The association between traumatic temporomandibular joint bony ankylosis and depressive disorder in growing rats.

BACKGROUND: The psychological symptoms of temporomandibular joint (TMJ) ankylosis were similar to that of depressive disorder, but there were no relevant evidences to confirm that the humans or animals with TMJ ankylosis had depressive disorder. The aim of this study was to investigate the association between TMJ ankylosis and depressive disorder in the rat model. METHODS: Thirty 3-week-old male Sprague-Dawley (SD) rats were used in this study. The damage of TMJ complexes and narrowed joint space were performed in the unilateral TMJ of test group to induce TMJ bony ankylosis (experimental sid

Enhanced Metabolic Syndrome Management Through Cannabidiol-Loaded PLGA Nanoparticles: Development and In Vitro Evaluation.

Cannabidiol (CBD) holds promise for managing metabolic diseases, yet enhancing its oral bioavailability and efficacy remains challenging. To address this, we developed polymeric nanoparticles (NPs), using poly(lactic-co-glycolic acid) (PLGA), encapsulating CBD using nanoprecipitation, aiming to create an effective CBD-nanoformulation for metabolic disorder treatment. These NPs (135-265 nm) demonstrated high encapsulation efficiency (EE% ≈ 100%) and sustained release kinetics. Their therapeutic potential was evaluated in an in vitro metabolic syndrome model employing sodium palmitate-induced He

Cannabis Hyperemesis Syndrome in Youth: Clinical Insights and Public Health Implications.

This review focuses on Cannabis Hyperemesis Syndrome (CHS) in youth, a condition linked to chronic cannabis use and characterized by cyclic vomiting, abdominal pain, and dehydration. The objectives were to explore CHS progression in youth and its impact on health, and to assess current treatment strategies. There are the three distinct phases of CHS: prodromal, hyperemetic, and recovery. During the prodromal phase, individuals experience early morning nausea and discomfort, often mistakenly alleviated by continued cannabis use. The hyperemetic phase is marked by severe vomiting, dehydration, a

Gestational THC exposure perturbates hippocampal mitochondrial respiration in the memory-impaired adolescent progeny: Is there a role for mitochondrial CB1 receptor?

Mitochondria are central to cellular energy metabolism, contributing to synaptic transmission and plasticity. The mitochondrial membranes present the cannabinoid type-1 receptor (mito-CB1R), which has been functionally linked to neuronal energy supply and cognitive processing. Prenatal exposure to Δ9-tetrahydrocannabinol (pTHC) has been associated with cognitive impairments associated with molecular cellular and functional abnormalities in several brain regions, including the hippocampus. This study aims at assessing whether, besides the memory impairment, pTHC exposure may result in mitochond

Chronic inflammatory pain and chronic THC vapor inhalation alter midbrain neuronal activity.

In an effort to reduce reliance on opioids for the treatment of pain in the clinic, ongoing work is testing the utility of cannabinoid drugs as a potential alternative for treatment of chronic pain. We tested chronic delta-9 tetrahydrocannabinol (THC) vapor inhalation effects on intrinsic and synaptic properties of ventrolateral periaqueductal gray (vlPAG) neurons in male and female rats treated with complete Freund's adjuvant (CFA). We report that chronic THC vapor inhalation modulates intrinsic and synaptic properties of vlPAG neurons, including reductions in action potential firing rate and

Psilocybin mitigates behavioral despair and cognitive impairment in treatment-resistant depression model using wistar kyoto rats.

Major depressive disorder (MDD) is a leading cause of disability that affects over 300 million people globally. Despite multiple antidepressant trials, approximately one-third of MDD patients remain symptomatic, progressing to treatment-resistant depression (TRD). This persistence possibly is due to the multifaceted etiology of TRD, encompassing biological, psychological, and environmental factors. Chronic stress, prevalent in modern life, significantly contributes to mental health disorders and complicates TRD treatment. This study investigated psilocybin as a potential TRD treatment using a

Activation of CB1R alleviates autism spectrum disorder-like behavior and synaptic impairments.

We previously found that enhancing the levels of 2-arachidonoylglycerol (2-AG) and anandamide (AEA) could improve autism spectrum disorder (ASD) symptoms. This study investigated the effect of cannabinoid type 1 receptor (CB1R) in ASD with pharmacological, genetic and brain-targeted intervention and the underlying mechanisms. Results showed that blocking CB1R counteracted the beneficial effects of boosting 2-AG or AEA on ASD-like behaviors in valproic acid (VPA)-exposed mice. Besides, CB1R knockout mice exhibited ASD-like behaviors and synaptic deficits. In CB1R-specific brain-targeted regulat

Cannabinoid Receptor Type 1 Availability in Individuals with a History of Childhood Trauma: A Positron Emission Tomography Study.

Early life adversity has a lasting impact on the endocannabinoid (eCB) system based on animal models. However, the impact of early life adversity such as childhood trauma (CT) on the eCB system has not been thoroughly studied. We assessed the availability of cannabinoid receptor type 1 (CB1R) in individuals with CT compared to healthy controls without CT (HCs). Cannabinoid receptor type 1 (CB1R) availability was compared in adults with CT (N = 22) and age- and sex-matched HCs (n = 22), using positron emission tomography (PET) imaging with the CB1R-specific radiotracer [11C]OMAR. A comprehensive review on schizophrenia: epidemiology, pathogenesis, diagnosis, conventional treatments, and proposed natural compounds used for management.

Schizophrenia (SCZ) is a complex, comprehensible mental condition that creates alienation from reality. SCZ is a mental disease, which is marked by progressive deficits in working memory, attention, and executive functioning. Because the disease's etiology is unknown, current psychotherapy and pharmacological treatments merely treat symptoms and do not provide a cure. SCZ symptoms include hallucinations, delusions, disorganized behavior, and lack of desire. Long-term use of antipsychotic (antagonists at multiple G-protein-coupled receptors) (mGPCR antagonists) medicines for therapy has negativ
