Gut microbiome composition in Huntington's disease: Stage-dependent differences between premanifest and manifest patients.

BACKGROUND: Huntington's disease (HD) is a progressive neurodegenerative disorder with substantial clinical heterogeneity. The gut microbiome has been proposed as a potential modulator of neurodegeneration, but its role in HD and across disease stages remains unclear. METHODS: This cross-sectional case-control study included 50 individuals with HD (35 manifest, 15 pre-manifest) and 36 age- and sex-matched cohabiting controls. Stool samples were analysed using 16S rRNA sequencing. Clinical, lifestyle, and dietary variables were recorded. Microbial diversity and differential taxonomic abundance

Wild-type C9orf72 drives proteasomal dysfunction and mutant aggregates via a Stat1-Isg15 axis in Huntington's disease.

Mutant C9orf72 has been extensively studied as a major genetic cause of amyotrophic lateral sclerosis and frontotemporal dementia, and is also known to generate Huntington's disease (HD)-like phenocopies. However, despite this strong disease association, the role of wild-type C9orf72 (wt-C9orf72) in neurodegeneration remains largely unexplored. HD is a neurodegenerative disease, and characterized by the accumulation of misfolded mutant Huntingtin (mHTT) and impaired proteostasis, yet the upstream mechanisms driving ubiquitin-proteasome system (UPS) dysfunction are not fully understood. Molecular insights of peroxisome proliferator-activated receptor-γ signalling in amyotrophic lateral sclerosis and Huntington's disease.

Progressive neuronal loss is a hallmark of neurodegenerative diseases like Huntingtons disease (HD) and Amyotrophic lateral sclerosis (ALS) which are caused by convergent mechanisms such as oxidative stress, mitochondrial dysfunction, neuroinflammation, impaired autophagy and dysregulated cell death pathways. Both conditions share significant disruptions in metabolic and inflammatory signalling despite having different genetic origins and clinical manifestations; underscoring the necessity of pathway-oriented treatment approaches. In the central nervous system, peroxisome proliferator-activate

In vivo mapping of striatal neurodegeneration in Huntington's disease with Soma and Neurite Density Imaging.

Huntington's disease (HD) is an inherited neurodegenerative disorder characterised by progressive cognitive and motor decline driven by basal ganglia (BG) atrophy. Clinical trials of novel disease-modifying therapies are ongoing, creating a need for sensitive non-invasive imaging biomarkers. Soma and Neurite Density Imaging (SANDI) is a multi-shell diffusion MRI model that estimates intracellular signal fractions from sphere-shaped soma and shows promise as a marker of neurodegeneration. The objectives of this study were to characterise HD-related microstructural abnormalities in the BG using

Blocking somatic repeat expansion and lowering huntingtin by RNAi synergize to attenuate Huntington's disease pathogenesis in mice.

Huntington's disease (HD) is a progressive neurodegenerative disorder with no approved therapies. Despite multiple clinical trials, huntingtin (HTT)-lowering strategies have yet to show meaningful clinical benefit. Both somatic expansion and toxic HTT species are key molecular drivers of HD, yet therapeutic strategies targeting these pathways have never been directly compared or evaluated in combination. Using therapeutic divalent siRNAs, we assessed the long-term impact of silencing MutS homolog 3 (MSH3), a critical regulator of somatic expansion, HTT, or both in Q111 HD mice (>110 CAGs), whi

Alternative splicing dysregulation in CAG repeat expansion diseases.

Alternative splicing of RNA is a highly regulated process that increases the complexity of gene expression, with disruption of splicing leading to significant disruption of cellular function and, ultimately, disease. This spliceopathy is exemplified by myotonic dystrophy type 1, a CTG repeat expansion disease, where dysregulation of alternative splicing drives core disease symptomatology. Recent studies across murine- and patient-derived disease models have demonstrated that similar alternative splicing changes are prevalent in CAG repeat expansion diseases, including Huntington's disease and

Intracerebroventricular Administration of Gangliosides for In Vivo Functional Studies in Mice.

Levels of gangliosides are reduced in several neurological conditions, and their restoration has shown therapeutic potential in animal models of Huntington's disease and Parkinson's disease. However, the full range of functions of gangliosides in the brain, along with their molecular targets and mechanisms of action following exogenous administration, remains ill-defined. Due to their poor blood-brain barrier (BBB) permeability, gangliosides face significant challenges when delivered through non-invasive routes. Direct intracerebroventricular (ICV) infusion circumvents this issue by bypassing

The Role of PolyQ in Global Conformational Dynamics and Stability of Huntingtin.

Huntington's disease (HD) is known for its abnormal polyQ stretch in the first exon of the HTT gene. HD is listed among the fatal autosomal dominant neurodegenerative disorders targeting the central nervous system and interfering with motor, cognitive, and psychiatric functions. The HTT gene codes for Huntingtin (Htt), a large three-domain protein with a polyQ stretch present in the N-terminal domain. In diseased cells, inclusion bodies are enriched in Htt fragments containing an extended polyQ region. The expansion of the polyQ correlates with aggregate size and onset of HD. However, the exac

The potential of vitamins in Huntington's disease: bridging mechanistic evidence with translational limitations and future strategies.

Neurodegenerative diseases are characterized by impairment of neuronal functions and neuronal loss. Huntington's disease is one of the neurodegenerative diseases caused by the formation of mutant Huntingtin protein aggregates in the brain. The other pathological reasons behind this disease are oxidative stress, mitochondrial dysfunction, and excitotoxicity, which lead to impairment of motor, cognitive, and psychiatric functions. The vitamins are considered an essential part of a healthy diet, as they tend to increase the nutritional value of the diet. Insufficient intake of macro- and micronut

The Role of MicroRNA in Diagnosis of Huntington's Disease: A Systematic Review.

Huntington's disease (HD) is an autosomal dominant genetic neurodegenerative disorder with features of progressive motor, cognitive, and psychiatric dysfunction. Current diagnosis relies largely on clinical presentation and genetic testing but lacks sensitivity to early disease diagnosis or progression monitoring. MicroRNAs (miRNAs) are short non-coding RNA molecules that regulate gene expression post-transcriptionally. They are increasingly being investigated as potential diagnostic biomarkers, especially given their detectability in biofluids like plasma and cerebrospinal fluid. This review

Dysfunction of Primary Cilium in Huntington's Disease: How Mutant Huntingtin Disrupts This Cellular Signaling Hub.

Primary cilia are solitary, antenna-like organelles that project from the surface of most vertebrate cells. They consist of a microtubule-based axoneme extending from a modified centriole (basal body) and enclosed by a lipid bilayer membrane. For several decades after their discovery, the functions of primary cilia had remained speculative; they were even considered vestigial structures. Currently, primary cilium is recognized as essential sensory and signaling structure involved in both chemo- and mechanosensation. Its anchoring at the centrosome surrounded by a radially organized network of

Network-based stratification of allele-specific expression reveals patient subgroups in Huntington's disease.

MOTIVATION: Huntington's disease (HD) exhibits substantial variability in age of onset and disease progression that is not fully explained by CAG repeat length alone. Part of this residual variation is heritable, implicating additional genetic mechanisms. cis-regulatory variation, genetic variants that alter transcription and splicing of nearby genes, represents one such mechanism that can be quantified through allele-specific expression (ASE) analysis. However, methods for integrating ASE profiles into patient stratification frameworks remain underdeveloped, particularly for rare diseases wit

A porcine spectral assay library to quantify brain proteome by DIA-MS.

Neurological disorders are the leading cause of health loss worldwide. The growing number of patients suffering from such conditions calls for improved strategies for their prevention, diagnosis, and therapy. To better understand human pathologies, relevant models and methodologies must be made available. In this study, we focused on a biomedical model capable of recapitulating the complexity of human pathology, the pig (Sus scrofa). Brain tissue and cerebrospinal fluid samples from a transgenic minipig model of Huntington's disease were subjected to multiple extraction and fractionation steps

Geometric properties of caudate and putamen mark the progression of Huntington's disease.

Volumetric MRI of the caudate and putamen is a robust biomarker of striatal degeneration in Huntington's disease (HD). Striatal degeneration may be reflected beyond volume alone, in additional shape and intensity alterations. We evaluated five novel morphometric biomarkers of the caudate and putamen-curvature, flatness, eccentricity, elongation, and focal assessment of divergence estimation (FADE; T1-weighted signal shift)-using cross-sectional and longitudinal MRI data from healthy controls (N = 100), individuals with premanifest HD (preHD; N = 100), and individuals with early manifest HD (ea

The pathophysiology of depression in Huntington's disease.

Depression affects Huntington's disease (HD) gene-expansion carriers at up to four times the general population rate, yet its neurobiological underpinnings remain inadequately understood, potentially contributing to persistent symptoms and suboptimal therapeutic outcomes, despite commonly used interventions. Understanding of the pathophysiological mechanisms remains constrained by fragmented, single-level approaches that do not capture interactions between biological systems. Here, we present an illustrative multilevel, reciprocal pathophysiological framework that integrates evidence across br

L-theanine from Camellia sinensis (L.) attenuates 3-NP-induced Huntington's disease-like alterations by remedying behavioral patterns and modulating neurotransmitters across three distinct brain regions: evidence from in-vivo and in-silico approaches.

Huntington's disease (HD) is a neurological condition with limited treatment. Its hallmarks are progressive loss of neurons, chorea, cognitive, motor, and metabolic impairments. The current study uses 3-nitropropionic acid (3-NP) to cause HD-like indications in rats. L-theanine (L-TH), an active component of Camellia sinensis, has already been shown to possess anti-inflammatory and anti-oxidant effects. The impact of the isolated phytoconstituent, L-TH, on 3-NP-induced changes in three distinct brain regions was assessed in the current study. The animals received a 7-day pretreatment consistin

Human-specific sequence features in HTT exon 1 promote toxic misprocessing via splicing factor SRSF7.

Expansion of CAG repeats in HTT exon 1 is the acknowledged driver of Huntington's disease. Alternative processing of HTT pre-mRNA generates the truncated HTT1a transcript, translated into a toxic peptide. While its dependence on CAG length is well documented, the role of adjacent sequences - particularly the Proline-Rich Domain (PRD) - remains unexplored. Using our HuntEx1-engineered mouse embryonic stem cell platform, we show that human PRD promotes HTT1a production, whereas its replacement with mouse PRD in an otherwise human exon 1 markedly reduces HTT1a levels. Mechanistically, we find tha

Nanotechnological and in vivo characterization of a new generation of brain-permeable nanoparticles for brain-targeted cholesterol delivery in Huntington's disease.

Reduced brain cholesterol synthesis is an early dysfunction that plays a major role in Huntington's disease (HD) pathogenesis. Because the blood-brain barrier (BBB) prevents cholesterol uptake from the circulation, surface-modified, biodegradable, and biocompatible nanoparticles (NPs) that enhance BBB crossing have emerged as promising tools for delivering cholesterol to the brain in HD mouse models. In these models, such NPs have been shown to rescue cognitive, motor, and neuropathological defects. Here, we describe a new formulation of nanoparticles primarily composed of cholesterol and surf

Evaluation of curcumin as an exploratory pharmacological tool for modulating dysregulated molecular pathways in cellular models of Huntington's disease.

BACKGROUND: Huntington's disease (HD) is a monogenic neurodegenerative disorder caused by expansion of CAG trinucleotide repeats in the HTT gene, resulting in an aggregation-prone mutant huntingtin protein (mHTT) and progressive neuronal dysfunction. HD pathogenesis involves disturbances in protein homeostasis, mitochondrial function, oxidative stress, epigenetic regulation, and inflammatory signalling. We investigated the effects of curcumin, a naturally occurring polyphenol, on these disease-relevant processes in vitro. METHODS AND RESULTS: Using established HD cellular models, we examined
