Mitochondria from huntington´s disease striatal astrocytes are hypermetabolic and compromise neuronal branching.

BACKGROUND: Deficits in mitochondrial bioenergetics and dynamics are strongly implicated in the selective vulnerability of striatal neurons in Huntington´s disease. Beyond these neuron-intrinsic factor, increasing evidence suggest that non-neuronal mechanisms, particularly astrocytic dysfunction involving disrupted homeostasis and metabolic support also contribute to disease progression. These findings underscore the critical role of metabolic crosstalk between neurons and astrocytes in maintaining striatal integrity. However, it remains unclear whether this impaired communication affects the

Natural products proposed for the management of Huntington's disease (HD): a comprehensive review.

Huntington's disease (HD), a neurodegenerative disease, typically begins in the prime of adulthood, followed by a gradual onset of specific mental abnormalities and cognitive and physical impairment. To the best of our knowledge, no medication exists to totally stop the progression of HD. Among numerous therapy techniques, extensive literature reviews have confirmed the medicinal importance of natural products in HD experimental models. This review provides a literature survey of natural compounds and medicinal plants used as neuroprotective agents against HD. Relevant studies were found in a

Decision tree for severity assessment of neurodegenerative diseases using possibility approach and gait dynamics.

Accurate and timely diagnosis of neurodegenerative disease (NDD) severity is crucial for clinical needs. Existing assessment methods for grading disease severity are mostly dependent on movement disorder specialist opinion resulting in subjectivity and inherent limitations. Gait instrumentation, on the other hand, can be used as a reliable tool to record various contrasting primary gait features. However, these features are agonized by individual's physical dimensions causing data dispersion. This study proposes normalized feature set, and a decision tree (DT) based model to evaluate NDD sever

Dysregulation of huntingtin interacting protein networks in human juvenile Huntington's disease brain.

BackgroundHuman Huntington's disease (HD) is a genetic neurodegenerative disorder caused by the mutant HTT gene containing CAG repeat expansions, resulting in motor dysfunction and behavioral deficits. CAG repeats of 40-53 occur in adult HD and 60-120 repeats occur in early onset juvenile HD, differing from the normal range of 5-35 repeats.ObjectiveThe HTT gene is translated to the huntingtin (HTT) protein that interacts with proteins in the development of HD. There have been few studies of HTT protein interactors in human HD brain. Therefore, this study evaluated the hypothesis that dysregula

Probing How Anti-huntingtin Antibodies Bind the Fibrillar Fuzzy Coat Using Solid-State NMR.

Antibodies are critical for the immune response and serve as important tools due to their ability to recognize specific amino acid sequences, or epitopes. Based on the latter, they are utilized as diagnostic tools in biological and biomedical research. Huntington's disease (HD) is a neurodegenerative condition caused by CAG repeat expansions in the huntingtin (HTT) gene and characterized by amyloid-like protein deposits in patients. Multiple anti-HTT antibodies are used in HD research for their ability to recognize specific HTT inclusions in both post-mortem tissue and in laboratory conditions

Avocado Oil Prevents Neurological and Behavioral Alterations in a Quinolinic Acid-Induced Model of Huntington's Disease.

Avocado oil is a nutraceutical that offers multiple health benefits due to its rich composition of fatty acids. Avocado’s antioxidant molecules can protect from neuronal damage, potentially preventing or slowing down the onset of neurodegenerative diseases. Within the search for therapeutic targets in neurodegenerative conditions, we focused on testing the neuroprotective effect of the components contained in avocado oil in a Huntington’s disease (HD) model. Quinolinic acid (QA) is a potent striatal excitotoxin that leads to axon-sparing neurodegeneration and induces N-methyl-D-aspartate recep

Self-driving microscopy detects the onset of protein aggregation and enables intelligent Brillouin imaging.

The process of protein aggregation, central to neurodegenerative diseases like Huntington's, is challenging to study due to its unpredictable nature and relatively rapid kinetics. Understanding its biomechanics is crucial for unraveling its role in disease progression and cellular toxicity. Brillouin microscopy offers unique advantages for studying biomechanical properties, yet is limited by slow imaging speed, complicating its use for rapid and dynamic processes like protein aggregation. To overcome these limitations, we developed a self-driving microscope that uses deep learning to predict t

Variation in the reported prevalence of Huntington's disease: a systematic review and guide to interpretation.

There is significant variation in the reported estimates of Huntington's disease (HD) prevalence in different settings. This systematic review was undertaken to describe and assess the sources of heterogeneity in estimated prevalence values, and to consider the role of quantitative synthesis in the context of such heterogeneity. Observational studies from which a prevalence estimate (point or period) or cumulative incidence of HD could be calculated between 1993 and 2024 were sought from Medline and Embase databases. The study features are described and the sources of heterogeneity are discuss

Intrastriatal Delivery of a Zinc Finger Protein Targeting the Mutant HTT Gene Allele Obviates Lipid Phenotypes in Brain and Plasma in Huntington's Disease Mice.

Reducing the burden of mutant Huntingtin (mHTT) protein in brain cells is a strategy for treating Huntington's disease (HD). However, it is still unclear what pathological changes can be reproducibly reversed by mHTT lowering and whether these changes can be measured in peripheral biofluids. We previously found that lipid changes that occur in brain with HD progression could be prevented by attenuating HTT transcription of the mutant allele in a genetic mouse model (LacQ140) with inducible whole body lowering. Here, we tested whether intrastriatal injection of a therapeutic capable of repressi

Evidence based molecular pathways, available drug targets, pre- clinical animal models and future disease modifying treatments of huntington's disease.

Huntington's disease is an autosomal dominant neurodegenerative disorder of variable progression. Its major features are motor dysfunction, cognitive decline, and psychiatric disturbances. The onset of HD in a patient occurs because of a polyglutamine-expanding mutation within the HTT gene, which leads to the formation of mutant huntingtin protein that aggregates and disrupts neuronal function. Epidemiologically, HD afflicts about 5-10 people per 100,000 throughout the world. However, among populations of European descent, its prevalence is increased. Even after much study into the disorder, m

Silent destruction: hidden muscle wasting and body decline in early Huntington's disease.

Huntington´s Disease (HD) is an autosomal dominant, neurodegenerative disorder with characteristic motor, behavioural, and cognitive impairment. Mutant Huntingtin may also affect peripheral tissue. We aimed to assess skeletal muscle (SMM) and fat mass, sarcopenia (EWGSOP2), and malnutrition in HD patients in early disease stages compared to age- and sex-matched healthy subjects. Body composition was evaluated by bioelectrical impedance analysis. The Unified HD Rating Scale and cognitive assessments were used for clinical characterization. Twenty early-stage HD patients (45% females) with a med

Development of the Movement Disorders Interpretation Bias Scale and psychometric evaluation in adults with Huntington's disease.

INTRODUCTION: Anxiety is widespread but undertreated in Huntington's disease (HD). Targeting negative interpretations of ambiguity may reduce anxiety in HD, but no valid measure of negative interpretation biases in HD exists. We developed the Movement Disorders Interpretation Bias Scale (MDIB) to measure negative interpretation biases in movement disorders (e.g., HD, Parkinson's disease [PD]) and evaluated its validity in adults with HD (preregistration: https://osf.io/26a3h). METHODS: We developed the MDIB based on qualitative interviews and expert review, then invited adults with HD (N = 65

Coniferaldehyde reverses 3-nitropropionic acid-induced Huntington's disease pathologies via PKM2 restoration and JAK2/STAT3 inhibition.

BACKGROUND: Huntington's disease (HD) is a fatal neurodegenerative disorder characterized by progressive motor decline and neuronal loss, with no curative disease-modifying therapies available. The mitochondrial toxin 3-nitropropionic acid (3-NP) is widely used to model HD-like pathologies. We investigated the therapeutic potential of coniferaldehyde (CFA), a natural phenolic compound with anti-inflammatory, antioxidant, and anti-radical properties, against 3-NP-induced neurodegeneration. Given the roles of oxidative stress, metabolic dysfunction, and neuroinflammation in HD, we hypothesize th

Huntington Disease and Chorea.

OBJECTIVE: This article presents a systematic approach to the diagnosis and management of choreiform disorders, focusing on Huntington disease (HD) as a model. LATEST DEVELOPMENTS: The availability of genetic testing and imaging biomarkers has led to changes in the definition of HD, which has shifted toward pathologic instead of clinical markers. A third vesicular monoamine transporter 2 inhibitor, valbenazine, is available for the management of chorea in HD. There has been an ongoing focus targeting the early neurodegenerative process in the development of disease-modifying strategies across

Along-Tract Diffusion Alterations in the Dentato-Rubro-Thalamic Tract Correlate With Motor and Cognitive Decline in Huntington's Disease.

Huntington's disease (HD) is a progressive neurodegenerative disorder caused by cytosine-adenine-guanine repeat expansion in the huntingtin (HTT) gene, leading to widespread brain atrophy and white matter degeneration. Although cortico-striatal pathways have been extensively studied, the dentato-rubro-thalamic tract (DRTT), a key cerebellar efferent pathway integrating motor and cognitive functions, remains largely unexplored, despite increasing evidence of cerebellar involvement in these functions. By investigating microstructural alterations along the DRTT, we aim to elucidate its role in HD

Huntingtin reduction results in altered nuclear structure and heterochromatic instability.

Huntington's disease (HD), a fatal neurodegenerative disease, arises due to a CAG repeat expansion in the huntingtin (HTT) gene. Non-pathogenic wild type HTT (wtHTT) is essential for neurodevelopment as well as many vital cellular functions within the adult brain; however, the consequences of wtHTT reduction in adulthood and particularly in extrastriatal regions of the brain have not been well characterized. Understanding the implications of wtHTT loss is essential as numerous genetic therapies for HD non-specifically reduce the expression levels of both mutant and wtHTT. The aim of the curren

Insights into Caspase-6 Mutations and Neuropathology in Huntington's Disease and Psychiatric Disorders: A Structural Genomics and Drug Repurposing Approach.

Huntington's disease (HD) is an inherited neurodegenerative condition that typically appears later in life, marked by progressive motor impairments, cognitive deterioration, and a range of psychiatric disturbances. Crucial to HD pathogenesis is the aberrant activation of caspase-6 (CASP6), which leads to the cleavage of mutant huntingtin (mHTT) protein, generating a toxic N-terminal fragment. CASP6 activity is elevated in human HD brains and is associated with the progression of neuropathology. This study aims to investigate the impact of deleterious mutations in CASP6 on the catalytic activit

Neuroprotective and gut health benefits of Ugni molinae in preclinical Huntington's disease models.

Huntington's disease (HD) is an autosomal-dominant inherited neurological disorder caused by an unstable trinucleotide CAG repeat expansion at the N-terminus of the IT-15 gene, which encodes the ∼350 kDa huntingtin protein (Htt). This mutation confers toxic properties, promoting neuronal dysfunction and death through multiple mechanisms. Effective treatments for HD are still lacking. Therefore, early-stage interventions could represent a promising approach to addressing the pathological aspects of HD. Recently, our laboratory explored the potential neuroprotective effect of murtilla (Ugni moli
