Base rates of low achievement scores in young adults: implications for specific learning disability assessment.

BACKGROUND: Specific learning disability (SLD) is a high-incidence diagnosis, with academic skill deficits at its core. Diagnostic models for SLD vary but they generally require data from norm-referenced achievement tests. Multivariate base rates for low scores on such tests have not been examined in young adults. METHODS: The young adult (ages 18-29) group from the Woodcock-Johnson IV (WJ-IV; 2014) normative sample (N = 1330) was used. We calculated base rates for low scores on the 11 subtests of the Standard Battery of the WJ-IV Tests of Achievement. We also calculated base rates for discre

Leveraging Preclinical Models to Understand Human Neurobiology and Behavior.

Rodent models have become indispensable tools for investigating the neurobiological mechanisms underlying human behavior and disease. Despite the substantial phenotypic differences between rodents and humans, shared evolutionary history, conserved genomic architecture, and analogous neural circuitry allow preclinical findings to meaningfully inform our understanding of human conditions. This chapter reviews the behavioral paradigms most commonly used to evaluate five core domains in rodent models: motor function, stress and anxiety-like behavior, repetitive and restrictive behaviors, social be

CasNano: An ultracompact CRISPR-Cas12f with a GC-rich PAM for epigenome editing with therapeutic potential.

Clinical use of CRISPR-Cas proteins for genome engineering is constrained by their large size, limiting in vivo delivery. To address this, we engineered an ultracompact and versatile platform from a minimally active, 400-amino acid Cas12f protein from Blautia lenta. A multi-stage strategy of structure-guided protein mutagenesis and systematic single guide RNA (sgRNA) optimization transformed BlCas12f into a "CasNano" as a potent genome targeting tool for mammalian cells. When a catalytically dead CasNano (dCasNano) is fused to a VPR domain, it activates endogenous genes with an efficiency and

Barriers and Opportunities in Anesthesia Care for Patients With Intellectual and Developmental Disabilities: A Qualitative Study.

Health disparities among individuals with intellectual and developmental disabilities (IDD) remain a significant public health concern requiring complex, interdisciplinary care tailored to communication, behavioral, and individualized needs. Despite frequent exposure to surgical and procedural interventions, nurse anesthesiology programs provide limited formal education to prepare clinicians for this population. Most importantly, the role of nurse anesthesiologists across the perioperative continuum remains underrepresented in discussions of IDD-focused care. This qualitative phenomenological

Clinical nursing faculty perceptions about teaching students to provide care for adults with developmental disability.

BACKGROUND: Adults with developmental disabilities (AWDD) experience health inequities that could be improved with nurses better prepared to address their unique needs. Lack of contact of nursing students with AWDD, which is pervasive in nursing education, contributes to the health inequities experienced by this population. This may be due in part to clinical nursing faculty members' perceptions about teaching students to provide care to this population. PURPOSE: This study explored the perceptions of clinical nursing faculty about teaching students to provide care for AWDD. DESIGN: This stu

Multiple Basal Cell Carcinomas in an Adolescent with an MLPA-Detected PTCH1-Region Deletion and a Gorlin-Goltz Phenotype.

Gorlin-Goltz syndrome, also known as nevoid basal cell carcinoma syndrome or basal cell naevus syndrome, is a rare hereditary cancer-predisposition disorder most often caused by disruption of the Hedgehog signaling pathway, particularly alterations of PTCH1. Classical disease is characterized by multiple or early-onset basal cell carcinomas, odontogenic keratocysts, palmoplantar pits, skeletal anomalies and intracranial calcifications. Deletions involving chromosome region 9q22.3 may create an overlapping but broader phenotype because the deleted interval may include PTCH1 together with neighb

Beyond benchmarks: a narrative review of evolving quality of life frameworks for children with neurodevelopmental and learning disabilities and the need for integrated digital twin approaches.

Beginning with an overview of the current state of Quality of Life (QoL), this narrative review provides a synthesis of thirty articles published between 2007 and 2026 to provide insight into the evolution of QoL frameworks, how QoL data tracking has been operationalized by these frameworks, and where the gaps are. While the literature has moved from a focus on deficits and impairment to a strengths-based approach centered around the child, and aligned with the principles of the United Nations Convention on the Rights of Persons with Disabilities (CRPD), one theoretical framework (Schalock & V

Special care dentistry: The quiet crisis at the margins of modern healthcare.

Special care dentistry (SCD) provides oral healthcare for adults who are unable to accept routine dental care due to physical, intellectual, medical, emotional, sensory, mental, or social impairments. In England, demand for SCD services is increasing in the context of population ageing, rising multimorbidity, improved tooth retention into later life, and persistent inequalities in access to National Health Service dental care. Recent national survey data highlight substantial levels of untreated dental disease, increasing oral-health-related impacts on quality of life, and significant barriers

mGlu7 expression is upregulated by Tcf4 deficiency in a model of Pitt Hopkins syndrome.

Pitt-Hopkins syndrome (PTHS) is a rare neurodevelopmental disorder caused by de novo mutations in the Transcription Factor 4 (TCF4) gene. Clinically, phenotypes of PTHS involve motor and cognitive domains as well as epilepsy and breathing abnormalities. These phenotypes exhibit significant overlap with another disorder, Rett syndrome (RTT), caused by mutations in the Methyl-CpG-Binding Protein 2 (MECP2) gene. We have shown that expression of metabotropic glutamate receptor 7 (mGlu7) is significantly decreased in brain samples of patients with RTT, and we showed therapeutic potential of increas

The Role of the Immunoglobulin Superfamily of Cell Adhesion Molecules in Intellectual Disability.

Immunoglobulin superfamily (IgSF) cell adhesion molecules (CAMs) are cell surface glycoproteins that play critical roles during multiple stages of synapse formation, including axon and dendrite growth and guidance, target recognition, axo-dendritic contact formation and its maturation into a functional synapse. Intellectual disability is a condition characterized by significant limitations in intellectual functioning and adaptive behaviors. Growing evidence indicates that deficits in synapse formation underlie the pathogenesis of intellectual disability. In this review, we discuss studies show

Linking visual span and reading speed in dyslexia.

This study examines visual processing in dyslexia using a psychophysical measure linked to reading speed, the visual span, defined as the number of letters reliably identified within a single fixation. We asked whether reduced visual span is associated with slower reading in dyslexia. In a laboratory study, adults with dyslexia and typical readers completed standardized assessments, reading speed, and visual span measurements. Individuals with dyslexia showed significantly slower reading speeds and narrower visual span profiles than typical readers. Visual span size was positively correlated w

Understanding the unfolded protein response in glioblastoma.

Glioblastoma is the most aggressive and lethal malignancy of the central nervous system and exhibits a complex interplay with endoplasmic reticulum (ER) stress (ERS). The multifunctional ER is the primary site of the unfolded protein response (UPR), which is activated under conditions of ERS to enhance protein-folding capacity and promote cell survival. However, persistent ERS can trigger apoptosis. Despite extensive research over the past decades, the mechanistic insights into the impact of ERS and the UPR on glioblastoma pathogenesis and progression remain unclear. The UPR operates through t

Association of socioeconomic factors on mental health in children and young people with intellectual developmental disabilities of genetic origin.

BACKGROUND: Children and young people (CYP) with intellectual developmental disability (IDD) have a high prevalence of mental health and behavioural problems, which might be influenced by socioeconomical status as well as the severity of their disabilities. This is however an understudied topic, and the present study investigated the association between mental health and socioeconomic deprivation as well as severity/type of disability in CYP with IDD of genetic origin. METHODS: Participants comprised a national cohort of 2778 participants with diagnosed genetic disorders associated with IDD,
