A Network Analysis of Anxiety-Depression Symptoms Among Chinese Adolescents: Similarities and Differences Between Problematic and Non-Problematic Internet Users.

Problematic Internet use is recognized as a significant public health concern among adolescents. Previous research has shown that adolescents with problematic Internet use tend to experience higher levels of anxiety and depression compared to non-problematic Internet users. Nevertheless, when it comes to differences in specific symptoms, the evidence is less clear-cut. This study conducted Gaussian graphical model and Bayesian network analysis to compare the network structures of anxiety symptoms and depression symptoms among adolescents with problematic and non-problematic Internet use. Composite grey matter fingerprints for genetic frontotemporal dementia.

BACKGROUND: Brain structural changes in frontotemporal dementia (FTD) can occur decades before symptom onset. Precise characterisation of grey matter changes is necessary for developing models of biomarker progression, while better understanding the trajectory of the pathology is invaluable for prognosis and detecting treatment effects as we enter the era of clinical trials. METHODS: Cortical and subcortical grey matter volume and thickness from structural MRI were assessed in a large cohort of 892 participants including presymptomatic and symptomatic carriers of mutations within the three ma

Living with an osteoporotic vertebral compression fracture: a qualitative exploration of patient and healthcare professional perspectives.

UNLABELLED: Osteoporotic spine fractures deeply affect patients' lives, yet little is known about patients' lived experiences. This qualitative study found that patients face delayed diagnosis, inconsistent care, and emotional distress, while professionals call for clearer communication and coordination. Results highlight the urgent need for more consistent, patient-centered osteoporosis care. PURPOSE: Osteoporotic vertebral compression fractures (OVCFs) are common and often debilitating, yet patient perspectives remain underexplored. This study aimed to investigate the lived experiences of p

From Evasion to Collapse: The Kinetic Cascade of TDP-43 and the Failure of Proteostasis.

Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are devastating neurodegenerative diseases that, despite the availability of symptomatic and modestly beneficial treatments, still lack therapies capable of halting disease progression. A histopathological hallmark of both diseases is the cytoplasmic deposition of TDP-43 in neurons, which is attributed to both intrinsic (e.g., mutations, aberrant cleavage) and extrinsic factors (e.g., prolonged oxidative stress, impaired clearance pathways). Mutations and certain PTMs (e.g., cysteine oxidation) destabilize RNA binding, promo

MTHFR and MTRR Polymorphisms Predict Sex-Dependent Psychotic Symptom Improvements, Not Metabolic Changes.

We investigated whether antipsychotic treatment response was influenced by the C677T and A1298C polymorphisms of methylenetetrahydrofolate reductase (MTHFR), and A66G of methyltetrahydrofolate-homocysteine methyltransferase reductase (MTRR)-genes central to folate and homocysteine metabolism and methylation, pathways often altered in schizophrenia patients. To our knowledge, no study has examined associations of C677T and A1298C with changes in schizophrenia symptom severity after antipsychotic treatment, while studies on metabolic outcomes remain sparse and inconsistent. The MTRR A66G has bee

Feasibility of machine learning classification of depression and anxiety symptoms among college students using 3D gait and sit-to-walk biomechanics.

BACKGROUND: Depression and anxiety are common and burdensome. Objective movement assessment may complement questionnaires by providing low burden screening signals. RESEARCH QUESTION: Can 3D gait and sit-to-walk biomechanics classify elevated depression and anxiety symptoms, and which movement yields better performance with few features? METHODS: Thirty young adults completed the Center for Epidemiologic Studies Depression Scale (CES-D) and the Generalized Anxiety Disorder scale (GAD-7), then performed gait and sit-to-walk in a laboratory with a sixteen camera optoelectronic motion capture s

Neurobiological mechanisms of electroconvulsive therapy in major depressive disorder: structure-function coupling with gene expression and molecular mechanism.

Major Depressive Disorder (MDD), a common and debilitating mental disorder, contributes considerably to global disease burden by increasing rates of comorbid physical illnesses, functional impairment, and suicide risk. Despite a spectrum of therapeutic modalities, a significant proportion of patients, particularly those with treatment-resistant depression, fail to achieve adequate symptom remission. Electroconvulsive Therapy (ECT) remains a pivotal intervention for refractory cases and is characterized by its rapid antidepressant efficacy; however, the underlying neurobiological mechanisms rem

Network analysis of problematic smartphone use, depression, and anxiety, and their relationships with mindfulness among medical students.

Previous studies have uncovered a negative correlation between problematic smartphone use (PSU) and both depression and anxiety, yet the investigation of PSU with depression and anxiety on a symptomatic level among medical students remains limited. This study aims to analyze the network structure of PSU and its relation to symptoms of depression and anxiety, as well as examine the role of mindfulness among these factors. A sample of 456 medical students was assessed using the Smartphone Addiction Scale - Short Version (SAS-SV), the nine-item Patient Health Questionnaire (PHQ-9), the seven-item

Enacted Restoration of Selfhood: A Kierkegaardian Perspective on Self-harm Among People with Mental Illness.

Non-suicidal self-harm, particularly the phenomenon of cutting, is gaining increasing academic attention. However, most existing literature approaches this topic from clinical, cultural, or historical perspectives, often neglecting an in-depth exploration of the lived experiences of individuals engaging in self-harm. Notably, within the context of mental illnesses, self-harm is often viewed merely as a symptomatic behavior and consequently addressed solely through symptomatic treatments. This article draws on data from the first and last authors' extensive projects focusing on individuals livi

Ganaxolone as a promising therapeutic agent for Alzheimer's disease: signaling pathways and mechanistic insights-a narrative review.

Alzheimer’s disease (AD) is a relentless neurodegenerative disorder characterized by progressive cognitive decline, memory loss, and synaptic failure. Its complex pathology involves amyloid-β plaque accumulation, tau hyperphosphorylation, chronic neuroinflammation, and oxidative stress hallmarks that drive neuronal dysfunction and death. Despite decades of research, current therapies deliver only modest symptomatic relief, underscoring the urgency for innovative strategies. Drug repurposing has emerged as a promising shortcut in this search, offering accelerated development timelines and estab

A brain-enriched circRNA blood biomarker can predict response to SSRI antidepressants.

Major Depressive Disorder (MDD) is a debilitating psychiatric disorder that is a leading cause of disability worldwide. Although treatment with antidepressants, such as Selective Serotonin Reuptake Inhibitors (SSRIs), has demonstrated clinical efficacy, the "trial and error" approach in choosing the most effective antidepressant treatment for each patient allows for only a subset of patients to achieve response to the first line of treatment. Circular RNAs (circRNAs), are highly stable and brain-enriched non-coding RNAs that are mainly derived from the backsplicing and covalent joining of exon

Facial emotion recognition deficits in different stages of schizophrenia: A comparative cross-sectional study.

BACKGROUND: The relationship of facial emotion recognition deficits (FERD) with the stage, severity, and treatment of schizophrenia is unclear. This study aimed to assess FERD across acute drug-free, chronic symptomatic, and asymptomatic stages of schizophrenia and its correlation with attention deficits, positive, and negative symptoms. METHODS: This was a cross-sectional study with 132 subjects. The patient group (n = 99) was divided into three groups: Group A: drug-free symptomatic acute schizophrenia (n = 33), Group B: chronic symptomatic schizophrenia (n = 33), Group C: chronic asymptoma

Classification and Treatment of Long-standing (Severe and Enduring) Anorexia Nervosa.

Long-standing anorexia nervosa is presently conceptualized as a persistent symptomatic state associated with global functional and interpersonal impairments. Understanding of effective treatments is incomplete; psychological therapy is at the core, but also important are psychosocial and recovery support that includes significant others, tailored treatment targets such as improved quality of life, a collaborative and flexible approach to higher levels of care, and pharmacologic agents that augment therapy and address concurrent mental health problems. Research is needed to develop agreed-upon

Early motor deficits, sleep dysfunction and reduction in dopaminergic neurons in a PARK7-/- zebrafish larval model of Parkinson's disease.

Parkinson’s Disease (PD) is the fastest-growing neurological disorder and only symptomatic treatment is available. Zebrafish are ideally suited for high-throughput screening of disease modifying drugs and mechanistic studies. Mutations in PARK7 are associated with early onset familial PD, however also idiopathic PD patients without known mutations in PARK7 show altered subcellular location and levels of its protein product DJ-1 in pathological tissues. Here, we show that PARK7-/- zebrafish already at their larval stage show a correlation between reduced number of dopaminergic neurons and motor

Current progress in pathway-targeted therapeutics for Alzheimer's disease: mechanistic insights and windows of opportunity.

Alzheimer’s disease (AD) remains a complex neurodegenerative condition characterized by progressive cognitive decline driven by amyloid-beta (Aβ) plaques, tau neurofibrillary tangles, and neuroinflammation. With extensive research, disease-modifying anti-amyloid monoclonal antibodies have now received U.S. FDA approval for early symptomatic disease; however, benefits remain modest and are accompanied by substantial monitoring and implementation constraints.. This review critically evaluates the current landscape of inhibitor-based therapeutics for AD, encompassing both clinical and preclinical
