Marked Improvement of Chronic Motor and Phonic Tics With Cariprazine in an Adult With Tourette Syndrome With Comorbid Bipolar Disorder: A Case Report.

Tourette syndrome (TS) is a neuropsychiatric disorder characterized by motor and phonic tics, often complicated by comorbid conditions and treatment-related adverse effects. We report a 61-year-old male with long-standing TS and comorbid bipolar disorder who experienced persistent, functionally impairing tics despite multiple prior therapies. Following initiation and titration of cariprazine, the patient demonstrated marked and sustained improvement in tic severity over approximately two years, with good tolerability and stable mood symptoms. This case highlights a potential role for cariprazi

Bidirectional associations between mental health conditions and cognitive impairment in patients with pain conditions of the back, neck, and spine: A population-based study.

BACKGROUND: Pain conditions (PCs) of the back, neck, and spine are frequently accompanied by psychiatric and cognitive comorbidities in older adults. However, the directionality and magnitude of the associations between psychiatric disorders and cognitive impairment in this population remain insufficiently characterized. OBJECTIVE: This population-based study examines the bidirectional relationship between Mental Health Conditions (MHCs) and Cognitive Impairment (CI) among patients with back, neck, and spine pain. It assesses whether MHCs-including depression, Bipolar Disorder (BD), Generaliz

Lifetime and acute risk of suicidality by profiles of early life adversity: an observational cohort study in a high-risk population.

Early life adversity in the form of child maltreatment (CM) is a transdiagnostic risk factor for suicidal ideation and behavior (SIB), yet its heterogeneity and relevance for lifetime versus acute SIB and psychotherapy outcomes remain unclear. In an observational cohort of 240 high-risk adults with borderline personality disorder (42.5%) or persistent depressive disorder (57.5%), lifetime and acute SIB were assessed using the Columbia-Suicide Severity Rating Scale. CM profiles were derived using a data-driven clustering approach based on the Childhood Trauma Questionnaire. Associations between

Relationship between mental disorders and non-traumatic cerebral hemorrhage: cross-sectional analysis and mendelian randomization.

BACKGROUND: The causality between mental disorders and spontaneous non-traumatic cerebral hemorrhage (NCH) has not been sufficiently explored. This study aims to investigate the associations of a broad spectrum of 16 mental disorders with NCH through Mendelian randomization (MR) analysis and to assess the association in a real-world study. METHODS: Univariate analysis and mediational MR analysis were employed to investigate the potential association between mental disorders and NCH. Genetic instruments associated with mental disorders were identified through a genome-wide association study (G

Global burden of depressive disorders from 1990 to 2021: temporal trends, COVID-19 Impact, and projections to 2050.

Depressive disorders constitute a leading global cause of disability, yet comprehensive evidence regarding their long-term trends, socioeconomic disparities, and the impact of the COVID-19 pandemic remains limited. This study aimed to investigate the global burden, epidemiological patterns, and risk factors of depressive disorders from 1990 to 2021, and to project future trends to 2050. Using Global Burden of Disease 2021 data, we analyzed spatial-temporal patterns, demographic disparities, and socioeconomic gradients with joinpoint regression, cross-lagged modeling, and SHAP analysis. The association between sleep spindles and cognitive performance in euthymic bipolar disorder.

Many people with bipolar disorder (BD) experience persistent cognitive deficits. Sleep spindles have been linked to cognitive ability in healthy populations and psychotic disorders. While there is preliminary evidence for altered spindle activity in BD, research directly examining the association between sleep spindle parameters and cognitive performance in this population is lacking. Therefore, our primary objective was to examine the association between fast spindle density and episodic memory performance in euthymic individuals with BD. As exploratory analyses, we looked at associations bet

Long-term impairments associated with gut-brain axis dysregulation following sublethal VX exposure in mice.

Exposure to organophosphorus (OP) compounds can induce transient cognitive, neurological, and somatic symptoms that may persist over time. OP poisoning mostly occurs from pesticides used in developing countries; however, several OP nerve agent (NA) events have been reported in the last decade. OP toxicity is based on cholinesterase inhibition, which leads to varying degrees of neurotoxicity. According to clinical reports, asymptomatic victims of OP exposure may experience long-term neurological sequelae. Given the continuous communication between the nervous and enteric systems, evaluating the

Octreotide Attenuates Anxiety-Like Behaviors and Cognitive Deficits in a Rat Model of Post-traumatic Stress Disorder.

Post-traumatic stress disorder (PTSD) is a long-lasting mental health condition characterized by persistent intrusive recollections, avoidance behaviors, mood and cognitive alterations, heightened arousal, and significant cognitive deficits, often accompanied by hypothalamic-pituitary-adrenal (HPA) axis dysregulation and neuroinflammation. This study evaluated the therapeutic potential of octreotide (OCT), a synthetic somatostatin analog, in a rat model of PTSD induced by foot-shock contextual fear conditioning. Male Wistar rats (n = 40) were randomly assigned to four groups (n = 10/group): Sa

Cognition and future depression: associations with risk in those with and without a history of depression.

BACKGROUND: Cognitive impairments are common in depression and often persist beyond mood resolution. However, the relationship between cognitive performance, its neurological underpinnings, and future depression risk is unclear, limiting strategies for primary and secondary prevention. OBJECTIVE: Our objective was to determine whether cognition associates with subsequent depression, both relapse and first-episode occurrences. METHODS: 1862 UK Biobank participants with a history of International Classification of Diseases (ICD)-10-defined depression in remission (RD) (mean (SD) age: 52.7 (7.1

Long-term cognitive effects after SCG-DBS in patients with treatment resistant depression: A 5-year follow-up.

INTRODUCTION: Cognitive symptoms represent a significant and persistent challenge in patients with treatment-resistant depression (TRD), substantially impacting their functional outcomes. Deep brain stimulation (DBS) targeting the subcallosal cingulate gyrus (SCG) has shown clinical efficacy. Nonetheless, its long-term effects on cognition remains insufficiently researched. This study systematically evaluates cognitive outcomes following SCG-DBS over an extended follow-up period. METHODS: This retrospective, exploratory secondary analysis used longitudinal data from 12 TRD patients treated wi

Glucocorticoid receptor antagonism in major depressive disorder with childhood trauma: a randomized controlled trial.

Childhood trauma (CT) is a key risk factor for major depressive disorder (MDD) onset and persistence. Hypothalamic-pituitary-adrenal (HPA) axis dysregulation may underlie this link, and preclinical studies suggest glucocorticoid receptor (GR) antagonism can reverse early life stress effects. This study tested whether the GR antagonist mifepristone reduces depressive symptoms in adults with MDD and CT. The RESET-medication study was a randomized, double-blind, placebo-controlled trial evaluating a 7-day course of mifepristone (1200 mg/day) or placebo in 158 adults with MDD and CT, assessed at b

Measuring psychomotor agitation via hand tremor in remitted major depressive disorder.

Psychomotor agitation (PmA), a common yet understudied symptom in Major Depressive Disorder (MDD), is characterized by heightened motor activity such as restlessness and fidgeting. PmA, like other symptoms of MDD, may persist into remission. This study examined the role of PmA in remitted MDD by using a novel smartphone based physiological measure of tremor. Participants (N = 157) consisted of healthy controls (N = 76) and those with remitted MDD (N = 81, to reduce the impact of current MDD symptoms on PmA), and tremor was assessed under the manipulation of position (palm-up, arm extended) and

Post-Stroke Epileptogenesis as a Dynamic Network Process: State-Dependent Biomarkers, Longitudinal Monitoring, and the Concept of an Irritable Brain State.

Post-stroke epilepsy (PSE) is one of the most common long-term neurological complications of stroke and represents a major contributor to disability, reduced quality of life, and increased healthcare burden. Although numerous risk factors and biomarkers have been proposed, many current risk models still emphasize lesion-related predictors and do not fully account for the temporal evolution of post-stroke network excitability. This narrative review aims to examine post-stroke epileptogenesis as a dynamic process of network reconfiguration and to summarize current evidence on biological, neuroph
