ROS-HIF1α-driven glycolytic reprogramming sustains ATP production in Huntington's disease.

Huntington's disease (HD) is a progressive neurodegenerative disorder in which mitochondrial dysfunction and impaired energy metabolism contribute to disease pathogenesis. Surprisingly, we find that ATP levels are not diminished but instead elevated in the striatum of R6/2 HD mice despite impaired TCA cycle intermediates and mitochondrial deficits. Integrative metabolomics, gene expression profiling, and pharmacological perturbation reveal that increased reactive oxygen species stabilize hypoxia-inducible factor-1α (HIF1α), driving enhanced glucose uptake and glycolytic flux. Single-cell transcriptomics and mouse model phenotyping for biomarker screen of peripheral blood in Huntington's disease.

Transcriptional dysregulation is among the most prominent molecular alterations in Huntington's disease (HD). It is not confined to the brain but also extends to peripheral tissues and cells, enabling minimally invasive screening strategies to identify transcriptional surrogates of the health status in HD mutation carriers. Nonetheless, transcriptomics approaches have failed to identify consistent candidates from peripheral blood, probably due to the low impact of the HD mutation in the transcriptional profiles of circulating cells, which can be masked by the high cellular complexity of this b

Clinical implications of loss of interruption variants for diagnosis, genetic counselling, and clinical trials in Huntington's disease.

Age of onset in Huntington disease (HD) is influenced by cis-acting genetic variants, particularly the loss of interrupting codons in the HTT CAG and CCG repeats (CAG-CCG LOI variant). The CAG-CCG LOI variant is not detectable by current diagnostic assays, leading to underestimation of CAG repeat length, misdiagnosis, and inaccurate prediction of risk of symptom onset in the reduced penetrance range. In clinical trials, unidentified CAG-CCG LOI variants may affect interpretation of results, particularly for small trials. Accurate ascertainment and reporting of the CAG-CCG LOI genotype therefor

Large scale functional and effective connectivity alterations cross the Huntington's disease integrated staging system.

Huntington's disease (HD) is a progressive neurodegenerative disease with severe motor, cognitive and behavioral symptoms. There is a recent impetus to develop treatments that slow progression before clinical signs emerge. Such early interventions require biomarkers sensitive to the very earliest HD progression. Here we applied cutting-edge fMRI analysis on data collected in the Track-On HD study to evaluate whether functional and effective connectivity obtained from model-free and model-based approaches can produce useful biomarkers of HD progression. We analyzed data from 231 participants wi

Striatal Neuron Excitability Is Regulated by Huntingtin in the Adult Brain.

Huntington's disease (HD) is a hereditary neurodegenerative disease that typically presents during midlife and is characterized by a combination of motor, cognitive, and psychiatric symptoms. HD is fatal and arises from a mutation in the huntingtin (HTT) gene, which results in decreased neuronal health followed by brain atrophy, with spiny projection neurons (SPNs) of the striatum being especially vulnerable to degeneration. HTT loss of function, caused by haploinsufficiency of the wild-type HTT gene (wtHTT), is an important feature of HD pathophysiology that has previously been understudied c

Transposable element small RNAs and large RNAs in aging brains and implications in Huntington's and Parkinson's disease.

Transposable elements (TEs) are implicated in aging and neurodegenerative disorders, but the impact on brain TE RNA dynamics in these phenomena is not fully understood. Therefore, we quantify TE RNA changes in aging postmortem human and mouse brains and in the neurodegenerative disorders Huntington's disease (HD) and Parkinson's disease (PD). We track TE small RNAs (smRNAs) to assess the relationship to TE large RNA (laRNA) expression patterns. Human brain transcriptomes from the BrainSpan Atlas display significant shifts of TE smRNA patterns, whereas aging mouse brains lack any TE RNA changes

Temporal single-cell atlas of full-length Huntington's disease mouse model defines stage-specific signatures of corticostriatal dysfunction.

BACKGROUND: Huntington's disease (HD) involves progressive corticostriatal dysfunction, yet the temporal dynamics and cell type-specific vulnerability patterns remain incompletely understood. While recent single-cell studies in rapidly progressing models have revealed early developmental and regional changes, temporal profiling distinguishing pathogenic mechanisms from normal aging in full-length HTT models remains lacking. Resolving stage-specific temporal dynamics across interconnected striatal and cortical neuronal populations over protracted time is essential for identifying drivers of cel

SQSTM1/p62 UFMylation Enhances Autophagic Clearance of Pathogenic Mutant Huntingtin.

Ubiquitin-fold modifier 1 (UFM1) covalently modifies protein substrates (UFMylation) and alters their biological functions. Genetic screening disclosed that enzymes in the UFMylation system play critical roles in regulating autophagy. However, it is still elusive which protein is UFMylated and how this modification modulates autophagy. Here, our quantitative proteomics and biochemical experiments identify SQSTM1/p62 as a UFMylation substrate and discover its two major UFMylation sites, K420 and K435. Mutating them to Arg (p622KR) completely abolishes the effect of p62 on autophagic activity. F

Neuroprotective Potential of Pinostrobin in a Rat Model of Huntington's Disease: Behavioural, Biochemical, and Molecular Docking Evidence.

Huntington's disease (HD) is an enormously destructive autosomal hereditary neurodegenerative disease that results in malfunction of motor, psychological, and cognitive deficits. The neurotoxin 3-nitropropionic acid (3-NPA) is known to induce HD-like signs in the in vivo rat model. The current research is aimed at defining the defensive properties of pinostrobin (PSB) against 3-NPA-induced HD in rats. Wistar rats were used as a test model, and PBS (10, 20, and 40 mg/kg, per oral) was administered before 3-NPA (10 mg/kg i.p.) treatment for 15 days. To assess neurodegeneration, behavioural tests

Long-Term Use of Tiapride and Tetrabenazine in Huntington's Disease: A 25-Year Retrospective Single-Center Analysis.

PURPOSE: Chorea is a key motor feature of Huntington's disease (HD). In Germany, Austria, and Switzerland, tiapride and tetrabenazine are established and formally approved for symptomatic management of chorea in HD. However, long-term real-world comparative data are lacking, and early documentation is often incomplete. The purpose of this study was to analyze prescription patterns, tolerability, and reasons for discontinuation of both medications in a retrospective real-world HD cohort and to examine outcomes of reinitiation and combination therapy. METHODS: We retrospectively reviewed medica

3-Nitropropionic Acid-Induced Huntington's Disease in Preclinical Models: Mechanisms, Peripheral Toxicities, Model Gaps, and Future Directions.

Huntington's disease (HD) is an autosomal dominant neurodegenerative disease caused by an expanded cytosine-adenine-guanine (CAG) repeat (> 36 repeats) in the huntingtin gene, leading to progressive motor dysfunction, cognitive decline, and psychiatric disturbances. Despite extensive research, the precise pathophysiological mechanisms underlying HD remain incompletely understood, necessitating reliable experimental models to investigate disease processes and identify therapeutic targets. Among preclinical models, 3-nitropropionic acid (3-NPA) is widely used to induce HD-like pathology in neuro

Data-driven model reveals increased stability of CAG-expanded huntingtin RNA due to MID1 binding.

RNA-binding proteins (RBP) are important regulators of RNA metabolism. In neurodegenerative disorders such as Huntington's Disease (HD), disrupted RBP-RNA interactions contribute to neuronal dysfunction. One such RBP, Midline 1 (MID1), has been shown to aberrantly associate with mutant huntingtin (Htt) RNA, enhancing its translation, yet the mechanism driving this effect remains unknown. Here, we develop a computational model to understand the role of MID1. Based on previously published data, our model predicts that MID1 increases the stability of the Htt RNA. We experimentally validate this p

Phosphoproteomic profiling reveals post-translational dysregulation in Huntington's disease patient-derived neurons.

Huntington's disease (HD) is a fatal neurodegenerative disorder caused by a CAG repeat expansion in the Huntingtin gene. Although transcriptomic and proteomic changes have been characterized in patient-derived neurons, the contribution of post-translational modifications, such as phosphorylation, remains poorly understood. Here, we present the first phosphoproteomic analysis by mass spectrometry (P-MS) of human induced neurons (iNs) directly reprogrammed from HD patient fibroblasts. We identified 177 phosphopeptides with significantly altered abundance in HD-iNs, mapping to phosphoproteins ass

Counseling patients in the predictive genetics clinic for Huntington's disease: A qualitative analysis of ethnographic observations.

Little research has been conducted on patients' experiences inside the predictive genetics clinic. In this clinic, a person with a familial risk of inheriting a rare disease seeks a test that will provide them with information about their future health. Counselors combine gentle challenges with neutral approaches to educate and counsel patients wanting to take the predictive test. We conducted ethnographic observations of predictive genetics clinics for Huntington's Disease in England and Wales. Four content categories were prominent throughout the analysis, which we present in this paper: con

Regulation of huntingtin-polyQ aggregation by glucose transporters and saccharide-binding receptors during aging in Drosophila.

Saccharides and responsive signaling pathways are important regulators of physiological and pathological processes. Saccharides can exert these functions via dedicated intracellular transporters and through binding to cellular sensors, such as saccharide-binding receptors. Glucose homeostasis and saccharide signaling are emerging as important regulators of neurodegeneration. Here, we utilized a Drosophila model of Huntington's disease to examine the role of Glut1 (homologous to multiple human glucose transporters) and saccharide-binding receptors. We find that Glut1 knockdown increases the amo

Which HTT transcript to lower?

Huntington's disease (HD) RNA therapeutics have focused on lowering canonical huntingtin (HTT), yet clinical benefit remains uncertain. Papadopoulou et al. and Bragg et al. suggest that HTT1a-engaging strategies can produce stronger molecular rescue than full-length HTT lowering that spares HTT1a, highlighting transcript-species coverage as a key variable in HTT-lowering pharmacology.

Mitochondrial Dysfunction as a Driver of Neurodegeneration in Parkinson's and Huntington's Disease: Molecular Insights and Emerging Interventions.

Mitochondrial dysfunction has emerged as a central contributor to the pathogenesis of major neurodegenerative disorders, such as Parkinson's and Huntington's disease. In Parkinson's disease, mitochondrial abnormalities are often linked to mutations in genes like PINK1 and Parkin, which regulate mitochondrial quality control, while α-synuclein aggregation further exacerbates mitochondrial damage. In Huntington's disease, mutant huntingtin protein impairs mitochondrial dynamics, transport, and ATP production, contributing to selective neuronal vulnerability. The convergence of mitochondrial impa

Recognizing repeat expansion disorders in clinical practice.

Repeat expansion disorders are caused by unstable DNA sequences that exceed pathogenic thresholds, disrupting normal gene function. These conditions often affect the nervous system, but may involve multiple organs, with presentations ranging from subtle cognitive or motor changes to overt neuromuscular or neurodevelopmental syndromes. Early symptoms can mimic common conditions, making clinical suspicion, family history, and awareness of intergenerational patterns essential. Molecular features, including repeat size, sequence, location and stability, determine disease severity and variability.

Early and progressive transcriptional remodeling of retrotransposons in Huntington's Disease Models.

Retrotransposons, including autonomous Long Interspersed Nuclear Elements (LINEs) and non-coding Short Interspersed Nuclear Elements (SINEs), are integral to genomic structure and plasticity, with emerging roles in neurodegeneration. In Huntington's disease (HD), the contribution of retrotransposable elements to pathogenesis is still poorly characterized. Here, we investigated the spatiotemporal expression profiles of LINE and SINE families in two HD mouse models (R6/2 and zQ175), across pre-symptomatic, symptomatic, and embryonic stages. Using quantitative PCR, we identified distinct, region-
