Sleep abnormalities are associated with greater cognitive deficits and disease activity in Huntington's disease: a 12-year polysomnographic study.

Increasing evidence suggests that the sleep pathology associated with neurodegenerative diseases can in turn exacerbate both the cognitive deficits and underlying pathobiology of these conditions. Treating sleep may therefore bear significant, even disease-modifying, potential for these conditions, but how best and when to do so remains undetermined. Huntington's disease, by virtue of being an autosomal dominant neurodegenerative disease presenting in mid-life, presents a key 'model' condition through which to advance this field. To date, however, there has been no clinical longitudinal study

Emerging nano-derived therapy for the treatment of dementia: a comprehensive review.

Dementia includes a variety of neurodegenerative diseases that affect and target the brain's fundamental cognitive functions. It is undoubtedly one of the diseases that affects people globally. The ameliorating the disease is still not known; the symptoms, however, can be prevented to an extent. Dementia encompasses Alzheimer's disease, Parkinson's disease, Huntington's disease, Lewy body dementia, mixed dementia, and various other diseases. The aggregation of β-amyloid protein plaques and the formation of neurofibrillary tangles have been concluded as the foremost cause for the onset of the d

Doxycycline: An essential tool for Alzheimer's disease.

The identification of active interventions in neurodegenerative disorders is a major challenge in neurology; the use of repurposed drugs may represent a valuable strategy. Tetracyclines, a second generation of antibiotic molecules, offer various potential applications. Following an anecdotal observation of the potential anti-amyloidogenic activity of iododoxorubicin, the search for chemical analogs with a better safety profile led to tetracyclines. Their heterocyclic structures with a planar conformation interfere with b-sheet amyloid formation. Thus, doxycycline, a derivative with favorable b

Continuous-time mediation analysis for repeatedly measured mediators and outcomes.

Mediation analysis aims to decipher the underlying causal mechanisms between an exposure, an outcome, and intermediate variables called mediators. Initially developed for fixed-time mediator and outcome, it has been extended to the framework of longitudinal data by discretizing the assessment times of mediator and outcome. Yet, processes in play in longitudinal studies are usually defined in continuous time and measured at irregular and subject-specific visits. This is the case in dementia research when cerebral and cognitive changes measured at planned visits in cohorts are of interest. We th

Potential common pathogenesis of several neurodegenerative diseases.

With the gradual advancement of research methods and technologies, various biological processes have been identified as playing roles in the pathogenesis of neurodegenerative diseases. However, current descriptions of these biological processes do not fully explain the onset, progression, and development of these conditions. Therefore, exploration of the pathogenesis of neurodegenerative diseases remains a valuable area of research. This review summarizes the potential common pathogeneses of Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, Huntington's disease, frontote

Progranulin function and regulation in the CNS.

Dysregulated progranulin expression is robustly associated with diseases of the central nervous system (CNS). Recent research has been progressing toward a mechanistic understanding of the role of progranulin in CNS disease pathophysiology. In this review we describe the consequences of dysregulated progranulin expression in experimental and disease states. Collectively, these studies reveal that progranulin has diverse roles as a cell signaling molecule that regulates lysosomal function, immune processes, and growth. Given the functional and pathological implications of aberrant progranulin e

Glial phagocytosis for synapse and toxic proteins in neurodegenerative diseases.

Glia, as resident immune and supportive cells of the central nervous system, play a critical role in maintaining brain homeostasis. One of their key homeostatic functions is phagocytic capacity in pruning synapses and removing cellular debris/protein aggregates, a process vital for synaptic plasticity and brain maintenance. However, these phagocytic functions are often dysregulated with aging and in neurodegenerative diseases (NDs), such as Alzheimer's disease, Parkinson's disease, Huntington's disease, amyotrophic lateral sclerosis, and frontotemporal dementia. This review aims to examine the

A descriptive analysis of rural seniors with femur and pelvis fractures and the impact of morphine milliequivalents on inpatient rehabilitation.

BACKGROUND: The US population ≥65 years old is rapidly growing. Geriatric disorders including hip fractures are becoming more numerous and the medical cost is significant. Hip fractures increase the risk of mortality in geriatric patients, but with improvements in medical care, this risk is declining over time. One important factor in the recovery and rehabilitation of hip fracture patients is pain management. Understanding the impact of opiate prescribing practices on outcomes in the rehabilitation setting can guide recommendations for the future. MATERIALS AND METHODS: Patients ≥65 years ol

High-fidelity and differential nonsense suppression in live cells and a frontotemporal dementia allele with human transfer RNAs.

Nonsense mutations generate premature termination codons (PTCs) that are responsible for 11% of genetic disease alleles. The arginine (Arg, CGA) to stop (UGA) mutation is the most common PTC. Humans encode >600 transfer RNA (tRNA) genes with many identical and similar copies. We developed a dual fluorescent reporter to quantify PTC readthrough in live cells and found single nucleotide mutations of human tRNAArg gene variants enabled differential nonsense suppression that depended on the tRNA sequence and the cell type. We investigated G36A variants of all six human tRNAArgUCG isodecoders, and
