Stress during puberty and adulthood pregnancy impact histone acetylation regulators in the hypothalamus.

Undergoing stressful events during puberty puts women at risk for a variety of negative outcomes, and this risk is heightened if they become pregnant later in life. We previously demonstrated that stress during puberty combined with pregnancy in adulthood led to a blunted response of the hypothalamic-pituitary-adrenal stress axis in humans and mice. We have begun to understand the mechanisms underlying this effect by examining the paraventricular nucleus of the hypothalamus (PVN), a key regulator of the HPA axis. Prior studies uncovered an increase in chromatin openness within the PVN of the a

BDE-209 toxicity: From spermiogenesis to sexual maturity in F1 male mice.

Most studies of enviromental toxic chemicals focused on the meiosis stage during spermatogenesis, however, the research on the spermiogenesis damage phenotype of BDE-209 is limited. This study aimed to evaluate the processes by which BDE-209 regulates the formation of acrosomes and mitochondrial sheath (MS), key structures during spermiogenesis and fertilization. ICR mice were divided into control, low, medium, and high-dose BDE-209 groups and treated for 42 days. A comprehensive method combining ultrastructural analysis, transcriptomics, molecular biology, and fertility experiments was adopte

m6A Methylation Mediated Autophagy and Nucleotide-Binding Oligomerization Domain-like Receptors Signaling Pathway Provides New Insight into the Mitigation of Oxidative Damage by Mulberry Leaf Polysaccharides.

m6A methylation modification is an important genetic modification involved in biological processes such as sexual maturation, antibacterial, and antiviral in aquatic animals. However, few studies have been conducted in aquatic animals on the relationship between m6A methylation modification and autophagy-inflammation induced by lipid metabolism disorders. In the present study, a high-fat (HF) group and HF-MLP group (1 g mulberry leaf polysaccharides (MLPs)/1 kg HF diet) were set up. The mid-hind intestines of Megalobrama amblycephala juveniles from the two groups were collected for MeRIP-seq a

Opposite-sex pairing alters social-induced GCaMP and dopamine activity in the insula of male prairie voles.

The prairie vole (Microtus ochrogaster) is a monogamous rodent species which displays selective social behaviors to conspecifics after establishing a pair-bonded relationship, specifically partner-directed affiliation and stranger-directed aggression. This social selectivity relies on the ability of an individual to respond appropriately to a social context and requires salience detection and valence assignment. The anterior insular cortex (aIC) has been implicated in stimulus processing and categorization across a variety of contexts, but its regulation of pair bond-induced social selectivity

Gender Dysphoria and Detransitioning in Adults: An Analysis of Nine Patients from a Gender Identity Clinic from Finland.

The aim of this study was to analyze the pathways of detransitioning, which is a rare, but serious complication of gender-affirming treatments (GATs). The patient group consisted of all patients who were referred to the Helsinki University Hospital's Gender Identity Clinic (GIC) wanting medical treatment for detransition from 2018 to 2019. A new assessment was made systematically and retrospective data were collected. The sample consisted of nine patients originally diagnosed with gender identity disorder (F64.0). Seven of them were assigned female at birth and two were assigned male at birth.

The effects of prepubertal ovariectomy and chronic stress on activity of brain limbic regions in adult mice.

Women are twice as likely than men to develop an anxiety disorder after stress exposure stress, a sex-specific vulnerability that arises after puberty. This suggests that pubertal hormones could contribute to the central changes induced by stress and leading to behavioral deregulations. The main brain regions involved in stress-induced psychopathologies are part of the limbic system, including the prefrontal cortex (PFC), the basolateral amygdala (BLA) and the ventral hippocampus (vHipp). Changes in their activity are often reported after stress, contributing to appearance of behavioral sympto

Mapping the immune cells of the testis: Key insights into sexual maturation.

BACKGROUND: The testicular immune microenvironment sustains homeostasis and immune privilege. However, the presence and functional significance of B cells within this specialized niche remain poorly characterized. Furthermore, the intricate crosstalk between resident immune populations and testicular stromal cells that may orchestrate the immunological and endocrine microenvironment during sexual maturation warrants systematic investigation. OBJECTIVE: To systematically delineate the immune cell atlas and developmental trajectory of testicular leukocytes before and after sexual maturation. M

Sex-dependent gene expression during puberty has potential mechanistic implications for the development of major depressive disorder.

Major Depressive Disorder (MDD) is a complex illness with heterogeneous symptom profiles, affecting 5% of the global population. Recent molecular studies across biological systems suggest that the manifestations of MDD are significantly shaped by sex. In response, this narrative review organizes up-to-date findings on sex-specific, MDD-related gene expression into multiple biological systems-primarily available from adult human and rodent studies. This adult-centric focus highlights a critical gap: when and how sex-dependent, MDD-associated gene expression patterns emerge, and contribute to ad

Effects on the middle adult stage of the Tohoku hynobiid salamander, Hynobius lichenatus, chronically γ-irradiated from the embryonic stage.

Tohoku hynobiid salamanders, Hynobius lichenatus (Amphibia, Caudata), were chronically irradiated with 137Cs γ-rays from the embryonic to the middle adult stages for over 3000 days, and the effects on survival, somatic growth (body weight increase), sexual maturation, and histological characteristics were examined. This article reports the effects observed in the middle adult stage (days 1955-3039). Irradiation at a dose rate of 34 μGy h-1 had no effect on any endpoint. At 150 or 500 μGy h-1, secondary sexual characteristics (tubercle at the anterior angle of the cloacal vent for males and ovi

Analysis of circular RNA (circRNA) characteristics and identification of key circRNAs in the hypothalamus during sexual maturation in female goats.

OBJECTIVE: Precocious puberty can shorten reproductive cycles, enhance reproductive capacity, and reduce feeding costs. Consequently, precocious livestock are widely utilized in cross-breeding. This study aims to elucidate the key molecular mechanisms by which circular RNA (circRNA) regulates the sexual maturation of sexually precocious goats. METHODS: In this study, we analyzed the circRNA expression profiles of hypothalamic tissue from Jining grey goats at four distinct postnatal developmental stages (1 day, 2 months, 4 months, 6 months). RESULTS: A total of 23,993 circRNAs were identified

The role of adipose tissue in puberty and reproductive health.

Adipose tissue is an endocrine organ that signals energy status to the hypothalamic-pituitary-gonadal axis to regulate reproductive function. Notably, in mammals, adipose tissue biology-adipose tissue expansion and body fat distribution-is closely linked to the onset of puberty. Some studies showed that early adipose tissue development continues into childhood or adulthood, indicating its potential impact on reproductive function. Factors such as maternal obesity, childhood body mass index gain, and adolescent obesity significantly contribute to early puberty onset and negative reproductive ev

High-Calorie Diet Exacerbates the Crosstalk Between Gestational Diabetes and Youth-Onset Diabetes in Female Offspring Through Disrupted Estrogen Signaling.

Background/Objectives: Recent global trends highlight a concerning rise in youth-onset type 2 diabetes (YOT2D), with a marked female preponderance. We aim to explore the crosstalk between gestational diabetes mellitus (GDM) and YOT2D in female offspring. Methods: In vivo, GDM mice were induced by Western diet (WD), and their female offspring were fed normal diet or WD within 3 to 8 weeks. We continuously detected the glucose metabolism disorders, serum estradiol level (ELISA), and the process of ovarian maturation. Meanwhile, the dynamic changes in ERα and insulin signal in liver were monitore

Circadian disruption impairs Leydig cell maturation and reproductive development in male rats.

Circadian desynchrony, caused by a misalignment between the internal biological clock and environmental light cues, is increasingly prevalent in adolescents due to irregular light exposure and social pressures. However, its impact on reproductive maturation remains poorly understood. In this study, the effects of chronic circadian disruption, induced by the 223 light regimen (two days of constant light, two days of constant darkness, and three days of a 14:10 h light-dark cycle), were examined in juvenile and peripubertal male rats (postnatal days 21-49). Gene expression profiles associated wi

Differences of sex development.

Differences of sex development (DSD) represent a group of congenital conditions that affect human sex development and maturation owing to discrepancies of chromosomal, gonadal and phenotypic sex. The Chicago consensus classifies DSD as sex chromosome DSD, 46,XY DSD and 46,XX DSD, with subclassifications according to gonadal determination into testes and ovaries and hormone-dependent differentiation of Müllerian and Wolffian embryonic structures into female-typical or male-typical internal and external sex organs. DSD may occur as an isolated condition or as part of a complex syndrome. Diagnosi

Advantages of outcrossing in Plasmodium falciparum: insights from genetic crosses using fluorescent labelled parasites.

Malaria parasites are obligately sexual hermaphrodite protozoans with gamete fusion occurring in the mosquito midgut, followed by meiosis and recombination. Malaria parasite populations show a spectrum of populations structures ranging from predominantly selfing to highly outcrossed. However, the fitness consequences of selfing and outcrossing for malaria parasites are poorly understood. This project was designed to investigate the dynamics of gamete fusion within the mosquito midgut and the relative fitness of selfed and outcrossed zygotes. We generated florescent-labelled clones of NF54 (mCh

A simultaneous hermaphrodite dwarf common carp (Cyprinus carpio carpio morpha hungaricus) from a natural thermal lake, Lake Hévíz, Hungary.

The fauna of Lake Hévíz is unique due to its status as the world's only natural warm-water medicinal lake with a peat bed. A population of dwarf common carp (Cyprinus carpio carpio morpha hungaricus) forms an isolated, self-sustaining community in the lake, adapted to extreme thermal and chemical conditions. During continuous research conducted since 2007, we identified a hermaphroditic individual for the first-time following sampling during the winter period in 2024. Macroscopic examination revealed testicular tissue distinguishable from ovarian tissue. Histological analysis indicated that th

Prevalence and relation to training of electrocardiographic findings in Caucasian children and adolescents 8- to 18-year-old practicing competitive sport.

AIMS: Training frequently induces electrocardiographic (ECG) changes that mimic heart diseases, requiring specific criteria for interpretation. Paediatric athletes represent a unique population as training-induced changes and those due to sexual maturation interact, and specific criteria may be needed. We aimed to assess the prevalence and its relation to training of ECG abnormalities in young athletes aged 8-18 years. METHODS AND RESULTS: We included 2458 young apparently healthy Caucasian athletes undergoing pre-participation screening. Electrocardiographic abnormalities were classified acc

Mast Cells Are Not Essential for Pubertal Mammary Gland Branching.

Mast cells are long-lived, tissue-resident immune cells of the myeloid lineage with cardinal functions in allergy and atopic disease. They are now increasingly recognized also for protective roles, for example against infections and venoms. Other functions originally assigned to mast cells in development and physiology, however, have been refuted, and for yet others, the true contribution of mast cells remains uncertain. Mast cells have been implicated in promoting ductal branching in the pubertal mammary gland, the organ that produces and secretes milk in mammals, but these findings are based

Reproductive system development in Monochamus alternatus hope subjected to maturation feeding.

Monochamus alternatus Hope (Coleoptera: Cerambycidae), a vector of the invasive pathogenic nematode Bursaphelenchus xylophilus (Steiner & Buhrer) Nickle, is a notorious forest pest in Asia. After emergence, adult M. alternatus must supplement nutrition to further develop and reach sexual maturity before they can engage in courtship and mating. However, there is a lack of information on the precopulation duration of the Chinese population of M. alternatus and the reproductive system development of both sexes subjected to maturation feeding. In this study, we investigated the precopulation durat

Steroid hormone-mediated epigenetic programming during puberty: uncovering links to depression.

DNA methylation (DNAm) is a key epigenetic modification that dynamically regulates eukaryotic development over time. DNAm has been found to influence a variety of biological processes in both normative and pathological states, such as depression. Since DNAm can serve as an interface between environmental influence and gene expression, it is a mechanism studied in the context of many pathologies, including psychiatric. Depression is a complex and heterogeneous disorder strongly influenced by puberty, as evidenced by increased rates in both sexes after sexual maturation. However, this effect is
