Hallucinogen-Psychosis Associations Are Confounded by Baseline Psychiatric History.

Background: Current drug policy debates often center on whether hallucinogen-related hospital admissions indicate a causal relationship with psychosis. This study examines the extent to which observed associations between hallucinogen use and psychosis are mediated by preexisting psychiatric conditions, providing important evidence for psychedelic policy formulation. Methods: We conducted a retrospective analysis using MarketScan Medicaid and commercial claims databases from 2015-2019. The population-based sample included individuals receiving substance use disorder treatment with documented s

Psilocybin restores behavior and 5-HT2A signaling while reducing microglial density after chronic traumatic brain injury in rats.

Traumatic brain injury (TBI) causes persistent neurobehavioral deficits and increases the risk of psychiatric disorders, including depression, anxiety, and cognitive dysfunction linked to disrupted neuroplasticity, neuroinflammation, and serotonergic (5-HT) signaling. No effective pharmacotherapies exist for chronic TBI. Psilocybin, a psychedelic 5-HT2A receptor agonist, shows promise due to its neuroplasticity-enhancing, anti-inflammatory, and antidepressant effects. Here, male rats received fluid-percussion or sham injury, followed one year later by a single psilocybin (1 mg/kg) or saline in

Music Playlist Use in Clinical Trials of Psychedelic Assisted Psychotherapy: A Systematic Review.

Recent studies have shown promising results for psychedelics in the treatment of psychiatric disorders. Both in clinical and recreational settings, contextual elements, such as music, can influence the experience of users and may be linked to positive outcomes. This systematic review identifies how music is being selected and used in psychedelisc-assisted psychotherapy (PAP). The search adopted PRISMA criteria and was conducted using PubMed, PsycNet, Web of Science, Scopus and Cochrane as databases. The use of music in PAP was compared, considering aspects such as the creation of a playlist, p

MFCC-DFT mapping of ligand recognition at the 5-HT2A receptor: energetic analysis of the interactions between serotonin, psychedelics, and antipsychotics.

Mental disorders represent a major global health problem, with depression being one of the most prevalent and disabling conditions worldwide. Growing evidence suggests that the serotonergic system, particularly the 5-HT2A receptor, plays an important role in modulating mood and cognitive processes, constituting a key pharmacological target for several psychoactive compounds. In this study, we investigated the molecular interaction profile between the 5-HT2A receptor and four pharmacologically relevant ligands, serotonin (5-HT), psilocybin/psilocin (PSILO), lysergic acid diethylamide (LSD), and

Hallucinogen-Assisted Psychotherapy for Trauma Disorders: A Bionian Lens.

There has been an increasing interest in the use of classic psychedelics (such as psilocybin) and 3,4-methylenedioxymethamphetamine (MDMA) for treating mental health conditions. Individuals often describe psychedelic sessions as among the most significant experiences in their lives, emphasizing the sense of awe, connectedness, and spiritual transformation taking place. While the psychedelic literature from the past two decades has mostly focused on using these drugs in the treatment of major depressive, anxiety, and substance use disorders, researchers have also investigated the utility of cla

Expectations and Motivations for Participation in Clinical Trials Utilizing Psychedelics for Treatment-Resistant Depression: A Qualitative Study.

PURPOSE: Expectations strongly shape participant experiences in clinical trials, contributing to placebo effects that complicate interpretation of outcomes. While quantitative tools exist to measure expectations, their subjective and multidimensional nature, especially for individuals with treatment-resistant depression (TRD), remains underexplored. This study aimed to explore the motivations and expectations of participants in psychedelic clinical trials. METHOD: Seventeen semi-structured interviews were conducted with individuals prior to screening for two TRD clinical trials involving 5-Me

Protocol for a qualitative mechanistic study of MDMA with a sample of psychoanalytic psychotherapists: A phenomenological investigation.

INTRODUCTION: Methylenedioxymethamphetamine (MDMA)-assisted psychotherapy (MDMA-AP) is a potentially transformative intervention for post-traumatic stress disorder (PTSD). Despite encouraging Phase III trial results, the psychological mechanisms underpinning MDMA's therapeutic effects remain insufficiently understood. Existing research on MDMA has emphasised neurobiological processes but has not adequately incorporated subjective, relational, and experiential processes that are likely to be central to optimising psychedelic-assisted treatments. In the absence of an established clinical psychol

Mechanistic insights toward dissociating therapeutic from psychedelic effects: bridging the gap between psychedelic research and mental health care.

Clinical trials investigating psychedelic compounds for depression and anxiety-related disorders are yielding promising preliminary results. Psychedelics produce profound alterations in brain function-such as suppression of the default mode network and thalamocortical dysregulation-leading to intense subjective experiences including ego dissolution and mystical-type states. While often described as meaningful or therapeutic, these effects can also be unpredictable or distressing. Because of these effects, which require labor-intensive, potentially cost-challenging preparation, administration, i

Psychedelic Therapy and the Role of Music: A Scoping Review of Quantitative Evidence on Subjective and Objective Outcomes.

PURPOSE: Psychedelics have received considerable attention due to their potential in treating psychiatric disorders. The "setting" during psychedelic-assisted therapy (PAT) is recognized as playing a central role in the experience, during which music features prominently. Although music is theorized as directing and shaping psychedelic sessions, its precise contribution to acute experience and therapeutic outcomes is unclear. This scoping review aimed to map quantitative research on the interplay of psychedelics and music by consolidating existing evidence, identifying gaps, and where possible

Community engagement as a foundation for implementation research for group psilocybin assisted therapy in New Mexico.

UNLABELLED: Informed by a community engagement process, we have developed a pragmatic, open-label, hybrid feasibility-implementation study of Group Psilocybin-Assisted Therapy (GPAT) for post-traumatic stress disorder (PTSD). As psychedelic-assisted therapies begin to enter the broader mental health arena, engaging communities in the design of research and care models is essential to ensuring that those most impacted have access to-and trust in-the treatments being developed. Guided by the principle of "nothing about us without us," the proposed study was designed from the outset to be communi

MDMA-assisted PTSD and Alcohol Therapy Trial (MPATHY): study protocol for a double-blind, randomised, controlled outpatient trial of MDMA-assisted integrated exposure-based therapy for comorbid post-traumatic stress disorder and alcohol use disorder.

INTRODUCTION: The treatment of comorbid post-traumatic stress disorder (PTSD) and alcohol use disorder (AUD) is significantly more challenging than the treatment of either disorder alone. While gold standard evidence-based treatments exist for this comorbidity, clinically significant improvements are only observed in approximately half of clinical trial participants. The use of adjunctive pharmacotherapies, such as 3,4-methylenedioxymethamphetamine (MDMA), may serve to optimise gold standard interventions. The primary aim of the MDMA-assisted PTSD and Alcohol Therapy Trial study is to examine

Psychedelics in Mental Health: Opportunities and Challenges.

Psychedelic and substance-assisted therapies are attracting renewed scientific, clinical, and public interest as potential interventions for mental health disorders, particularly treatment-resistant depression, post-traumatic stress disorder, and substance use disorders. However, as the field moves from early efficacy trials toward clinical implementation, major questions remain regarding therapeutic mechanisms, psychological support models, safety, ethics, accessibility, and the role of subjective experience. This editorial introduces the Brain and Behavior special issue "Psychedelics in Ment

Combined DMT-harmine formulation reduces negative self-referential emotions during social self-evaluation: a randomized placebo-controlled trial in healthy volunteers.

RATIONALE AND OBJECTIVES: Negative self-referential emotions such as embarrassment and shame play a key role in the psychopathology of psychiatric disorders but are often insufficiently addressed by standard treatments. Psychedelics such as ayahuasca - an Amazonian plant decoction containing the psychedelic N,N-dimethyltryptamine (DMT) and harmala alkaloids, have been proposed to positively modulate self-referential processing. The aim of this study was to investigate the effects of an ayahuasca-inspired harmine+DMT formulation and harmine on embarrassment and shame. METHODS: In this randomiz

Psilocybin produces amplified acute responses and sex-specific long-term increases in sensorimotor gating in the mGlu5 knockout mouse model of schizophrenia.

Psilocybin is a serotonergic psychedelic drug with emerging therapeutic applications, yet its actions under glutamatergic dysfunction, and relevance to schizophrenia and associated psychiatric disorders, remain unclear. We examined the acute and long-lasting effects of psilocybin (1 mg/kg, intraperitoneal) on behavioral and neural outcomes in the metabotropic glutamate receptor 5 (mGlu5) knockout (KO) mouse model of schizophrenia. The mGlu5 KO mice displayed psilocybin-induced hyperlocomotion, whereas wild-type (WT) mice did not. Additionally, male mGlu5 KO mice showed an amplified psilocybin-

Polysubstance use disorders among US adults.

Polysubstance use disorders ( ≥ 2 substance use disorders (SUDs)) are associated with high morbidity and mortality. We analyzed data from 92,233 adult participants in the 2022-2023 US National Surveys on Drug Use and Health to estimate past-year prevalence of polysubstance use disorders and to examine their associations with age of substance use initiation. Multivariable logistic regression and Poisson regression were applied. Age- and sex-adjusted past-year prevalence of 2 SUDs was 19.2-44.9% (95% CIs=11.1-62.3%) among adults with any SUD. Age- and sex-adjusted past-year prevalence of ≥3 SUDs

From monotherapy to sequential models: An updated scoping review on ibogaine's role in treatment for psychiatric disorders.

BACKGROUND: Preliminary evidence examining ibogaine in neuropsychiatric and substance use disorders is accumulating. As patient interest and off-label use grow, clinicians require an understanding of emerging dosing strategies, investigational protocols, and reported adverse effects to appropriately counsel patients and contextualize potential risks and benefits. METHODS: Databases were searched for human studies of ibogaine, noribogaine, or 5-MeO-DMT with relevant clinical outcomes, prioritizing randomized controlled trials (RCTs), microdosing paradigms, and sequential protocols.
