Unexpected off-targeting effects of anti-huntingtin ribozymes and siRNA in vivo.

Gene transfer strategies to reduce levels of mutant huntingtin (mHtt) mRNA and protein by targeting human Htt have shown therapeutic promise in vivo. Previously, we have reported that a specific, adeno-associated viral vector (rAAV)-delivered short-hairpin RNA (siHUNT-2) targeting human Htt mRNA unexpectedly decreased levels of striatal-specific transcripts in both wild-type and R6/1 transgenic HD mice. The goal of this study was to determine whether the siHUNT-2-mediated effect was due to adverse effects of RNA interference (RNAi) expression in the brain. To this end, we designed two catalyti

Noninvasive measurement of protein aggregation by mutant huntingtin fragments or alpha-synuclein in the lens.

Many diverse human diseases are associated with protein aggregation in ordered fibrillar structures called amyloid. Amyloid formation may mediate aberrant protein interactions that culminate in neurodegeneration in Alzheimer, Huntington, and Parkinson diseases and in prion encephalopathies. Studies of protein aggregation in the brain are hampered by limitations in imaging techniques and often require invasive methods that can only be performed postmortem. Here we describe transgenic mice in which aggregation-prone proteins that cause Huntington and Parkinson disease are expressed in the ocular

RNA-binding protein TLS is a major nuclear aggregate-interacting protein in huntingtin exon 1 with expanded polyglutamine-expressing cells.

Formation of intracellular aggregates is the hallmark of polyglutamine (polyQ) diseases. We analyzed the components of purified nuclear polyQ aggregates by mass spectrometry. As a result, we found that the RNA-binding protein translocated in liposarcoma (TLS) was one of the major components of nuclear polyQ aggregate-interacting proteins in a Huntington disease cell model and was also associated with neuronal intranuclear inclusions of R6/2 mice. In vitro study revealed that TLS could directly bind to truncated N-terminal huntingtin (tNhtt) aggregates but could not bind to monomer GST-tNhtt wi

Bradykinesia is not a "systematic" feature of adult-onset Huntington's disease; implications for basal ganglia pathophysiology.

Our goal was to determine whether bradykinesia is present in choreic adult-onset Huntington's disease (HD) patients, and determine the impact of chorea on their voluntary movements. We recorded whole-body involuntary movements (WBIM) and voluntary motor acts simultaneously, using a magnetic tracker system, in 15 choreic HD patients and 15 healthy age- and gender-matched control subjects. Participants were asked to perform two distinct tasks; a manual-tracking (MT) task yielding a measure of chorea intrusion during accurate movements, and a rapid alternating movement (RAM) task, yielding measur

Inositol 1,4,5-tripshosphate receptor, calcium signalling and Huntington's disease.

Huntington's disease (HD) is an autosomal-dominant neurodegenerative disorder that has no cure. HD primarily affects medium spiny striatal neurons (MSN). HD is caused by polyglutamine (polyQ) expansion (exp) in the amino-terminal region of a protein huntingtin (Htt). The connection between polyQ expansion in Htt(exp) and MSN neurodegeneration remains elusive. My laboratory discovered that mutant Htt(exp) protein specifically binds to the carboxy-terminal region of the type 1 inositol 1,4,5-trisphosphate receptor (InsP3R1), an intracellular Ca2+ release channel. Moreover, we found that Htt(exp)

Cutting-edge patents in Alzheimer's disease drug discovery: anticipation of potential future treatments.

Neurodegenerative disease broadly includes many different diseases, such as Alzheimer's, Parkinson's, amyotrophic lateral sclerosis, multiple sclerosis, Huntington's, dementias with Lewy bodies, post-traumatic brain injury, and stroke. Although few common physiopathological changes have been discovered among these conditions, the semiology (if known), the triggered molecular pathways that lead to the observed pathologies, and the symptomatology are essentially different. These differences entail that the treatments, both current and future, have disease-specific indications. This idea led us t

fMRI reveals altered auditory processing in manifest and premanifest Huntington's disease.

Structural alterations of the basal ganglia occur in patients with Huntington's disease (HD). The aim of this exploratory study was to assess auditory processing mechanisms by functional MRI (fMRI) in patients with premanifest (pHD) and manifest HD to gain more insight in possible alterations in basal ganglia-thalamic circuits. Sixteen HD and 18 pHD as well as corresponding age- and gender-matched controls were included. The pHD group was divided into two subgroups close (cpHD; <10 years) and far pHD (fcHP; >10 years), according to their estimated age of disease onset (eAO). Tone perception an

Response inhibition in Huntington's disease-a study using ERPs and sLORETA.

Huntington's disease (HD) is an autosomal dominant inherited neurodegenerative disorder, with neurodegeneration mainly affecting the striatum. We investigated executive functions related to response inhibition in (HD) and healthy controls by means of event-related potentials (ERP) in a simple Go/Nogo-task. In Nogo as opposed to Go trials two fronto-central ERP components are elicited: the Nogo-N2 and Nogo-P3. These components are supposed to depend on (medial) prefrontal regions, especially the anterior cingulate cortex (ACC). The results show that the Nogo-N2 did not differ between the groups

Orienting of spatial attention in Huntington's Disease.

To explore the functioning of spatial attention in Huntington's Disease (HD), 14 HD patients and 14 age-matched controls performed a cued response time (RT) task with peripheral cues. In Experiment 1, cues were not informative about the future target location, thus eliciting a purely exogenous orienting of attention. At short stimulus-onset asynchrony (SOA), controls showed an initial facilitation for cued locations, later replaced by a cost (inhibition of return, IOR). Patients had a larger and more persistent validity effect, with delayed IOR, resulting from a larger cost for uncued targets.

[Mechanisms of neuronal death in Huntington's disease. Second part: therapeutic challenges].

Huntington's disease is caused by an abnormal CAG expansion within the gene encoding Huntingtin which induces a major cortico-striatal degeneration as well as motor and cognitive impairments. Since the discovery of the present mutation, a number of experimental data have been collected to uncover the physiopathological consequences of mutated Huntingtin expression. Here, we review the therapeutic challenges of Huntington's disease.

Role of Nrf2-dependent ARE-driven antioxidant pathway in neuroprotection.

The promoter regions of many detoxification enzymes contain a cis-acting enhancer known as the antioxidant response element (ARE). NF-E2-related factor 2 (Nrf2) is considered as one of the major transcription factors for the ARE. Nrf2-dependent transcriptional activation by means of the ARE is known to coordinate the upregulation of these antioxidant enzymes involved in combating oxidative stress and has been shown to be protective against neural toxicants. The mitochondrial complex II inhibitor malonate causes striatal damage reminiscent of Huntington's disease and is known to involve oxidati

Cortical expression of brain derived neurotrophic factor and type-1 cannabinoid receptor after striatal excitotoxic lesions.

An involvement of one particular neurotrophin, namely, the brain-derived neurotrophic factor (BDNF), has been demonstrated in the pathophysiology Huntington's disease. Type-1 cannabinoid (CB1) receptor has been postulated to upregulate BDNF gene transcription. To better understand the relationship between CB1 and BDNF levels in a situation where the striatum is degenerating, we studied, by dual label immunofluorescence, the distribution of CB1 and BDNF in cortical neurons projecting to the striatum in our rat quinolinic acid model of striatal excitotoxicity. We completed our study with quantit

NF-kB/NOS cross-talk induced by mitochondrial complex II inhibition: implications for Huntington's disease.

Nuclear factor-kB (NF-kB) is a family of DNA-binding proteins that are important regulators involved in immune and inflammatory responses, as well as in cell survival and apoptosis. In the nervous system NF-kB is activated under physiological and pathological conditions including learning and memory mechanisms and neurodegenerative diseases. NF-kB is activated in neurons in response to excitotoxic, metabolic and oxidative stress and there is a body of evidence to suggest that glutamate induces NF-kB by the main ionotropic glutamate receptors. In the present study, 3 nitroproprionic acid (3NP),

Hypocretin-1 and secondary signs in Huntington's disease.

Huntington's disease is characterised by unwanted movements, psychiatric disturbances and cognitive decline. Less well recognised symptoms and signs are weight loss, autonomic dysfunction and sleep disorders. In this study we focus on hypocretin-1 and hypothalamus functions. We found a reduction by about 30% in hypocretin signalling in patients with HD. However, it remains unclear whether this moderate decrease in hypocretin signalling contributes to clinical symptoms.

Commentary on "Duplex-guided balloon angioplasty and stenting for occlusive and stenotic lesions of the infrainguinal arteries".

Alois Alzheimer is known for his seminal work in recognizing a form of presenile dementia. His early interests were natural history and botany. He started his medical education in Berlin and attended the universities of Wurzburg and Tübingen. Nissl and Alzheimer worked together on extensive investigation of the pathology of the nervous system, especially the anatomy of the cerebral cortex. In 1902 Emil Kraepelin invited Alzheimer to work with him in the university psychiatric clinic in Heidelberg. In 1903 both moved to work in the university psychiatric clinic in Munich. It was during these ye

Using the Best Interests Standard to decide whether to test children for untreatable, late-onset genetic diseases.

A new analysis of the Best Interests Standard is given and applied to the controversy about testing children for untreatable, severe late-onset genetic diseases, such as Huntington's disease or Alzheimer's disease. A professional consensus recommends against such predictive testing, because it is not in children's best interest. Critics disagree. The Best Interests Standard can be a powerful way to resolve such disputes. This paper begins by analyzing its meaning into three necessary and jointly sufficient conditions showing it: 1. is an "umbrella" standard, used differently in different conte

Detection of Huntington's disease decades before diagnosis: the Predict-HD study.

OBJECTIVE: The objective of the Predict-HD study is to use genetic, neurobiological and refined clinical markers to understand the early progression of Huntington's disease (HD), prior to the point of traditional diagnosis, in persons with a known gene mutation. Here we estimate the approximate onset and initial course of various measurable aspects of HD relative to the time of eventual diagnosis. METHODS: We studied 438 participants who were positive for the HD gene mutation, but did not yet meet the diagnostic criteria for HD and had no functional decline. Predictability of baseline cogniti
