Integrated analysis of nutrition profiles and dried blood spot proteomics reveals patterns associated with autism spectrum disorder.

OBJECTIVES: To investigate whether dietary behavior-derived nutritional profiles and proteomic analysis of dried blood spots (DBS) could identify patterns associated with autism spectrum disorder (ASD). METHODS: A total of 29 children with ASD (mean age, 56.0 ± 8.7 months; 23 boys) and 27 control children (mean age, 60.6 ± 9.5 months; 20 boys) participated in the study. Three-day food records were used to assess nutrient and food group intakes. Blood samples were collected as DBS. To evaluate group separation, partial least squares discriminant analysis was applied to food group data, nutrien

Sex differences in 5-year incidence and prevalence of physical illnesses following early childhood autism diagnosis.

BACKGROUND: Sex differences in the clinical presentation of autism are established, but evidence on early-life co-occurring physical illnesses in early-diagnosed autistic individuals is scarce. This nationwide cohort study examined sex-stratified incidence of physical illnesses within 5 years after autism spectrum disorder (ASD) diagnosis in children ≤5 years. METHODS: Using Taiwan's National Health Insurance Research Database (2000-2019), we identified 36,656 autistic males and 9024 autistic females newly diagnosed before age 6 and followed for five years. Adjusted odds ratios and hazard rat

Structured interactive play for autism spectrum disorder screening in toddlers: a primary care-based cohort study.

BACKGROUND: Early identification of autism spectrum disorder (ASD) is crucial for improving outcomes, yet diagnosis is often delayed. Primary care faces challenges, including the limited accuracy of existing tools and scarce access to specialist assessments. This prospective cohort study evaluated the feasibility of a novel, structured examiner-child interactive play protocol as an early behavioral screening tool for ASD. METHODS: We recruited 260 toddlers aged 18-36 months. At baseline, all children participated in a structured interactive play session. Their behaviors (e.g., eye contact, tu

Maternal Extracellular Vesicles During Pregnancy and Autism Risk in Children.

BACKGROUND: Differences in extracellular vesicles (EVs), bioactive nanoparticles involved in intercellular signaling, have been reported in those with autism. However, little is known about the association between maternal EVs during pregnancy and the likelihood of autism in offspring. This study evaluated the association of the concentration and cargo material of EVs in prenatal maternal plasma with childhood autism likelihood. METHODS: Participants in the Nulliparous Pregnancy Outcomes Study provided maternal plasma at 15-23 weeks' gestational age. EVs were isolated by ultracentrifugation,

Cortical surface-based descriptors for childhood autism classification using segmentation-guided cortical reconstruction from sMRI.

OBJECTIVES: Autism spectrum disorder (ASD) involves subtle and heterogeneous brain-structural differences that are not usually visible on routine structural magnetic resonance imaging (sMRI). This study investigated a segmentation-guided sMRI feature-classification framework for childhood ASD analysis. METHODS: Pediatric T1-weighted sMRI scans were obtained from the publicly available Autism Brain Imaging Data Exchange II (ABIDE-II) dataset. Preprocessing used the FMRIB Software Library (FSL) Brain Extraction Tool (BET), N4 bias-field correction, intensity normalization, and Advanced Normaliz

Night-to-Night Changes in REM Sleep Are Associated With Symptom Severity in Children With Autism Spectrum Disorder: A Two-Night Home Polysomnography Study.

Sleep disturbances are highly prevalent in autism spectrum disorder (ASD), yet conventional single-night polysomnography (PSG) provides limited insight into dynamic sleep regulatory processes. Using the Synaptic Homeostasis Hypothesis as a conceptual framework, we quantified night-to-night changes (Δ; Night 2 - Night 1) in sleep architecture across two consecutive nights of home-based PSG and examined their association with ASD symptom severity. Seventy-nine children with ASD, stratified into mild ASD (M-ASD, n = 35) and moderate-to-severe ASD (MS-ASD, n = 44) subgroups using the Childhood Aut

Specificities of auditory behavior in children with autism spectrum disorder.

Auditory behavior reflects an individual's response to environmental sounds. Atypical responses to auditory input may indicate developmental difficulties, particularly in children with autism spectrum disorder (ASD). This study aimed to identify auditory behavior in children with ASD in relation to autism severity, age, treatment onset, and history of ear disease. The sample included 30 children with ASD (21 female and 9 male), classified into three groups according to ASD severity: mild (n = 3), moderate (n = 4), and severe (n = 23). Auditory behavior was assessed using the Questionnaire on A

The relationship between stereotypical movement disorder and neuroinflammation.

OBJECTIVE: Stereotypic movement disorder (SMD) is a neurodevelopmental condition frequently co-occurring with autism spectrum disorder (ASD), yet its underlying pathophysiology remains poorly understood. Emerging evidence suggests that neuroinflammatory mechanisms may contribute to the etiology of neurodevelopmental disorders. Based on this, we aimed to investigate the potential role of neuroinflammation, specifically interleukin-1β (IL-1β) and interleukin-6 (IL-6), in the etiopathophysiology of both primary SMD and ASD. METHODS: This cross-sectional study included children and adolescents ag

Washed microbiota transplantation improves clinical symptoms, gut microbiota, and metabolic profiles in autism spectrum disorder in a twin cohort.

OBJECTIVE: Autism spectrum disorder (ASD) is a heterogeneous neurodevelopmental condition characterized by impaired social communication, repetitive behaviors, and restricted interests. Dysregulation of the microbiota-gut-brain axis is closely associated with the pathogenesis of ASD. Washed microbiota transplantation (WMT) has emerged as a promising intervention for ASD, but existing cohort studies lack genetically identical controls, making it difficult to distinguish intervention-related changes from genetic and environmental confounding factors. This twin-paired controlled study adopted a s

Design and content validation of AUTIVA: an ecobiopsychosocial instrument for childhood autism.

There are no community-based surveys for the development or assessment of public policies with an ecobiopsychosocial approach for autistic children. This article reports the development of an instrument assessing characteristics of Chilean autistic children, in order to understand how autism manifests in different environments. This study consisted of two stages: Firstly, after a comprehensive literature review, an instrument was designed by health and education professionals alongside autistic individuals and primary caregivers. Secondly, content validation was performed using the Content Val

Maternal Thyroid Dysfunction During Pregnancy and Childhood Autism or Other Developmental Outcomes.

Overt maternal hypothyroidism during pregnancy has been associated with cognitive impairment in offspring, but less is known about effects of subclinical hypothyroidism and other maternal thyroid conditions on autism spectrum disorder (ASD). This study examined the relationships between maternal thyroid dysfunction during pregnancy and child neurodevelopmental outcomes. Participants were from a high ASD likelihood pregnancy cohort (MARBLES). Maternal thyroid dysfunction (hypothyroid or hyperthyroid compared to euthyroid) was measured in up to one blood sample per trimester. Thyroid stimulating

Serum Biomarkers of Neuroaxonal and Astroglial Damage in Autism Spectrum Disorder: Relationship With Symptom Severity, Behavioural Dimensions and Age.

PURPOSE: Autism spectrum disorder (ASD) is a neurodevelopmental condition characterized by persistent deficits in social communication and the presence of restrictive, repetitive patterns of behaviour. Although its exact aetiology remains multifaceted and partially understood, recent clinical interest has shifted towards neurobiological substrates, specifically neuroaxonal and astroglial integrity. This study aims to compare serum levels of Neurofilament Light Chain (NfL), Glial Fibrillary Acidic Protein (GFAP), Tau and S100B between children with ASD and healthy controls, while investigating

Brief online parent-led early intervention for childhood autism: A randomised-controlled trial.

BACKGROUND: Child behavioural and emotion regulation, and social communication; and parental confidence and mental health, all contribute to developmental outcomes for autistic children; however, there are limited programmes which integrate support across these domains. METHODS: We report on a parallel, assessor-masked randomised controlled trial used to evaluate a brief, parent-led intervention, ParentWorks-Spectrum. Parents of 100 children aged 2-5 years with autism Level 2/3 and behavioural difficulties were randomly assigned (1:1) to ParentWorks-Spectrum (n = 49) or Waitlist (n = 51). Par

Altered alpha-band regulation during naturalistic theory of mind processing in autistic children.

BACKGROUND: Theory of Mind (ToM) deficits are a core feature of autism, yet the neural dynamics associated with social-cognitive processing in naturalistic contexts remain unclear. Alpha-band oscillations have been implicated in the regulation of internal attention and may be involved in supporting mentalizing during real-world social interactions. METHODS: Electroencephalography (EEG) was recorded from 41 autistic children and 34 typically developing (TD) children aged 3-10 years while they passively viewed a nonverbal animated film. Investigation of the Neurocognitive Characteristics of Parents of Children Diagnosed With Autism Spectrum Disorder: A CANTAB-Based Study.

Autism spectrum disorder (ASD) is a highly heritable neurodevelopmental condition whose genetic underpinnings may manifest as subclinical cognitive and behavioral traits in first-degree relatives, including parents of diagnosed individuals. Assessing parental cognitive profiles is valuable for identifying Broad Autism Phenotype (BAP) endophenotypes. Accordingly, this study examined possible cognitive endophenotypes by comparing neurocognitive profiles of parents (mothers and fathers) of children with ASD versus parents of neurotypical children. This cross-sectional study included 124 parents o
