Split-GFP Reporter-Based Detection of Chlamydia trachomatis Inclusion Damage.

Chlamydia trachomatis, an obligate intracellular pathogen responsible for trachoma and widespread urogenital infections, replicates exclusively within the confines of a membrane-bound vacuole known as the inclusion. This compartment shields the bacterium from host cellular immune detection; thus, clarifying how inclusion integrity is maintained and protected from vacuole-destabilizing host attacks may identify new therapeutic strategies. To enable such insights, we recently developed split-GFP-based microscopic reporters that can detect early forms of membrane damage and track individual bacte

Experimental Evolution of Drug Resistance for Antibiotic Target Discovery in Chlamydia trachomatis.

The recent application of experimental and computational drug discovery workflows has identified hundreds of new molecules that display potent, and in some cases highly selective, activity against bacteria of the genus Chlamydia. The full value of these novel antichlamydial molecules, however, depends critically on our ability to determine their molecular targets and modes of action. A particularly powerful and widely applied strategy for identifying candidate targets involves selecting for bacterial mutants that have acquired resistance to a compound's inhibitory activity, followed by determi

8-year vaccine protection following a single dose of Vi-tetanus toxoid conjugate vaccine in children in Nepal (TyVOID): a prospective cohort follow-up study of the TyVAC Nepal randomised controlled trial.

BACKGROUND: In low-income and middle-income countries, where typhoid fever remains a major public health problem, WHO recommends the use of typhoid conjugate vaccine (TCV). Here, we report vaccine effectiveness up to 8 years following a single dose of TCV in Nepal. METHODS: TyVOID was a prospective cohort study that extended the follow-up of children enrolled in a phase 3, double-blind, randomised controlled trial in Lalitpur, Nepal (TyVAC; Nov 20, 2017, to Oct 20, 2021). Children aged 9 months to 15 years were randomly assigned (1:1) to receive Vi-tetanus toxoid conjugate vaccine (Vi-TT) or

Monitoring Chlamydia trachomatis Development in Polarized Endocervical Epithelial Cells.

Understanding Chlamydia trachomatis infection requires physiologically relevant models tailored to various research questions. One of the main sites of C. trachomatis infection is the columnar epithelium of the endocervix; therefore, a model that accurately mimics the environment of endocervical epithelium is essential. Polarized cell models provide a valuable solution, offering a cost-effective yet biologically relevant alternative to conventional 2D culture systems and more complex and costly 3D or animal models. Immortalized human endocervical epithelial cells (A2EN cells) form a polarized

Chlamydia trachomatis Infection in a Microphysiologic Cell Culture Model.

Chlamydia trachomatis is an obligate intracellular bacterium that infects the columnar epithelium of the human endocervix. While conventional two-dimensional cell cultures and animal models have been instrumental in advancing our understanding of C. trachomatis biology, they are limited in capturing the multicellularity, architecture, and physiological microenvironment of the human cervix. This chapter describes the use of a three-dimensional (3D) microphysiologic model to study Chlamydia trachomatis infection. The model is inexpensively made without specialized equipment and is designed to re

A Conditional Knockout Mutant Approach to Study Essential Genes in Chlamydia in Cell Culture and In Vivo.

Identification and characterization of essential Chlamydia-specific factors are vital for the generation of effective therapeutics and vaccines. Despite delays in the development of genetic tools for Chlamydia, significant advancements in Chlamydia genetic manipulation have been made over the last decade and a half, now allowing for the targeted interruption or deletion of chromosomal genes. However, because of the obligate intracellular nature of the bacteria and its unique biphasic developmental cycle, it remains challenging to inactivate essential genes and study the function of their encod

A longitudinal analysis of Clostridioides difficile virulence profiles across three geographic regions in Israel (2020-2022).

BACKGROUND: Clostridioides difficile infection (CDI) is one of the leading infectious diarrhea worldwide. C. difficile pathogenicity is mediated by various virulence factors, including toxins and biofilm formation. This study included 313 C. difficile isolates that were previously collected from four medical centers. We aimed to investigate the prevalence, distribution, and associations of virulence factors in C. difficile strains from both healthcare-associated (HA)- and community-acquired (CA)-CDI patients across Israel from 2020 to 2022. METHODS: Multi-locus sequence typing (MLST) and whol

The Phases of Clostridioides difficile Infection (CDI) in the United States: The History and Epidemiology of CDI From 1935 to Present.

This review combines historical reports and surveillance data to contextualize Clostridioides difficile infection (CDI) in the United States as a phased epidemic, currently in the deceleration phase. C. difficile was first isolated in 1935. During a prolonged emergence phase, the organism attracted limited clinical attention. In 1978, C. difficile was established as the cause of antibiotic-associated pseudomembranous colitis and from 1978 to a peak in 2010, CDI was in an acceleration phase characterized by fluoroquinolone-resistant, toxin-overproducing strains, large hospital outbreaks, and in

Structural insights and therapeutic targets in Acinetobacter baumannii capsule biosynthesis.

Gram-negative bacteria produce nonulosonic acids, a family of nine-carbon sugars that mimic host sialic acids and may contribute to bacterial virulence. The nonulosonic acid C8-epimeric 5,7-di-N-acetyllegionaminic acid (8-epi-Leg5,7Ac2) is a component of the K-locus 49 (KL49) capsule of hypervirulent Acinetobacter baumannii, and its epimerization is proposed to be mediated by ElaA, ElaB, and ElaC. We identified a KL49 strain that induces lethal sepsis in mice and found deletion of elaA, elaB, or elaC abolished lethality and markedly reduced capsule production. Crystal structures of ElaB, ElaA,

Matrix extension to bovine feces and evaluation of semi-automated DNA extraction methods for the detection of Campylobacter jejuni in bovine and canine fecal samples using gyrA PCR.

We evaluated a semi-automated method for DNA extraction, expanded to herbivore feces a gyrA PCR method for the detection of Campylobacter jejuni in canine feces, added an internal positive control (IPC) DNA to specimens before DNA extraction, and multiplexed the gyrA PCR assay with the IPC assay. The addition of IPC DNA and multiplexing of the IPC assay did not affect the performance of the gyrA PCR assay. The method was repeatable with no significant difference in results across 6 d (F = 0.715; p = 0.614 with R2 = 0.818). In a single-laboratory randomized method test evaluating detection in b

Sampling and Host-Strain Interactions Shape Detection of Staphylococcus aureus Transmission in Hospitals.

BACKGROUND: In principle, whole-genome sequencing of Staphylococcus aureus in hospitals is most effective when used prospectively, but its practical limits are unclear. METHODS: We performed large-scale genomic surveillance across 2 interconnected hospitals, sequencing 4779 S. aureus isolates from admission screening and clinical cultures. Integration of genomic and epidemiologic data identified 361 transmission events undetected by standard surveillance. RESULTS: Despite dense sampling, most events (90%) were detected only at readmission, indicating that transmission escapes recognition dur

Surface-displayed whole-cell glucose oxidase from engineered Pichia pastoris exhibits enhanced intestinal repair and barrier restoration in a polymicrobial-infected mouse model.

Pathogenic infections pose significant threats to livestock production, necessitating antibiotic-free alternatives. This study engineered a high-yield whole-cell glucose oxidase (GOD) in Pichia pastoris via yeast cell surface display technology by optimizing anchor proteins (Gcw21), gene copy numbers (3 copies), and auxiliary proteins (HAC1) co-expression. The engineered strain yielded a GOD activity of 9322 U/g dry cell weight. The whole-cell GOD exhibited superior thermal stability, acid resistance, and tolerance to proteases and metal ions compared to free GOD. In vitro, its catalytic produ

DksA-dependent stringent stress response drives virulence and gastrointestinal persistence of Klebsiella pneumoniae.

Successful gastrointestinal (GI) colonization by bacterial pathogens requires adaptation to nutrient competition and host-derived stresses in the gut, with adaptation via the bacterial stringent stress response playing a critical role. Epidemiological data suggest that the GI tract serves as a reservoir from which K. pneumoniae can spread and cause invasive disease or transmit to another host. DksA is a conserved stringent response transcriptional regulator that was identified in an in vivo transposon mutagenesis screen as an important K. pneumoniae gut determinant. However, its role in K. pne

Host-microbiome interactions and enteric pathogen infections-from early-life to long-term impacts on gut-brain communication.

Interactions of the host with the microbes that constitute the intestinal microbiota are important for maintaining health and well-being within the gastrointestinal tract and beyond. Disruptions to these interactions, such as those that follow enteric pathogen infection or administration of antibiotics, can lead to persistent pathophysiology, particularly if disruptions occur during early development in neonatal life. Here, we highlight the impacts of altered host-microbe interactions following insults across the life span and discuss emerging mechanisms of action promoting these disruptions (
