Camillo Golgi and the pathology of Huntington's disease: An unresolved controversy.

In 1874, Camillo Golgi published the neuropathological study of a patient with chronic chorea and dementia, demonstrating impairments in  the frontal-parietal and temporal cortices, striatum, and cerebellum. This study anticipated by fifty years those traditionally recognized as the first to link choreic movements with cortico-striatal involvement, but regardless of the author's intention, has also come to be considered the first on the neuropathology of Huntington's disease. This has given rise to a dispute between one position in favor of this diagnosis based on the clinical-anatomical corre

Inhibiting Cytosine-Adenine-Guanine (CAG) repeat expansions as a therapeutic strategy for Huntington's disease.

Huntington's Disease (HD) became the first disease mapped to a single chromosome and associated with mutations in the huntingtin (HTT) gene, specifically expansions in the trinucleotide cytosine-adenine-guanine (CAG) within exon 1. The onset and progression of disease has been linked, inversely, to the inherited CAG repeat length in the mutated allele but also to an ongoing somatic expansion. However, HD remains an unmet medical need. A rational but not yet successful therapeutic approach has been to inhibit the mutant HTT mRNA/protein although recent trials have shown signs of early promise.

The HEK293T cells manage overload by the overexpressed full-length Htt variants via proteasome activation.

Huntington's disease (HD) is an autosomal dominant neurodegenerative disease caused by a pathological mutation that results in the abnormal expansion of more than 37 consecutive trinucleotide repeats (CAG) in the HTT gene. These repeats encode the polyglutamine tract (polyQ tract) in the huntingtin protein (Htt). Progressive lethal Huntington's chorea is characterized by impaired motor activity and marked cerebral atrophy. The disease affects neurons in specific areas of the central nervous system, mainly GABAergic neurons in the striatum and cortex. It is believed that the neuron-specific pro

Exploring the interaction between nucleotide excision repair pathways and Huntington disease: Implications for neurodegeneration and phenotypic overlap.

Under normal conditions DNA is constantly exposed to various endogenous and exogenous factors that induce DNA damage. To maintain and ensure its integrity, eukaryotic cells possess several complex DNA repair mechanisms. Each of these mechanisms is activated in response to DNA damage. Nucleotide excision repair (NER) is one of the most important and versatile DNA repair pathways, targeting the removal of bulky DNA lesions. Environmental agents (ultraviolet, component of solar radiation, ionizing radiation, chemotherapy) or cellular factors (oxidation, alkylation, hydrolysis and reactive oxygen

Longitudinal investigation of structural and resting-state effective connectivity alterations in a non-human primate model of Huntington's disease.

Huntington's disease (HD) is a genetic neurodegenerative disorder caused by expanded CAG repeats in the huntingtin gene which produce a mutant huntingtin (mHTT) protein that contributes to progressive striatal, cortical, and white-matter atrophy, resulting in motor dysfunction and cognitive decline. Recently, a non-human primate (NHP) model of HD was developed via stereotaxic delivery of an adeno-associated viral vector expressing 85 CAG repeats (85Q) into the striatum. This model recapitulates several neuropathological changes and symptoms observed in people with HD (PwHD) including chorea an

Development and Validation of a Brief Fall Questionnaire (FALL-HD) for Patients with Huntington's Disease.

BACKGROUND: Huntington's disease (HD) is an autosomal dominant neurodegenerative disorder caused by a mutation in the huntingtin gene on chromosome 4, leading to progressive cognitive decline, motor impairment, and functional disability. Falls represent the leading cause of nursing home placement in HD. Despite the significant clinical implications, validated instruments for fall assessment, including fall frequency, are lacking in HD. OBJECTIVES: To construct and evaluate the psychometric properties of a questionnaire for assessing fall status, including fall frequency, in ambulatory patient

Impairment of lysosomal quality control in Huntington disease.

Huntington disease (HD) is a neurodegenerative disease caused by a polyglutamine expansion (polyQ) in the Huntingtin protein (muHTT), which makes it prone to misfolding and aggregation. muHTT aggregates sequester a wide variety of proteins essential for cell homeostasis, including chaperones and transcription factors, and their depletion may contribute to HD pathogenesis. Lysosomes are the main hubs for degradative and signaling activities in cells, and their functionality is crucial for cell homeostasis, especially for neurons. Different forms of cellular stresses, including proteotoxic stres

Mechanism of trinucleotide repeat expansion by MutSβ-MutLγ and contraction by FAN1.

Triplet repeat expansion underlies multiple pathologies, including Huntington's disease, often arising in somatic non-dividing tissues such as the brain. Despite identification of genetic modifiers, mechanistic insights remain limited. Using purified human proteins, we show that MutLγ (MLH1-MLH3), stimulated by MutSβ (MSH2-MSH3), incises DNA opposite an extrahelical loop on the 5' side. This activity, with a moderate sequence preference, generates DNA nicks enabling Polδ-mediated displacement synthesis with the loop as a template, leading to expansion. PCNA confines these MutLγ incisions near

Brain organoid models of Huntington's disease shift the focus towards neurodevelopment.

Huntington's disease (HD) is traditionally viewed as an age-related disorder. Emerging evidence suggests that mutant huntingtin (mHTT) disrupts early neurodevelopment, although the contribution of developmental alterations to the late disease onset remains to be clarified. Leveraging human pluripotent stem cell-derived brain organoids, we and others are exploring how mHTT affects the developing human brain. These models reveal impaired neural progenitor organization and function, accompanied by a mitochondrial stress response, indicating reduced capacity to manage cellular stress. Enhancing mi

BDNF Mitigates Early Oxidative Stress and Promotes Release of Neuroprotective Mitochondria From Striatal Astrocytes of zQ175 Huntington's Disease Mice.

Huntington's disease (HD) is a neurodegenerative disorder characterized by mutant huntingtin (mHTT) aggregation, striatal neuron degeneration, astrocyte dysfunction, and accompanied by oxidative stress and mitochondrial impairment. We previously demonstrated that the neurotrophin brain-derived neurotrophic factor (BDNF) exerts antioxidant and anti-inflammatory effects in cultured rat astrocytes. Given the critical role of astrocytes in maintaining redox homeostasis, we investigated oxidative stress in cortical and striatal astrocytes from the zQ175 knock-in mouse model of HD and examined the r

The Evolving Landscape of Stem Cell Therapies for Huntington's Disease.

Huntington's disease (HD) is a fatal, inherited neurodegenerative disorder marked by the progressive and selective loss of spiny projection neurons (SPNs), resulting in a characteristic triad of motor, cognitive, and psychiatric symptoms. Despite ongoing research, no disease-modifying treatments are available, and existing therapies are limited to symptomatic relief. Stem cell-based approaches represent a promising avenue to restore striatal circuitry by replacing lost neurons and/or delivering trophic support to preserve the remaining neural tissue. In this review, we present a critical analys

Consolidation deficits in episodic memory define distinct clinical and neurodegenerative profiles in Huntington's disease.

BACKGROUND: Huntington's disease (HD) is primarily associated with executive dysfunction, but episodic memory impairment is also present. Traditionally, these memory deficits have been attributed to retrieval difficulties linked to fronto-striatal dysfunction, rather than to disruptions in encoding or consolidation processes. However, the specific nature and diversity of memory impairments in HD remain underexplored. OBJECTIVE: To characterize the profile of episodic memory impairment in HD, identify distinct cognitive phenotypes, and examine their clinical, neuroanatomical, and biomarker cor

Dendritome mapping reveals the spatial organization of striatal neuron morphology.

Dendritic arbors are essential for neuronal computation and signal propagation, yet large-scale single-neuron morphology studies remain challenging. Here we present a systems biology approach, termed 'dendritome mapping', to profile the dendritic morphology of genetically defined single neurons in mice, unveiling striatal medium spiny neuron (MSN) morphological territories and aging-associated or disease-associated alterations. We generated 3,762 three-dimensional-reconstructed and reference-atlas-mapped striatal D1-type and D2-type MSNs, revealing distinct impacts of D1/D2 genotypes and stria

Bromelain Alleviates 3-Nitropropionic Acid-Induced Huntington's Disease-Like Symptoms in Rats Through the Interplay of SIRT1/PGC-1α and FOXO3a Pathways.

Bromelain, a combination of pineapple-derived enzymes, has demonstrated neuroprotective effects owing to its antioxidant and anti-inflammatory attributes. The current study intended to explore bromelain's neuroprotective impact against 3-nitropropionic acid (3-NP)-induced Huntington's disease (HD)-like symptoms in rats. Four groups of rats were randomly allocated: group 1 received saline; bromelain (40 mg/kg/i.p.) was given to groups 2 and 4 daily, whereas groups 3 and 4 received daily doses of 3-NP (10 mg/kg/i.p.) for 14 days. On the molecular level, bromelain administration resulted in upreg

Psychosocial Impacts of Huntington's Disease on Individuals, Relatives and Family Systems: A Thematic Synthesis.

Huntington's disease (HD) has prevalent, life-altering consequences for affected individuals, relatives, familial caregivers and systemic functioning. However, the shared psychosocial impacts of HD across family systems are inadequately understood, and a synthesis of evidence regarding these experiences is currently lacking. This thematic synthesis provides an up-to-date integration of qualitative research describing psychological, social and relational difficulties experienced by HD families. A systematic search across PsycINFO, CINAHL, MEDLINE and Scopus identified nine qualitative studies.

Prevalence of living alone with dementia and other progressive neurological conditions: findings from primary care data in England.

BACKGROUND: Living alone with a neurodegenerative condition such as dementia may result in inequalities in access to support and resources. However, estimates of the number of people living alone vary widely in research studies. We aimed to estimate the prevalence of living alone with dementia, Parkinson's disease, motor neurone disease (MND), and Huntington's disease in England using electronic healthcare records. METHODS: Individuals identified in the Clinical Practice Research Datalink (CPRD) database were categorised as living alone or living with others based on medical codes and the num
