Enterotoxigenic Escherichia coli (ETEC), Shigella, and Campylobacter have been identified as major causes of diarrheal diseases worldwide. In addition to overt disease and death, they are responsible for stunting in children with the risk of lifelong consequences on health and economic opportunities. All three of these bacterial pathogens, which collectively account for approximately 30 % of the cases of diarrheal diseases, are recognized as antimicrobial resistance (AMR) threats. In spite of the dangers these pathogens represent for both children and adults, there is as yet no licensed vaccin

UNLABELLED: We performed phenotypic susceptibility testing and whole-genome sequencing to investigate in vitro mecillinam activity against multidrug-resistant (MDR) Shigella. Ninety-five isolates (53 [56%] Shigella sonnei and 42 [44%] Shigella flexneri) were included. The MIC50 and MIC90 were 1 and 2 µg/mL, respectively, by agar dilution, and all zone diameters were ≥15 mm by disc diffusion testing. No known clinically relevant mecillinam resistance mutations (including cysB) were detected. The favorable MIC profile and absence of resistance markers make pivmecillinam an agent worth considerin

BACKGROUND: Berberine (BBR), an isoquinoline alkaloid, has been applied clinically to treat dysentery caused by Shigella for decades. Nevertheless, the precise mechanisms behind its anti-Shigella effect have not been fully elucidated.
PURPOSE: This study aims to investigate the mechanism of BBR on antibacterial activity against S. flexneri infection.
METHODS: We initially reproduced the mouse model of Shigella flexneri-induced dysentery, and then, assessed the therapeutic effect of BBR. In vitro, we measured the inhibitory effect of BBR against S. flexneri and the GTPase activity of FtsZ (fi

MicroRNAs have recently emerged as major players in host -bacterial pathogen interactions, either as part of the host defense mechanism to neutralize infection or as a bacterial arsenal aimed at subverting host cell functions. Here, we identify the newly evolved human microRNA miR-6762-5p as a new player in the host-Shigella interplay. A microarray analysis in infected epithelial cells allowed the detection of this miRNA exclusively during the late phase of infection. Conditional expression of miR-6762-5p combined with a transcriptome analysis indicated a role in cytoskeleton remodeling. Likew

Shigella are Gram-negative non motile facultative anaerobic bacteria belonging to family Enterobacterales. They are a leading cause of diarrhoea and dysentery and constitute a significant public health problem in both the developing and developed nations. In Asia alone, 125 million infections and 14,000 deaths are estimated to be due to shigellosis annually. Bacteremia caused by Shigella spp is rare. We hereby present a case of bloodstream infection caused by fluoroquinolone resistant, extended spectrum beta lactamase producing Shigella flexneri strain in a five-year-old malnourished child tha

OBJECTIVE: This research aims to investigate the epidemiology and genetic changes of Shigella species in the cities of Qazvin and Tehran from 2003 to 2023, with a focus on assessing antibiotic resistance among the isolates. This study is based on the analysis of 80 Shigella isolates, which were obtained from patients' feces.
RESULTS: Using pulsed-field gel electrophoresis (PFGE) as the primary typing method, the isolates were classified into 13 distinct clusters, revealing a dominant pattern that has persisted for over twenty years. This persistence may be attributed to factors such as poor h

The conserved invasion plasmid antigen D (IpaD) protein demonstrates broad protective capabilities against bacillary dysentery caused by Enteroinvasive Escherichia coli (EIEC) and Shigella. However, the instability of the IpaD protein at room temperature limits its therapeutic potential. The stabilization and efficient production of functional recombinant proteins remain critical challenges in therapeutic and vaccine development. This study presents a novel fluorescence fusion strategy for producing a stable IpaD-EGFP recombinant protein using a flexible linker (GGGGS)₃. The fusion technique e

Ubiquitination plays vital roles in modulating pathogen-host cell interactions. RNF213, a E3 ligase, can catalyze the ubiquitination of lipopolysaccharide (LPS) and is crucial for antibacterial immunity in mammals. Shigella flexneri, an LPS-containing pathogenic bacterium, has developed mechanisms to evade host antibacterial defenses during infection. However, the precise strategies by which S. flexneri circumvents RNF213-mediated antibacterial immunity remain poorly understood. Here, through comprehensive biochemical, structural and cellular analyses, we reveal that the E3 effector IpaH1.4 of

No licensed vaccines are available for the largely antibiotic-resistant Shigella or enterotoxigenic Escherichia coli (ETEC), the two most common bacteria causing children's diarrhea and travelers' diarrhea. Virulence heterogeneity is a key obstacle to developing vaccines against Shigella or ETEC. By applying a multiepitope fusion antigen (MEFA) vaccinology platform, we recently constructed epitope- and structure-based polyvalent proteins to induce cross-protective antibodies against heterogeneous Shigella or ETEC strains. In this study, we combined a polyvalent Shigella protein with two polyva

Shigella is a leading cause of diarrheal morbidity and mortality in young children from low- and middle-income countries. Here, we aimed to verify the ability of the generalized modules for membrane antigens (GMMA)-based Shigella sonnei candidate vaccine 1790GAHB to elicit an anti-protein antibody response. Serum samples from previous clinical trials in adults (a dose-escalation study and its extension in France, a vaccine efficacy study after human challenge in the United States, and a study in Kenya) were investigated using pan-proteome microarrays consisting of 3,150 full-length or fragment

Shigella is a leading cause of moderate-to-severe diarrhea, primarily affecting children in impoverished regions. Disease incidence is highest in toddlers 1-2 years of age. Acquisition of immunity over time reduces the risk of infection. However, the evolution of childhood immunity resulting from Shigella exposure and the host responses that mediate protection remain poorly understood. Gaining insight into the immunological features that develop over time and prevent subsequent Shigella infection is critical for guiding vaccine design. We examined the antigenic repertoire and magnitude of humo

Shigella is an important human pathogen that has no licensed vaccine. Despite decades of seminal work suggesting that its pathogenicity relies on inflammatory cell death of macrophages, the in vivo role of macrophages in controlling Shigella infection remains poorly understood. Here, we use a zebrafish model of innate immune training to investigate the antibacterial role of macrophages following a non-lethal Shigella infection. We found that macrophages are crucial for zebrafish larvae survival during secondary Shigella infection. Consistent with signatures of trained immunity, we demonstrate

Shigella flexneri, a formidable gram-negative bacterium, that triggers the most contagious form of shigellosis through bloody diarrhea, presenting a significant threat. As a severe foodborne pathogen, it underscores the need for intensified research into more effective prevention strategies and heightened public health awareness. The traditional methods used to detect S. flexneri are time-consuming and inconvenient. As a result, there is a need for accurate and rapid diagnostics in food and water samples. The experimental methods involved modifying S. flexneri-specific HGD-aptamer and employin

INTRODUCTION: Diarrheagenic bacteria are among the major contributors to the global diarrheal burden. The absence of up-to-date data on the etiologies of diarrhea due to limited active surveillance and clinical laboratory capacity makes the burden more severe. Shigella, Salmonella, and Campylobacter are among the most common bacterial etiologies of childhood diarrhea. The present study aimed to provide epidemiological data on these bacterial etiologies in under-five-year-old children in Ethiopia.
METHODOLOGY: A cross-sectional health facility-based study was conducted from December 2020 to Au

Endomembrane breaching is a crucial strategy employed by intracellular pathogens enclosed within vacuoles to access the nutrient-rich cytosol for intracellular replication. While bacteria use various mechanisms to compromise host membranes, the specific processes and factors involved are often unknown. Shigella flexneri, a major human pathogen, accesses the cytosol relying on the Type Three Secretion System (T3SS) and secreted effectors. Using in-cell correlative light and electron microscopy, we tracked the sequential steps of Shigella host cell entry. Moreover, we captured the T3SS, which pr
