Metabolite-targeted LC-HRMS quantification of emerging indazole-based synthetic cannabinoids in urine samples from an addiction treatment cohort.

The continuous emergence of structurally diverse indazole-based synthetic cannabinoids (SCs) challenges routine analytical detection because parent compounds are rapidly metabolized and often absent in urine. Therefore, sensitive metabolite-targeted analytical methods are required for reliable exposure assessment. In this study, we adapted and fully validated a targeted high-resolution LC-HRMS method for the quantitative determination of eight contemporary indazole-based SC markers in human urine and applied it to 50 authentic urine specimens from treatment-seeking individuals attending an add

Disruption of Reelin signaling in a dual-hit mouse model of schizophrenia: impact of postnatal Δ9-tetrahydrocannabinol exposure in a maternal immune activation model.

Since the discovery of the first antipsychotic, pharmacological treatment of schizophrenia (SCZ) has primarily relied on dopamine D2 receptor antagonist. However, variability in treatment responses highlights the need for novel biomarkers and therapeutics targets. Here, we developed a dual-hit mouse model combining maternal immune activation (MIA) induced by polyinosinic-polycytidylic acid (Poly(I:C)) with adolescent exposure to Δ9-tetrahydrocannabinol (THC), the main psychoactive component of cannabis. We evaluated the face validity of this model and investigated potential alterations in the

Effects of Δ9-tetrahydrocannabinol on affiliative social behaviour: A systematic review of rodent studies.

INTRODUCTION: Affiliative social behaviour is critical for survival and well-being and is shaped by complex neurobiological systems, including the endocannabinoid system (ECS). Δ9-tetrahydrocannabinol (THC), the primary psychoactive compound in cannabis, is a partial agonist at cannabinoid receptors and directly modulates ECS signaling. Given increasing cannabis use globally, understanding the effects of THC on social functioning is essential. METHODS: We conducted a systematic review of preclinical studies indexed in MEDLINE and PsycINFO to examine the effects of THC on social behaviour in w

Prenatal Δ9-tetrahydrocannabinol exposure rewires glucocorticoid-hippocampal crosstalk and impairs memory under stress in adolescent rat offspring.

BACKGROUND: Stress is a major determinant of psychopathology, with coping and cognitive impairment arising from abnormal glucocorticoid activity and brain-derived neurotrophic factor (BDNF)-dependent neuroplasticity in the hippocampus. The endocannabinoid system modulates stress and hippocampal functioning since early development, thus its early-life disruption may program vulnerability to stress-related cognitive deficits. METHODS: We investigated whether prenatal exposure to tetrahydrocannabinol (pTHC; 2 mg/kg, gestational days 5-20) alters spatial learning and memory in adolescent offsprin

Polysubstance Use Disorders in Individuals with Cannabis Use Disorder: Results from a Nationally Representative Sample (National Epidemiologic Survey on Alcohol and Related Conditions).

OBJECTIVE: Cannabis use disorder (CUD) is one of the most common substance use disorders (SUDs) worldwide and is frequently associated with high rates of polysubstance use; however, despite rising rates of polysubstance use disorders (PUD), the characteristics of individuals with both CUD and PUD remain unclear. This study, therefore, aims to examine social and clinical characteristics of adults diagnosed with CUD and comorbid PUD. It also aims to assess whether psychiatric disorders are linked to higher odds of PUD among individuals with CUD. METHODS: Using a nationally representative U.S. d

Cannabidiol metabolites identified by LC-QTOF after controlled dosing.

Cannabidiol (CBD) is a non-intoxicating cannabinoid found in cannabis and often used for its purported therapeutic benefits. In the form of Epidiolex®, CBD is an FDA-approved treatment for seizure disorders in children. After the 2018 Farm Bill removed hemp (cannabis with <0.3% THC) from the Controlled Substance Act in the United States, non-pharmaceutical CBD became widely available on the retail market. With increased use of CBD, it is important to measure CBD in various biological matrices. In urine, previous studies have measured 7-hydroxy-CBD and 7-carboxy-CBD, analogous to the major meta

Clinical outcomes from a mid-western opioid treatment program during covid-19 emergency regulations: a brief report on the effect of tetrahydrocannabinol (THC) use on take home methadone access.

The study examines whether tetrahydrocannabinol (THC) use alone affected sustainability of weekly take-home methadone medication among a small sample of 33 patients attending a single opioid treatment program, under special exception by state and federal governments during the Covid-19 pandemic. Kaplan Meier analyses of survival rates in months of eligibility for take-home methadone showed that the majority of patients with continuing THC use remained eligible for 10 months despite continued THC use. Majority were employed, insured, and housed stably, but Log Rank Tests on these predictor vari

Self-disturbance in first-episode psychosis: Theoretical framework and potential cannabis interactions - a systematic review.

Cannabis use represents a significant environmental risk factor for psychotic disorders, with emerging evidence suggesting complex interactions between cannabinoid exposure, dissociative experiences, and fundamental disturbances of self-experience (self-disorders) in early psychosis. This systematic review examines the phenomenological and neurobiological relationships among dissociation, cannabis use, and self-disturbance in first-episode psychosis (FEP) patients. Following PRISMA guidelines, we conducted a comprehensive search of four databases (PubMed, Scopus, PsycINFO, Web of Science) from

Adolescent cannabis use and psychological distress from 2013 to 2023: A population-based study in Ontario, Canada.

BACKGROUND AND AIMS: Epidemiologic research suggests that adolescent cannabis use is associated with psychological distress (i.e. depression and anxiety symptoms); however, most studies have relied on 20th-century data, when cannabis was significantly less potent than today. This study aimed to estimate the association between adolescent cannabis use and psychological distress using contemporary population-based data and examine the roles of time [as a proxy for increasing Δ9-tetrahydrocannabinol (THC) potency], sex and age of initiation. DESIGN: Representative cross-sectional survey conducte

Intrinsic brain activity and multiscale mechanisms of auditory verbal hallucinations in schizophrenia: A systematic review and meta-analysis.

Auditory verbal hallucinations (AVH) represent one of the most debilitating symptoms in schizophrenia. The amplitude of low-frequency fluctuation (ALFF) and fractional ALFF (fALFF), derived from resting-state fMRI, serve as robust metrics for intrinsic brain activity; however, the network-level architecture and biological substrates underlying AVH-related ALFF/fALFF alternations have not yet been systematically elucidated. In this study, we conducted a comprehensive systematic review and meta-analysis of ALFF/fALFF studies in schizophrenia patients with AVH, integrating neurochemical mapping a

Resilience and associated factors within the mental health profile of incarcerated adults in Portugal: a cross-sectional study.

BACKGROUND: Mental disorders are widely recognised as a major contributor to morbidity in prisons across the European Union, with evidence consistently indicating a markedly higher mental health burden among incarcerated individuals compared with community populations. Prison research has mainly focused on mental illness, often neglecting protective aspects of mental health from a salutogenic perspective. In Portugal, previous studies have examined specific mental health outcomes in prison populations, such as suicidal behaviour, personality pathology, trauma-related symptoms, or substance use

Cannabinoids and drug-drug pharmacokinetic interactions: Deciphering the risks.

The relationship between cannabinoids and mental health has become a major focus of scientific inquiry and public discourse. Cannabinoids are diverse chemical compounds from the Cannabis species that have been explored for their therapeutic applications in treating chronic pain, psychiatric and neurological conditions, such as depression, schizophrenia, epilepsy and anxiety. Additionally, cannabinoids are frequently used for recreational purposes. The evolving legislation surrounding cannabis has added a layer of complexity to the understanding of its pharmacological effects. Societal attitude

Effects of prescribed medical cannabis and alcohol on real-world driving performance (CAN-TRACK): a study protocol for a two-phase trial.

BACKGROUND: Medical cannabis is now commonly prescribed for a range of chronic health conditions. Many medical cannabis products contain delta-9-tetrahydrocannabinol (THC), the intoxicating component in cannabis, though it is unclear whether these products produce impairment when used as prescribed and at therapeutic doses. With current Australian laws prohibiting driving with any amount of THC in one's system, this trial aims to generate novel data on the impact of prescribed medical cannabis on real-world driving performance to inform road safety policy. METHODS: This is a two-phase trial,

Psychotic Risk Associated With Cannabinoid Use: A Case Report of Ekbom-Like Delusional Infestation.

Chronic use of high-potency cannabinoids has been consistently associated with an increased risk of psychosis, particularly with early onset, daily use and prolonged exposure. However, atypical somatic presentations may be underrecognised and can create diagnostic challenges in clinical practice. We reported the case of a 49-year-old man with long-standing daily cannabis use who developed delusional infestation characterised by persistent pruritus, tactile hallucinations and a fixed somatic delusional belief. Dermatological and neurological assessments, routine laboratory investigations, and c

The effect of daily aerobic cycling exercise on cannabis withdrawal: An inpatient randomised controlled trial.

Aerobic exercise can reduce withdrawal symptoms from tobacco and other drugs but its potential to alleviate cannabis withdrawal remains unexplored. This study investigated whether aerobic exercise can reduce the severity of cannabis withdrawal. Forty-six adults were randomised to (i) 35 min of aerobic cycling exercise (n=24), or (ii) stretching (n=22) each day for seven days. Withdrawal, cravings, exertion, free-fatty acids, triglycerides, and cannabinoids were measured during sessions. We hypothesised that exercise would cause release of THC from fat stores and reduce withdrawal in this way.

Selective upregulation of fatty acid-binding protein 5 within the basolateral amygdala blunts stress-induced reinstatement of cocaine-seeking behavior in mice.

Anandamide (AEA), an endogenous cannabinoid, is thought to exert an inhibitory role in the basolateral amygdala complex (BLA), dampening hypothalamus-pituitary-adrenal (HPA) axis activation and reducing stress-related fear and anxiety behaviors. Stress-induced reductions in AEA mediated signaling have been associated with amygdala hyperexcitability contributing to the reinstatement of cocaine-seeking behavior. Fatty acid-binding protein 5 (FABP5) serves as the major intracellular transporter of AEA, facilitating its degradation by fatty acid amide hydrolase (FAAH). In vivo studies demonstrate
