KCTD10 Modulates DSS-induced Ulcerative Colitis via Regulating Intestinal Stem Cell Function.

The incidence of inflammatory bowel disease (IBD) continues to rise annually, which severely threatens public health. While estimates indicate that current therapies cause a 30% IBD clinical treatment failure, the precise pathogenesis remains unclear. Here, we found that a key embryonic development regulator, KCTD10, is correlated with the severity of IBD. We observed enhanced KCTD10 expression during enterocolitis. Intestinal epithelial cell (IEC)-specific knockout of KCTD10 in mice impaired intestinal epithelial barrier function, activated intestinal adaptive immune response, and disrupted i

Epidemiological comparison of cirrhosis to other long-term health conditions in the United Kingdom.

BACKGROUND: Although cirrhosis is a serious long-term health condition (LTC), it tends to receive little attention from healthcare policymakers. Here, we sought to describe how cirrhosis compares to other LTCs to inform priority-setting discussions. METHODS: We analysed data from the Our Future Health (OFH) study, a cohort of ∼1.7 million individuals from England, United Kingdom. Self-reported diagnosis status for cirrhosis and 15 benchmark LTCs (including type 1 diabetes, type 2 diabetes, Parkinson's disease, chronic kidney disease, schizophrenia, and stage 4 cancer) were ascertained from th

Beyond the pelvis: redefining endometriosis as a systemic inflammatory disorder. A systematic review and meta-analysis.

BACKGROUND: Endometriosis has traditionally been defined as a disease localized to the pelvis. However, its clinical presentation is often accompanied by systemic comorbidities that extend far beyond the pelvic cavity, a phenomenon that challenges the traditional disease paradigm. OBJECTIVE AND RATIONALE: This study aimed to comprehensively assess the associations between endometriosis and autoimmune diseases, allergic conditions, chronic pain syndromes, and endocrine disorders through a systematic review and meta-analysis, thereby providing evidence for redefining endometriosis as a systemic

Decoding autoimmune diseases through the adaptive immune receptor repertoires.

The adaptive immune receptor repertoire (AIRR), composed of T-cell receptors (TCRs) and B-cell receptors (BCRs), underpins antigen-specific immunity through extensive diversity generated by V(D)J recombination and junctional modification. Advances in high-throughput AIRR sequencing have enabled comprehensive characterization of clonal diversity, gene usage patterns, and antigen-driven selection at unprecedented resolution. In autoimmune diseases, breakdown of immune tolerance permits the persistence and expansion of autoreactive lymphocyte clones, and is associated with characteristic repertoi

Long-lived plasma cells in rheumatic and selected antibody-mediated autoimmune diseases: biology, evidence, and therapeutic opportunities.

Persistent autoantibody production after conventional immunosuppression or B-cell depletion has focused attention on antibody-secreting cells that survive without ongoing proliferation. Long-lived plasma cells (LLPCs) are indispensable for durable protective immunity, but autoreactive members of this compartment may also maintain pathogenic antibodies in selected autoimmune diseases. Interpretation is complicated because longevity is usually inferred from population kinetics, anatomical persistence, transcriptional state, or treatment resistance rather than demonstrated by lineage tracing in h

Effectiveness of exercise training on hippocampal mitochondrial regulation in Alzheimer's disease: A mechanistic systematic review.

Alzheimer's disease (AD) is associated with hippocampal mitochondrial dysfunction, including impaired mitophagy, biogenesis, dynamics, bioenergetics, and redox homeostasis, which may contribute to amyloid-β (Aβ) accumulation, tau pathology, and cognitive decline. This systematic review examined whether exercise training is associated with changes in hippocampal mitochondrial regulation and cognitive function in transgenic AD rodent models. Following PRISMA 2020 guidelines, PubMed, EMBASE, and Web of Science were searched up to July 2, 2026. Twenty-one studies met inclusion criteria. Exercise m

Homocysteine and related B-vitamins throughout our International Conferences: a historical perspective.

In this invited keynote review, I reflect on biochemical, clinical, and epidemiological evidence on the role of folate and vitamin B12 in one-carbon metabolism and their relationship with homocysteine, integrating over five decades of research. One-carbon metabolism represents a central biochemical network linking folate-dependent one-carbon transfer reactions with vitamin B12-dependent remethylation of homocysteine to methionine. Disruptions in this pathway result in elevated homocysteine concentrations and impaired methylation capacity, providing a mechanistic link between micronutrient stat

One protein, many faces: TRAF1 from signaling hub to disease diversity.

Tumor necrosis factor receptor-associated factor 1 (TRAF1) is a unique member of the TRAF family, distinguished by its lack of an N-terminal RING domain. At the transcriptional level, TRAF1 expression is regulated by diverse mechanisms, including methylation, RNA-binding domain (RBD) proteins, and microRNAs. Post-translationally, TRAF1 undergoes modifications such as M1-linked ubiquitination and phosphorylation. In immune signaling pathways, TRAF1 usually displays functional duality: it activates the canonical NF-κB pathway by enhancing cIAP recruitment downstream of TNFR1, whereas at death do

Beta-2 adrenergic receptor in CD8+ T cells inhibits GVHD through transcriptional modulation of differentiation and exhaustion.

Allogeneic hematopoietic cell transplantation (allo-HCT) is an effective therapy for selected hematologic diseases. However, graft-versus-host disease (GVHD) limits broader application of allo-HCT. The catecholaminergic sympathetic nervous system modulates immune cells. Beta-2 adrenergic Receptor (β2AR) acts as an immune checkpoint, yet its role in CD8+ T cells remains poorly understood. We used an MHC-mismatched model of acute GVHD to study how β2AR activity on donor CD8+ T cells affects GVHD severity. Host mice receiving β2AR+/+ (WT) CD8+ T cells demonstrated less severe GVHD, lower inflamma

Selective NRF2 target induction despite falling NFE2L2 transcription across the MASLD spectrum: implications for marginal donor grafts and KEAP1-targeted therapy.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) affects one-third of adults, and one in five progresses to metabolic dysfunction-associated steatohepatitis (MASH), which can cause liver failure, burdening hepatology clinics and transplant waiting lists. NRF2-KEAP1 axis is the principal regulator of hepatic antioxidant defence, yet its status across the disease spectrum is unclear. We mapped NRF2-KEAP1 transcriptional signatures across control, steatosis and MASH livers. METHODS: Eight GEO datasets (590 biopsies: 154 control, 134 steatosis, 302 MASH) were

Elastin-derived peptide VGVAPG induces redox-associated pro-inflammatory activation and early disease-associated microglia (DAM) molecular profile in a human microglia-neuron in vitro model involving NEU1/TLR4-related signaling.

The elastin-derived hexapeptide Val-Gly-Val-Ala-Pro-Gly (VGVAPG) exhibits well-documented biological activity; however, its role in brain inflammation remains insufficiently characterized. This is an important issue, as VGVAPG levels increase with age and have been implicated in processes contributing to neurodegeneration. The present study aimed to evaluate the ability of aging-related VGVAPG to activate human microglial (HMC3) cells in vitro and to elucidate the underlying molecular mechanisms. Our results demonstrated no cytotoxic effect of VGVAPG. However, further analyses revealed NF-κB n

Deubiquitination of WBP11 governs OPLAH-mediated glutathione biosynthesis to suppress triple programmed cell death in prostate cancer.

Prostate cancer (PC) is one of the most common male cancers. Cancer cells are well known for their capacity to avoid programed cell death (PCD) via a variety of molecular pathways, allowing them to survive oncogenic stress and therapeutic challenges. Although oncogenic splicing factors are frequently dysregulated in tumors, their functional interplay with PCD regulatory networks in PC remains largely uncharacterized. Here, we demonstrate that WBP11 exerts a pro-tumorigenic effect by promoting PC cell proliferation, while its genetic silencing triggers concurrent induction of apoptosis, pyropto

Mcu upregulation triggers mitochondrial oxidative stress and cardiomyocyte injury after myocardial infarction yet is suppressed by Fto and Ythdf2 in an m6A-dependent manner.

Mitochondrial calcium uniporter (Mcu) is essential for mitochondrial calcium homeostasis, yet its function and upstream regulatory mechanism in myocardial infarction (MI) remain unclear. This study was designed to define the pivotal regulatory pathway controlling Mcu expression post-MI and verify whether Mcu can act as a therapeutic target for MI. Mendelian randomization was used to verify the genetic link between Mcu and MI. In vitro gain- and loss-of-function assays together with m6A-seq, MeRIP-qPCR, RIP-qPCR and dual-luciferase assays were applied to explore the m6A-dependent regulatory mec

Lycorine ameliorates MASH by targeting ACSL4 to regulate lipid metabolism and inhibit ferroptosis.

Metabolic dysfunction-associated steatohepatitis (MASH), a progressive stage of metabolic dysfunction-associated steatotic liver disease (MASLD), has emerged as one of the most prevalent chronic liver diseases worldwide, yet therapeutic options remain limited. In this study, we found that lycorine effectively alleviated MASH both in vitro and in vivo. RNA-seq analysis revealed that lycorine suppressed lipid uptake and mitigated ferroptosis in MASH models. Using drug affinity responsive target stability coupled with mass spectrometry (DARTS-MS), we identified acyl-CoA synthetase long-chain fami

CXC chemokines as context-dependent regulators of immunity, homeostasis and cancer.

Chemokines are a large family of small secreted proteins that mainly orchestrate immune cell trafficking and tissue organization. Among them, the CXC chemokine family plays a central role in regulating immune surveillance, inflammation, angiogenesis and stem cell dynamics. Despite their relatively simple conserved fold - characterized by a β-sheet core and a C-terminal α-helix - CXC chemokines exhibit remarkable functional diversity. This diversity arises from structural plasticity, including concentration-dependent monomer-dimer equilibria and interactions with glycosaminoglycans (GAGs), whic
