Propofol at a Sedative Dose Potently Activates the G1-S Phase Transition to Promote Neural Stem Cell Proliferation via Down-regulated FoxO3a-p27 Pathway.

INTRODUCTION: Sedative doses of propofol have recently been reported to exert positive effects on the developing brain, potentially enhancing later cognitive function, but the underlying mechanism remains unclear. This study aims to investigate changes in cell cycle status and reveal the dominant upstream molecular cascade following sedative-dose propofol treatment of primary neural stem cells (NSCs) in vitro. METHODS: Primary C57BL/6 murine NSCs were isolated and cultured. Proliferation was assessed using CCK-8 assays, direct cell counting, and EdU labeling after exposure to 10 μM propofol.

Assessment of lipemia interference on phenobarbital quantification using particle-enhanced turbidimetric inhibition immunoassay with Dimension clinical chemistry system and comparison to high-performance liquid chromatography.

Phenobarbital, an anti-seizure medication with a narrow therapeutic window, requires therapeutic drug monitoring (TDM) for safe and effective use. Lipemia is a well-known interferent in turbidimetric immunoassays, yet its impact on phenobarbital quantification compared with high-performance liquid chromatography (HPLC) has not been fully characterized. We evaluated the effect of lipemia on phenobarbital measurement using a particle-enhanced turbidimetric inhibition immunoassay (PETINIA; Dimension Xpand Plus clinical chemistry system, Siemens) versus HPLC. Simulated lipemic plasma was prepared

Discovery of an intravenous drug injection site: A meticulous cutaneous examination provides the forensic dermatologic clue to a diagnosis of homicide.

An unsuspected homicide in a decedent was considered when the meticulous cutaneous examination of the deceased resulted in the discovery of an intravenous injection site. A 24-year-old woman was found dead in her home. Initial assessment by the crime scene investigator and detective could not determine either the cause or the manner of death. Law enforcement personnel returned to the scene after the assistant medical examiner found a single intravenous needle puncture site during her cutaneous examination of the left antecubital fossa of the dead woman. Additional evidence located near the dec

In Silico to In Cerebro-Development of the Orexin Receptor Antagonist "Photorexant" for Photomodulation In Vitro and in Mouse Brains.

In this study, we report the first photoswitchable small molecule dual ligands for the orexin receptor subtypes 1 (OX1R) and 2 (OX2R) as molecular tools for optical control of orexin signaling. The orexin system is critical for several physiological processes and has been increasingly implicated in the pathogenesis of various psychiatric disorders. We implemented a prospective structure-based design approach, generating azobenzene-containing derivatives of the FDA-approved dual orexin receptor antagonist suvorexant with a computational workflow that featured a novel fragmentation and azobenzen

Association of a routine dexmedetomidine-remifentanil anesthetic strategy with POD3 morning cortisol change in older patients undergoing radical esophagectomy.

BACKGROUND: Older patients undergoing radical esophagectomy may experience marked postoperative endocrine stress, inflammation, pain, and short-term cognitive decline. We evaluated whether a routine anesthetic strategy combining dexmedetomidine with remifentanil was associated with a smaller POD3 serum cortisol increase than remifentanil- or dexmedetomidine-based strategies alone. METHODS: This single-center retrospective cohort screened 426 consecutive patients aged ≥65 years who underwent elective radical esophagectomy from January 2023 to December 2025. Of these, 347 had verifiable pathway

Inhaled β-myrcene alleviates insomnia in mice by modulating 5-HT1A/PKA signaling and the gut microbiota.

BACKGROUND AND OBJECTIVES: Insomnia is a major public health issue that impairs daily functions and quality of life. Sleep-onset difficulty, a common complaint among patients, is accompanied by reduced central serotonin (5-HT) function. β-myrcene from lavender has sedative-hypnotic effects via serotonergic pathways, but most previous studies used intraperitoneal injection, which causes stress and limits clinical translation. Here, we asked whether non-invasive inhalation of β-myrcene produced similar effects in a mouse model of insomnia with low 5-HT function, and explored the underlying mecha

Combined Effect of Hypnotic-Cognitive Therapy With Therapeutic Exercise on Quality of Life in Individuals With Fibromyalgia: A Randomized Controlled Trial Protocol.

BACKGROUND: A multidisciplinary approach is viewed as the primary treatment strategy for fibromyalgia (FM). Therapeutic exercise is an important component of this approach. Hypnotic cognitive therapy (HYP-CT), a relatively new intervention strategy, has not traditionally been incorporated in FM treatment, although it has shown beneficial effects in other chronic pain conditions. This study aims to investigate the potential benefits of a combined approach that incorporates HYP-CT with therapeutic exercise in the rehabilitation of individuals with FM. MATERIALS AND METHODS: One hundred and four
