Correlation between bacterial culture and IS900 qPCR to detect Mycobacterium avium subsp. paratuberculosis in the feces of sika deer.

Mycobacterium avium subsp. paratuberculosis (MAP) infection in deer causes paratuberculosis (PTb; Johne's disease), a slowly progressive chronic granulomatous enteritis. Accurate and rapid detection of MAP shedding by subclinical animals is essential for effective control of infection in deer herds. The VetMAX MAP 2.0 qPCR assay targeting IS900 was developed for cattle, sheep, and goats. However, the performance of the assay in sika deer (Cervus nippon) has not been evaluated. Here, we describe the use of the VetMAX MAP 2.0 qPCR assay on fecal samples from sika deer for the detection of MAP an

Clinical and ecological significance of bacterial dormancy in human pathogens.

SUMMARYBacteria can enter a transient state of reduced metabolic activity to survive temporary unfavorable conditions. This phenomenon, sometimes referred to as dormancy, growth arrest, or quiescence, enables pathogen survival inside and outside of their host, with major clinical consequences for the management of infections. From spores to persister cells, these states associated with bacterial dormancy share molecular mechanisms for controlling cellular activity and for responding to related environmental signals. In this review, we provide a synthesis of the diversity of dormant states in h

Characterization and genomic analysis of a jumbo Aeromonas hydrophila phage BMBAH1, representing a candidate novel viral genus.

Aeromonas hydrophila is an opportunistic pathogen of fish, humans, and livestock, and has a severe negative impact on aquaculture development. Phage therapy is considered an alternative strategy for controlling bacterial infections and contamination. In this study, a novel phage, Aeromonas hydrophila BMBAH1, was isolated. Transmission electron microscopy showed that phage BMBAH1 had a long tail and an icosahedral head. Biological characterization indicated that phage BMBAH1 remained highly active across a broad range of temperatures and pH values, exhibiting long-term and significant inhibitor

Termite sanitary behaviour and their associated bacteria protect against latent Xylaria and entomopathogenic Metarhizium.

Fungus-growing termites harbour diverse microbial communities in their combs and guts. However, culture-based surveys, cross-colony and cross-regional comparisons and assessments of their antimicrobial activities against entomopathogens and fungal weeds remain limited. Through culture-dependent isolation, we surveyed the bacterial and fungal diversity associated with the combs and guts of termites in the genera Macrotermes and Odontotermes, recovering 600 non-Termitomyces fungal isolates representing 79 genera, as well as 59 bacterial isolates. Pathogenicity tests on Macrotermes gilvus reveale

Gut Microbiota Composition in Helicobacter pylori-Positive Adolescents With Autism Spectrum Disorder: An Exploratory Compositional Analysis.

Alterations in the gut microbiota have been implicated in autism spectrum disorder (ASD), but reported microbial profiles remain inconsistent, potentially because of heterogeneity in factors affecting gut microbial composition. Because Helicobacter pylori infection is associated with alterations in the gut microbiota, we investigated whether ASD was associated with gut microbial composition in a cohort of H. pylori-positive adolescents. This cross-sectional study included seven adolescents with ASD and 80 controls before H. pylori eradication therapy. Gut microbiota was analyzed using 16S rRNA

Biodegradable Hybrid Scaffolds Integrating Synergistic Chemical and NIR-II Photothermal Antibacterial Activity.

Biodegradable orthopedic materials have attracted increasing attention due to their excellent biocompatibility, controllable degradation behavior, and capacity for osteointegration, enabling effective bone regeneration while eliminating the need for secondary implant-removal surgery. However, bacterial colonization and subsequent dense biofilm formation on implanted scaffolds often lead to persistent infections, posing a major barrier to clinical translation. The development of antibacterial-functionalized biodegradable scaffolds is therefore crucial for advancing next-generation orthopedic im

Preparation of antibacterial and hydrophobic cotton fabrics with nanosilver-attached SiO2 particles.

The preparation of antibacterial cotton fabrics is crucial for preventing bacterial infections. This study demonstrates a novel textile finishing strategy that uniquely combines Ag@SiO2 nanoparticles with hexadecyltrimethoxysilane (HDTMS) modification to simultaneously confer antimicrobial property and hydrophobicity to cotton fabric with long-term stability. Ag@SiO2 nanoparticles were synthesized with stannous chloride (SnCl2) as the reducing agent, followed by cotton fabric modification with both HDTMS and Ag@SiO2 nanoparticles. This resulting Cot/Ag@SiO2 fabric exhibited excellent antibacte

Decoding the genomic repertoire of fosfomycin resistance genes in staphylococci.

Fosfomycin is regaining clinical relevance for multidrug-resistant staphylococcal infections, yet the diversity, evolutionary origin and nomenclature of staphylococcal fosfomycin resistance genes remain inconsistently defined. We surveyed 17,024 publicly available genomes representing 14 clinically common Staphylococcus species to curate a catalogue of fosfomycin-modifying fos homologues. By integrating amino-acid identity, phylogenetic topology, genomic context and population-level distribution, we established a reproducible classification framework that resolved species- or lineage-associate

Microbe Profile: Acinetobacter baumannii: persist and resist.

Acinetobacter baumannii is a Gram-negative opportunistic pathogen associated with a wide variety of healthcare-associated infections, including ventilator-associated pneumonia, urinary tract infections, bacteraemia and wound infections. It adopts a 'persist and resist' pathogenic strategy to establish infection in the host. It can form robust biofilms and survive in a desiccated state for months at a time, driving its persistence in the hospital-built environment. It also has a remarkable capacity to rapidly develop antimicrobial resistance, with >90% of contemporary clinical isolates being mu

Unexplained Colistin Resistance in Klebsiella pneumoniae: Insights into sRNA-Mediated Post-Transcriptional Regulation.

Colistin is a last-resort antibiotic used to treat infections caused by multidrug-resistant Gram-negative pathogens, including Klebsiella pneumoniae. Resistance to colistin can be explained by canonical mechanisms involving chromosomal mutations that activate lipid A modification pathways or by acquisition of plasmid-mediated mcr genes. These mechanisms modify the net charge of the bacterial outer membrane, reducing the binding affinity of colistin. However, an increasing number of genomic surveillance studies report colistin-resistant K. pneumoniae isolates that lack detectable alterations in
