Synbiotic intervention with Bifidobacterium bifidum and prebiotics reduces enterohemorrhagic Escherichia coli colonization.

This study evaluated the protective effect of combining Bifidobacterium bifidum with the prebiotics inulin and isomalto-oligosaccharides against enterohemorrhagic Escherichia coli infection and investigated the underlying mechanisms, using an integrated strategy of in vitro screening followed by in vivo validation. Among ten prebiotics screened, IMO and inulin were identified as optimal for promoting B. bifidum proliferation and acetate production (>2.5-fold), with IMO further enhancing bacterial adhesion to Caco-2 cells (four-fold). In a murine infection model, B. bifidum alone reduced total

Conjugation frequency of ESBL- and pAmpC- E. coli in broiler chickens in vivo and in vitro.

BACKGROUND: Plasmid-mediated conjugation is a major form of horizontal gene transfer (HGT), facilitating dissemination of antimicrobial resistance (AMR) and the emergence of multi-drug-resistant (MDR) strains. In poultry, Escherichia coli producing extended-spectrum β-lactamases (ESBL) and plasmid-mediated AmpC β-lactamase (pAmpC) enzymes are common and contribute to antibiotic resistance. Additionally, plasmid-mediated colistin resistance gene mcr-1 in poultry requires attention, as it is a last-resort antibiotic in human medicine. Although plasmid-mediated conjugation is known to play a role

Enteric Infections, Dysbiosis, and Metabolic Dysfunction: The Role of Diarrheagenic Pathogens in Insulin Resistance.

Type 2 diabetes and insulin resistance are increasingly recognized as conditions influenced not only by genetic and lifestyle factors but also by infectious and microbial exposures. Diarrheagenic pathogens, including enterotoxigenic, enteroaggregative, and enterohemorrhagic Escherichia coli, as well as other enteric microorganisms, disrupt the gut microbiota and compromise intestinal barrier integrity. These alterations promote dysbiosis, increased intestinal permeability, and systemic exposure to lipopolysaccharides and other microbial products, leading to metabolic endotoxemia and chronic lo

Human immunodeficiency virus-associated gut microbiome impacts systemic immunodeficiency and susceptibility to opportunistic gut infection.

The gut microbiome of people living with human immunodeficiency virus (PLWH) has been characterized, but its role in influencing host immunity and associated clinical features are unclear. Here we used shotgun metagenomics to characterize the faecal microbiome of two geographically distinct cohorts of PLWH and healthy controls in Israel and Ethiopia. We uncovered disease-specific, geographically divergent microbial patterns including a shift from Bacteroides to Prevotella species in an Israeli cohort and multiple Enterobacteriaceae species including Escherichia coli and Klebsiella quasivariico

Ring-tailed coatis (Nasua nasua) are major carriers of ESBL-producing and diarrheagenic E. coli in an Ecological Park in São Paulo, Brazil.

Escherichia coli is widely distributed in the intestinal microbiota of vertebrates, and the presence of multidrug-resistant (MDR) and extended-spectrum β-lactamase (ESBL)-producing strains in wild animals in urban areas is a result of anthropogenic factors. A total of 232 E. coli isolates were obtained from samples of rectal swabs from 149 coatis. Among the strains, 106 (45.7 %) were classified as MDR, and 47 (20.3 %) showed the ESBL phenotype. Among the 149 animals, 84 (56.4 %) were carriers of MDR strains, and 33 (22.1 %) carried ESBL-producing strains. Regarding beta-lactamase-encoding gene

Sequencing of the invasive E. coli strain BEN2908 isolated from poultry: A comparative investigation of genomic regions shared with intestinal and extraintestinal model E. coli strains.

Extraintestinal pathogenic Escherichia coli (ExPEC) cause disease outside the gut and include avian pathogenic E. coli (APEC), a leading cause of bacterial infections in poultry. Among their highly diverse types, strain BEN2908 stands out for its significant invasive ability across various human and avian cell types. Aiming to investigate further aspects of this strain and its plasmid, we sequenced and assembled the complete genome of BEN2908 and compared it to 22 E. coli strains, including other invasive strains such as adherent and invasive E. coli (AIEC) LF82 and NRG857c, by constructing a

Identification of specific metabolic capacities associated with major extraintestinal pathogenic Escherichia coli lineages.

Bacterial niche colonization relies on multiple factors, among which the metabolic capacity to utilize specific substrates is pivotal. As a gut commensal of humans and other vertebrates acting as an intestinal and extraintestinal opportunistic pathogen, Escherichia coli faces such environmental pressures. We therefore aimed to identify (i) metabolic patterns associated with E. coli lifestyle (commensal versus extraintestinal pathogenic E. coli [ExPEC]) and (ii) ExPEC-specific metabolic features that could help explain the emergence and success of major pandemic clones. Using a pangenomic frame

Impact of plant-based antibiotic alternative supplemented feed on the gut microbiota of Bábolna Tetra-SL chickens experimentally infected with Salmonella enterica and Escherichia coli.

BACKGROUND: Salmonella enterica and Escherichia coli-associated enteric disturbances contribute to health and productivity losses in poultry and pose zoonotic concerns. In the context of antimicrobial resistance, phytogenic feed additives may offer antibiotic-sparing strategies by modulating the intestinal microbiota. We evaluated a fenugreek (Trigonella foenum-graecum) and turmeric (Curcuma longa) extract combination for its microbiota-modulating effects in a controlled dual-challenge model. Vaginal microbiota and urinary tract infections in women: the protective role of vaginal lactobacilli in counteracting urinary tract infections occurrence and recurrence.

Urinary Tract Infections (UTIs) are common clinical conditions. Women are affected by UTIs more than men and their probability of experiencing such conditions is high (60%). The vagina and its defensive system play a key role in the pathogenesis of UTIs, making the vaginal microbiota (VMB) a target for controlling UTIs and their recurrence (rUTIs). Current therapy consists of low-dose antibiotics for rUTIs prevention. The increasing incidence of antimicrobial resistance documented to date has rendered this strategy less attractive. The initial step of UTIs pathogenesis involves colonization of

Mind the gap! Typing Escherichia coli O157:H7 in the pre- and post-genomic revolution eras.

Shiga toxin-producing Escherichia coli (STEC) serotype O157:H7 emerged in the 1980s as a cause of foodborne outbreaks of infectious intestinal disease. Over the last decade, whole-genome sequencing (WGS) has replaced phage typing (PT) as the typing method of choice to detect outbreaks and monitor trends of STEC O157:H7. The aim of this study was to correlate historical PT data with WGS outputs and to describe the continuous trends in STEC O157:H7 notifications in England over a 40-year period from 1985 to 2024.At the UK Health Security Agency (UKHSA), both PT and WGS typing schemes were in use

AI-based monitoring of broiler vocal repertoire dynamics reveals robust developmental and diurnal patterns but limited disease sensitivity.

Broiler vocalizations may contain relevant information about the welfare and health status; however, there is limited information on how disease affects vocal patterns. This study aimed 1) to investigate whether specific vocalization changes emerge in response to two important poultry diseases: intestinal (Eimeria spp.) and respiratory (infectious bronchitis virus, IBV; combined with avian pathogenic Escherichia coli, APEC), and 2) to characterize changes in broiler chicken vocalizations over age (14-41 days of age), and across time of the day (morning: 6:00 AM-12:00 PM; afternoon: 12:00 PM-6:

Identification, immunological function, and antimicrobial mechanisms of CXCL12 in turbot (Scophthalmus maximus).

The chemokine CXCL12, as a pivotal immune regulator, plays a crucial role in teleost fish against bacterial infection, but current research on its specific mechanisms remains scarce. This study systematically identified two copies of the CXCL12 in turbot (Scophthalmus maximus) (SmCXCL12a and SmCXCL12b), investigated their expression patterns, and revealed their immune functions and antibacterial mechanisms. Firstly, phylogenetic and syntenic analyses indicated that the SmCXCL12a/b were relatively conserved and exhibited high homology with orthologs in other teleost fish, including Japanese flo

Description of OXA-244 carbapenemase-producing Escherichia coli in farm animals in The Netherlands, 2024.

BACKGROUND: To minimize the risk of livestock becoming a reservoir of carbapenemase-producing Enterobacterales (CPE), it is prohibited to use carbapenems in food-producing animals in Europe. Nonetheless, CPE have been detected in several EU countries in pigs, poultry and cattle. OBJECTIVES: To detect and characterize CPE from farm animals in the Netherlands obtained within the European monitoring programme for antimicrobial resistance in animals and food. METHODS: Caecal samples from livestock animals are screened for the presence of CPE according to the recommended EURL-AR protocol and supp

Punicalagin alleviates enterotoxigenic Escherichia coli-induced intestinal mucosal injury via Krüppel-like factor 4.

Previous studies have shown that Enterotoxigenic Escherichia coli (ETEC) causes damage to the intestinal mucosal injury, and punicalagin (PUN) can help to ameliorate this damage. This study systematically investigates the protective effects of punicalagin on ETEC-induced intestinal damage and delineates the molecular targets implicated in its action. Experimental findings demonstrate that punicalagin administration significantly attenuates (P < 0.05) ETEC-induced weight loss, mucosal architecture disruption, and goblet cell depletion in a murine model. In parallel, punicalagin markedly enhance

Intestinal colonization of weaner pigs by extended-spectrum-β-lactamase-encoding Escherichia coli classified for differential host-association using a phylogenetics-based approach.

Intestinal bacteria in healthy humans and animals are important reservoirs of antimicrobial resistance (AMR) genes. Certain Escherichia coli lineages dominate the global spread of AMR, but the extent of their host specificity and its impact on transmission remains unclear. Here, 1,198 E. coli isolates from cattle, chickens, humans, and pigs across Germany, the UK, Spain, and Vietnam were classified for host-association using a phylogeny-based bioinformatic approach, with non-host-associated isolates defined as 'generalists'. To validate this classification, 17 extended-spectrum beta-lactamase

Locus of Adhesion and Autoaggregation (LAA) pathogenicity island genes hes and sisA are involved in virulence and biofilm formation in LEE-negative Shiga toxin-producing Escherichia coli (STEC).

Shiga toxin-producing Escherichia coli (STEC) is a major foodborne pathogen capable of causing severe diseases such as hemolytic uremic syndrome (HUS), particularly in children. While the locus of enterocyte effacement (LEE) has long been associated with STEC virulence, an increasing number of LEE-negative strains are being linked to human infections. Some strains harbor the locus of adhesion and autoaggregation (LAA), a pathogenicity island comprising genes implicated in adhesion and biofilm formation. In this study, we investigated the role of two LAA-associated genes, hes and sisA, in the v

Human enterotoxigenic Escherichia coli (ETEC) infections elicit antibodies that broadly neutralize mucinases of pathogenic Escherichia coli and Shigella.

Enterotoxigenic Escherichia coli (ETEC) and Shigella are the most common bacterial diarrheal pathogens among young children of low-middle income regions. Enteric pathogens must overcome formidable host defenses, including the protective barrier formed by intestinal mucus. ETEC produce a virulence protein called EatA, a member of the Serine Protease Autotransporter of the Enterobacteriae (SPATE) family, where the secreted passenger domain (EatAp) specifically degrades MUC2, the major mucus secreted by goblet cells of the human intestine. Notably, some Shigella spp., as well as other diarrheagen
