Metabolic Syndrome and Incidence of Parkinson Disease: A Community-Based Longitudinal Study and Meta-Analysis.

BACKGROUND AND OBJECTIVES: The association between metabolic syndrome (MetS) and incident Parkinson disease (PD) remains equivocal. We aimed to investigate the association of MetS and its components with the risk of PD and to explore the role of genetic background in the MetS-PD association. METHODS: This prospective cohort study included PD-free adults aged 37-73 years from the UK Biobank. MetS was defined as presence of 3 or more of the following: elevated waist circumference (≥102 cm for men; ≥88 cm for women), hypertension (systolic blood pressure ≥130 mm Hg, diastolic blood pressure ≥85

[Efficacy and safety of bempedoic acid in the prevention of cardiovascular events: results of the CLEAR Outcomes study and subgroup analyses].

Bempedoic acid is a lipid-lowering drug recently introduced into clinical practice. The CLEAR Outcomes randomized controlled clinical trial has confirmed the efficacy of bempedoic acid in reducing low-density lipoprotein cholesterol (LDL-C) levels and has shown that this reduction translates into a significant decrease in major adverse cardiovascular events (MACE). The clinical benefits were consistent with the benefits obtained with other lipid-lowering drugs that have the same efficacy in reducing LDL-C levels. Furthermore, the CLEAR Outcomes trial confirmed the safety of bempedoic acid. Thi

Phenolic acids from Anisopus mannii modulates phosphofructokinase 1 to improve glycemic control in patients with type 2 diabetes: A double-blind, randomized, clinical trial.

Phenolic acid-rich fraction from Anisopus mannii (PhAM) contains abundance of ferulic acid, gallic acid, protocatechuic acid, and syringic acid. Among other glycolytic enzymes, in vitro, PhAM counteracted the binding of sodium orthovanadate to phosphofructokinase 1 (PFK-1), improving its activities. In a rat model of diet-induced diabetes, PhAM monotherapy reduced HbA1c by an average of 0.63 % and fasting plasma glucose by 25 mg/dl. This herb rescued β-cells from streptozotocin-mediated destruction, thereby improving glycemic control. Supported by the preclinical trial, eighty-five patients wi

Perioperative Risks and Outcomes in Asian American Patients with Type 2 Diabetes Mellitus and/or Metabolic Syndrome: a Systematic Scoping Review.

Asian Americans (AA) have an increased risk of developing type 2 diabetes mellitus (T2DM) and metabolic syndrome (MetS) compared to non-Hispanic White Americans, yet over half of AA patients with T2DM are underdiagnosed or untreated. Surgical stress, known to exacerbate hyperglycemia in T2DM, is also associated with increased morbidity and mortality. Thus, AA patients may be at elevated risk of experiencing poor outcomes following surgery. This review aims to summarize the available literature on the perioperative (defined as before, during, and after surgery) risk and outcomes of T2DM in AA s

Shedding light on eating disorders in adolescents with type 1 diabetes: insights and implications.

Eating disorders (EDs) are complex medical conditions that pose a considerable health burden for individuals with type 1 diabetes mellitus (T1DM). EDs in individuals with T1DM are linked to poor metabolic control, which heightens the risk of diabetes complications. Consequently, regular screening for EDs is essential. This study investigates the prevalence of EDs in adolescents with T1DM, investigating the associations with diabetes duration, pubertal stage, glycemic control, and diabetes-related complications. In this cross-sectional study, 350 adolescents (155 males, 195 females) with T1DM,

Emulating Six Cardiovascular Outcome Trials of Antidiabetic Drugs in Individuals with Type 2 Diabetes: Analyses Based on Real-World Databases from Korea and Taiwan.

With the increasing volume of clinical evidence derived from large-scale Asian real-world data (RWD) and the growing interest in its use in regulatory decision-making, it is important to conduct benchmarking exercises that evaluate whether studies using Asian RWD can generate similar conclusions as randomized controlled trials (RCTs). We aimed to assess whether observational studies based on Korea and Taiwan RWD can yield comparable results with trials by emulating six cardiovascular outcome trials (CVOTs) of antidiabetic drugs in individuals with type 2 diabetes (T2D). We emulated six CVOTs u

Glycaemic control in labour with diabetes: GILD, a scoping study.

BACKGROUND: Diabetes in pregnancy is common, affecting 5-10% of pregnant women. Poor glycaemic control in labour is associated with neonatal hypoglycaemia and other adverse outcomes for mother and baby, but tight glucose control is burdensome, intrusive and may not always be necessary. The ideal intrapartum glucose target level is unknown, traditionally 'tight' control (target 4-7 mmol/l) has been recommended; however, this increases the risk of maternal hypoglycaemia. OBJECTIVE: To determine the feasibility of a randomised clinical trial to compare clinical and cost-effectiveness of permissi

Testing a New Diabetes Adaptive Weight management Network (NewDAWN): A protocol for a randomised controlled trial.

BACKGROUND: The NHS Path to Remission (PtR) offers a total diet replacement (TDR) programme to help people newly-diagnosed with type 2 diabetes (T2D) lose weight. It is very effective for people who participate, but most eligible people do not take part. AIM: To assess whether offering a range of weight loss programmes can increase uptake of, and persistence with, weight loss and lead to a higher proportion of the population achieving remission of T2D compared with offering PtR only. METHOD: 1788 people diagnosed with T2D in the last six years, who are willing to try to lose weight to achiev
