Cinnamomum zeylanicum essential oil inactivates Shigella flexneri by inhibiting biofilm formation, disrupting cell membrane integrity, and suppressing virulence gene expression.

Shigella flexneri, a foodborne pathogen that causes shigellosis, poses a significant risk to human health and food safety. Cinnamomum zeylanicum essential oil (CZEO) is known to inhibit microbial growth, but its possible antibacterial and antibiofilm mechanisms against S. flexneri remain unclear. The chemical composition of CZEO was identified via GC-MS. The minimum inhibitory concentration of CZEO against S. flexneri was 200 μg/ml. CZEO-treated S. flexneri exhibited an extended lag phase, indicating delayed bacterial growth, and demonstrated potent bactericidal activity in fresh coconut water

Burden of Shigella among children with diarrhea in the Americas: A systematic review and meta-analysis.

INTRODUCTION: Shigella is a leading cause of diarrhea worldwide. While the burden of Shigella has been shown to be highest in Africa and Asia, recent studies have also shown considerable burden in the Americas. With several pediatric Shigella vaccines in clinical development, policymakers in the region will eventually consider whether a Shigella vaccine is appropriate for their setting. METHODS: We conducted a systematic review and meta-analyses to summarize the burden (characterized by prevalence, incidence, and attributable fraction estimates) of Shigella diarrhea among children under 72 mo

Bacterial dysentery complicated with Crohn's disease: A case report.

RATIONALE: Crohn's disease is easily confused with bacillary dysentery in clinical manifestations. Crohn's disease is a chronic disease that can involve both intestinal and extraintestinal areas, and severe patients have prolonged and poor prognosis, so early diagnosis and treatment are crucial for patients. This case is a combination of bacillary dysentery and Crohn's disease, which emphasizes the differentiation between bacillary dysentery and Crohn's disease, and expects clinicians to pay attention to and discover the early manifestations of Crohn's disease. PATIENT CONCERNS: A young male

Correlation of fecal calprotectin levels with the detection of treatable enteric pathogens in children with severe acute diarrheal disease in Botswana.

Diarrheal disease is a leading cause of death among young children globally. Current guidelines recommend supportive treatment of acute diarrhea and using antimicrobials only with presence of blood in the stool. Select enteric pathogens, including Shigella, commonly cause disease in high-burden settings; targeted treatment of these pathogens could decrease morbidity and mortality. In settings with limited access to microbiological testing, practical diagnostics are needed to differentiate treatable causes of pediatric diarrhea. Evolving evidence suggests fecal calprotectin (fCal) could help di

Characterization of Shigella virulence factor intracellular spread A (IcsA, or VirG) functional epitopes against S. flexneri 2a and S. sonnei invasion and adherence.

There are no licensed vaccines against Shigella, a leading cause of children's diarrhea and travelers' diarrhea. To develop a cross-protective vaccine against heterogeneous Shigella species and serotypes, we attempted to apply an epitope- and structure-based vaccinology platform, multiepitope fusion antigen, to construct an optimal polyvalent chimeric immunogen with functional epitopes from the key Shigella virulence determinants. With Shigella invasion plasmid antigens B and D functional epitopes identified in recent studies, in this study, we focused on Shigella intracellular spread protein

Phenotypic and genotypic relationships between Shigella species and Escherichia coli O37:H10 isolates in Addis Ababa, Ethiopia.

OBJECTIVE: This study aimed to investigate the genotypic and phenotypic differences between Shigella species and E. coli O37:H10, as well as their antimicrobial resistance (AMR) and virulence factors, in children aged under five with diarrhea in Addis Ababa, Ethiopia. RESULTS: Using whole genome sequencing (WGS), all 28 S. sonnei identified with antisera were also verified as S. sonnei, while all nine isolates of S. flexneri were found to be E. coli O37:H10. One S. flexneri sample, identified serologically, was excluded from WGS analysis because of low genome coverage. Most resistant genes we

Macrophages orchestrate elimination of Shigella from the intestinal epithelial cell niche via TLR-induced IL-12 and IFN-γ.

Bacteria of the genus Shigella replicate in intestinal epithelial cells and cause shigellosis, a severe diarrheal disease that resolves spontaneously in most healthy individuals. During shigellosis, neutrophils are abundantly recruited to the gut and have long been thought to be central to Shigella control and pathogenesis. However, how shigellosis resolves remains poorly understood due to the longstanding lack of a tractable and physiological animal model. Here, using our newly developed Nlrc4-/-Casp11-/- mouse model of shigellosis, we unexpectedly find no major role for neutrophils in limiti

Neutralizing antibodies induced by IpaB and IpaC mRNA vaccines inhibit Shigella flexneri invasion.

Shigellosis, caused by Shigella species, remains a significant global health burden due to its high morbidity and mortality, particularly in developing countries, and frequent antimicrobial resistance. Thus far, various Shigella vaccine candidates have been tested, but many have shown limited immunogenicity, high reactogenicity, or insufficient cross-serotype protection due to serotype-specific O-polysaccharide (OPS) targeting. In this study, we evaluated mRNA vaccine candidates targeting the highly conserved Shigella invasion plasmid antigens (IpaB and IpaC), aiming to provide a preventive st

Shigella type-III secretion system effectors counteract the induction of host inflammation and cell death.

Many enteric bacterial pathogens deliver virulence effectors to counteract host innate immune responses, such as inflammation and cell death, and colonize the intestinal epithelium. However, host cells recognize the disruption of their innate immune signaling by bacterial effectors and induce alternative immune responses, collectively termed "effector-triggered immunity", to clear bacterial pathogens. Here, we describe a mechanism of cell death induction via effector-triggered immunity and the bacterial countermeasures of the pathogen Shigella flexneri. Shigella delivers the OspI effector to i

Sexually transmitted enteric infections in men who have sex with men.

SUMMARYSexual transmission of enteric infections (STEIs) among men who have sex with men (MSM) has been reported since the 1960s and is increasingly recognized since the widespread adoption of multiplex molecular diagnostics. However, the overall burden of sexually transmitted enteric infections has been difficult to ascertain, as the public health response to these infections and identification of transmission networks fall between the traditional groupings of sexually transmitted and foodborne diseases. The global emergence of extensively drug-resistant Shigella and Campylobacter infections

Shigella PCR-positive Gastroenteritis in Children: Insights From a Comparison Between Culture-positive and Culture-negative Cases.

BACKGROUND: Detection of Shigella in stool by polymerase chain reaction (PCR) is rapid and highly sensitive but presents challenges in distinguishing between disease-associated and asymptomatic Shigella carriage. This study compared children with gastroenteritis who had Shigella identified in their stool by PCR-testing and a positive stool culture to those with Shigella identified by PCR-testing but a negative culture. METHODS: All stool samples of outpatients and inpatients (<18 years) tested by multiplex-PCR at the Soroka University Medical Center laboratory between January 2020 and July 20

Antibodies elicited by altSonflex1-2-3 GMMA vaccine are bactericidal against a panel of drug-resistant Shigella clinical isolates.

Shigellosis significantly impacts global health, particularly affecting vulnerable populations in low- and middle-income countries, with over 270 million annual infections. It also causes morbidity in specific high-risk groups in high-income countries. Antibiotic treatment is increasingly compromised by multidrug-resistant strains, highlighting the urgent need for a Shigella vaccine. We developed a four-component O-antigen-based vaccine candidate targeting Shigella sonnei and Shigella flexneri 1b, 2a, and 3a serotypes, named altSonflex1-2-3, to provide broad protection against most globally pr

High-content high-resolution microscopy and deep learning-assisted analysis reveals host and bacterial heterogeneity during Shigella infection.

Shigella flexneri is a Gram-negative bacterial pathogen and causative agent of bacillary dysentery. S. flexneri is closely related to Escherichia coli but harbours a virulence plasmid that encodes a type III secretion system (T3SS) required for host cell invasion. Widely recognised as a paradigm for research in cellular microbiology, S. flexneri has emerged as important to study mechanisms of cell-autonomous immunity, including septin cage entrapment. Here, we use high-content high-resolution microscopy to monitor the dynamic and heterogeneous S. flexneri infection process by assessing multipl

Enhanced virulence and stress tolerance are signatures of epidemiologically successful Shigella sonnei.

Shigellosis is a leading cause of diarrhoeal deaths, with Shigella sonnei increasingly implicated as a dominant agent. S. sonnei is divided into five monophyletic lineages, yet most infections are caused by a few clonal sub-lineages within Lineage 3 that are quite distinct from the widely used Lineage 2 laboratory strain 53G. Factors underlying the success of these globally dominant lineages remain unclear in part due to a lack of complete genome sequences and animal models. Here, we utilise a novel reference collection of representative Lineage 1, 2 and 3 isolates and find that epidemiologica

Utilizing the effectiveness of phage cocktail to combat Shigella and Salmonella infections and their polymicrobial biofilm control activity.

BACKGROUND: Shigella and Salmonella are major foodborne and waterborne pathogens responsible for acute gastrointestinal infections and significant global morbidity and mortality. Both species are capable of forming bacterial biofilms in the food processing industry, a key survival mechanism that significantly reduces the effectiveness of antibacterial drugs. The global rise in antimicrobial resistance (AMR) necessitates the urgent development of new strategies. Bacteriophages, particularly phage cocktails, provide a potential alternative because of their host specificity and ability to degrade

Real-Time Intracellular Bacterial Metabolism During Shigella Infection Using Hyperpolarized [2-13C]Pyruvate.

Intracellular bacterial infections are challenging to study metabolically, as traditional methods often rely on indirect indicators. Hyperpolarized magnetic resonance spectroscopy (MRS) enables noninvasive, real-time metabolic monitoring, while being able to distinguish between pathogenic bacterial metabolism and host metabolism. Using dDNP-NMR, we demonstrate real-time conversion of hyperpolarized [2-13C]pyruvate to [1-13C]acetate within Shigella-infected HeLa cells. The conversion of hyperpolarized [1,4-13C2]fumarate to [1,4-13C2]malate was used to verify host cell integrity during infection

Extended spectrum β-lactamase and integron genes among Shigella flexneri and Shigella sonnei isolated from children with diarrhea in shiraz, Southwest Iran.

Shigellosis, a bacterial infection marked by dysentery, is typically a self-limiting disease and can be effectively managed with oral rehydration. However, antibiotics may help reduce the severity and duration of Shigellosis despite rising antimicrobial resistance. This study examines the prevalence of ESBL-producing isolates and the presence of blaTEM, blaCTX-M, blaSHV, intI1, and intI2 genes among Shigella sonnei and Shigella flexneri isolates from children with diarrhea in Shiraz, southwest Iran. From October 2019 to March 2020, 50 S. flexneri and 50 S. sonnei were isolated from children un
