From Patient Selection to Surveillance: Artificial Intelligence Applications in Radioiodine Therapy - A Systematic Review.

UNLABELLED: Dietary phytochemicals exhibit anticancer activity, and increasing evidence suggests that the combinations of co-occurring compounds may provide greater therapeutic efficacy than individual compounds. Eruca sativa Mill. (rocket salad) contains both erucin and kaempferol; however, their synergistic anticancer activity against non-small cell lung cancer (NSCLC) has not been systematically investigated. Network pharmacology, molecular docking, and molecular dynamics (MD) simulations were performed to identify molecular targets of erucin and kaempferol. Their synergistic anticancer act

MicroRNAs in polyglutamine diseases: Mechanistic insights, circulating biomarkers, and emerging microRNA-based therapeutic strategies.

MicroRNAs (miRNAs) have emerged as critical regulators in the pathogenesis of polyglutamine (PolyQ) diseases-a group of fatal neurodegenerative disorders caused by CAG repeat expansions, such as Huntington's disease, spinocerebellar ataxias, dentatorubral-pallidoluysian atrophy, and spinal and bulbar muscular atrophy. This review synthesizes recent advances in miRNA dysregulation across all nine PolyQ diseases, focusing on studies published since 2019. We examine how specific miRNAs modulate core pathogenic cascades-including mutant protein aggregation, transcriptional dysregulation, mitochond

Protection of multiple aspects of Alzheimer's disease pathology using dietary supplementation with taurine.

As Alzheimer’s disease (AD) continues to rise amongst the aging population, preventative measures such as dietary or lifestyle changes represent an attractive option to mitigate the burden. Taurine, known for its antioxidant and anti-inflammatory properties, may also play a neuroprotective role. This study investigates the protective effects of taurine supplementation in 5xFAD mice. Taurine was administered through drinking water at doses of 0, 500, 1000, 2000, 4000 mg/kg/day, with no change in water consumption or body mass was observed. Postmortem markers of neuroinflammation using cytokine

CoxMDS: multiple data splitting for high-dimensional mediation analysis with survival outcomes in epigenome-wide studies.

Causal mediation analysis investigates whether the effect of an exposure on an outcome operates through intermediate variables known as mediators. Although progress has been made in high-dimensional mediation analysis, current methods do not reliably control the false discovery rate (FDR) in finite samples, especially when mediators are moderately to highly correlated or follow non-Gaussian distributions. These challenges frequently arise in DNA methylation studies. We introduce CoxMDS, a multiple data splitting method that uses Cox proportional hazards models to identify putative causal media

Implant Therapy in Patients With Neurodegenerative Diseases-A Scoping Review.

OBJECTIVES: The increasing longevity of populations has resulted in a growing number of older adults requiring prosthodontic care, including those with neurodegenerative diseases (NDs). Neurodegenerative diseases pose significant challenges to prosthodontic care, and it remains unclear whether implant therapy in this population achieves outcomes comparable to those observed in the general older population. MATERIALS AND METHODS: A first systematic review was reframed to a scoping review using a PCC framework with Population (individuals with neurodegenerative diseases, who were partially or c

Evaluating Alzheimer's disease with the TMS-EEG perturbation complexity index.

The Perturbation Complexity Index-State Transitions (PCIST) measures the complexity of the brain's response to transcranial magnetic stimulation (TMS) using electroencephalography (EEG) and is sensitive to consciousness, such as minimally conscious states. Individuals with early-stage Alzheimer's disease (AD) show dysfunction of conscious processes, such as attention, working memory, episodic memory, and executive function, with relatively spared unconscious processes, such as procedural memory, operant conditioning, and priming. We sought to test the hypothesis that PCIST would be reduced in

From Cerebrovascular Injury to Vascular Cognitive Impairment and Dementia: Therapeutic Potential of Stem Cell-Derived Extracellular Vesicles.

Vascular cognitive impairment and dementia (VCID) encompass a spectrum of cognitive syndromes ranging from mild cognitive impairment to vascular dementia, accounting for approximately 15-20% of all dementia cases and representing the second most common form of dementia. Despite its high prevalence and clinical burden, effective therapeutic strategies remain lacking. Increasing evidence indicates that vascular dysfunction plays a central role in the pathogenesis of VCID by compromising cerebrovascular integrity, impairing endothelial function, and disrupting neurovascular coupling, which collec

APP-mediated intracellular signaling rescues sleep impairment and blood-brain barrier leakage in Alzheimer's disease mouse model.

INTRODUCTION: Amyloid beta peptide (Aβ) accumulation in the brain is an Alzheimer´s disease (AD) hallmark. Sleep disturbances hamper Aβ production and clearance, thereby exacerbating the Aβ burden. The mechanisms involved remain unclear. We reported that amyloid precursor protein (APP), the Aβ source, possesses intracellular signaling that attenuates Aβ production and prevents cognitive decline in AD mice. Our follow-up study assessed whether enhancing APP-mediated signaling affected sleep. METHODS: We expressed a membrane-tethered APP intracellular domain (mAICD) and a variant lacking the Gα

The Neuroprotective Effects of Agomelatine in a 3‑Nitropropionic Acid-Induced Rat Model of Huntington's Disease via Brain-Derived Neurotrophic Factor (BDNF)/tropomyosin-Related Kinase Receptor Type B (TrKB)/Phosphoinositide 3‑Kinase (PI3K)/Protein Kinase B (AKT) Activation and Rho-Associated Protein Kinase 1 (ROCK1) Pathway Inhibition.

The accumulation of CAG nucleotide duplicates in the huntingtin (HTT) gene triggers a neurological ailment described as Huntington's disease (HD), which is an irreversible, progressive, and inherited condition and affects both motor and cognitive abilities, resulting in a range of symptoms, including irregular gestures (chorea, dyskinesia), psychological disorders, and advanced dementia. Agomelatine is a novel antidepressant and melatonin analog. It exerts a synergistic pharmacological mechanism, combining stimulation of both MT1/MT2 melatonergic receptors with inhibition of 5-HT2C receptors.

Global Recognition of Traumatic Brain Injury as a Chronic and Notifiable Condition: A Post-WHA78 Advocacy Commentary.

BACKGROUND: Traumatic brain injury (TBI) is a leading cause of disability but one of the least recognized health problems in the world, affecting up to 69 million people annually. The associated lifelong disability in survivors, the loss of economic productivity, and being a risk factor for dementia consume 0.5% of global economic activity. Yet TBI is still largely invisible in national surveillance systems and not well represented in chronic disease frameworks. Consequently, governments are not equipped to provide proportional financing of acute care and long-term care of survivors, nor to bu

Neurovascular Impairment in Type 2 Diabetes Mellitus: The Role of Adipocyte-Derived Exosomes.

Type 2 diabetes mellitus (T2DM) is a major metabolic disorder characterized by chronic hyperglycemia with far-reaching morbidities. Among these, diabetes-related cerebrovascular complications such as ischemic and hemorrhagic stroke, cerebral blood vessel disease, and vascular dementia are significant contributors to morbidity and mortality. Adipose tissue is a metabolically active endocrine organ that becomes dysfunctional in T2DM and communicates with distant tissues via secreted factors, including extracellular vesicles such as exosomes (EXs), phospholipid bilayer-enclosed nanosized particle

Variant Prioritization by Pedigree-Based Haplotyping.

Whole genome sequence (WGS) data provides opportunities for comprehensive evaluation of variants that may influence complex traits. However, prioritizing the large number of variants, particularly those in non-coding regions, is a challenge. Here we present an approach that uses pedigree-based haplotyping to identify the risk haplotype and resulting set of prioritized variants in a region of interest (ROI) defined by identity-by-descent (IBD) sharing among familial cases. The approach is applicable for use in both a full range of pedigree sizes and for the full allele frequency spectrum of var

Statistical analysis of correlated expression data from high throughput experiments.

Data obtained from high throughput experiments often exhibit complex dependencies among features. These dependencies arise from various sources, including genetic correlation, batch effects, technical replicates, and shared biological pathways. Ignoring these dependencies can lead to inflated false discovery rate (FDR), reduced statistical power, and biased biological interpretations. Properly accounting for these dependencies is crucial for accurate detection of biological signals. We propose a new method called Analysis of Correlated Expressions (ACE) to compare the mean expression of featur

Target product profiles for treatments to delay or prevent symptomatic Alzheimer's disease.

Despite advances in understanding the mechanisms, risk factors and treatment strategies for Alzheimer's disease (AD), no approved therapies exist to prevent or delay onset in at-risk individuals or those with elevated biomarkers who do not yet show symptoms. Multiple candidate interventions are now being evaluated in clinical trials in these settings, raising key questions around which populations are most appropriate and what criteria should guide regulatory and clinical decision-making. Data are expected within 1-2 years, underscoring the need for stakeholder alignment on clinically meaningf
