Mechanistic Evaluation of Psilocybin in Neurodegenerative Disorders: Relevance of 5-HT2A Receptors and Downstream Pathways.

More than fifty million individuals worldwide are experiencing neurodegenerative disorders, which include Parkinson's disease, Alzheimer's disease, amyotrophic lateral sclerosis, Huntington's disease, multiple sclerosis, and frontotemporal dementia. These disorders have a significant role in the gradual degradation of cognition and motor skills. Most current treatment approaches have little effect on changing the course of the disease and are largely symptomatic. Psilocybin, a naturally occurring tryptamine alkaloid from psilocybin mushrooms, has become a promising medicinal agent because of i

Analysis of dementia strategic plans in US states.

State dementia strategic plans provide policy and practice recommendations to reduce the risk, progression, and impact of dementia. Yet there is little systematic understanding of the contents of state plans nor how they aim to influence dementia strategy. This article analyzes state dementia plans, identifying five underspecified dementia policy areas in recent state plans and inconsistency in how states monitor the implementation of plans for effectiveness.

Higher-order executive functions are associated with financial altruism in older adults.

Objective: Executive function abilities are important for optimal financial decision making and have been shown to decline in early Alzheimer's disease (AD). Greater financial altruism has been associated with a cognitive profile characteristic of early AD that largely implicates non-executive functioning abilities. This study investigated the associations between executive functions and "blind giving" in older age. Method: One hundred and thirty-two older adults without dementia (M age = 69.6; M MMSE = 28.3) completed all-digital executive functioning tasks from the Delis-Kaplan Executive Fun

Comparing operational definitions of screening-based cognitive impairment in the Tohoku Medical Megabank cohort: Focus on digital engagement.

BackgroundDementia prevention is a global health priority, yet most large-scale population studies come from Western settings. Japan, a rapidly aging society, provides a unique non-Western epidemiological context for characterizing cognitive decline.ObjectiveTo assess whether the Tohoku Medical Megabank (TMM) cohorts replicate established dementia-related associations, and to identify correlates that are statistically significant across three prespecified operational classifications.MethodsWe analyzed data from >81,000 participants in the TMM cohorts. Participants were classified using (i) sel

Caregiver-provider relationship quality and health outcomes in family caregivers of people living with dementia.

OBJECTIVES: Family caregivers of individuals with Alzheimer's Disease and Related Dementias (ADRD) work with healthcare providers to ensure effective care coordination on behalf of the care recipient. However, challenges within the caregiver-healthcare provider relationship are common and may contribute to poor caregiver health outcomes. This study examined the impact of perceived quality of the relationship between family caregivers and healthcare providers and the caregivers' daily mental health symptom burden (depressive symptoms, anxiety, and stress). METHODS: The study included 228 famil

Brain-derived tau in plasma and cerebrospinal fluid after surgery: a prospective cohort substudy.

BACKGROUND: Postoperative increases in plasma total tau (t-tau) protein have raised concerns that surgery can cause CNS injury. Brain-derived tau (BD-tau) is a novel biomarker that quantifies CNS-derived tau. We assessed BD-tau concentrations in plasma and CSF after surgery to investigate neuronal injury and mechanisms driving increases in plasma t-tau. METHODS: This was a substudy of a previous prospective cohort study enrolling adult patients undergoing hip and knee arthroplasty. We measured plasma and CSF BD-tau and t-tau before neuraxial anaesthesia (baseline) and at 4, 8, 24, 32, and 48

Identification of Succinylation-Related Transcriptional Signatures in Alzheimer's Disease: Integrated Bioinformatics Analysis and Clinical Validation.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder involving mitochondrial dysfunction, oxidative stress, proteostasis imbalance, and immune-inflammatory alterations. Protein succinylation is closely linked to mitochondrial metabolic status, but peripheral blood transcriptomic signatures related to succinylation in AD remain insufficiently characterized. This study aimed to identify peripheral blood transcriptional candidate biomarkers associated with a predefined succinylation-related gene set in AD. The GSE63061 dataset was used as the discovery cohort, and GSE63060 served

Exercise promotes lipophagy and mitigates cognitive impairment in Alzheimer's disease through downregulating microglial TMEM9.

BACKGROUND: Autophagy-lysosomal dysfunction contributes to lipid droplet (LD) accumulation, which drives pathological progression in Alzheimer's disease (AD). Exercise is reported to restore autophagy-lysosomal function and attenuate cognitive impairment in AD, but its regulation of LD deposition remains unknown. METHODS: 5×FAD transgenic mice underwent a three-month exercise intervention. Cognitive and emotional behaviors were assessed using the Morris water maze, novel object recognition, and open field test. LD deposition was evaluated by oil red O and BODIPY staining. De novo formation of familial Alzheimer's disease-relevant Aβ strains occurs exclusively in the human brain.

In familial Alzheimer's disease (FAD), mutations in the amyloid precursor protein gene (APP) accelerate the aggregation of Aβ peptide, leading to deposition of amyloid plaques within the brain. Conformational variants of Aβ aggregates ("Aβ strains") possess distinct pathological properties and may contribute to disease heterogeneity. While the Swedish APP mutation produces wild-type (WT) Aβ, the Arctic APP mutation generates E22G-mutant Aβ. To clarify whether different FAD mutations are sufficient to specify the formation of distinct Aβ strains, we performed Aβ seeding studies in AppNL-F knock

Atherosclerotic cardiovascular disease risk is associated with AT(N)V neuroimaging biomarkers and cognitive impairment in the health and aging brain study-health disparities cohort.

BACKGROUND: Cardiovascular risk is increasingly recognized as relevant to cognitive aging and dementia-related brain changes, yet its relationship to multimodal neuroimaging biomarkers of amyloid, tau, neurodegeneration, and vascular injury (AT(N)-V) remains incompletely understood in racially and ethnically diverse cohorts. We examined whether ten-year atherosclerotic cardiovascular disease (ASCVD) risk was associated with AT(N)-V neuroimaging biomarkers and cognitive domain scores. METHODS: This cross-sectional study included 1,626 Health and Aging Brain Study-Health Disparities (HABS-HD) p

Epigenetics in Alzheimer's disease: from molecular mechanisms to emerging novel biomarkers.

Alzheimer's disease (AD) is an age-related progressive neurodegenerative disorder that represents the leading cause of dementia worldwide, affecting over 50 million people. This global burden is expected to rise dramatically over the coming decades because of global population aging, representing a public health concern. Despite significant advances in understanding AD pathophysiology, early diagnosis remains challenging, emphasizing the urgent need for novel, reliable diagnostic and predictive biomarkers capable of identifying individuals at risk of progression during the preclinical stages o

Angiotensin II Receptor Blockers and the Risk of Alzheimer's Disease Progression: A Population-Based Cohort Study.

We investigated the effects of angiotensin II receptor blockers (ARBs) on the progression of Alzheimer's disease (AD). This retrospective cohort study used data from the Korean nationwide claims data to identify patients with AD aged 60-79 years in 2010. ARB users and nonusers were 1:1 propensity score matched. Cox proportional hazards regression evaluated AD progression, defined as memantine initiation and mortality. Active-comparator and dose-response analyses were conducted. ARB use was associated with lower risks of memantine initiation (hazard ratio [HR] = 0.79; 95% confidence interval [C

A Data-Driven Operational Definition of Square-Wave Jerks and Its Diagnostic Utility in Progressive Supranuclear Palsy.

BACKGROUND: The Movement Disorder Society diagnostic criteria for progressive supranuclear palsy (PSP) list frequent square-wave jerks (SWJ) as a suggestive oculomotor feature. A standardised operational definition of SWJ is however presently lacking. OBJECTIVE: To create a data-driven optimal SWJ definition and to assess its diagnostic utility in PSP. METHODS: We prospectively recruited PSP n = 25, Parkinson's disease (PD) n = 32, Alzheimer's disease (AD) n = 35, and controls n = 88. SWJ were recorded with a visual fixation task using video-oculography. Multiple candidate definitions of SWJ

An integrated human forebrain organoid reveals microglia-mediated CD8+ T cell recruitment and neuroimmune dysfunction in Alzheimer's disease pathology.

Genetic evidence implicates immune dysfunction in Alzheimer's disease (AD), yet the neuroimmune mechanisms operating in human tissue remain poorly defined. Here, we establish a modular human forebrain organoid platform that systematically integrates human induced pluripotent stem cell-derived microglia and CD8+ T cells to reconstitute multicellular AD pathology. This system enables functional interrogation of both innate and adaptive immune components in a human-relevant context. Using this platform, we demonstrate that microglia mediate amyloid-β clearance and neuronal maturation but also dri

Why diabetes matters in dementia studies: Failure to account for diabetes status masks regional mitochondrial DNA copy number changes in human hippocampus, amygdala, and cerebellum in Alzheimer's disease.

INTRODUCTION: Diabetes is a major risk factor for Alzheimer's disease (AD), and both diseases involve mitochondrial dysfunction. Changes in mitochondrial DNA (mtDNA) can influence energy metabolism, cellular signaling, and epigenetic programming, thereby affecting cellular health. We hypothesized that AD is associated with reduced mtDNA copy number (mtDNA-CN) in vulnerable brain regions, and that diabetes modifies these changes. METHODS: MtDNA-CN was quantified by absolute quantitative polymerase chain reaction (qPCR) in post-mortem hippocampus, amygdala, and cerebellum from cognitive control
