AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-scale association studies and other computational methods, including network-based analyses of multi-omics data. Here, as an example application, we describe an

Overinvestment in ultra‑rare APOE variants and highly speculative apoE-inhibitor docking risks undermining translational progress in sporadic Alzheimer's disease.

Misclassification of ultra-rare and isoform-defining APOE variants, together with unsupported attempts to dock small-molecule 'inhibitors' to apolipoprotein E (apoE), continues to appear in the structural bioinformatics literature. Such inconsistencies stem from a broader misunderstanding of apoE as a highly flexible, two-domain protein with extensive intrinsically disordered regions (IDRs), a context essential for interpreting sequence variation. A recurring issue is the treatment of the pathogenic ε4 and protective ε2 substitutions as independent rare variants rather than as the canonical AP

Differential atrophy along the longitudinal axis of the hippocampus in Alzheimer's disease and suspected non-Alzheimer's disease pathophysiology (SNAP).

PURPOSE: Cerebrospinal fluid (CSF) biomarkers are increasingly used to support Alzheimer's disease (AD) diagnosis: Alzheimer's Disease continuum (ADc) is defined by abnormal amyloid-beta (Aβ), while Suspected Non-AD Pathophysiology (SNAP) refers to normal Aβ with elevated tau. Given the differential properties along the hippocampal longitudinal axis, this study aimed to evaluate anterior-posterior subregional hippocampal volume differences among ADc, SNAP, and biomarker-negative participants, and their associations with CSF biomarkers and clinical presentation. METHODS: Data from 1242 partici

Clonal hematopoiesis in Alzheimer's brain: Protective, pathogenic, and context-dependent?

Clonal hematopoiesis is emerging as a surprising modifier of Alzheimer's disease. Recent findings suggest that mutant myeloid cells may enter or expand within the brain, adopting either inflammatory or reparative states. We propose that their effects depend on the mutation, timing, clone size, brain niche, and disease stage.

Zika virus infection induces a persistent accumulation of Alzheimer's disease-like Tau phosphorylation in adult immunocompetent mice in association with memory and social behavior impairments.

Clinical and epidemiological data support the link between viral encephalitis and neurodegeneration but its causal mechanism remains mostly unknown. Zika virus (ZIKV) is an emerging, neurotropic flavivirus susceptible to induce cognitive impairments in infected adults. In this work we have analyzed the capacity of ZIKV to induce the accumulation of pathological phosphorylated Tau protein (pTau), a major driver of neurodegenerative disorders such as Alzheimer's disease, throughout the infection of adult immunocompetent mice. The capacity of ZIKV to induce pTau in vivo, was analyzed in the long

Network toxicology screening of plastic-associated chemicals reveals candidates with structural mimicry to Alzheimer's disease-relevant molecules and predicted neurotoxicity.

Alzheimer's disease (AD) pathogenesis involves environmental factors, with plastic-associated chemicals emerging as potential neurotoxicants. This study employed a computational toxicology framework to screen plastic-associated chemicals for potential interference with AD-related pathways. From a compiled library of 12,751 plastic-associated chemicals, 2,315 compounds with high blood-brain barrier (BBB) permeability were identified. Using Morgan fingerprints and Tanimoto similarity (threshold ≥ 0.6), we pinpointed chemicals structurally analogous to known AD-active molecules, including 3,5-Dim

Therapeutic Gene Editing of APOE4 in Sporadic Alzheimer's Disease via Prime Editor 7.

The apolipoprotein E4 (APOE4) allele is the strongest genetic risk factor for sporadic Alzheimer's disease (AD), driving Aβ accumulation, tau pathology, and synaptic dysfunction. Allele-specific correction of APOE4 represents a promising therapeutic strategy to mitigate disease progression. In this study, we developed an APOE4-specific prime editing strategy based on an optimized APOE4-targeting pegRNA, enabling precise and efficient conversion of the APOE4 allele to the lower-risk APOE3 variant. We found that PE7 targeting the APOE4 allele achieved robust and specific editing without detectab

Reducing overconfident errors in clinical prediction models.

Machine Learning (ML) models are increasingly being used in clinical workflows. Evaluation of these models tends to focus on global performance metrics, which can obscure error patterns and lead to bias in clinical decision making. Here, we propose Proximal Error-Based Confidence Adjustment (PECA), a framework designed explicitly to improve the safety of ML predictions by reducing a model's confidence in regions of the feature space associated with historical prediction errors. Rather than optimizing global metrics alone, PECA targets confident misclassifications by attenuating prediction conf

The missing improvers: Tau pathology, neuroplasticity, and the case for tau-informed patient selection before amyloid immunotherapy.

Amyloid immunotherapy with lecanemab and donanemab has been approved on the basis of statistically significant slowing of cognitive and functional decline in early Alzheimer's disease (1,2). Yet clinicians treating individual patients face the everyday challenge of estimating whether a given patient is declining at the rate that would be expected for them, and whether their trajectory reflects a response to treatment. I argue that this difficulty arises in large part because the field cannot yet routinely stratify patients by tau pathology before treatment, even though tau burden is among the

Preliminary Development of a Hybrid Aptamer and Antibody Fluorescent Lateral Flow Assay for Ultrasensitive Detection of Tau441 Protein in Serum: Toward Point-of-Care Diagnostics for Alzheimer's Disease.

A hybrid DNA aptamer and antibody-Europium nanoparticle (Eu NP) fluorescent sandwich lateral flow (LF) assay for Tau441 is described which works well in human serum with an apparent visible detection limit for Tau 441 of ~ 10 pg/ml or 1 pg of Tau protein. The same LF assay appears to detect phosphorylated threonine 217 Tau 441 (phospho Thr-217 Tau) consistently to an even lower level of 1 pg/ml or 100 fg of total phosphorylated Tau protein in human serum. The aptamer was designed in silico on the Xelari computational platform to target an epitope centered around alanine 390 distal from the thr

The influence of mutant genes APP and PSEN1 on hippocampal asymmetry in 5xFAD mice, transgenic model of Alzheimer disease.

Alzheimer's disease (AD) is an age-related neurodegenerative pathology and one of the primary causes of dementia associated with memory and cognitive impairment. The dentate gyrus (DG) of the hippocampus plays a crucial role in learning and memory encoding, and it undergoes significant changes in AD. To study the underlying mechanisms of AD and to test new therapeutic agents, mouse models of this pathology are developed. One of the most popular and relevant models for AD research is the 5xFAD transgenic mouse strain carrying mutant human genes, specifically: amyloid beta precursor protein (APP

Stability of plasma pTau217/Aβ42 ratio with delayed blood processing.

INTRODUCTION: Blood-based biomarkers may improve access to Alzheimer's disease diagnosis, but use may be limited in clinics without on-site processing capability. We evaluated the stability of the plasma phosphorylated tau 217 (pTau217)/amyloid beta 42 (Aβ42) ratio after delayed whole-blood processing. METHODS: We compared blood processed within 2 h or held at 4°C for approximately 24 or 48 h before centrifugation and analysis using the Fujirebio Lumipulse G1200. Stability was assessed using ordinary least squares regression, error index, Bland-Altman analysis, and Deming regression. RESULTS

More than just accuracy: A pathway-level framework for implementing blood and digital biomarkers for Alzheimer's disease care.

Blood biomarkers are transforming Alzheimer's disease diagnostics by enabling a scalable, minimally invasive assessment of pathology. Food and Drug Administration clearances and European market certifications of different plasma tests mark a shift toward biological diagnosis in routine care. However, clinical implementation is not guided by diagnostic accuracy alone but will depend on improved patient-centered and pathway-level outcomes, including diagnostic confidence, the appropriate use of CSF and PET assessments, time to diagnosis, amyloid-targeting treatment eligibility assessment, and tr

Effectiveness of exercise training on hippocampal mitochondrial regulation in Alzheimer's disease: A mechanistic systematic review.

Alzheimer's disease (AD) is associated with hippocampal mitochondrial dysfunction, including impaired mitophagy, biogenesis, dynamics, bioenergetics, and redox homeostasis, which may contribute to amyloid-β (Aβ) accumulation, tau pathology, and cognitive decline. This systematic review examined whether exercise training is associated with changes in hippocampal mitochondrial regulation and cognitive function in transgenic AD rodent models. Following PRISMA 2020 guidelines, PubMed, EMBASE, and Web of Science were searched up to July 2, 2026. Twenty-one studies met inclusion criteria. Exercise m

Statistical modeling and inference for Jack's Alzheimer's disease biomarker cascade with practical and computational considerations.

Alzheimer's Disease (AD) research increasingly recognizes the importance of characterizing the joint and relative dynamics of biomarker trajectories during the preclinical phase. Jack's model stands as the most well-known theoretical framework of these processes. While it offers conceptual guidance on modeling, it is not a directly implementable model for observed data. Key challenges include: (i) biomarker measurements are often noisy, depend on individual characteristics, and often exhibit complex temporal and cross-sectional correlations; (ii) data are sampled sparsely and may cover only a

Single cell RNA sequencing and emerging transcriptomic technologies in Alzheimer's disease cerebral organoids: a mini review.

Alzheimer's disease (AD), the most common cause of dementia worldwide, presents a formidable challenge due to its complex, multifactorial etiology, and limited translational success of existing models. Recent advances in stem cell biology have enabled the generation of three-dimensional human cerebral organoids which exhibit multicellular complexity and self-organisation, recapitulating key aspects of fetal brain development in vitro. This mini review highlights the landscape of AD modelling in human cerebral organoids. While studies using single cell RNA sequencing (scRNA-seq) form the core o
