Novel selective morpholine trace amine-associated receptor 1 partial agonists show promising preclinical effects for neuropsychiatric disorders and are well tolerated in healthy volunteers.

Trace amine-associated receptor 1 (TAAR1) is a novel target for antipsychotic and potentially mood-stabilizing and anti-addictive drugs, offering a new mechanism by modulating dopaminergic, serotonergic, and glutamatergic neurotransmission. TAAR1 agonists from a prior amino oxazolines series were studied preclinically and clinically, but development of partial agonist RO5263397 was halted due to poor metabolization by N-glucuronidation in individuals with UGT2B10 splice site mutations. A medicinal chemistry program subsequently identified new potent and selective TAAR1 ligands from the morphol

Long-term Effects of Early Life Adversity On Brain Dopamine and Serotonin Receptor Systems Involved In Cocaine Reinforcement In Adult Macaques: a Positron Emission Tomography Study.

UNLABELLED: Early life adverse (ELA) experiences such as child maltreatment (MALT) are associated with physical and mental illness, including substance use disorders (SUDs), but underlying neurobiological mechanisms remain unclear. This study examined long-term effects of infant MALT on adult brain serotonin (5HT) and dopamine (DA) receptors in corticolimbic regions involved in reward and emotional control, using positron emission tomography (PET) imaging, a translational infant MALT macaque model of cocaine use disorder (CUD) risk and a COC self-administration (SA) paradigm. The study focused

Dynamic Alterations in the Blood Transcriptome Characterize Drug Use Behavior and Co-Morbidities in Cocaine Use Disorder: A Preliminary Study.

Individuals with cocaine use disorder (CUD) who attempt abstinence experience craving and relapse that can benefit from multimodal treatment monitoring. Longitudinal studies linking behavioral manifestations in CUD to the blood transcriptome are not only limited but also computationally complex. Therefore, we developed an analytical pipeline to investigate the connection between drug use behaviors during abstinence and change in the blood transcriptome. We conducted a longitudinal study with CUD (n = 12 subjects) and collected behavioral metrics and blood RNA-seq at baseline, 3, 6, and 9 month

Cocaine use disorder, mental health diagnoses, and serious mental illness characteristics in mental health treatment.

Over half of treatment-seeking individuals with a cocaine use disorder have a co-occurring mental health disorder. Mental health disorder symptoms may impact an individual's functioning so severely that they are classified as a serious mental illness (SMI). However, there is variability in defining SMI, including the duration of symptoms, functional impairment, or specific diagnoses such as schizophrenia. This study answered the following questions among a national sample of adults with cocaine use disorder who received services from mental health treatment facilities from 2013 to 2021 (N = 35

Tailoring cariprazine for dual disorders via secondary pharmacophore optimization.

Cariprazine is an approved drug used to treat schizophrenia, bipolar disorder, and (more recently) major depression. These serious mental illnesses are often comorbid with psychostimulant use disorders, for which no pharmacotherapeutic is currently approved. Based on preclinical data and multiple case studies, cariprazine demonstrates significant potential as a treatment for such dual disorders. Notably, this drug acts as a high-affinity (Ki = 0.22 nM) partial agonist (Emax = 45.1%) of the dopamine D3 receptor (D3R), a molecular target implicated in a variety of substance use disorders. Unlike

Polygenic risk scores reveal genetic underpinnings of comorbidity between substance use disorders and ADHD.

One in five people attending services with a substance use disorder (SUD) also has comorbid attention-deficit/hyperactivity disorder (ADHD). While some evidence shows that shared heritability might partly explain this frequent comorbidity, studies on this topic are scarce. Therefore, the goal of this study was to test whether ADHD and SUD have shared genetic influences. A total of 1,513 SUD treatment-seeking individuals of European ancestry were included in the study. Polygenic risk scores (PRS) were applied to test a) the association of the PRS for ADHD with multiple SUD-related phenotypes, a

A multicenter study to validate the Brief Cognitive Status Examination as a screening tool for the detection of cognitive impairment in a substance use disorder population over 45 years of age in a Spanish population.

Cognitive impairments are common among individuals with substance use disorders (SUD) and can significantly affect treatment outcomes, including retention and abstinence rates. Traditional neuropsychological assessments are often impractical for routine clinical use due to time and resource limitations, underscoring the need for brief, reliable cognitive screening tools. This study aims to evaluate two such tools, the Montreal Cognitive Assessment (MoCA) and the Brief Cognitive Status Examination (BCSE), in comparison to the Neuropsychological Battery of Fundació ACE (NBACE), considered the go

Compulsive sexual behavior in cocaine use disorder: Treatment outcomes and clinical challenges.

BACKGROUND: Emotion regulation (ER) difficulties, overall psychopathological burden and poor impulse control may explain the comorbidity between compulsive sexual behavior (CSB) and cocaine use disorder (CUD). Although some studies have assessed the prevalence of CSB in CUD patients, more data are needed to better understand their relationship. This study first examined whether ER difficulties at treatment entry for CUD predict CSB after outpatient treatment. Second, it explored whether CSB and psychopathological burden are associated with worse treatment outcomes after inpatient and outpatien

Functional Ultrasound Imaging-Based Mapping of Cocaine Induced Neural Changes in Awake Mice.

INTRODUCTION: Cocaine is one of the most commonly used addictive drugs with adverse medical consequences. Its exposure involves rapid neurodynamic changes that are challenging to capture in awake subjects. This study aimed to characterize acute cocaine-induced alterations in neuronal activity and functional connectivity (FC) using high-resolution functional ultrasound (fUS) and to develop a machine-learning-based framework for state classification. METHODS: Using fUS with superior spatiotemporal resolution to fMRI, we measured cerebral blood volume (CBV) as a proxy for neuronal activity and a
