Estimated burden of acute gastroenteritis and foodborne gastroenteritis caused by non-typhoidal Salmonella enterica, Shigella and norovirus in Gansu Province, Northwest China.

BACKGROUND: Information on the burden of disease caused by foodborne pathogens in Northwest China is limited. This study aims to estimate the burden of acute gastroenteritis (AGE) and foodborne salmonellosis, shigellosis and norovirus gastroenteritis in Gansu Province. METHODS: Using data from population surveys conducted during 2011-2015, the burden of AGE was estimated in Gansu Province. By determining the number of pathogen-specific cases from sentinel hospital surveillance conducted during 2014-2019, with adjustments applied for healthcare-seeking behavior and stool specimen submission ba

Development and evaluation of a dual-target glycoconjugate against Shigella sonnei.

INTRODUCTION: Shigellosis morbidity and mortality, combined with the increase in multidrug-resistant infections, make Shigella vaccine development a global imperative. Glycoconjugate vaccines that couple immunogenic O-antigen to protein derived from Shigella may provide broader protection across Shigella species and serogroups. Such an approach also circumvents immunotolerance arising from repeated use of the same carrier. Here we use bioconjugation, exploiting an oligosaccharyltransferase (OST) enzyme to couple O-antigen and carrier protein in vivo, to generate a Shigella-specific dual-target

Rare Fatal Event of Multidrug-resistant Shigella Septicemia and Cryptococcal Meningitis in an Immunocompromised Host.

Bacteriemia due to Shigella spp. is a rare event. However, an individual with impaired immunity remains at risk of such invasion where the infective dose is as low as 10 viable bacilli. This case report describes one such fatal event where a 40-year-old immunocompromised male presented to a tertiary-level healthcare facility with a history of diarrhea for 10 days and seizures. Prompt microbiological workup established multidrug-resistant Shigella flexneri septicemia and cryptococcal meningitis.

Expanded prevalence of group 4 capsule expression and its genetic requirements in a broad array of Shigella strains.

UNLABELLED: The expression of a group 4 capsule (G4C) has been identified in Shigella sonnei and Shigella flexneri serotype 6 and demonstrated to play a significant role in pathogenic processes in S. sonnei. However, it has not been examined phenotypically in S. flexneri 6. Despite the impact of G4C, no systematic analysis of its expression or the presence of the genomic determinants has been performed across a large collection of S. sonnei and S. flexneri serotypes 2a and 6 or any of the other >46 Shigella strains and serotypes that cause disease. Our examination of the genomes of 1,219 geogr

Bacteriophage Sf14 survival under simulated gastrointestinal conditions.

Shigella flexneri is an enteric pathogen responsible for causing shigellosis, a type of bacillary dysentery of significant global health concern. The increasing prevalence of antibiotic resistance has driven the search for alternative treatments, including bacteriophage (phage) therapy. However, the effectiveness of orally administered phages for enteric pathogens depends on their ability to survive passage through the gastrointestinal tract. In this study, the ability of bacteriophage Sf14 to maintain infectivity was evaluated under simulated gastrointestinal conditions using a modified INFOG

Clinical development of human vaccine candidates against Shigella flexneri: a systematic review of clinical trials.

Shigella flexneri remains a leading cause of shigellosis, particularly in low- and middle-income countries, where it contributes substantially to childhood morbidity, mortality, and the growing burden of antimicrobial resistance. Despite decades of vaccine research, no licensed vaccine against S. flexneri is currently available. This systematic review aimed to summarize and critically evaluate the clinical evidence regarding the safety, immunogenicity, and efficacy of human S. flexneri vaccine candidates. A comprehensive literature search was conducted in major electronic databases following P

Bridging the translational gap: an operational classification and quantitative framework for accelerating enteric vaccine research using animal models.

Enteric bacterial infections, notably shigellosis and salmonellosis, represent a staggering global health burden. Vaccine development is severely hampered by host-restriction, creating a persistent "translational gap" where preclinical data fails to correlate with human outcomes. Traditionally, model selection has been largely qualitative. This review provides a comprehensive analysis of the research landscape, evaluating the physiological fidelity of non-human primates, the surgical precision of rabbit and guinea pig models, and specialized induced rodent systems, including the streptomycin‑i

Microalgal food supplements are becoming increasingly popular due to their promising biological effects and high nutritional value, evidenced in in vitro, in vivo, and human studies. Some products of this kind have, however, raised controversies concerning their safety. At the same time, not much is known about the frequency of adverse events following the use of microalgal supplements, potential factors that may influence them, and general characteristics and behaviours of the consumer group. The present study aimed to fill this gap and surveyed a group of Polish consumers of microalgal produ

Impact of left ventricular end-diastolic diameter size within 24 hours of hospital admission on outcome events in patients with ST-elevation myocardial infarction.

BACKGROUND: The prognosis of patients with ST-elevation myocardial infarction (STEMI) is closely related to the left ventricular end-diastolic diameter (LVEDD) size. However, the associations between LVEDD, pre-admission risk factors, admission biochemical markers, and medication use warrant further investigation. The prognostic impact of LVEDD evaluation within 24 hours of admission in STEMI patients has not been extensively studied. METHODS: We analyzed the association between LVEDD measurements within 24 hours of admission and a composite endpoint of cardiovascular events and mortality in

A laboratory system for rearing Simuliidae (Diptera) under simulated lotic environments.

BACKGROUND: Black flies (Diptera: Simuliidae) are ecologically important components of lotic ecosystems and a public health concern due to their aggressive biting behavior, pathogen transmission, and induction of allergic and inflammatory reactions in humans and livestock. Despite their relevance, experimental research on Simuliidae remains limited compared with other dipteran vectors, largely because their immature stages require specific aquatic conditions that are difficult to reproduce in the laboratory. Larval development depends on continuous water flow, high dissolved oxygen, and suspen

Seroprevalence of IgG antibodies against measles in adolescents and adults from Metropolitan Lima and Callao, Peru, 2024-2025.

INTRODUCTION: The EndoFLIP (for its acronym in English: endoluminal functional luminal probe) is a system that measures in real time the compliance index of the gastroesophageal junction and secondary esophageal peristalsis, based on the principle of impedance planimetry. Although this technology is relatively new and its diagnostic usefulness is still being evaluated, its use in clinical practice is increasingly recognized, fundamentally in those patients where there is no conclusive manometric diagnosis of esophageal motor pathology, mainly esophagogastric junction outflow obstruction and ac

PLGA-based nanovaccine delivering chimeric IpaD-StxB protein induces robust immune response against Shigella infection.

BACKGROUND AND AIM: Shigellosis is a major cause of morbidity and mortality worldwide, especially among children under five in low- and middle-income countries. Despite its public health burden, there is no licensed vaccine. Recombinant and nanoparticle-based vaccine approaches offer promising alternatives to conventional methods. This study aimed to develop a chimeric nanovaccine based on the fusion of Shigella flexneri IpaD and StxB, encapsulated in PLGA nanoparticles, and to evaluate its immunogenic potential in a murine model. METHODS: The recombinant IpaD-StxB (IdSb) protein was expresse

A public resource of 15 genomically characterized representative strains of Shigella sonnei.

Shigella sonnei is rapidly emerging as the dominant agent of shigellosis, an enteric disease responsible for a significant burden of morbidity and mortality worldwide. Whole-genome sequencing of S. sonnei isolated over the last three decades has revealed phylogenomic diversity within the population and the emergence of multiple lineages associated with distinct epidemiological patterns such as resistance to critical antimicrobials and/or transmission within different groups. However, most experimental work on S. sonnei biology and pathogenicity has focused on a single laboratory strain (53G),

Microneedle array patch delivery of Shigella flexneri 2a GMMA.

Shigella infections have long been a significant contributor to global diarrheal mortality, yet no approved Shigella vaccines are currently available. Generalized Modules for Membrane Antigens (GMMA) has emerged as an innovative platform for developing vaccines against Shigella. Here, for the first time, this technology was combined with a microneedle array patch (MAP), a transdermal vaccine delivery system, as an attractive method to overcome the drawbacks of parenteral administration routes. First, we demonstrated that two types of MAPs, the coated (C-MAP) and powder-attached (P-MAP) formats
