Enhancing Mental Wellness in Kenyan Adolescents and Young People: Protocol for a Platform Development and Usability Study.

BACKGROUND: Mental health disorders are a leading cause of disability among adolescents and young people in Kenya, yet access to appropriate and timely support remains severely limited due to shortages of trained professionals, stigma, financial barriers, and geographical inequities. Digital mental health interventions, particularly those leveraging AI, present a promising opportunity to expand access to culturally relevant, low-stigma mental wellness support in low- and middle-income country (LMIC) settings. However, evidence on the feasibility, usability, and acceptability of AI-powered ment

Exercise-induced epigenetic remodeling in COPD skeletal muscle dysfunction: From sedentary behavior to molecular rehabilitation.

Chronic obstructive pulmonary disease (COPD) is increasingly recognized as a multisystem disorder in which extrapulmonary manifestations substantially contribute to disease burden. Among these, skeletal muscle dysfunction (SMD) represents a major determinant of exercise intolerance, reduced physical activity, impaired quality of life, and mortality. Growing evidence suggests that physical inactivity is not merely a consequence of advanced COPD but may act as an early and potentially modifiable driver of skeletal muscle deterioration, being detectable even in patients with mild disease and pote

Factors Associated with Functional Disability and Walking Capacity in Adults with Chronic Low Back Pain: A Cross-Sectional Study.

Background/Objectives: Disability in chronic low back pain (CLBP) is multifactorial, yet the independent associations of pain, fear-avoidance beliefs, and comorbidity burden with self-reported disability and performance-based walking capacity remain unclear, particularly in Saudi Arabia. This study examined factors independently associated with functional disability and walking capacity in adults with CLBP and explored whether disability was independently associated with walking capacity after accounting for demographic and clinical factors. Methods: This cross-sectional study included 339 adu

Human iPSC-Derived Brain Cells, Organoids, and Organoids-on-Chips: Advancing Neurological Disease Modeling to Improve Clinical Outcome.

Neurological disorders are the leading cause of overall disease and disability worldwide, affecting more than 3.4 billion people globally and over 180 million Americans. Yet there are no curative treatments available for any of the cardinal neurological diseases, such as Alzheimer's disease (AD), Parkinson's disease (PD), amyotrophic lateral sclerosis-frontotemporal dementia (ALS-FTD) and Huntington's disease (HD). Many compounds that demonstrate excellent activity in preclinical models fail in clinical trials. The most compelling reason for this failure is attributed to inherent limitations o

Gluten-Free and Gluten-Restricted Diets in Multiple Sclerosis: A Systematic Review of Disease Activity and Patient-Reported Outcomes.

Background: Dietary modification is widely used by people with multiple sclerosis (MS), and gluten avoidance is a common self-directed strategy. Biological plausibility has been proposed through gut-barrier and immune mechanisms, but whether gluten restriction improves clinical outcomes or objective disease activity remains uncertain. Objective: To systematically identify, appraise, and synthesise evidence on gluten-free and gluten-restricted diets in MS, with particular attention to whether observed effects can be attributed specifically to gluten. Methods: The protocol was registered with PR

Evaluation of a novel body temperature sensor (CORE) in dogs under general anesthesia following premedication with dexmedetomidine or acepromazine: A pilot study.

Core body temperature monitoring in anesthetized dogs is a vital part of physiological evaluation. Depending on the procedure and patient positioning, consistent core temperature measurements can be difficult to obtain. The objective of this study was to evaluate the reliability and clinical validity of a wireless, non-invasive, superficial heat flux-based thermometric device (CORE, greenTEG AG, Rümlang, Switzerland). Three healthy beagle dogs were premedicated with either dexmedetomidine or acepromazine prior to induction with intravenous propofol and isoflurane maintenance. They were monitor

Digital health and artificial intelligence in paediatric respiratory medicine: what clinicians need to know and what is coming next.

Digital health and artificial intelligence (AI) are increasingly part of paediatric respiratory clinics, from home-based symptom monitoring and remote consultations to AI-supported test interpretation and large language model (LLM)-supported communication. These tools promise to capture real-world disease and treatment patterns, support self-management, and enhance clinical decisions. Yet for most of these tools, the paediatric evidence base remains limited and fragmented, and the clinical benefit observed in practice is often modest. What has emerged is that effectiveness depends less on the

Role of vitellogenins on the mitochondrial unfolded protein response.

Sex-specific differences influence aging and disease onset. Using Caenorhabditis elegans , we previously found that hermaphroditic nematodes activate a robust mitochondrial unfolded protein response (UPR mt ) in the intestines, while female nematodes do not. We hypothesized that the accumulation of vitellogenins in females was suppressing the UPR mt . Loss of vitellogenins via RNAi enhanced the UPR mt in hermaphrodites but had no impact on the UPR mt of females unless a combination of vitellogenins was simultaneously knocked down. Furthermore, combinatorial loss of vitellogenins did not restor

Hypoxia-inducible factor 2α does not regulate cortical brain development.

Dynamic changes in physiological oxygen levels are crucial for proper cortical brain development. In this context, oxygen is important not only for its role in energy metabolism but also as a signaling molecule. While much is already known about its most prominent signaling factor hypoxia-inducible factor (HIF)-1α, little is known about its functionally similar isoform HIF-2α during cortical development. Therefore, we utilized HIF-2α conditional knock-out mice (Emx1 and Nestin driven), which were kept under maternal hypoxia (10% pO2), physioxia (21% pO2), or hyperoxia (75% pO2) starting at E14

Leucine-rich repeat kinase 2 is involved in the microtubule-mediated regulation of intraocular pressure.

Leucine-rich repeat kinase 2 (LRRK2) is known as the most common gene responsible for familial Parkinson disease (PD). This protein plays a key role in the neurodegenerative pathways associated with PD. LRRK2 also plays critical roles in actin dynamics and microtubule cytoskeletal assembly in the cells. In addition to PD, actin and microtubule cytoskeletons are associated with the onset and development of glaucoma by regulating the intraocular pressure (IOP) in humans and animals. However, the role of LRRK2 in regulating IOP remains unclear. Hence, this study investigated the contribution of L
