Genome-wide analyses identify 60 loci associated with liver triglyceride content.

We performed a large-scale genome-wide association study meta-analysis of magnetic resonance imaging-derived liver triglyceride content across 71,488 European-ancestry participants from the UK Biobank, approximately doubling the sample size of previous genome-wide association studies1. We identified 60 independent loci, including 31 not previously reported, and estimated the single nucleotide polymorphism-based heritability of liver triglyceride content at 25%. Integrative fine-mapping and gene-prioritization analyses nominated candidate effector genes for 48 loci. Of the 31 newly identified v

Dramatic regression of mass-forming gastric IgG4-related disease with inebilizumab: a case report.

BACKGROUND: Gastric immunoglobulin G4-related disease (IgG4-RD) is exceedingly rare, with fewer than 50 cases reported worldwide. Its mass-forming presentation frequently mimics malignancy, leading to high rates of unnecessary surgical resection. Inebilizumab, a CD19-directed monoclonal antibody and the first FDA-approved therapy for IgG4-RD, has not previously been described in gastric IgG4-RD. We report the first case of mass-forming gastric IgG4-RD treated with inebilizumab, achieving clinical, endoscopic, and histologic remission without surgical intervention. CASE PRESENTATION: A 52-year

Rituximab and corticosteroids for M-protein-negative PGNMID: case report.

BACKGROUND: We report a case of a 69-year-old patient negative for serum M-protein who was diagnosed with proliferative glomerulonephritis with monoclonal immunoglobulin deposits (PGNMID) via renal biopsy. CASE PRESENTATION: A 69-year-old male presented with hematuria, edema, and rapidly progressive renal failure. Serum M-protein was negative, but renal biopsy confirmed IgG3-κ PGNMID with intense C3 deposition and an MPGN-like pattern. Rituximab was initiated, but due to a pulmonary infection, only one of the planned rituximab doses was administered. After infection resolution, the patient's

Unraveling the dual role of fibroblast growth factor 5 (FGF5) in cardiorenal diseases through multi-omics causal inference.

BACKGROUND: As populations age worldwide, multimorbidity has become an increasingly prominent public health challenge. Among age-associated conditions, cardiovascular diseases (CVDs) are among the most prevalent and life-threatening, and they frequently coexist with other chronic disorders, particularly chronic kidney disease (CKD). Pleiotropic molecules may provide an important entry point for elucidating comorbidity mechanisms and identifying therapeutic opportunities. However, the clinical translational value of different pleiotropic candidates exhibits substantial heterogeneity, and their

Proteomic signatures characterized by inflammatory signaling and metabolic processes have divergent associations with 30-year incidence of coronary artery disease in type 1 diabetes: the Pittsburgh Epidemiology of Diabetes Complications cohort.

BACKGROUND: While inflammation is hypothesized to be a possible driver of the excess coronary artery disease (CAD) observed in type 1 diabetes, prior biomarker studies have focused on candidate proteins, overlooking the complex systemic processes underlying atherosclerosis. We thus applied a high-throughput proteomics approach to identify proteins prospectively associated with CAD in type 1 diabetes. We further hypothesized inflammatory proteins would partially mediate the effect of glycemic exposure on CAD risk. METHODS: We measured 184 circulating proteins (Olink CVD II and III panels) in e

Circulating 16 micronutrients concentration and risk of abdominal aortic aneurysm: a two-sample mediation Mendelian randomization analysis.

BACKGROUND: Abdominal aortic aneurysm (AAA) is a severe cardiovascular disease involving various genetic and metabolic factors. Micronutrients play critical roles in cardiovascular health, yet their specific impacts on AAA remain unclear. This study aims to explore the effects of micronutrients calcium and zinc on AAA risk mediated by plasma proteins using the two-sample Mendelian Randomization (MR) approach. METHODS: Utilizing large-scale GWAS data from the UK Biobank; FinnGen databases and Decode database, we assessed the causal relationships between 16 micronutrients concentration and AAA

An interferonopathy associated with a monoallelic variant in TRIM28.

BACKGROUND: In this work, we describe the clinical and functional changes in a family with a novel type I interferonopathy phenotype which we suggest could be associated with a TRIM28 variant, presenting with chilblains, skin ulceration, lipodystrophy and calcinosis. TRIM28 has multiple roles in regulating human health and disease, which include DNA repair and anti-viral response. The latter includes negative regulation of IRF7, one of the master regulators of the interferon response. METHODS: Whole exome sequencing and targeted panel sequencing was performed on DNA extracted from peripheral

Cerebral blood flow as a biomarker of resilience in cognitively unimpaired APOE ɛ4 carriers.

INTRODUCTION: It remains unclear whether medial temporal lobe (MTL) hyperperfusion in cognitively unimpaired apolipoprotein E (APOE) ɛ4 carriers reflects resilience or dysfunction. We examined whether cortical MTL cerebral blood flow (CBF) relates to cognition differently in APOE ɛ4 carriers and noncarriers with and without AD pathophysiological changes. METHODS: One hundred forty-eight cognitively unimpaired older adults underwent pseudo-continuous arterial spin labeling MRI to quantify MTL CBF and assessment of memory and executive function. In a subset (n = 126), AD pathology was indexed u

Preliminary study on expression and functions of Toxoplasma gondii SRS67 and SRS20A proteins.

BACKGROUND: Toxoplasma gondii surface proteins play critical roles in parasite invasion, proliferation, and host immune modulation, and represent promising targets for anti-toxoplasmosis intervention. SRS67 and SRS20A are poorly characterized surface proteins of T. gondii, and their biological functions and host regulatory effects remain largely unclear. METHODS: In the present study, we prepared recombinant SRS67 and SRS20A proteins and generated corresponding polyclonal antibodies to explore their functional characteristics and protective potentials. RESULTS: Immunofluorescence analysis lo

Blood and Cervical Immune Biomarkers Associated With HIV Susceptibility Are Compromised Rapidly After Sexual Trauma.

PROBLEM: Sexual violence compromises women's health and increases susceptibility to pathogens. Yet immunobiological consequences of acute sexual trauma remain incompletely understood. METHODS: We conducted a cross-sectional study among women without HIV, comparing women exposed to forced vaginal penetration in the past 4 days (Acute Cases, n = 10), within the past 12 weeks (Cases, n = 10) and never exposed (Controls, n = 12). Sixteen immune biomarkers were measured by ELISA in plasma and cervical swabs. Blood CD4 and CD8 T cells were quantified by flow cytometry. Group differences were analyz

Bone-Targeting Antibodies for Skeletal Precision Therapy.

Therapeutic antibodies have transformed the treatment of cancer, inflammation, infection, and disorders of bone biology by recognizing disease-associated molecular targets with high specificity. However, achieving therapeutically effective antibody concentrations at the site of disease remains a major challenge, as systemic administration does not inherently result in efficient tissue-specific delivery. Bone is a particularly demanding destination because mineralized matrix, limited vascular access, marrow complexity, and active remodeling can restrict local antibody exposure. Recent advances

Metabolic reprogramming of NK cells in chronic brucellar arthritis: role of the HIF-1α/mTOR axis.

BACKGROUND: Brucellar arthritis is the most common complication of brucellosis, and its chronic progression is closely associated with immune cell dysfunction. Natural killer (NK) cells play a central role in anti-infective immunity; however, their functional status and regulatory mechanisms in brucellar arthritis remain poorly understood. METHODS: Peripheral blood and synovial tissue samples were collected from healthy controls, as well as from patients with acute-phase and chronic-phase brucellar arthritis. Single-cell RNA sequencing (scRNA-seq), flow cytometry, quantitative real-time PCR (

Brain imaging in dystrophinopathies.

Duchenne and Becker muscular dystrophies (DMD, BMD) are caused by genetic variants in the DMD gene coding dystrophin proteins. In addition to the impairment of muscular function, these patients also present frequent cognitive and neuropsychiatric disorders due to impaired expression of several brain dystrophins: Dp427, Dp140 and Dp71. Cerebral imaging has shed new light on the impact of dystrophinopathies on brain structure, function, and connectivity. We review the brain imaging results obtained in patients and divers animal models, showing presence of mild cortical atrophy, slight modificati

Optimization of 68Ga-lactosylated human serum albumin (LSA) for PET-based quantitative evaluation of hepatic functional reserve in liver fibrosis models.

Accurate preoperative assessment of hepatic functional reserve (HFR) and future remnant liver function is essential for reducing postoperative morbidity and mortality after liver surgery, yet current clinical methods remain indirect and provide limited spatial and quantitative information. Here, we developed an asialoglycoprotein receptor (ASGPR)-targeted tracer for positron emission tomography (PET), [68Ga]Ga-NOTA-lactosylated human serum albumin ([68Ga]Ga-NOTA-LSA), and systematically optimized the degree of lactosylation for hepatic imaging. Comparative evaluation of lactosylated human seru

Serum evaluation of synovial fluid biomarkers in osteoarthritis.

ObjectiveIn our previous study, we identified forty-seven differentially expressed proteins in the synovial fluid of osteoarthritis (OA) patients. Given that serum testing is less invasive and remains the first-line screening tool, this study aimed to explore whether these synovial fluid-derived protein candidates could be detected in serum and whether they might serve as translatable biomarkers for OA.MethodsUsing antibody array technology, we analyzed the expression levels of these forty-seven proteins in serum samples from forty OA patients and forty healthy controls.ResultsCompared to heal

Nuclear SUN2 coordinates endothelial cell-matrix interactions to regulate blood vessel homeostasis and barrier function.

Vascular endothelial cells respond to environmental forces to remodel vessels during development and to achieve homeostasis, and mis-regulated responses lead to vascular dysfunction and disease. The nucleus participates in transmitting signals to cell-matrix junctions via the Linker of Nucleoskeleton and Cytoskeleton (LINC) complex that resides in the nuclear envelope, but how this information is regulated and relayed is incompletely understood. We found that the LINC complex protein SUN2 is required for proper endothelial cell-matrix interactions that occur far from the nucleus and affect ang

Beyond Hemodynamics: Uremic Toxins, Post-Translational Modifications, and Inflammatory-Metabolic Remodeling in Uremic Cardiomyopathy.

Uremic cardiomyopathy has traditionally been attributed to pressure overload, volume overload, hypertension, and neurohormonal activation in chronic kidney disease. However, growing experimental and clinical evidence indicates that nonhemodynamic mechanisms also contribute to myocardial remodeling in uremia. These mechanisms include retention of uremic toxins and protein-bound solutes, mediators of mineral metabolism, gut-derived metabolites, inflammatory signaling, mitochondrial dysfunction, and post-translational protein modifications. In this review, we discuss uremic cardiomyopathy as a sy
