Anterior temporal lobe, word comprehension, and physiology of atrophy in semantic primary progressive aphasia.

Peak focal atrophy in the anterior temporal lobe (ATL) highlights the critical role of this area for word comprehension in semantic variant primary progressive aphasia (svPPA). However, the assumption that peak atrophy sites are specific markers of dysfunctional brain sites, and therefore reliable variables for clinicopathologic correlations, has not been rigorously tested. Using structural MRI and FDG-PET, we assessed atrophy and hypometabolism in 32 individuals with PPA (11 svPPA) and 10 healthy controls. Word comprehension was measured using the Peabody Picture Vocabulary Test. Voxel-based

Single-nucleus and spatial transcriptomic profiling of human temporal cortex and white matter reveals key associations with AD pathology.

Alzheimer's disease, the leading cause of dementia in the elderly, is a neurodegenerative disorder that has been studied to uncover therapeutic pathways through its molecular and cellular hallmarks. Here, we present a comprehensive investigation of cellular heterogeneity from the temporal cortex region of 40 individuals, comprising healthy donors and individuals with differing AD pathology. Using single-nucleus transcriptomic analysis of 430,271 nuclei from both gray and white matter of these individuals, we identified cell type-specific subclusters in both neuronal and glial cell types with v

Regional brain aging patterns reveal disease-specific pathways of neurodegeneration.

The heterogeneity of brain aging is a hallmark of neurological and psychiatric disorders, yet machine-learning tools used to characterize this process, including the 'brain age' paradigm, have largely relied on global metrics that lack the specificity to map these complex patterns. Here, we introduce BrainAgeMap, an interpretable deep learning framework that generates fine-grained, voxel-wise maps of brain-predicted age difference (brain-PAD) from T1-weighted magnetic resonance imaging scans. We provide converging lines of evidence for the framework's clinical, prognostic, and neurobiological

Anatomical Progression of Neuropathology in FTLD-TDP Type C and Linkage to Annexin A11.

OBJECTIVE: Frontotemporal lobar degenerations (FTLD)-TDP type C (TDP-C) is distinguished from other FTLD-TDP subtypes by 3 unique features: (1) invariable onset in the anterior temporal lobe (ATL), (2) phosphorylated TDP-43 (pTDP) neurites in cortex, and (3) colocalization of all pTDP deposits with annexin A11 (ANXA11). This article provides a whole-brain anatomical account of TDP-C disease progression in relation to clinical, imaging, and neuropathologic patterns. METHODS: Thirty-two cases with TDP-C were studied, including neuropathologic findings and longitudinal magnetic resonance imaging

Hippocampal Development and Epilepsy: Insights from Organoid Models.

The hippocampus is a crucial component of the human brain. It is located on the medial side of the temporal lobe and is connected to the limbic system, influencing memory and cognitive function. The critical functions of the hippocampus have a profound impact on an individual's overall ability to maintain daily life functioning. In adults, hippocampal damage impairs cognitive functions, including memory, learning, and emotional regulation. It is associated with conditions such as memory impairment, Alzheimer's disease, various forms of dementia, depression, and stress-related disorders. Damage

Chemogenetic manipulation of learning-tagged neurons is sufficient to rescue progressive memory deficits in a mouse model of Alzheimer's disease.

INTRODUCTION: Alzheimer's disease (AD) primarily affects episodic memory, which relies on the medial temporal lobe, including the hippocampus and lateral entorhinal cortex (LEC). However, it remains unclear whether memory deficits in AD reflect disrupted encoding of new experiences or impaired retrieval of previously stored information. METHODS: APPJ20 transgenic mice were used to investigate memory deficits. Neuronal populations activated during the learning phase of associative and non-associative tasks were tagged to express the excitatory chemogenetic receptor hM3Dq. Chemogenetic activati

Excitation-inhibition imbalance as a common thread linking early Alzheimer's disease with temporal lobe epilepsy.

Growing evidence suggests that network hyperexcitability is a pivotal yet under-recognized pathology linking early Alzheimer's disease (AD) with mesial Temporal Lobe Epilepsy (mTLE). This narrative review synthesises pre-clinical and clinical data showing how disruption of excitation-inhibition balance, driven chiefly by the loss or dysfunction of parvalbumin- and somatostatin-positive GABAergic interneurons (INs), emerges early in AD and fosters subclinical epileptiform activity that hastens cognitive decline. We integrate findings that degeneration of Ventral Tegmental Area dopaminergic proj

Immune dysfunction in Alzheimer disease.

Emerging evidence highlights the crucial role of peripheral immune cells in maintaining brain homeostasis and their influence on the pathology of Alzheimer disease (AD). Genome-wide association studies have identified numerous AD risk variants in genes expressed by immune cells, implicating innate and adaptive immune pathways in disease progression. Advances in neuroimmunology have revealed that immune cell crosstalk involving T cells, B cells, monocytes and/or macrophages and neutrophils can modulate the hallmark features of AD, including amyloid plaque accumulation, tau pathology and chronic

Integrative gene co-expression network analysis reveals protein-coding and LncRNA genes associated with Alzheimer's disease pathology.

Alzheimer's disease (AD) is a complex neurodegenerative disorder marked by widespread molecular changes, many of which remain poorly understood. While AD pathology progresses through specific brain regions, it is unclear whether these regions are affected similarly. Long non-coding RNAs (lncRNAs), emerging as key cellular regulators, remain largely uncharacterized in AD. Understanding how lncRNAs interact with protein-coding genes across brain regions could shed light on AD mechanisms and progression. To investigate this, we performed consensus weighted gene co-expression network analysis on 3

MAPT mutation-induced behavioral variant frontotemporal dementia in an Asian patient: a multimodal biomarker case report resolving diagnostic challenges with Alzheimer's disease.

BACKGROUND: The clinical phenotypic overlap between frontotemporal dementia (FTD) and Alzheimer's disease (AD) frequently leads to misdiagnosis, while biomarkers (e.g., Aβ-PET) and genetic testing provide critical differential diagnostic evidence. Although MAPT gene mutations represent common genetic etiologies of FTD, their occurrence in Asian populations remains underreported. Specifically, FTD caused by the MAPT IVS10 + 16C>T mutation shows limited documentation in Asian populations, with its phenotypic heterogeneity and treatment responses remaining poorly characterized. METHODS: We prese

Mapping the neural correlates of the effect of psycholinguistic variables on picture naming performance: a FDG-PET study across neurodegenerative diseases.

BACKGROUND: Picture naming performance is influenced by the properties of the stimuli and of the words to be retrieved, such as word length and lexical frequency. Significant inconsistencies, however, remain regarding the brain regions mediating these effects in neurodegenerative patients. In the present study, we addressed this issue by correlating regional cerebral metabolism with several naming-related variables in a large cohort of neurodegenerative patients, who are likely to exhibit naming impairments due to different mechanisms of cognitive dysfunction. METHODS: A total of 178 patients

Mesial temporal tau pathology impacts basal forebrain degeneration in early Alzheimer's disease.

INTRODUCTION: The cholinergic basal forebrain system, particularly the nucleus basalis of Meynert (Ch4), is selectively vulnerable to amyloid beta (Aβ) and tau in Alzheimer's disease (AD). Their interplay may be a critical driver of AD progression, but remains poorly understood. METHODS: Data from 779 older individuals in the Australian Imaging, Biomarker, and Lifestyle study were analyzed, all of whom underwent 18F-NAV4694 Aβ and 18F-MK6240 tau positron emission tomography and magnetic resonance imaging. RESULTS: The co-occurrence of Aβ and mesial-temporal (MTL) tau pathologies was associat

Daily temporal organization of inflammation and cognition-related factors and antioxidant enzymes are modified by an intracerebroventricular injection of amyloid-beta peptide (1-42) aggregates in the rat temporal cortex.

Alzheimer's dementia (AD) is a neurodegenerative disorder that causes memory loss and dementia in older adults. The neuropathological hallmarks of AD include amyloid plaques, neurofibrillary tangles, oxidative damage, neuroinflammation, synaptic loss and neuronal cell death. The accumulation of Aβ in the brain plays a key role in the pathogenesis of AD. Elevated levels of Aβ causes an increase in oxidative damage and neuroinflammation both are considered key factors in the progression of AD. Also patients with Alzheimer's show alterations in their circadian rhythms. Our objectives were (a) to

A deep dive into statistical modeling of RNA splicing QTLs reveals variants that explain neurodegenerative disease.

Genome-wide association studies (GWASs) have identified thousands of putative disease-causing variants with unknown regulatory effects. Efforts to connect these variants with splicing quantitative trait loci (sQTLs) have provided functional insights, yet sQTLs reported by existing methods cannot explain many GWAS signals. We show that current sQTL modeling approaches can be improved by considering alternative splicing representation, model calibration, and covariate integration. We then introduce MAJIQTL, a pipeline for sQTL discovery. MAJIQTL includes two statistical methods: a weighted multi

Transcranial photobiomodulation therapy in older women regarding cognitive functions: a systematic review.

Transcranial photobiomodulation therapy (tPBM) is a promising non-invasive treatment that uses red or near-infrared light to modulate biological functions and elicit therapeutic effects. This systematic review aimed to summarise the evidence on the effectiveness of tPBM in improving cognitive function in older women, in experimental animal models and in humans, by analyzing the optimal tPBM parameters and behavioral and neurobiological outcomes. tPBM offers advantages specific to older women as a non-invasive brain stimulation technique, applied to a sensitive population with increased risk of

Advances in the genetics and pathology of Lewy body dementia.

Lewy body dementia is a heterogeneous disease that is underdiagnosed and poorly understood. Pathologically, Lewy body dementia is characterised by the accumulation of intraneuronal aggregates of misfolded α-synuclein, known as Lewy bodies and Lewy neurites. The genetic architecture of Lewy body dementia is complex, involving both common genetic variants with small risk effects and rare genetic variants with large effects. Alzheimer's disease pathology frequently coexists with Lewy body pathology and influences the clinical presentation. A deeper understanding of the pathophysiological pathways

Disentangling the inverse relationship between cancer and Alzheimer's or Parkinson's disease: A systematic review on Mendelian randomization studies.

INTRODUCTION: Although studies have reported an inverse relationship between cancer and neurodegenerative diseases such as Alzheimer's disease (AD) and Parkinson's disease (PD), findings remain inconsistent. Observational studies are limited by survival bias and reverse causation. To better understand the relationship, we conducted a systematic review of Mendelian randomization (MR) studies examining both directions-assessing cancer as a risk factor for AD or PD, as well as AD or PD as exposures influencing cancer risk. METHODS: We systematically reviewed MR studies investigating the causal r

Quantifying motor-cognitive reserve using a novel multi-modal stress test.

Reserve is a physiological capacity used under demanding situations. The concept was developed to account for the discrepancy between pathology and clinical manifestation. In neuroscience, motor, brain and cognitive reserves are abstract measures, conceptually defined yet elusive to quantify. Reserve is indirectly assessed using proxies such as years of education and brain volume, limiting its utility. Moreover, the dichotomy in definitions of cognitive and motor reserves is artificial, as daily function requires an intricate network of connections between these domains. Here, we assessed the
