BACKGROUND: Subjective cognitive decline (SCD) may represent an early stage of cognitive impairment. Although poor oral health has been linked to systemic inflammation and dementia, its association with SCD remains unexplored. This study investigated the relationship between poor oral health, delayed dental care, and SCD in a nationally representative sample of U.S. adults. METHODS: We analyzed data from 94,812 adults aged ≥ 45 years using the 2020–2022 Behavioral Risk Factor Surveillance System (BRFSS). Poor oral health was defined as loss of ≥ 6 teeth; delayed dental care was defined as no d

INTRODUCTION: Synapse loss, a key feature of behavioral variant frontotemporal dementia (bvFTD), can now be visualized in vivo using positron emission tomography (PET) that targets synaptic vesicle glycoprotein 2A (SV2A). Comparing this new imaging method with the clinical standard of glucose metabolism using 1 8F-fluorodeocyglucose (1 8F-FDG) PET might improve diagnostic accuracy and disease monitoring in bvFTD.
METHODS: We measured synaptic vesicle glycoprotein 2A (SV2A) binding using 1 8F-SynVesT-1 PET and glucose metabolism with 1 8F-FDG PET in 10 patients with bvFTD and 10 age-matched he

Chorea can arise from genetic, metabolic, pharmacologic, and autoimmune causes. In clinical practice, however, non-genetic causes are rare. The most common genetic cause is a CAG repeat expansion in HTT, leading to Huntington's disease (HD). Beyond HD, systematic studies have been lacking and many individuals with non-HD chorea remain without a molecular diagnosis. We conducted whole-exome and genome sequencing analysis on 190 non-HD chorea cases, leveraging data from the All of Us Research Program (n = 134), UK Biobank (n = 26), and a clinically ascertained multicenter Spanish cohort recruite

BACKGROUND: Late-onset Alzheimer´s disease (LOAD) is a heterogeneous disorder influenced by complex genetic factors. We previously described intermediate alleles (IAs; 27–35 CAG repeats) in the huntingtin (HTT) gene as potential modifiers in around 6% of AD population. The caudate nucleus, the most affected region in Huntington’s disease, is highly sensitive to these HTT CAG repeats. We hypothesized that HTT IAs induce gene expression deregulation, including altered microRNA (miRNA) profiles, leading to altered disease progression. METHODS: We investigated the impact of HTT IAs on LOAD progres

BackgroundNeuropsychiatric symptoms (NPS) are highly prevalent in dementia. While biomarkers reflecting amyloid, tau, neuronal injury, and glial activation are increasingly used in dementia research, their association with NPS remains unclear.ObjectiveTo identify and characterise associations between biomarkers and NPS in people with young-onset dementia (YOD).MethodsWe conducted a systematic review and meta-analysis of studies examining associations between validated biomarkers and NPS in individuals with Alzheimer's disease (AD), frontotemporal dementia (FTD), Huntington's disease (HD), and

Frontotemporal dementia (FTD) is an early onset dementia characterized by neuropathology and changes to patient behaviour. Haploinsufficiency of the gene progranulin (GRN) is a major cause of FTD, for which there are no effective therapies. Corrective gene therapies that restore GRN expression are of clinical interest, but current in vivo systems have limitations. We developed a novel strain of mice expressing a human GRN transgene bearing a four base pair deletion in exon 5 (GRNc.388_391delCAGT) that causes FTD. Characterization of mice expressing the mutant transgene (GRNmEx5) indicates that

Aging is the predominant risk factor for neurodegenerative diseases, yet the mechanisms linking biological aging to selective neuronal degeneration remain incompletely understood. Accumulating evidence indicates that aging progressively disrupts epigenetic regulation, manifested as increased epigenetic noise in DNA methylation, histone modifications, and chromatin accessibility, which undermines transcriptional precision and the stability of neuronal identity. Recent advances in single-cell and spatial epigenomics further suggest that these age-associated epigenetic alterations are not merely

Neurodegenerative diseases are increasingly recognized as disorders associated with metabolic dysfunction with arginine metabolism emerging as a significant contributor. Arginase, by regulating the balance between arginine and ornithine, is positioned at the crossroads of multiple arginine metabolic pathways, thereby controlling a variety of cellular processes essential for proper brain homeostasis. Chronic disruption of these pathways may lead to dysfunction of neurons and glia ultimately resulting in the induction of neurodegenerative processes. In this review, based on data from patients an

Neurodegenerative diseases (NDDs) represent a growing cause of mortality and disability among aging populations worldwide. However, the relationship between cumulative, multidomain environmental exposures, and NDD mortality remains poorly characterized. This study aimed to examine the association between comprehensive environmental quality and NDD mortality in the United States. We used the environmental quality index (EQI) from 2000 to 2005 to characterize cumulative environmental exposure. Associations between EQI quintiles and NDD mortality, including dementia, Parkinson's disease (PD), amy

Alzheimer's disease (AD) is a common age-related neurodegenerative disorder with extremely low drug development success rates, making nutritional intervention a promising strategy. Cerebral energy metabolism dysfunction is a core pathological feature of AD. Odd-chain fatty acids (OCFAs) can generate propionyl-CoA via β-oxidation to replenish the impaired tricarboxylic acid (TCA) cycle. This study characterized the lipid composition of OCFAs-enriched algal oil by UPC2-Q-TOF-MS, evaluated its neuroprotective effects on Caenorhabditis elegans (C. elegans) models with AD, Parkinson's disease (PD),

BACKGROUND: Rare neurological diseases often present with severe cognitive impairment but face significant challenges in access to specialized care. Telemonitoring and telerehabilitation are promising approaches to improve the standard of care in common neurological diseases and could prove even more beneficial to patients with rare neurological diseases.
AIM: To review the available evidence on the usability and validity of cognitive telemonitoring or telerehabilitation in rare neurological diseases.
METHODS: We performed a scoping review, examining all studies published in the last 25 year

Alzheimer's disease (AD) causes a characteristic spatiotemporal pattern of neurodegeneration. The factors that account for this pattern of degeneration, including associated neuronal dysfunction and dysmorphia, are unclear. Previously, we generated a model of AD using the nematode Caenorhabditis elegans with the AD risk variant of apolipoprotein E (APOE4). We showed that the soma of HSN class neurons degenerate in early-adult animals. Here, we performed behavioral analyses to deduce the effect of APOE4 on the function of distinct neuronal circuits. We found evidence that APOE4 induces dysfunct

Age-related macular degeneration (AMD) is caused by the degeneration of photoreceptors and retinal pigment epithelium (RPE) along with drusen deposition and is the leading cause of vision loss in older adults. Both these structures within the central nervous system (CNS) utilize common neuro-inflammatory mechanisms because the retina is an outgrowth of the brain. Like the brain, the eye has its own physical characteristics and surface molecules as well as a tendency towards specific immune reactions. Numerous distinct neurodegenerative diseases like Alzheimer's disease (AD), Parkinson's diseas

Recent genetic studies have shown somatic expansion of the CAG repeat is the key process driving Huntington's disease (HD) pathogenesis. Recognition of insertion-deletion loops (IDLs), lesions prone to form within the CAG repeat, by Mutsβ (MSH3/MSH2) is thought to be the primary event in the expansion process. This starts a cascade that leads to error-prone repair and incorporation of additional CAG units into the repeat. In vitro data shows MSH3 binds IDLs through a DNA-binding pocket formed by MSH3 residues Y245/K246. In this study, we investigated the significance of this DNA-binding motif

Biomarker-based technologies for predicting age of onset are currently being developed for carriers and individuals at 50% risk of autosomal dominant neurodegenerative diseases such as Huntington's disease, frontotemporal dementia and spinocerebellar ataxias. Qualitative interview studies indicate that carriers and individuals at 50% risk expect that onset predictions would be valuable for life planning but would also impact mental health. Ethically responsible implementation of biomarker-based onset prediction in research and clinical settings requires maximizing the utility and minimizing ne

INTRODUCTION: Positron emission tomography (PET) without usable or accompanying magnetic resonance imaging (MRI) is typically excluded in quantitative analyses of Alzheimer's disease, potentially limiting study generalizability. We investigated participant features predicting data exclusion in magnetic resonance (MR)-dependent analyses and evaluated an existing MR-free PET pipeline to quantify these missing data.
METHODS: Imaging, clinical, cognitive, and sociodemographic data were analyzed for 2119 individuals in a multi-site cohort. Agreement between MR-dependent and MR-free Centiloids (CL)

The blood-brain barrier (BBB) plays a central role in brain function and is increasingly implicated in neurodegenerative disease. Major neurodegenerative disorders, including Alzheimer's disease (AD), frontotemporal dementia (FTD), and Huntington's disease (HD), share overlapping pathological features. Yet, the extent to which these diseases converge or diverge at the level of BBB-associated cell types remains poorly understood. Here, we performed a comparative analysis of vessel-enriched human brain transcriptomic datasets across AD, FTD, and HD to define shared and disease-specific neurovasc
