis IgG) was negative, yet developed marked eosinophilia. The patient had a childhood history of
