Sustained consummatory reward processing deficits in remitted melancholic depression.

BACKGROUND: Depression is believed to be associated with functional alterations in both the anticipation and the consumption phase of the reward processing. It may also manifest in depressed individuals during remission. Still, depression is a heterogeneous syndrome, and reward-related impairments could be more evident in those having melancholic depression. Therefore, the present study aimed to examine whether neural abnormalities are present in individuals with remitted melancholic depression during the reward anticipation and reward consumption phases. METHODS: Thirty-one participants with

Systematic Review of Methylenetetrahydrofolate Reductase (MTHFR) 677C>T and 1298A>C Variants and Treatment-Resistant Depression: Insights for Precision Psychiatry.

INTRODUCTION: Treatment-resistant depression (TRD) is a major clinical challenge, affecting approximately one-third of patients with major depressive disorder (MDD). Genetic factors, particularly genetic variants in the MTHFR gene, have been implicated in antidepressant response variability. The MTHFR 677C>T (rs1801133) and 1298A>C (rs1801131) variants are associated with altered folate metabolism, increased homocysteine levels, and potential disruptions in neurotransmitter synthesis, which may contribute to TRD. This systematic review aimed to evaluate the association between MTHFR variants a

Sleep, Steps, and Screens: Between- and within-person effects of digital markers of daily life behaviors on smartphone-based assessments of cognitive functioning in depression.

Cognitive impairment represents a core feature of major depressive disorder (MDD), often persisting after mood symptoms remit and not addressed by usual antidepressant treatments. Despite its relevance, cognition is typically assessed with infrequent tests in clinical settings, overlooking its contextual nature. Smartphones and wearables enable ecologically valid, repeated measurements of cognition and daily life behaviors that may impact it. We examined whether sleep duration, step count, and smartphone screen time are associated with cognitive functioning in MDD. We conducted secondary analy

Psychological distress among emergency medical services personnel: implications for public health preparedness and system resilience.

INTRODUCTION: Emergency Medical Services (EMS) personnel are integral to disaster and emergency response systems. Recurrent exposure to high-acuity incidents, irregular shifts, and operational pressures may increase vulnerability to psychological distress, with potential implications for workforce sustainability and system resilience. This study assessed the severity distribution and predictors of stress, anxiety, and depression among EMS personnel in the Makkah region of Saudi Arabia. METHODS: A cross-sectional survey was conducted between January and April 2024 among certified EMS profession

Pathway-specific regulation of the paraventricular thalamic nucleus in depressive-like behavior.

Bipolar disorder is one of major psychiatric disorders afflicting about 1% of the world population. While genetic factors have been increasingly recognized as contributing to the disorder, the underlying etiology of bipolar disorder, such as associated brain regions and cellular pathology, are yet to be elucidated. Our previous studies using a genetic bipolar disorder animal model indicated potential role of the paraventricular thalamic nucleus (PVT) in recurrent depressive-like episodes that exhibit phenotypic similarity to the depression phase of bipolar disorder. Notably, chronic activation

Maternal health and autism risk: parsing direct and indirect genetic effects using 3-generation family linkage.

Despite autism's prominent genetic etiology and early-life origins, parsing genetic effects contributing to the condition into those that operate directly (via allelic transmission to offspring) vs. indirectly (via influencing prenatal environment) remains challenging. We examined this using a novel design leveraging 3-generation family linkage in Danish national registers. The cohort included all children born in Denmark from 1998-2015 and their relatives identified through 3-generation family linkage. The analytic sample comprised full maternal cousin pairs, including parallel (children of m

Therapeutic effects of TRPC4/5 antagonism in a rat model of bipolar disorder.

Bipolar disorder, a chronic neuropsychiatric condition, is characterized by alternating episodes of mania and depression. Although acute manic episodes have become increasingly manageable with the advent of third-generation neuroleptics, the primary therapeutic challenge remains the prevention of recurrences and the effective management of often refractory depressive phases. Emerging evidence implicates transient receptor potential (TRP) channels in mood regulation through their roles in calcium signaling, synaptic plasticity, and neuroinflammation. Among these, TRPC5 has gained attention as a

Cognition and future depression: associations with risk in those with and without a history of depression.

BACKGROUND: Cognitive impairments are common in depression and often persist beyond mood resolution. However, the relationship between cognitive performance, its neurological underpinnings, and future depression risk is unclear, limiting strategies for primary and secondary prevention. OBJECTIVE: Our objective was to determine whether cognition associates with subsequent depression, both relapse and first-episode occurrences. METHODS: 1862 UK Biobank participants with a history of International Classification of Diseases (ICD)-10-defined depression in remission (RD) (mean (SD) age: 52.7 (7.1

White matter abnormalities in bipolar II and unipolar depression - Evidence from fixel-based analysis.

BACKGROUND: Challenges in correctly identifying bipolar disorder II (BD-II) during depressive states have led to poor clinical outcomes. BD-II-specific diffusion-weighted imaging studies are lacking. This study addresses current knowledge gaps by comparing white matter (WM) integrity in BD-II and unipolar depression (UD) using fixel-based analysis (FBA). METHOD: Primary analyses compared fibre density (FD) among 33 individuals with BD-II, 50 with UD, and 51 healthy controls (HC) in 11 a priori tracts. Sensitivity analyses among unmedicated participants examined whether medication status affec

Incident and Recurrent Suicide Attempts in Adolescents With Major Depressive Disorder: Baseline Correlates and Prospective Predictors in a One-Year Follow-Up.

BackgroundSuicidal behavior in adolescents with major depressive disorder (MDD) remains a critical public health concern. While prior research has identified numerous correlates of suicide attempts, few longitudinal studies have distinguished between factors associated with incident versus recurrent suicidal behavior.MethodsIn this prospective follow-up study, adolescents diagnosed with MDD were assessed at baseline (T0) and followed for one year (T1). Suicide attempts were evaluated longitudinally, and participants were categorized according to baseline and follow-up suicide attempt status. M

Subclinical neuropsychiatric trait variation in parents of children with autism spectrum disorder: a cohort study.

BACKGROUND: Many neuropsychiatric conditions share overlapping features underpinned by shared genetics. In this study, we examined the co-aggregation of sub-threshold variation in neuropsychiatric phenotypes in biological parents of autistic children to provide insights into the intergenerational transmission of genetic liability for autism. METHODS: Autistic, neuropsychiatric (i.e., anxiety, depression, ADHD), and cognitive traits were characterized in biological parents of children enrolled in a longitudinal developmental study: 189 families of autistic children and 100 families with no aut
