Differential temporal links of serum interleukin‑6 with posttraumatic stress disorder and depression after trauma.

This study investigated the longitudinal associations between serum interleukin-6 (sIL-6) levels measured shortly after physical injury and the subsequent development of post-traumatic stress disorder (PTSD) and depressive disorder across two years. Adults with moderate to severe physical injuries were prospectively enrolled from a regional trauma center. Within one month of hospitalization, fasting blood samples were collected for sIL-6 measurement, and detailed socio-demographic and clinical data were obtained. PTSD was assessed using the Clinician-Administered PTSD Scale for DSM-5 (CAPS-5),

From prediction to mechanism: rethinking PTSD treatment non-response through an iron-inflammation framework.

Precision psychiatry has improved prediction of treatment response across psychiatry, but translating biomarkers into treatment strategy remains difficult. Using PTSD as a model, we outline an iron-inflammation framework linking treatment-predictive neural circuitry to an upstream biological mechanism, illustrating how neuroimaging may support biologically informed stratification and targeted adjunctive treatments.

Efficacy of eye movement desensitization and reprocessing for anxiety: An umbrella review and meta-meta-analysis of randomized controlled trials.

BACKGROUND: Anxiety disorders are highly prevalent, and many patients do not remit with first-line treatments. Eye Movement Desensitization and Reprocessing (EMDR) is effective for post-traumatic stress disorder (PTSD), but meta-analytic evidence on anxiety outcomes remains fragmented. OBJECTIVE: To synthesize meta-analyses of randomized controlled trials on the efficacy of EMDR in reducing anxiety symptoms. METHODS: Web of Science, PubMed, PsycINFO, and Scopus were searched from inception to April 2026 (PROSPERO CRD420261373395). Two reviewers independently selected studies, extracted data,

Exercise Interventions for PTSD: A Systematic Review and Network Meta-Analysis of Overall Effects, Comparative Effectiveness, and Ranking Uncertainty.

Background/Objectives: Exercise has been investigated as an adjunctive intervention for post-traumatic stress disorder (PTSD), but the comparative effectiveness of exercise modalities remains uncertain. This study evaluated the effects of exercise and movement-based interventions and whether the available evidence supports a reliable hierarchy of exercise modalities. Methods: PubMed, APA PsycINFO, the Cochrane Central Register of Controlled Trials (CENTRAL), and ScienceDirect were searched through 31 January 2026 for randomized controlled trials in adults with PTSD or clinically significant sy

Potential effectiveness of war-adapted brief prolonged exposure for PTSD prevention: preliminary study among Nova Festival survivors during ongoing conflict.

BACKGROUND: The 7 October 2023, terrorist attack at the Nova Festival in southern Israel caused deaths, injuries, hostage-taking and prolonged life-threatening exposure, creating an urgent need for early interventions to reduce post-traumatic morbidity. Although prolonged exposure is an established post-traumatic stress disorder (PTSD) treatment, its feasibility and benefit during ongoing threat remain unclear. AIMS: This preliminary study evaluated a war-adapted brief prolonged exposure protocol (WBPE) delivered shortly after trauma in Nova Festival survivors with acute stress disorder. MET

Living under fire: longitudinal insights into civilian coping and recovery during armed conflict.

This longitudinal study examines the psychological impact of the 2023 Israel-Hamas war on Jewish Israeli civilians, focusing on changes in mental health, social support, and institutional trust over time. Data were collected at two time points: one month after the outbreak of war (T1; November 2023) and approximately 16 months later (T2; March 2025). Participants (N=409 at T1; N=293 at T2) completed validated measures of anxiety, post-traumatic stress disorder (PTSD), social support, trust, and emotion regulation. Results indicate a significant decrease in anxiety and PTSD symptoms over time a

Use of the arousal-based individual screening model for studying post-traumatic stress disorder: Findings and future directions.

Post-traumatic stress disorder (PTSD) is a debilitating psychiatric condition arising in a subset of trauma-exposed susceptible individuals, yet the pathophysiological determinants of this individual variability remain largely unresolved. We developed the arousal-based individual screening (AIS) model with the aim of filling key gaps in the preclinical field. The AIS model identifies susceptible and resilient mice of both sexes by applying z-normalization of post-trauma changes in acoustic startle reactivity (ASR) induced by a single 24-hour restraint. Susceptible mice develop a long-lasting,

Measurement-Based Care for PTSD.

Festschrift for Jon Conte's research idea. Effective treatments for post-traumatic stress disorder (PTSD) in children and adults exist but are not widely available. Among those who have access, there is variability in response: some do not engage, some are better within a few sessions and some are not better at the endpoint. Even among trained providers, there is drift from adherent delivery over time. Clients with PTSD typically receive whatever the provider has to offer. An alternative approach is measurement-based care (MBC) that focuses on outcomes versus treatment model or content. MBC is

Stage- and context-dependent modulation of fear memory by oxytocin: Neurobiological mechanisms and implications for PTSD.

Post-traumatic stress disorder (PTSD) is a multifactorial psychiatric condition characterized by re-experiencing, avoidance, hyperarousal, negative alterations in cognition and mood, and disturbances in social and emotional functioning. Within this broader clinical syndrome, persistent traumatic memories and excessive fear generalization represent important mechanistic and therapeutic domains. Despite substantial advances in understanding its neurobiological basis, current treatments remain suboptimal, with limited efficacy and high dropout rates. The neuropeptide oxytocin (OXT), a key modulat

Ketamine administration reduced conditioned defensive behaviour and upregulated protein expression associated with neuroplasticity in a planarian model of aversive memory reactivation.

Traumatic memories that are resistant to modification or extinction are a characteristic feature of post-traumatic stress disorder (PTSD). The process of fear memory reconsolidation has been identified as a potential therapeutic target for the treatment of PTSD. Ketamine, an NMDA receptor antagonist with dissociative properties, has been identified as a drug with high therapeutic potential for this target, as it destabilises traumatic memory while enabling rapid modification and update of the memory to no longer be threatening. However, its exact mechanisms of action on reducing the impact of

A cross-sectional study of the prevalence and correlates of post-traumatic stress among people attending acceptance and commitment therapy for chronic pain.

OBJECTIVE: Experiences of trauma are common among people with chronic pain, although estimates of comorbid post-traumatic stress vary widely across contexts. Acceptance and commitment therapy (ACT), which targets psychological flexibility to improve pain-related functioning, is increasingly delivered for people with pain. To inform treatment delivery, understanding of the prevalence and correlates of post-traumatic stress among those attending ACT-based treatment for pain is needed. This study aimed to identify the prevalence of post-traumatic stress and its associations with key pain-related

Reduced Somatomotor Cortical Thickness in Women With PTSD After Childhood Sexual Abuse: A Structural Correlate Distinct From Adult Sexual Dysfunction.

BACKGROUND: Childhood sexual abuse (CSA) is a major risk factor for posttraumatic stress disorder (PTSD) and adult sexual dysfunction in women. Although adverse childhood experiences have been linked to altered brain structure, cortical alterations specifically associated with CSA and PTSD in women remain poorly characterized, and their relationship to adult sexual functioning is unknown. METHODS: Women with CSA and current PTSD (n = 52), women with CSA without current PTSD (n = 32), and control women without CSA or PTSD (n = 48) underwent structural magnetic resonance imaging. Voice and speech as digital biomarkers of mental health: a scoping review of clinical interpretability, data collection and translational readiness.

BACKGROUND: Voice and speech encode acoustic, linguistic, and interactional information associated with mental health. Translating these signals into clinically meaningful digital biomarkers depends not only on predictive performance but on how speech is elicited, how the target state is defined, whether findings generalize beyond the original dataset, and whether features can be measured reliably in real-world settings. OBJECTIVE: To map voice, speech, language, and communication biomarkers studied across mental health outcomes and to examine three dimensions of translational readiness: clin

Adolescent M-CSF Administration Provides Durable Protection Against Trauma-Induced PTSD-Like Phenotypes in Male Mice Through Microglia-Dependent Mechanisms.

Post-traumatic stress disorder (PTSD) is a severe psychiatric condition for which effective preventive strategies are lacking. Innate immune preconditioning has shown promise for the prevention of PTSD, yet the transient nature of such protection when induced in adulthood poses a substantial limitation. Using male C57BL/6J mice, we tested whether administration of macrophage colony-stimulating factor (M-CSF, 100 µg/kg, intraperitoneally) during adolescence, a developmental period characterized by heightened microglial plasticity, could establish enduring resilience against trauma-induced PTSD-

Complex post-traumatic stress disorder in refugees, asylum seekers, and internally displaced people: a systematic review and meta-analysis.

Background: Forcibly displaced populations, including refugees, asylum seekers, and internally displaced persons, are frequently exposed to severe, prolonged and repeated trauma from which escape is difficult or impossible. Such exposures increase the risk for complex post-traumatic stress disorder (CPTSD), defined by core PTSD symptoms alongside disturbances in self-organisation (DSO), including affective dysregulation, negative self-concept, and relational difficulties. However, the prevalence of CPTSD in forcibly displaced populations has not yet been quantitatively synthesised, and the add

In-session cortisol reactivity as a predictor of treatment response in Korean patients with PTSD.

Post-traumatic stress disorder (PTSD) is associated with hypothalamic-pituitary-adrenal (HPA) axis dysregulation, and in-session cortisol reactivity during exposure-based therapy may carry prognostic value. This study examined whether within-session salivary cortisol reactivity during Written Exposure Therapy (WET) predicts treatment response in Korean patients with PTSD. In a secondary analysis of a non-randomized waitlist-controlled trial (N = 41), cortisol was collected before and 15 min after the writing task at each of five sessions. Treatment response was defined using the reliable chang

Two distinct projection pathways converge onto the same BLA neurons to contribute to fear memory retrieval.

Studying the neural circuits that mediate context-induced fear memory retrieval is of importance to develop new therapeutic approaches to prevent post-traumatic stress disorder relapse. The basolateral amygdala (BLA) is crucial for context-induced retrieval of fear memory. Previous studies indicate that projection neurons from the ventral hippocampus CA1 (vCA1) to the BLA (vCA1-BLA neurons) mediate context-induced retrieval of fear memory. However, it remains unclear whether other projection neurons to the BLA are involved in context-induced retrieval of fear memory and how they interact with

Balancing Innovation and Evidence: Reflections on Structured Protocols in MDMA-Assisted Therapy Versus a Principle-Guided, Patient-Directed Approach.

There is growing interest in incorporating MDMA (3,4-Methylenedioxymethamphetamine) into structured, evidence-based cognitive-behavioral treatment protocols for post-traumatic stress disorder. Yet this risks overlooking the distinctive qualities of MDMA-assisted therapy, which has already demonstrated strong clinical effects, high response rates, and low dropout using a principle-guided, patient-directed therapeutic model. This model integrates MDMA administration into a broader therapeutic process, including nondrug preparatory and integrative sessions, designed to support the unique, nonline

A Case of Complex Post-Traumatic Stress Disorder Manifesting With Psychogenic Non-Epileptic Seizures.

In cases where complex post-traumatic stress disorder (CPTSD) co-occurs with psychogenic non-epileptic seizures (PNES), it is essential to emphasize that severe interpersonal trauma is strongly associated with PNES. Failure to recognize this association may result in misdiagnosis, inappropriate treatment (including polypharmacy), and potential iatrogenic harm.

Associations of post-traumatic stress disorder and metabolic syndrome with accelerated brain aging in South African adults: A case-control study.

UNLABELLED: Post-traumatic stress disorder (PTSD) and metabolic syndrome (MetS) are both associated with childhood trauma (CT) and accelerated brain aging. We examined the putative moderating effects of CT exposure on the associations of PTSD diagnosis and MetS status with brain aging in cases (n = 97) compared to trauma-exposed controls (n = 87). Brain age predictions were generated from T1-weighted magnetic resonance image scans using a pre-trained machine learning pipeline (brainageR). A measure of relative brain aging known as "predicted age difference" (PAD) was calculated by subtracting
